1Straw strip mulching in a semiarid rainfed agroecosystem achieves winter wheat yields similar to those of full plastic mulching by optimizing the soil hydrothermal regime显示文摘Straw strip mulching(SM)is a new mulching technology.From 2012 to 2018,SM’s effects on soil moisture and temperature and production performances were compared with other mulching practices,using three treatments:full-cover plastic mulch(PM),no mulch with wheat sown in rows as the control(CK),and SM with 50%to 59%of the field area mulched.Compared with CK,on average over six growing seasons,SM and PM increased grain yield by 27.0%and 21.7%,straw yield by 21.6%and 22.6%,kernels ha;by 26.6%and 19.0%,net income by 29.8%and-25.0%,soil temperature at 5 cm by-1.5°C and 0.2°C from overwintering to maturity,and soil water storage at 0–200 cm by 25 and 22 mm,respectively.The increase in soil moisture in SM and PM was greater in the early period(overwintering to jointing)than in the later period(booting to maturity)and at 0 to 120 cm than at 120–200 cm in the early period.Although the mean evapotranspiration of whole growth period across six seasons was similar among treatments,SM and PM increased water consumption during the key formation period of yield components after overwintering by 16 and 32 mm,respectively,while reducing it before overwintering.Compared with CK,SM and PM had the effects of warming during overwintering and cooling after jointing.By increasing water consumption after overwintering and ratio of transpiration to evapotranspiration and providing favorable soil temperature for multiple growth stages and more sufficient soil moisture,SM and PM promoted vegetative growth and increased kernels ha^(-1),the main mechanisms by which SM and PM increased grain yield relative to CK.Relative to PM,SM is a more economically beneficial and environment-friendly technology for dryland wheat production.Yuwei Chai Qiang Chai Rui Li Yawei Li Changgang Yang Hongbo Cheng Lei Chang Shouxi Chai 2022The Crop Journal2022,10,3:6
2Stalagmite-inferred Holocene precipitation in northern Guizhou Province, China, and asynchronous termination of the Climatic Optimum in the Asian monsoon territory显示文摘An absolute-dated, bi-decadal-resolution, stalagmite oxygen-isotopic time series from Shigao Cave reveals the evolution of summer monsoon precipitation over the past 9.9 ka BP in northern Guizhou Province, Southwest China. The 18O-inferred climate conditions are divisible into three distinct stages: (1) a maximum humid era from 9.9-6.6 ka BP; (2) a gradual declining precipitation interval between 6.6-1.6 ka BP; and (3) a relatively low precipitation time window after 1.6 ka BP. Consistency of contemporaneous stalagmite Holocene 18O records between Shigao and other caves in the Indian and East Asian monsoon realms support the effect of primary orbital solar forcings on monsoonal precipitation. However, statistical analysis shows a significant spatial asynchroneity of the Holocene Optimum termination in the Asian monsoon territory. The Holocene Optimum ended at 7.2-7.4 ka BP in Oman, located in the Indian monsoon region, and at 5.6-5.8 ka BP in Central China, in the East Asian monsoon zone. In Southwest China, the termination occurred between these periods, at 6.6-7.0 ka BP, and was influenced by both monsoon systems. 3SARS-CoV-2 ORF10 suppresses the antiviral innate immune response by degrading MAVS through mitophagy显示文摘The global coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused severe morbidity and mortality in humans.It is urgent to understand the function of viral genes.However,the function of open reading frame 10(ORF10),which is uniquely expressed by SARS-CoV-2,remains unclear.In this study,we showed that overexpression of ORF10 markedly suppressed the expression of type I interferon(IFN-I)genes and IFN-stimulated genes.Then,mitochondrial antiviral signaling protein(MAVS)was identified as the target via which ORF10 suppresses the IFN-I signaling pathway,and MAVS was found to be degraded through the ORF10-induced autophagy pathway.Furthermore,overexpression of ORF10 promoted the accumulation of LC3 in mitochondria and induced mitophagy.Mechanistically,ORF10 was translocated to mitochondria by interacting with the mitophagy receptor Nip3-like protein X(NIX)and induced mitophagy through its interaction with both NIX and LC3B.Moreover,knockdown of NIX expression blocked mitophagy activation,MAVS degradation,and IFN-I signaling pathway inhibition by ORF10.Consistent with our observations,in the context of SARS-CoV-2 infection,ORF10 inhibited MAVS expression and facilitated viral replication.In brief,our results reveal a novel mechanism by which SARS-CoV-2 inhibits the innate immune response;that is,ORF10 induces mitophagy-mediated MAVS degradation by binding to NIX.Xingyu Li Peili Hou Wenqing Ma Xuefeng Wang Hongmei Wang Zhangping Yu Huasong Chang Tiecheng Wang Song Jin Xue Wang Wenqi Wang Yudong Zhao Yong Zhao Chunqing Xu Xiaomei Ma Yuwei Gao Hongbin He 2022Cellular & Molecular Immunology2022,19,1:4
