| 2 | Diversity, structure and function of the coiled-coil domains of plant NLR immune receptors显示文摘Plant nucleotide-binding, leucine-rich repeat receptors(NLRs) perceive pathogen avirulence effectors and activate defense responses.Nucleotide-binding, leucine-rich repeat receptors are classified into coiled-coil(CC)-containing and Toll/interleukin-1 receptor(TIR)-containing NLRs. Recent advances suggest that NLR CC domains often function in signaling activation, especially for induction of cell death. In this review, we outline our current understanding of NLR CC domains, including their diversity/classification and structure, their roles in cell death induction, disease resistance, and interaction with other proteins. | 3 | Construction of chlorogenic acid-containing liposomes with prolonged antitumor immunity based on T cell regulation显示文摘As a potential cancer immunotherapeutic agent,chlorogenic acid(CHA)has entered phase II clinical trials in China as a lyophilized powder formulation for treating glioma.However,the in vivo instability of CHA necessitates daily intramuscular injections,resulting in patient noncompliance.In this study,CHA-phospholipid complex(PC)-containing PEGylated liposomes(CHA-PC PEG-Lipo,named as CPPL),with CHA-PC as the drug intermediate,were prepared to lower the administration frequency.CPPL demonstrated excellent physicochemical properties,enhanced tumor accumulation,and inhibited tumor growth even when the administration interval was prolonged to 4 days when compared to a CHA solution and CHA-PC loaded liposomes(CHA-PC Lipo,labeled as CPL),both of which only demonstrated antitumor efficacy with once-daily administration.Further evaluation of the in vivo antitumor immune mechanism suggested that the extended antitumor immune efficacy of CPPL could be attributed to its distinct immune-stimulating mechanism when compared with CHA solution and CPL,such as stimulating both CD4+and CD8+T cell infiltration,inhibiting myeloid-derived suppressor cell expression,reducing the expression of Th2 related factors,and notably,increasing the memory T cells in tumor tissues.This CHA-containing formulation could reduce the frequency of in vivo CHA administration during cancer treatment via T cells,especially memory T cell regulation. | Yun Zhang Yanfang Yang Jun Ye Yue Gao Hengfeng Liao Junzhuo Zhou Yu Feng Dongdong Liu Yingying Meng Xiaoguang Chen Lili Gao Yuling Liu | 2021 | Science China(Life Sciences)2021,64,7: | 2 |
| 4 | The Establishment of Infectious Clone and Single Round Infectious Particles for Coxsackievirus A10显示文摘Coxsackievirus A10(CVA10)is one of the major etiological agents of hand,foot,and mouth disease.There are no vaccine and antiviral drugs for controlling CVA10 infection.Reverse genetic tools for CVA10 will benefit its mechanistic study and development of vaccines and antivirals.Here,two infectious clones for the prototype and a Myc-tagged CVA10 were constructed.Viable CVA10 viruses were harvested by transfecting the viral m RNA into human rhabdomyosarcoma(RD)cells.Rescued CVA10 was further confirmed by next generation sequencing and characterized experimentally.We also constructed the vectors for CVA10 subgenomic replicon with luciferase reporter and viral capsid with EGFP reporter,respectively.Co-transfection of the viral replicon RNA and capsid expresser in human embryonic kidney 293 T(HEK293 T)cells led to the production of single round infectious particles(SRIPs).Based on CVA10 replicon RNA,SRIPs with either the enterovirus A71(EVA71)capsid or the CVA10 capsid were generated.Infection by EVA71 SRIPs required SCARB2,while CVA10 SRIPs did not.Finally,we showed great improvement of the replicon activity and SRIPs production by insertion of a cis-active hammerhead ribozyme(HHRib)before the 50-untranslated region(UTR).In summary,reverse genetic tools for prototype strain of CVA10,including both the infectious clone and the SRIPs system,were successfully established.These tools will facilitate the basic and translational study of CVA10. | Min Wang Jingjing Yan Liuyao Zhu Meng Wang Lizhen Liu Rui Yu Ming Chen Jingna Xun Yuling Zhang Zhigang Yi Shuye Zhang | 2020 | Virologica Sinica2020,35,4: | 1 |

