Knockout of glutathione peroxidase 5 down-regulates the piRNAs in the caput epididymidis of aged mice显示文摘The mammalian epididymis not only plays a fun dame ntal role in the maturati on of spermatozoa, but also provides protecti on agai nst various stressors. The foremost among these is the threat posed by oxidative stress, which arises from an imbalance in reactive oxygen species and can elicit damage to cellular lipids, proteins, and nucleic acids. In mice, the risk of oxidative damage to spermatozoa is mitigated through the expression and secretion of glutathione peroxidase 5(GPX5)as a major luminal scavenger in the proximal caput epididymidal segment.Accordingly, the loss of GPX5^-/-mediated protection leads to impaired DNA integrity in the spermatozoa of aged Gpx57 mice.To explore the underlying mechanism,we have conducted transcriptomic analysis of caput epididymidal epithelial cells from aged(13 months old)Gpx5^-/-m mice. This analysis revealed the dysregulation of several thousand epididymal mRNA transcripts,in eluding the downregulation of a subgroup of piRNA pathway gen es,in aged Gpx5^-/-mice.In agreeme nt with these fin dings,we also observed the loss of piRNAs,which potentially bind to the P-element-induced wimpy testis(PlWI)-like proteins PIWIL1 and PIWIL2.The absence of these piRNAs was correlated with the elevated mRNA levels of their putative gene targets in the caput epididymidis of Gpx5^-/-mice.Importantly,the oxidative stress response genes tend to have more targeting piRNAs,and many of them were among the top increased genes upon the loss of GPX5^-/-.Taken together, our findings suggest the existence of a previously uncharacterized somatic piRNA pathway in the mammalian epididymis and its possible invoIvement in the aging and oxidative stress-mediated responses. | Chen Chu Lu Yu Joelle Henry-Berger Yan-Fei Ru Ayhan Kocer Alexandre Champroux Zhi-Tong Li Miao He Sheng-Song Xie Wu-Bin Ma Min-Jie Ni Zi-Mei Ni Yun-Li Guo Zhao-Liang Fei Lan-Tao Gou Qiang Liu Samanta Sharma Yu Zhou Mo-Fang Liu Charlie Degui Chen Andrew L Eamens Brett Nixon Yu-Chuan Zhou Joel R Drevet Yong-Lian Zhang | 2020 | Asian Journal of Andrology2020,22,6: | 2

Do reactive oxygen species damage or protect the heart in ischemia and reperfusion?Analysis on experimental and clinical data显示文摘The role of reactive oxygen species(ROS)in ischemic and reperfusion(I/R)injury of the heart has been discussed for more than 40 years. It has been demonstrated that reperfusion triggers a multiple increase in free radical generation in the isolated heart.Antioxidants were found to have the ability to mitigate I/R injury of the heart. However,it is unclear whether their cardioprotective effect truly depends on the decrease of ROS levels in myocardial tissues.Since high doses and high concentrations of antioxidants were experimentally used,it is highly likely that the cardioprotective effect of antioxidants depends on their interaction not only with free radicals but also with other molecules. It has been demonstrated that the antioxidant N-2-mercaptopropionyl glycine or NDPH oxidase knockout abolished the cardioprotective effect of ischemic preconditioning.Consequently, there is evidence that ROS protect the heart against the I/R injury. | Leonid N.Maslov Natalia V.Naryzhnaya Maria Sirotina Alexandr V.Mukhomedzyanov Boris K.Kurbatov Alla A.Boshchenko Huijie Ma Yi Zhang Feng Fu Jianming Pei Viacheslav N.Azev Vladimir A.Pereverzev | 2023 | The Journal of Biomedical Research2023,37,4: | 1

