1Overexpression of a dominant-negative mutant of SIRT1 in mouse heart causes cardiomyocyte apoptosis and early-onset heart failure显示文摘SIRT1,a mammalian ortholog of yeast silent information regulator 2(Sir2),is an NAD+-dependent protein deacetylase that plays a critical role in the regulation of vascular function.The current study aims to investigate the functional significance of deacetylase activity of SIRT1 in heart.Here we show that the early postnatal hearts expressed the highest level of SIRT1deacetylase activity compared to adult and aged hearts.We generated transgenic mice with cardiac-specific expression of a dominant-negative form of the human SIRT1(SIRT1H363Y),which represses endogenous SIRT1 activity.The transgenic mice displayed dilated atrial and ventricular chambers,and died early in the postnatal period.Pathological,echocardiographic and molecular phenotype confirmed the presence of dilated cardiomyopathy.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling analysis revealed a greater abundance of apoptotic nuclei in the hearts of transgenic mice.Furthermore,we show that cardiomyocyte apoptosis caused by suppression of SIRT1 activity is,at least in part,due to increased p53acetylation and upregulated Bax expression.These results indicate that dominant negative form of SIRT1(SIRT1H363Y)overexpression in mouse hearts causes cardiomyocyte apoptosis and early-onset heart failure,suggesting a critical role of SIRT1 in preserving normal cardiac development during the early postnatal period.MU WenLi ZHANG QingJun TANG XiaoQiang FU WenYan ZHENG Wei LU YunBiao LI HongLiang WEI YuSheng LI Li SHE ZhiGang CHEN HouZao LIU DePei 2014Science China(Life Sciences)2014,57,9:12
2Mo_2NiB_2基金属陶瓷的制备与性能显示文摘采用单质硼粉、镍粉和钼粉结合反应硼化烧结法制备了Mo2Ni B2基金属陶瓷,研究了Mo2Ni B2基金属陶瓷在烧结过程中的物相转变和尺寸变化以及烧结温度和保温时间对其力学性能和显微组织的影响.结果发现:随着烧结温度升高,材料物相逐渐由单质相变为二元硼化物相和三元硼化物相,并且材料的尺寸先发生细微收缩,再在硼化反应过程中逐渐增加,最后在液相烧结过程中逐渐减小;随着烧结温度升高,Mo2Ni B2基金属陶瓷的抗弯强度和硬度先增加后减小,在1290℃达到最大,分别为1346.5 MPa和83.7 HRA,并且硬质相颗粒逐渐粗化;保温时间对材料性能的影响与烧结温度一致,但在保温30min时抗弯强度最大(1453.3 MPa),保温60 min时硬度最大(83.7 HRA).易善杰 尹海清 殷小龙 曲选辉 ZHENG Qingjun 2015工程科学学报2015,37,4:8

4The Effect of 2,5-hexanedione on Myelin Protein Zero Expression,and Its Mitigation Using Ginkgo Biloba Extract显示文摘Objective To investigate the role of myelin protein zero (P 0) in 2,5-hexanedione (2,5-HD)-induced peripheral nerve injury,and the protective effect of Ginkgo biloba extract (Egb761) on 2,5-HD-induced toxic peripheral neuropathy.Methods After 4 weeks of treatment with 2,5-HD at different doses (50,100,200,400 mg/kg) in rats,changes in the levels of P 0 in rat sciatic nerves was investigated,and the effect of Egb761 on 2,5-HD-induced toxic peripheral neuropathy was studied.Results The blood-nerve barrier (BNB) permeability of the sciatic nerve increased,and the expression of P 0 mRNA and P 0 protein decreased in a dose-dependent manner after treatment with 2,5-HD for 4 weeks.Pretreatment with Egb761 protected against BNB interruption,and inhibited P 0 mRNA and protein reduction during 2,5-HD treatment.Pretreatment with Egb761 significantly reduced loss of body weight (P<0.01) and mitigated gait abnormalities (2.85±0.22) induced by 400 mg/kg 2,5-HD (P<0.01).It also reduced the signs of neurotoxicity induced by 2,5-HD.Conclusion 2,5-HD inhibited the expression of P 0 in a dose-dependent manner,and this may be an important mechanism by which toxic peripheral neuropathy is induced by 2,5-HD.Egb761 has a protective effect against 2,5-HD-induced peripheral neurotoxicity in rats.ZHAO Lei LIU QingJun CHEN Hong DUAN HuaWei BIN Ping LIU Qing NIU Yong DAI YuFei ZHENG YuXin 2011Biomedical and Environmental Sciences2011,24,4:3