4AEG-1 expression correlates with CD133 and PPP6c levels in human glioma tissues显示文摘Astrocyte elevated gene-1(AEG-1) is associated with tumor genesis and progression in a variety of human cancers.This study aimed to explore the significance of AEG-l in glioma and investigate whether it correlated with radioresistance of glioma cells.Immunohistochemical staining showed that the intensity of AEG-l,CD133 and PPP6 c protein expression in glioma tissues increased significantly,mainly in the cytoplasm.The expression rate of AEG-l,CD133 and PPP6c were 85.9%(67/78),60.3%(47/78) and 65.8%(51/78),respectively.AEG-l expression was correlated with age(r=0.227,P=0.045),clinical stage(r=0.491,P<0.001) and clinical grade(r=0.450,P<0.001).No correlation was found between AEG-l expression and other clinicopathologic parameters(P>0.05).The expression of AEG-1 was positively correlated with the expression of CD133(r=0.240,P = 0.035) and PPP6c(r= 0.250,P = 0.027).In addition,retrieved data on TCGA implied co-occurrence of genomic alterations of AEG-l and PPP6 c in glioblastoma.Our findings indicate that AEG-l is positively correlated with CD133 and AEG-l expression.It may play an important role in the progression of glioma and may serve as potential novel marker of chemoresistance and radioresistance.Jia Guo Xin Chen Ruxing Xi Yuwei Chang Xuanwei Zhang Xiaozhi Zhang 2014The Journal of Biomedical Research2014,28,5:3

13Neurodegeneration-associated FUS is a novel regulator of circadian gene expression显示文摘Background:Circadian rhythms are oscillating physiological and behavioral changes governed by an internal molecular clock,and dysfunctions in circadian rhythms have been associated with ageing and various neurodegenerative diseases.However,the evidence directly connecting the neurodegeneration-associated proteins to circadian control at the molecular level remains sparse.Methods:Using meta-analysis,synchronized animals and cell lines,cells and tissues from FUS R521C knock-in rats,we examined the role of FUS in circadian gene expression regulation.Results:We found that FUS,an oscillating expressed nuclear protein implicated in the pathogenesis of amyotrophic lateral sclerosis(ALS)and frontotemporal dementia(FTD),exerted a novel feedback route to regulate circadian gene expression.Nr1d1-encoded core circadian protein REV-ERBαbound the Fus promoter and regulated the expression of Fus.Meanwhile,FUS was in the same complex as PER/CRY,and repressed the expression of E box-containing core circadian genes,such as Per2,by mediating the promoter occupancy of PSF-HDAC1.Remarkably,a common pathogenic mutant FUS(R521C)showed increased binding to PSF,and caused decreased expression of Per2.Conclusions:Therefore,we have demonstrated FUS as a modulator of circadian gene expression,and provided novel mechanistic insights into the mutual influence between circadian control and neurodegeneration-associated proteins.Xin Jiang Tao Zhang Haifang Wang Tao Wang Meiling Qin Puhua Bao Ruiqi Wang Yuwei Liu Hung-Chun Chang Jun Yan Jin Xu 2018Translational Neurodegeneration2018,7,1:1

16CD97 negatively regulates the innate immune response against RNA viruses by promoting RNF125-mediated RIG-I degradation显示文摘The G protein-coupled receptor ADGRE5(CD97)binds to various metabolites that play crucial regulatory roles in metabolism.However,its function in the antiviral innate immune response remains to be determined.In this study,we report that CD97 inhibits virus-induced type-I interferon(IFN-I)release and enhances RNA virus replication in cells and mice.CD97 was identified as a new negative regulator of the innate immune receptor RIG-I,and RIG-1 degradation led to the suppression of the IFN-I signaling pathway.Furthermore,overexpression of CD97 promoted the ubiquitination of RIG-I,resulting in its degradation,but did not impact its mRNA expression.Mechanistically,CD97 upregulates RNF125 expression to induce RNF125-mediated RIG-I degradation via K48-linked ubiquitination at Lys181 after RNA virus infection.Most importantly,CD97-deficient mice are more resistant than wild-type mice to RNA virus infection.We also found that sanguinarine-mediated inhibition of CD97 effectively blocks VSV and SARS-CoV-2 replication.These findings elucidate a previously unknown mechanism through which CD97 negatively regulates RIG-I in the antiviral innate immune response and provide a molecular basis for the development of new therapeutic strategies and the design of targeted antiviral agents.Huasong Chang Peili Hou Xuefeng Wang Aibiao Xiang Hao Wu Wenjing Qi Rukun Yang Xue Wang Xingyu Li Wenqi He Guimin Zhao Weiyang Sun Tiecheng Wang Daniel Chang He Hongmei Wang Yuwei Gao Hongbin He 2023Cellular & Molecular Immunology2023,20,12:0