Synthesis of Iron-Carbide Nanoparticles:Identification of the Active Phase and Mechanism of Fe-Based Fischer-Tropsch Synthesis显示文摘Despite the extensive study of the Fe-based Fischer-Tropsch synthesis(FTS)over the past 90 years, its active phases and reaction mechanisms are still unclear due to the coexistence of metals, oxides, and carbide phases presented under realistic FTS reaction conditions and the complex reaction network involving CO activation,C-C coupling, and methane formation. To address these issues,we successfully synthesized a range of pure-phase iron and iron-carbide nanoparticles(Fe,Fe_(5)C_(2),Fe_(3)C,and Fe_(7)C_(3))for the first time.By using them as the ideal model catalysts on high-pressure transient experiments,we identified unambiguously that all the iron carbides are catalytically active in the FTS reaction while Fe_(5)C_(2) is the most active yet stable carbide phase, consistent with density functional theory(DFT)calculation results. The reaction mechanism and kinetics of Fe-based FTS were further explored on the basis of those model catalysts by means of transient high-pressure stepwise temperature-programmed surface reaction(STPSR)experiments and DFT calculations. Our work provides new insights into the active phase of iron carbides and corresponding FTS reaction mechanism, which is essential for better iron-based catalyst design for FTS reactions. | Huabo Zhao Jin-Xun Liu Ce Yang Siyu Yao Hai-Yan Su Zirui Gao Mei Dong Junhu Wang Alexandre I.Rykov Jianguo Wang Yanglong Hou Wei-Xue Li Ding Ma | 2021 | CCS Chemistry2021,3,11: | 1

Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 14 | Thermal damage and ablation behavior of plasma sprayed Ba_(2)SmTaO_(6) coatings irradiated by high power continuous laser显示文摘Ba_(2)SmTaO_(6) laser protection coatings of≈200μm thickness were deposited onto stainless steel surfaces by air plasma spraying, and the laser irradiation resistance of the coatings was investigated. For laser irradiation with a laser power density less than 1000 W/cm^(2),the coatings kept intact. For a laser power density exceeding 1500 W/cm^(2),the Ba_(2)SmTaO_(6) coatings underwent recrystallization, grain growth occurred, and certain spray morphology features disappeared by melting. In the case of a laser power density of 2000 W/cm^(2) applied for 10 s,the incident laser parameter was beyond the coatings protection threshold, and the coating peeled off.The samples back surface temperature kept unchanged within the first 1s of laser irradiation, indicating thatBa_(2)SmTaO_(6) coatings have excellent laser protection capability and can limit the rise of the substrate temperature. However, the low thermal conductivity of Ba_(2)SmTaO_(6) leads to a detrimental laser energy concentration at the beginning of the laser irradiation period on the sample front surface, resulting in a rapid increase of the surface temperature up to the melting point. | Jiayi Zheng Zhuang Ma Yanbo Liu Lihong Gao Alexandr A.Rogachev | 2021 | Progress in Natural Science:Materials International2021,31,4: | 0

New patterns of the tree beta diversity and its determinants in the largest savanna and wetland biomes of South America显示文摘Clear and data-driven bioregionalizations can provide a framework to test hypotheses and base biodiversity conservation. Here we used occurrence and abundance data in combination with objective analytical methods to propose two bioregionalization schemes for tree species of the Cerrado and the Pantanal in South America. We also evaluated the contribution of three sets of determinants of the occurrence-and abundance-based subregions. We compiled data on tree species composition from 894 local assemblages based on species occurrences, and from 658 local assemblages based on species abundances. We used an unconstrained community-level modelling approach and clustering techniques to identify and map tree subregions for the occurrence and the abundance data sets, separately.Hierarchical clustering analyses were conducted to investigate floristic affinities between the subregions and to map broader floristic regions. We used multinomial logistic regression models, deviance partitioning, and rank-sum tests to assess the main subregion correlates. We identified 18 occurrence-and four abundance-based subregions in the Cerrado-Pantanal.The hierarchical classifications grouped the occurrence-based subregions into nine floristic zones and abundance-based subregions into two broad floristic zones.Variation in subregions were explained mainly by environmental factors and spatial structure in both occurrence and abundance data sets. The occurrence-and abundance-based subregions are complementary approaches to disentangle macroecological patterns and to plan conservation efforts in the Cerrado and the Pantanal. Our findings based on occurrence data revealed more complex and interdigitated boundaries between subregions of tree species than previously reported. The environment, historical stability, and human effects act in a synergetic way on the distribution of the subregions. Finally, the relevance of contemporary environmental factors to the subregion patterns we found alert us to the profound impact global warming may have on the spatial organization of the Cerrado-Pantanal tree flora. | Karla J.P.Silva-Souza Maíra G.Pivato Vinícius C.Silva Ricardo F.Haidar Alexandre F.Souza | 2023 | Plant Diversity2023,45,4: | 0