5Revisiting the role of delta-1-pyrroline-5-carboxylate synthetase in drought-tolerant crop breeding显示文摘The delta-1-pyrroline-5-carboxylate synthetase(P5CS)gene exercises a protective function in stressed plants.However,the relationship between proline accumulation caused by P5CS and abiotic stress tolerance in plants is not always clear,as P5CS overexpression has been reported to repress plant growth under normal conditions in several reports.We re-evaluated the role of P5CS in drought-tolerant rice breeding by expressing the AtP5CS1 and feedback-inhibition-removed AtP5CS1(AtP5CS1^(F128A))genes under the regulation of an ABA-inducible promoter to avoid the potential side effects of P5CS overexpression under normal conditions.ABA-inducible AtP5CS1 and AtP5CS1^(F128A) increased seedling growth in a nutrient solution(under osmotic stress)and grain yield in pot plants.However,the evidently deleterious effects of AtP5CS1 on grain quality,tiller number,and grain yield in the field indicated the unsuitability of P5CS for drought-tolerance breeding.Xuanjun Feng Yue Hu Weixiao Zhang Rongqian Xie Huarui Guan Hao Xiong Li Jia Xuemei Zhang Hanmei Zhou Dan Zheng Ying Wen Qingjun Wang Fengkai Wu Jie Xu Yanli Lu 2022The Crop Journal2022,10,4:2
6Zinc-based metal organic framework with antibacterial and anti-inflammatory properties for promoting wound healing显示文摘The synergistic effect of antibacterial and anti-inflammatory is needed to overcome the problem of wound healing difficulties.Based on the favorable antibacterial and anti-inflammatory effect of zinc ions(Zn^(2+))and the physicochemical properties of metal organic frameworks(MOFs),we prepared nanosized zinc-based MOF:Zn-BTC with the ability to slowly release Zn^(2+).In cellular levels,Zn-BTC possessed lower toxicity to fibroblasts and enhanced capacity of cell proliferation and migration.It also had good bactericidal effect on multiple drug-resistant bacteria by reducing 41.4%MRSA and 47.2%Escherichia coli.In addition,Zn-BTC also displayed the ability of lowering the expression of antioxidant genes:superoxide dismutase 1,superoxide dismutase 2 and interleukin 6,and enhancing the expression of wound healing genes:transforming growth factors-b and type I collagen.Finally,it also demonstrated that Zn-BTC could effectively improve the skin wound healing of SD rats and had no toxicity on major organs.The favorable biocompatibility,antibacterial and anti-inflammatory properties of Zn-BTC gave a new insight of designing novel MOFs for promoting skin wound healing.Yuting Chen Jinhong Cai Dachang Liu Shuhan Liu Doudou Lei Li Zheng Qingjun Wei Ming Gao 2022Regenerative Biomaterials2022,9,1:2

9A pH-responsive mesoporous silica nanoparticles-based drug delivery system with controlled release of andrographolide for OA treatment显示文摘Andrographolide(AG)has favorable anti-inflammatory and antioxidative capacity.However,it has low bioavailability due to high lipophilicity and can be easily cleared by the synovial fluid after intra-articular injection,leading to low therapeutic efficiency in osteoarthritis(OA).Herein,we designed a nano-sized pH-responsive drug delivery system(DDS)for OA treatment by using modified mesoporous silica nanoparticles(MSNs)with pH-responsive polyacrylic acid(PAA)for loading of AG to form AG@MSNs-PAA nanoplatform.The nanoparticles have uniform size(120 nm),high drug loading efficiency(22.3860.71%)and pH-responsive properties,beneficial to sustained release in OA environment.Compared with AG,AG@MSNs-PAA showed enhanced antiarthritic efficacy and chondro-protective capacity based on IL-1b-stimulated chondrocytes and anterior cruciate ligament transection-induced rat OA model,as demonstrated by lower expression of inflammatory factors and better prevention of proteoglycan loss.Therefore,the AG@MSNs-PAA nanoplatform may be developed as a promising OA-specific and on-demand DDS.Mingwei He Zainen Qin Xiaonan Liang Xixi He Bikang Zhu Zhenhui Lu Qingjun Wei Li Zheng 2021Regenerative Biomaterials2021,8,4:1
10Involvement of a Putative Bipartite Transit Peptide in Targeting Rice Pheophorbide a Oxygenase into Chloroplasts for Chlorophyll Degradation during Leaf Senescence显示文摘Leaf senescence is one of the major factors contributing to the productivity and the grain quality in crops. The regulatory mechanism of leaf senescence remains largely unknown. Here, we report the identification and characterization of a rice early senescence 1(eas1)mutant, which displayed an early leaf senescence phenotype, accompanying by dwarfism and reduced tiller number, eventually leading to the reduction of grain yield. Map-based cloning revealed that the nuclear gene EAS1 encodes a pheophorbide a oxygenase(PaO), a key enzyme for chlorophyll breakdown. A highly conserved