1Challenges in the diagnosis and treatment of gestational trophoblastic neoplasia worldwide显示文摘Gestational trophoblastic neoplasia(GTN) is a rare tumor that originates from pregnancy that includes invasive mole, choriocarcinoma(CCA), placental site trophoblastic tumor and epithelioid trophoblastic tumor(PSTT/ETT). GTN presents different degrees of proliferation, invasion and dissemination, but, if treated in reference centers, has high cure rates, even in multi-metastatic cases.The diagnosis of GTN following a hydatidiform molar pregnancy is made according to the International Federation of Gynecology and Obstetrics(FIGO)2000 criteria: four or more plateaued human chorionic gonadotropin(hCG)concentrations over three weeks; rise in hCG for three consecutive weekly measurements over at least a period of 2 weeks or more; and an elevated but falling hCG concentrations six or more months after molar evacuation. However,the latter reason for treatment is no longer used by many centers. In addition,GTN is diagnosed with a pathological diagnosis of CCA or PSTT/ETT. For staging after a molar pregnancy, FIGO recommends pelvic-transvaginal Doppler ultrasound and chest X-ray. In cases of pulmonary metastases with more than 1cm, the screening should be complemented with chest computed tomography and brain magnetic resonance image. Single agent chemotherapy, usually Methotrexate(MTX) or Actinomycin-D(Act-D), can cure about 70% of patients with FIGO/World Health Organization(WHO) prognosis risk score ≤ 6(low risk), reserving multiple agent chemotherapy, such as EMA/CO(Etoposide,MTX, Act-D, Cyclophosphamide and Oncovin) for cases with FIGO/WHO prognosis risk score ≥ 7(high risk) that is often metastatic. Best overall cure rates for low and high risk disease is close to 100% and > 95%, respectively. The management of PSTT/ETT differs and cure rates tend to be a bit lower. 2Dietary supplementation with free methionine or methionine dipeptide mitigates intestinal oxidative stress induced by Eimeria spp. challenge in broiler chickens显示文摘Background: This study evaluated the effects of Eimeria spp. challenge and dietary supplementation with free methionine or methionine dipeptide on animal performance;expression of genes associated with the immune system, antioxidant system, and amino acid transport in the jejunum;and redox status of the jejunum of broiler chickens.Methods: A randomized, 2 × 3 factorial design was used, in which Eimeria spp. challenge was the first factor(Eimeria-challenged, EC, or unchallenged, UC, broilers) and methionine supplementation was the second factor(non-supplemented, NS;free DL-methionine, DL-Met;and methionine dipeptide, DL-methionyl-DL-methionine, DLMMet). At 14 days of age, chickens were inoculated orally with sporulated oocysts of Eimeria acervulina, Eimeria praecox, Eimeria maxima, and Eimeria mitis. Birds were killed by cervical dislocation 144 h post-inoculation(PI), and the jejunum was collected for biochemical and molecular analyses.Results: EC broilers had a 13% lower feed intake(FI), 37% lower body weight gain(BWG), and 39% higher feed conversion ratio(FCR) than UC broilers. Chickens fed the DL-Met diet had higher BWG(about 12% higher) and better FCR(about 12% lower) than chickens fed the NS diet. EC chickens had lower relative weight of the bursa of Fabricius(51.8%) and higher relative weights of the spleen and whole intestine(53.6% and 26.3%, respectively) than UC chickens. Eimeria spp. challenge led to an increase in the levels of oxidative substances, such as nitrite and thiobarbituric acid reactive substances(TBARS), in the jejunum of chickens 144 h PI. Among UC chickens, those fed the DL-Met diet had higher total antioxidant capacity(TAC) and lower catalase(CAT) and superoxide dismutase(SOD) activities. EC chickens that received the NS diet had higher carbonylated protein content(CP). This result was associated with their lower TAC and catalase activity. The lower TAC in EC chickens might have been due to reduced expression of catalase(CAT) and superoxide dismutase 1(SOD1) genes. Chickens fed the DL-Met and DLMMet diets had lower nitrite content. Eimeria spp. challenge suppressed neutral amino acid transporter 1(B0 AT1),peptide transporter 1(PEPT1), toll-like receptor 5(TLR5), interleukin 2(IL2), and occludin(OCLN) gene expression and enhanced cationic amino acid transporter 1(CAT-1) and interferon gamma(IFNG) gene expression. 3Effects of estrogen receptor modulators on cytoskeletal proteins in the central nervous system显示文摘Estrogen receptor modulators are compounds of interest because of their estrogenic agonistic/antagonistic effects and tissue specificity. These compounds have many clinical applications, particularly for breast cancer treatment and osteoporosis in postmenopausal women, as well as for the treatment of climacteric symptoms. Similar to estrogens, neuroprotective effects of estrogen receptor modulators have been described in different models. However, the mechanisms of action of these compounds in the central nervous system have not been fully described. We conducted a systematic search to investigate the effects of estrogen receptor modulators in the central nervous system, focusing on the modulation of cytoskeletal proteins. We found that raloxifene, tamoxifen, and tibolone modulate some cytoskeletal proteins such as tau, microtuble-associated protein 1(MAP1), MAP2, neurofilament 38(NF38) by different mechanisms of action and at different levels: neuronal microfilaments, intermediate filaments, and microtubule-associated proteins. 