| 6 | The novel peptide Nb PPI1 identified from Nicotiana benthamiana triggers immune responses and enhances resistance against Phytophthora pathogens显示文摘In plants, recognition of small secreted peptides, such as damage/danger-associated molecular patterns(DAMPs), regulates diverse processes,includingstressandimmune responses. Here, we identified an SGPS(Ser-Gly-Pro-Ser) motif-containing peptide, Nicotiana tabacum NtPROPPI, and its two homologs in Nicotiana benthamiana, NbPROPPI1 and NbPROPPI2. Phytophthora parasitica infection and salicylic acid(SA) treatment induced NbPROPPI1/2 expression. Moreover, SignalP predicted that the 89-amino acid NtPROPPI includes a 24-amino acid N-terminal signal peptide and NbPROPPI1/2-GFP fusion proteins were mainly localized to the periplasm. Transient expression of NbPROPPI1/2 inhibited P.parasiticacolonization,andNbPROPPI1/2 knockdown rendered plants more susceptible to P. parasitica. An eight-amino-acid segment in the NbPROPPI1 C-terminus was essential for its immune function and a synthetic 20-residue peptide, NbPPI1, derived from the C-terminus of NbPROPPI1 provoked significant immune responses in N. benthamiana. These responses led to enhanced accumulation of reactive oxygen species, activation of mitogen-activated protein kinases, and up-regulation of the defense genes Flg22-induced receptor-like kinase(FRK) and WRKY DNA-binding protein 33(WRKY33). The NbPPI1-induced defense responses require Brassinosteroid insensitive1-associated receptor kinase 1(BAK1). | 10 | Observation of regular pulse train in a narrow-band optoelectronic oscillator显示文摘We have experimentally observed a new operating state of a regular pulse train in a narrow-band optoelectronic oscillator(OEO) system, where the direct-current(DC) bias of the Mach-Zehnder modulator is set at the maximum value of the transmission transfer function instead of the usual quadrature point. The observed quasi-steady-state pulse train is distinctly periodic, with a period of 10.5 μs and a center frequency of 10 GHz, and resembles a mode-locked OEO in its waveform. The formation of regular pulses here may arise from the dynamic balance of nonlinearity and narrow-band filter effects, with the transient characteristics of the pulses arising mainly from instabilities between the gain and cavity loss. | 12 | Study on the Deformation Behavior of Mg-Gd-Y-Zn-Mn Wrought Magnesium Alloys by Visco-plastic Self-consistent Modeling显示文摘The plastic deformation behavior of new Mg-Gd-Y-Zn-Mn magnesium alloys gains great necessity to clarify and understand the mechanism deeply. In the present work,the tensile mechanical property test and visco-plastic self-consistent (VPSC) model are used to investigate the activities of deformation modes of VW84M and VW94M magnesium alloys during the tensile deformation. The results show that the mechanical properties of the above extruded alloys are similar but VW94M has higher strength than VW84M after the same aging process. Compared with the extruded alloys,the as-aged alloys have significantly higher activation of pyramidal slip at the later stage of plastic deformation. In addition,the as-aged VW94M alloy with higher strength has the largest activity of pyramidal slip. | 13 | U(VI) adsorption behavior onto polypyrrole coated 3R-MoS2 nanosheets prepared with the molten salt electrolysis method显示文摘To improve the separation capacity of uranium in aqueous solutions, 3R-MoS2 nanosheets were prepared with molten salt electro- lysis and further modified with polypyrrole (PPy) to synthesize a hybrid nanoadsorbent (PPy/3R-MoS2). The preparation conditions of PPy/3R- MoS2 were investigated and the obtained nanosheets were characterized with scanning electron microscope (SEM), high resolution transmis- sion electron microscope (HRTEM), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectro- scopy (XPS). The results showed that PPy/3R-MoS2 exhibited enhanced adsorption capacity toward U(VI) compared to pure 3R-MoS2 and PPy;the maximum adsorption was 200.4 mg/g. The adsorption mechanism was elucidated with XPS and FTIR: (1) negatively charged PPy/3R-MoS2 nanosheets attracted by an electrostatic interaction;(2) exposed C, N, Mo, and S atoms complexed with U(VI) through co- ordination;(3) Mo in the complex partly reduced the adsorbed U(VI) to U(IV), which further regenerated the adsorption point and continu- ously adsorbed U(VI). | 14 | Boosting Li-ion storage in Li2MnO_(3) by unequal-valent Ti4+-substitution and interlayer Li vacancies building显示文摘Lithium rich layered oxide(LRLO) has been considered as one of the promising cathodes for lithium-ion batteries(LIBs). The high voltage and large capacity of LRLO depend on Li2MnO_(3)phase. To ameliorate the electrochemical performance of Li2MnO_(3), also written as Li(Li1/3Mn2/3)O_(2), we propose a strategy to substitute Mn4+and Li+in Mn/Li transition metal layer with Ti4+, which can stabilize the structure of Li2MnO_(3)by inhibiting the excessive oxidation of O_(2)-above 4.5 V. More significantly, the unequal-valent substitution brings about the emergence of interlayer Li vacancies, which can promote the Li-ion diffusion based on the enlarged interlayer and increase the capacity by activating the Mn3+/4+redox. We designed Li0.7[Li1/3Mn2/3]0.7Ti0.3O_(2)with high interlayer Li vacancies, which presents a high capacity(290 m Ah/g at 10 m A/g) and stable cycling performance(84% over 60 cycles at 50 m A/g).
