| 1 | Hi-TOM: a platform for high-throughput tracking of mutations induced by CRISPR/Cas systems显示文摘The CRISPR/Cas system has been extensively applied to make precise genetic modifications in various organisms. Despite its importance and widespread use, large-scale mutation screening remains time-consuming, labour-intensive and costly. Here, we developed Hi-TOM(available at http://gffzz103f3ce5d9b14419h99659kobvb0k6590.ffgz.tsg.suse.edu.cn/hi-tom/), an online tool to track the mutations with precise percentage for multiple samples and multiple target sites. We also described a corresponding next-generation sequencing(NGS) library construction strategy by fixing the bridge sequences and barcoding primers. Analysis of the samples from rice, hexaploid wheat and human cells reveals that the Hi-TOM tool has high reliability and sensitivity in tracking various mutations, especially complex chimeric mutations frequently induced by genome editing. Hi-TOM does not require special design of barcode primers,cumbersome parameter configuration or additional data analysis. | 5 | Manipulating mRNA splicing by base editing in plants显示文摘Precursor-mRNAs(pre-mRNA) can be processed into one or more mature m RNA isoforms through constitutive or alternative splicing pathways. Constitutive splicing of pre-mRNA plays critical roles in gene expressional regulation, such as intronmediated enhancement(IME), whereas alternative splicing(AS) dramatically increases the protein diversity and gene functional regulation. However, the unavailability of mutants for individual spliced isoforms in plants has been a major limitation in studying the function of mRNA splicing. Here, we describe an efficient tool for manipulating the splicing of plant genes. Using a Cas9-directed base editor, we converted the 5′ splice sites in four Arabidopsis genes from the activated GT form to the inactive AT form. Silencing the AS of HAB 1.1(encoding a type 2 C phosphatase) validated its function in abscisic acid signaling, while perturbing the AS of RS31 A revealed its functional involvement in plant response to genotoxic treatment for the first time. Lastly,altering the constitutive splicing of Act2 via base editing facilitated the analysis of IME. | 7 | Hierarchical method of task assignment for multiple cooperating UAV teams显示文摘The problem of task assignment for multiple cooperating unmanned aerial vehicle(UAV) teams is considered. Multiple UAVs forming several small teams are needed to perform attack tasks on a set of predetermined ground targets. A hierarchical task assignment method is presented to address the problem. It breaks the original problem down to three levels of sub-problems: target clustering, cluster allocation and target assignment. The first two sub-problems are centrally solved by using clustering algorithms and integer linear programming, respectively, and the third sub-problem is solved in a distributed and parallel manner, using a mixed integer linear programming model and an improved ant colony algorithm. The proposed hierarchical method can reduce the computational complexity of the task assignment problem considerably, especially when the number of tasks or the number of UAVs is large. Pre-treated shark skin was used as casting template to prepare a flexible female die of silicone rubber by soft die formation. A waterborne epoxy resin was then used to graft long-chains of drag reduction agent and prepare a synthetic drag reduction shark skin with nano-long chain drag reduction interface and lifelike micro-grooves. Replication precision analysis shows that this technology could replicate the complicated three-dimensional morphology of a biological drag reduction surface with high precision. | 12 | Mercury oxidation and adsorption characteristics of potassium permanganate modified lignite semi-coke显示文摘The adsorption characteristics of virgin and potassium permanganate modified lignite semi-coke(SC)for gaseous Hg0 were investigated in an attempt to produce more effective and lower price adsorbents for the control of elemental mercury emission.Brunauer-EmmettTeller(BET)measurements,X-ray powder diffraction(XRD)and X-ray photoelectron spectroscopy(XPS)were used to analyze the surface physical and chemical properties of SC,Mn-SC and Mn-H-SC before and after mercury adsorption.The results indicated that potassium permanganate modification had significant influence on the properties of semi-coke,such as the specific surface area,pore structure and surface chemical functional groups.The mercury adsorption efficiency of modified semi-coke was lower than that of SC at low temperature,but much higher at high temperature.Amorphous Mn7+,Mn6+ and Mn 4+ on the surface of Mn-SC and Mn-H-SC were the active sites for oxidation and adsorption of gaseous Hg0,which oxidized the elemental mercury into Hg2+ and captured it.Thermal treatment reduced the average oxidation degree of Mnx+ on the surface of Mn-SC from 3.80 to 3.46.However,due to the formation of amorphous MnOx,the surface oxidation active sites for gaseous Hg0 increased,which gave Mn-H-SC higher mercury adsorption efficiency than that of Mn-SC at high temperature. | Huawei Zhang Jitao Chen Peng Liang Li Wang | 2012 | Journal of Environmental Sciences2012,24,12: | 12 |
| 13 | Effect of tip geometry and tip clearance on aerodynamic performance of a linear compressor cascade显示文摘The tip leakage flow between a blade and a casing wall has a strong impact on compressor pressure rise capability, efficiency, and stability. Consequently, there is a strong motivation to look for means to minimize its impact on performance. This paper presents the potential of passive tip leakage flow control to increase the aerodynamic performance of highly loaded compressor blades. Experimental investigations on a linear compressor cascade equipped with blade winglets mounted to the blade tips have been carried out. Results for a variation of the tip clearance and the winglet geometry are presented. Current results indicate that the use of proper tip winglets in a compressor cascade can positively affect the local aerodynamic field by weakening the tip leakage vortex. Results also show that the suction-side winglets are aerodynamically superior to the pressure-side or combined winglets. The available literature on the RCF testing of thermal spray coatings under various condition services is considerable; it is generally difficult to synthesize all of the result to obtain a comprehensive understanding of the parameters which has a great effect on a thermal spray coating's resistance of RCF. The effects of service conditions(lubrication states, contact stresses, revolve speed, and slip ratio) on the changing of thermal spray coatings' contact fatigue lifetime is introduced systematically. The effects of different service condition on RCF failure mechanism of thermal spray coating from the change of material structure integrity are also summarized. Moreover, In order to enhance the RCF performance, the parameter optimal design formula of service condition and material structure integrity is proposed based on the effect of service condition on thermal spray coatings' contact fatigue lifetime and RCF failure mechanism. The shortage of available literature and the forecast focus in future researches are discussed based on available research. In the present paper, attention is given to new experimental techniques by incorporating a damagemodified constitutive model into the SHPB technique and combining the Hopkinson pressure bar(HPB) technique with WPIA. First, to distinguish the response due to dynamic constitutive behavior and the response due to dynamic damage evolution, the SHPB method incorporating a damage-modified constitutive model is developed, including an explicit damage-modified Zhu–Wang–Tang model and an implicit damage-modified constitutive model. Second, when the SHPB results become invalid, a method of combining new Lagrange inverse analyses with the HPB technique is developed, including cases of the HPB arranged in front of a long specimen and behind the specimen.