18Synthesis, Characterization and Luminescence Properties of Rare Earth Complexes with Bis(Pyridinyl-Amide)显示文摘A ligand, N1,N4-di(pyridin-2-yl)succinamide (L) and its lanthanide(Ⅲ) complexes (RE= La, Eu, Tb, Gd, Yb ) were synthesized and characterized in detail. The results indicate that the composition of the binary complexes is determined as [REL(H2O)2(NO3)2] NO3, that the complexes are 1∶1 electrolytes in DMF, and that the Tb 3+ complex has brightly green fluorescence in a solid state. At the same time, the energy levels of the excited triplet states for the six ligands were determined to be 22989 cm -1 . The fact that the ligand sensitize Tb 3+ complexes was explained by the relative energy gap between the lowest triplet energy level of the ligand (T) and 5DJ of Tb 3+ or Eu 3+ . When 2000 cm -1 <ΔE(T-5D4)<3000 cm -1 , the luminescent intensity of the Tb 3+ complex is stronger. When 3000 cm -1 <ΔE(T-5D1), the luminescent intensity of the Eu 3+ complex is weaker or close to nil. 19A cell marker-based clustering strategy(cmCluster)for precise cell type identification of scRNA-seq data显示文摘Background:The precise and efficient analysis of single-cell transcriptome data provides powerful support for studying the diversity of cell functions at the single-cell level.The most important and challenging steps are cell clustering and recognition of cell populations.While the precision of clustering and annotation are considered separately in most current studies,it is worth attempting to develop an extensive and flexible strategy to balance clustering accuracy and biological explanation comprehensively.Methods:The cell marker-based clustering strategy(cmCluster),which is a modified Louvain clustering method,aims to search the optimal clusters through genetic algorithm(GA)and grid search based on the cell type annotation results.Results:By applying cmCluster on a set of single-cell transcriptome data,the results showed that it was beneficial for the recognition of cell populations and explanation of biological function even on the occasion of incomplete cell type information or multiple data resources.In addition,cmCluster also produced clear boundaries and appropriate subtypes with potential marker genes.The relevant code is available in GitHub website(huangyuwei301/cmCluster).Conclusions:We speculate that cmCluster provides researchers effective screening strategies to improve the accuracy of subsequent biological analysis,reduce artificial bias,and facilitate the comparison and analysis of multiple studies.Yuwei Huang Huidan Chang Xiaoyi Chen Jiayue Meng Mengyao Han Tao Huang Liyun Yuan Guoqing Zhang 2023Quantitative Biology2023,11,2:0
20Transcriptomic but not genomic variability confers phenotype of breast cancer stem cells显示文摘Background:Breast cancer stem cells(BCSCs)are considered responsible for cancer relapse and drug resistance.Understanding the identity of BCSCs may open new avenues in breast cancer therapy.Although several discoveries have been made on BCSC characterization,the factors critical to the origination of BCSCs are largely unclear.This study aimed to determine whether genomic mutations contribute to the acquisition of cancer stem-like phenotype and to investigate the genetic and transcriptional features of BCSCs.Methods:We detected potential BCSC phenotype-associated mutation hotspot regions by using whole-genome sequencing on parental cancer cells and derived serial-generation spheres in increasing order of BCSC frequency,and then performed target deep DNA sequencing at bulk-cell and single-cell levels.To identify the transcriptional program associated with BCSCs,bulk-cell and single-cell RNA sequencing was performed.Results:By using whole-genome sequencing of bulk cells,potential BCSC phenotype-associated mutation hotspot regions were detected.Validation by target deep DNA sequencing,at both bulk-cell and single-cell levels,revealed no genetic changes specifically associated with BCSC phenotype.Moreover,single-cell RNA sequencing showed profound transcriptomic variability in cancer cells at the single-cell level that predicted BCSC features.Notably,this transcriptomic variability was enriched during the transcription of 74 genes,revealed as BCSC markers.Breast cancer patients with a high risk of relapse exhibited higher expression levels of these BCSC markers than those with a low risk of relapse,thereby highlighting the clinical significance of predicting breast cancer prognosis with these BCSC markers.Conclusions:Transcriptomic variability,not genetic mutations,distinguishes BCSCs from non-BCSCs.The identified 74 BCSC markers have the potential of becoming novel targets for breast cancer therapy.Mengying Tong Ziqian Deng Mengying Yang Chang Xu Xiaolong Zhang Qingzheng Zhang Yuwei Liao Xiaodi Deng Dekang Lv Xuehong Zhang Yu Zhang Peiying Li Luyao Song Bicheng Wang Aisha Al-Dherasi Zhiguang Li Quentin Liu 2018Cancer Communications2018,38,1:0
