1Characterization of transient groundwater flow through a high arch dam foundation during reservoir impounding显示文摘Even though a large number of large-scale arch dams with height larger than 200 m have been built in the world, the transient groundwater flow behaviors and the seepage control effects in the dam foundations under difficult geological conditions are rarely reported. This paper presents a case study on the transient groundwater flow behaviors in the rock foundation of Jinping I double-curvature arch dam, the world's highest dam of this type to date that has been completed. Taking into account the geological settings at the site, an inverse modeling technique utilizing the time series measurements of both hydraulic head and discharge was adopted to back-calculate the permeability of the foundation rocks,which effectively improves the uniqueness and reliability of the inverse modeling results. The transient seepage flow in the dam foundation during the reservoir impounding was then modeled with a parabolic variational inequality(PVI) method. 2Umbilical cord mesenchymal stem cell transplantation for the treatment of Duchenne muscular dystrophy显示文摘Due to their relative abundance,stable biological properties and excellent reproductive activity,umbilical cord mesenchymal stem cells have previously been utilized for the treatment of Duchenne muscular dystrophy,which is a muscular atrophy disease.Three patients who were clinically and pathologically diagnosed with Duchenne muscular dystrophy were transplanted with umbilical cord mesenchymal stem cells by intravenous infusion,in combination with multi-point intramuscular injection.They were followed up for 12 months after cell transplantation.Results showed that clinical symptoms significantly improved,daily living activity and muscle strength were enhanced,the sero-enzyme,electromyogram,and MRI scans showed improvement,and dystrophin was expressed in the muscle cell membrane.Hematoxylin-eosin staining of a muscle biopsy revealed that muscle fibers were well arranged,fibrous degeneration was alleviated,and fat infiltration was improved.These pieces of evidence suggest that umbilical cord mesenchymal stem cell transplantation can be considered as a new regimen for Duchenne muscular dystrophy.Xiaofeng Yang Yifeng Xu Naiwu Lu Yibin Zhang Hongmei Wang Xin Lu Jiping Cui JinxuZhou Hong Shan Yanxiang Wu Xinping Liu 2011Neural Regeneration Research2011,6,10:4
3Genome-wide association study and transcriptome analysis reveal new QTL and candidate genes for nitrogen-deficiency tolerance in rice显示文摘The development of rice cultivars with improved nitrogen use efficiency(NUE)is desirable for sustainable agriculture.Achieving this goal depends in part on understanding how rice responds to low soil nitrogen(N)and identifying causative genes underlying this trait.To identify quantitative trait loci(QTL)or genes associated with low N response,we conducted a genome-wide association study(GWAS)using a diverse panel of 230 rice accessions and performed a transcriptomic investigation of rice accessions with differential responses to low N stress at two N levels.We detected 411 GWAS-associated genes in 5 QTL and 2722 differentially expressed genes in response to low N,of which 24 were identified by both methods and ranked according to gene annotations,literature queries,gene expression,and genetic diversity analysis.The large-scale datasets obtained from this study reveal low N-responsive characteristics and provide insights towards understanding the regulatory mechanisms of N-deficiency tolerance in rice,and the candidate genes or QTL would be valuable resources for increasing rice NUE via molecular biotechnology.Qing Li Xueli Lu Changjian Wang Lan Shen Liping Dai Jinli He Long Yang Peiyuan Li Yifeng Hong Qiang Zhang Guojun Dong Jiang Hu Guangheng Zhang Deyong Ren Zhenyu Gao Longbiao Guo Qian Qian Li Zhu Dali Zeng 2022The Crop Journal2022,10,4:3