4Initial floristic composition of rehabilitated gullies through bioengineering in the Mixteca Region, Sierra Madre del Sur, Mexico显示文摘Re-vegetation plays a fundamental role for erosion control and plant recovery in lands affected by gully erosion. Bioengineered practices facilitate the gullies rehabilitation. Objectives of the research were: 1) Identify taxonomically the pioneer vegetation on each gully section; 2) Characterize vegetation distribution preferences and 3) Assess structural/functional traits to recognize erosion control key species. Bioengineering was applied in a watershed belonging to Sierra Madre del Sur, at Oaxaca, Mexico, on eight gullies, with local support and minimal investment. 'La Mixteca' is a poor ecological and socio-economic region, comparable to other regions of the world. The Initial Floristic Composition(IFC) inventory is the baseline of the successional process. The transect method was used to determine the colonization of species. Cover abundance of registered species was estimated using the semi-quantitative scale of Braun-Blanquet. This procedure was repeated in five different positions(floor, hillslopes and tops), in the cross section of the gully. Throughcorrespondence analysis and clustering, the distribution of species was analyzed. Adequate responses were obtained in soil retention(quantity) and plant cover(existence and diversity); as measurable indicators of the bioengeneering works efficiency. Occupation of soil by native species from the Tropical Deciduous Forest was favored using live barriers. We detected species guilds with spatial distribution preferences in the gullies cross section. Plant cover characterization includes: native colonizer species, herbaceous, shrubby and trees of the forest community bordering the gully area, with cover abundance and structural/functional traits, useful to protect degraded areas. This spatial occupation process of plants responds to a secondary succession in gullies, where the proposed IFC model is correctly represented through bioengineering. 8The detrimental effects of lipopolysaccharideinduced neuroinflammation on adult hippocampal neurogenesis depend on the duration of the pro-inflammatory response显示文摘Adult hippocampal neurogenesis is a finely tuned process regulated by extrinsic factors. Neuroinflammation is a hallmark of several pathological conditions underlying dysregulation of neurogenesis. In animal models, lipopolysaccharide(LPS)-induced neuroinflammation leads to a neurogenic decrease mainly associated to the early inflammatory response. However, it is not well understood how the neuroinflammatory response progresses over time and if neurogenesis continues to be diminished during the late neuroinflammatory response. Moreover, it is unknown if repeated intermittent administration of LPS along time induces a greater reduction in neurogenesis. We administered one single intraperitoneal injection of LPS or saline or four repeated injections(one per week) of LPS or saline to young-adult mice. A cohort of new cells was labeled with three 5-bromo-2-deoxyuridine injections(one per day) 4 days after the last LPS injection. We evaluated systemic and neuroinflammation-associated parameters and compared the effects of the late neuroinflammatory response on neurogenesis induced by each protocol. Our results show that 1) a single LPS injection leads to a late pro-inflammatory response characterized by microglial activation, moderate astrocytic reaction and increased interleukin-6 levels. This response correlates in time with decreased neurogenesis and 2) a repeated intermittent injection of LPS does not elicit a late pro-inflammatory response although activated microglia persists. The latter profile is not accompanied by a continued longterm hippocampal neurogenic decrease. 9Species of Lasiodiplodia associated with mango in Brazil显示文摘Mango(Mangifera indica)is a major tropical fruit species cultivated in Brazil.The objective of this study was to identify species of Lasiodiplodia associated with dieback and stem-end rot of mango in the semi-arid region of Northeastern Brazil,and compare the species in relation to mycelial growth,pathogenicity and virulence.A total of 120 isolates of Lasiodiplodia were used and identifications were made using a combination of morphology and phylogenetic analysis based on partial translation elongation factor 1-αsequence(EF1-α)and internal transcribed spacers(ITS).The following species were identified:Lasiodiplodia crassispora,L.egyptiacae,L.hormozganensis,L.iraniensis,L.pseudotheobromae,L.theobromae and Lasiodiplodia sp..Lasiodiplodia theobromae was the most frequently isolated species,which represented 41%of all the isolates.Only this species had been previously reported on mango in Brazil,while the other species represent the first report associated with mango tree diseases in this country.Lasiodiplodia crassispora is reported for the first time associated with mango diseases worldwide.There were significant differences in mycelial growth rates among the Lasiodiplodia species and also in the optimum temperature for growth.All species of Lasiodiplodia were pathogenic on mango fruit.There were significant differences in virulence among the species,wherein L.hormozganensis and Lasiodiplodia sp.were the most virulent,while the least virulent were L.iraniensis,L.pseudotheobromae,L.crassispora and L.egyptiacae.Marília W.Marques Nelson B.Lima Marcos Antônio de Morais Jr Maria Angélica G.Barbosa Breno O.Souza Sami J.Michereff Alan J.L.Phillips Marcos P.S.Câmara 2013Fungal Diversity2013,,4:2