Thr residue of PaO was mutated into Ile in the eas1 mutant. Phylogenetic analysis indicates that PaO is an evolutionarily conserved protein, and EAS1 is 68% identical to the Arabidopsis ACCERLERATED CELL DEATH(ACD1) protein. Unlike ACD1 that contains a single transit peptide, EAS1 contains two putative transit peptides at its N-terminus, which are essential for its functionality, suggesting that targeting of EAS1 to the chloroplast is likely mediated by a putative bipartite transit peptide. Consistently, only a short version of EAS1 lacking the first putative transit peptide, but not the full-length EAS1,was capable of rescuing the Arabidopsis acd1 mutant phenotype. 17ODOR DISCRIMINATION BY MITRAL CELLS IN RAT OLFACTORY BULB USING MICROWIRE ARRAY RECORDING显示文摘Response features of mitral cells in the olfactory bulb were examined using principal component analysis to determine whether they contain information about odorant stimuli.Using microwire electrode array to record from the olfactory bulb in freely breathing anesthetized rats,we recorded responses of different mitral cells to saturated vapor of anisole(1 M),carvone(1 M),isobutanol(1 M),citral(1 M)and isoamyl actate(1 M).The responses of single mitral cells to the same odorant varied over time.The response profiles showed similarity during certain amount of period,which indicated that the response was not only depended on odor itself but also associated with context.Furthermore,the responses of single mitral cell to different odorants were observed with difference in firing rate.In order to recognize different odorant stimuli,we apply four cells as a sensing group for classification using principal component analysis.Features of each cell’s response describing both temporal and frequency characteristics were selected.The results showed that five different single molecular odorants can be distinguished from each other.These data suggest that action potentials of mitral cells may play a role in odor coding.JUN ZHOU QI DONG LIUJING ZHUANG QINGJUN LIU SHAOMIN ZHANG XIAOXIANG ZHENG PING WANG 2012Journal of Innovative Optical Health Sciences2012,5,1:0
18Obstructor,a Frankliniella occidentalis protein,promotes transmission of tomato spotted wilt orthotospovirus显示文摘Tomato spotted wilt orthotospovirus(TSWV)causes substantial economic losses to vegetables and other crops.TSWV is mainly transmitted by thrips in a persistent and proliferative manner,and its most efficient vector is the western flower thrips,Frankliniella occidentalis(Pergande).In moving from the thrips midgut to the salivary glands in preparation for transmission,the virions must overcome multiple barriers.Although several proteins that interact with TSWV in thrips have been characterized,we hypothesized that additional thrips proteins interact with TSWV and facilitate its transmission.In the current study,67 F occidentalis proteins that interact with GN(a structural glycoprotein)were identified using a split-ubiquitin membrane-based yeast 2-hybrid(MbY2H)system.Three proteins,apolipoprotein-D(ApoD),orai-2-like(Orai),and obstructor-E-like isoform X2(Obst),were selected for further study based on their high abundance and interaction strength;their interactions with Gn were confirmed by MbY2H,yeastβ-galactosidase and luciferase complementation assays.The relative expressions of ApoD and Orai were significantly down-regulated but that of Obst was significantly up-regulated in viruliferous thrips.When interfering with Obst in larval stage,the TSWV acquisition rate in 3 independent experiments was significantly decreased by 26%,40%,and 35%,respectively.In addition,when Obst was silenced in adults,the virus titer was significantly decreased,and the TSWV transmission rate decreased from 66.7%to 31.9%using the leaf disk method and from 86.67%to 43.33%using the living plant method.However,the TSWV acquisition and transmission rates were not affected by interference with the ApoD or Orai gene.The results indicate that Obst may play an important role in TSWV acquisition and transmission in Frankliniella occidentalis.Xiaobin Zheng Yanran Wan Min Tao Jiangjiang Yuan Kun Zhang Jing Wang Youjun Zhang Pei Liang Qingjun Wu 2023Insect Science2023,30,3:0