11In situ synthesis and unidirectional insertion of membrane proteins in liposome-immobilized silica stationary phase for rapid preparation of microaffinity chromatography显示文摘Cell membrane affinity chromatography has been widely applied in membrane protein(MP)-targeted drug screening and interaction analysis.However,in current methods,the MP sources are derived from cell lines or recombinant protein expression,which are time-consuming for cell culture or purification,and also difficult to ensure the purity and consistent orientation of MPs in the chromatographic stationary phase.In this study,a novel in situ synthesis membrane protein affinity chromatography(iSMAC)method was developed utilizing cell-free protein expression(CFE)and covalent immobilized affinity chromatography,which achieved efficient in situ synthesis and unidirectional insertion of MPs into liposomes in the stationary phase.The advantages of iSMAC are:1)There is no need to culture cells or prepare recombinant proteins;2)Specific and purified MPs with stable and controllable content can be obtained within 2 h;3)MPs maintain the transmembrane structure and a consistent orientation in the chromatographic stationary phase;4)The flexible and personalized construction of cDNAs makes it possible to analyze drug binding sites.iSMAC was successfully applied to screen PDGFRβinhibitors from Salvia miltiorrhiza and Schisandra chinensis.Micro columns prepared by in-situ synthesis maintain satisfactory analysis activity within 72 h.Two new PDGFRβinhibitors,salvianolic acid B and gomisin D,were screened out with KD values of 13.44 and 7.39μmol/L,respectively.In vitro experiments confirmed that the two compounds decreasedα-SMA and collagen Ⅰ mRNA levels raised by TGF-βin HSC-T6 cells through regulating the phosphorylation of p38,AKT and ERK.In vivo,Sal B could also attenuate CCl_(4)-induced liver fibrosis by downregulating PDGFRβdownstream related protein levels.The iSMAC method can be applied to other general MPs,and provides a practical approach for the rapid preparation of MP-immobilized or other biological solid-phase materials.Yanqiu Gu Rong Wang Panpan Chen Shengnan Li Xinyi Chai Chun Chen Yue Liu Yan Cao Diya Lv Zhanying Hong Zhenyu Zhu Yifeng Chai Yongfang Yuan Xiaofei Chen 2022Acta Pharmaceutica Sinica B2022,12,9:1
12Finite element analysis for general elastic multi-structures显示文摘A finite element method is introduced to solve the general elastic multi-structure problem, in which the displacements on bodies, the longitudinal displacements on plates and the longitudinal displacements on beams are discretized using conforming linear elements, the rotational angles on beams are discretized using conforming elements of second order, the transverse displacements on plates and beams are discretized by the Morley elements and the Hermite elements of third order, respectively. The generalized Korn's inequality is established on related nonconforming element spaces, which implies the unique solvability of the finite element method. 14GelNB molecular coating as a biophysical barrier to isolate intestinal irritating metabolites and regulate intestinal microbial homeostasis in the treatment of inflammatory bowel disease显示文摘Inflammatory bowel disease(IBD)is a chronic,immune-mediated inflammatory disease characterized by the destruction of the structure and function of the intestinal epithelial barrier.Due to the poor remission effect and severe adverse events associated with current clinical medications,IBD remains an incurable disease.Here,we demonstrated a novel treatment strategy with high safety and effective inflammation remission via tissue-adhesive molecular coating.The molecular coating is composed of o-nitrobenzaldehyde(NB)-modified Gelatin(GelNB),which can strongly bond with-NH_(2)on the intestinal surface of tissue to form a thin biophysical barrier.We found that this molecular coating was able to stay on the surface of the intestine for long periods of time,effectively protecting the damaged intestinal epithelium from irritations of external intestinal metabolites and harmful flora.In addition,our results showed that this coating not only provided a beneficial environment for cell migration and proliferation to promote intestinal repair and regeneration,but also achieved a better outcome of IBD by reducing intestinal inflammation.Moreover,the in vivo experiments showed that the GelNB was better than the classic clinical medication-mesalazine.Therefore,our molecular coating showed potential as a promising strategy for the prevention and treatment of IBD.Qijiang Mao Haoqi Pan Yiyin Zhang Yi Zhang Qiuwen Zhu Yi Hong Zhengze Huang Yang Li Xu Feng Yifeng Fang WenChao Chen Pengfei Chen Bo Shen Hongwei Ouyang Yuelong Liang 2023Bioactive Materials2023,,1:1

16梯度多孔材料部分填充圆管强化换热的研究显示文摘基于GPM(梯度多孔材料)的优良特性,本工作设计了GPM部分填充圆管强化换热的结构。当Rp(多孔材料填充率)为0.8时,结合管内充分发展段的速度分布图、管内流动阻力系数和平均Nu数,分别研究了dp(填充梯度孔径)多孔材料、ε(梯度孔隙率)多孔材料对于管内流体流动以及传热特性的影响规律,并将其与HPM(均匀多孔材料)填充圆管时的结果做了对比分析。在此基础上,比较了各种多孔材料填充结构的换热与流动综合评价性能(简称PEC)。结果表明,通过合理设计GPM的结构,能够在强化换热的基础上,有效地降低圆管流动阻力,从而得到较好的综合评价性能(PEC)。王柏村 王鹏飞 Yifeng Hong 许忠斌 2016热能动力工程2016,31,8:1
