1Programmed death-1 expression is associated with the disease status in hepatitis B virus infection显示文摘AIM: To define the potential role of programmed death-1/programmed death-ligand (PD-1/PD-L) pathway in different hepatitis B virus (HBV) infection disease status; we examined the expression of PD-1 on antigen specific CD8+ T cells in peripheral blood of patients with chronic hepatitis B (CHB) and acute exacerbation of hepatitis B (AEHB) infection. METHODS: The PD-1 level on CD8+ T lymphocytes and the number of HBV specifi c CD8+ T lymphocytes in patients and healthy controls (HCs) were analyzed by staining with pentameric peptide-human leukocyte antigen2 (HLA2) complexes combined with flow cytometry. Real-time quantitative polymerase chain reaction (PCR) was used to measure the serum HBV-DNA levels. RESULTS: The level of PD-1 expression on total CD8+ T cells in CHB patients (13.86% ± 3.38%) was significantly higher than that in AEHB patients (6.80%± 2.19%, P < 0.01) and healthy individuals (4.63% ± 1.23%, P < 0.01). Compared to AEHB patients (0.81% ± 0.73%), lower frequency of HBV-specific CD8+ T cells was detected in chronic hepatitis B patients (0.37% ± 0.43%, P < 0.05). There was an inverse correlation between the strength of HBV-specific CD8+ T-cell response and the level of PD-1 expression. Besides, there was a significant positive correlation between HBV viral load and the percentage of PD-1 expression on CD8+ T cells in CHB and AEHB subjects (R = 0.541, P < 0.01). However, PD-1 expression was not associated with disease flare-ups as indicated by alanine aminotransferase (ALT) levels (R = 0.066, P > 0.05). CONCLUSION: Our results confirm previous reports that HBV specific CD8+ T-cell response in the peripheral blood is more intense in patients with AEHB than in chronic hepatitis B with persistent viral infection. 3Brain-derived neurotrophic factor mediates macrophage migration inhibitory factor to protect neurons against oxygen-glucose deprivation显示文摘Macrophage migration inhibitory factor(MIF)is a chemokine that plays an essential role in immune system function.Previous studies suggested that MIF protects neurons in ischemic conditions.However,few studies are reported on the role of MIF in neurological recovery after ischemic stroke.The purpose of this study is to identify the molecular mechanism of neuroprotection mediated by MIF.Human neuroblastoma cells were incubated in Dulbecco’s modified Eagle’s medium under oxygen-glucose deprivation(OGD)for 4 hours and then returned to normal aerobic environment for reperfusion(OGD/R).30 ng/mL MIF recombinant(30 ng/mL)or ISO-1(MIF antagonist;50μM)was administered to human neuroblastoma cells.Then cell cultures were assigned to one of four groups:control,OGD/R,OGD/R with MIF,OGD/R with ISO-1.Cell viability was analyzed using WST-1 assay.Expression levels of brain-derived neurotrophic factor(BDNF),microtubule-associated protein 2(MAP2),Caspase-3,Bcl2,and Bax were detected by western blot assay and immunocytochemistry in each group to measure apoptotic activity.WST-1 assay results revealed that compared to the OGD/R group,cell survival rate was significantly higher in the OGD/R with MIF group and lower in the OGD/R with ISO-1 group.Western blot assay and immunocytochemistry results revealed that expression levels of BDNF,Bcl2,and MAP2 were significantly higher,and expression levels of Caspase-3 and Bax were significantly lower in the MIF group than in the OGD/R group.Expression levels of BDNF,Bcl2,and MAP2 were significantly lower,and expression levels of Caspase-3 and Bax were significantly higher in the ISO-1 group than in the OGD/R group.MIF administration promoted neuronal cell survival and induced high expression levels of BDNF,MAP2,and Bcl2(anti-apoptosis)and low expression levels of Caspase-3 and Bax(pro-apoptosis)in an OGD/R model.These results suggest that MIF administration is effective for inducing expression of BDNF and leads to neuroprotection of neuronal cells against hypoxic injury.Su Hwan Bae Mi Ran Yoo Ye Yeong Kim In Kyung Hong Mi Hee Kim Seung Hak Lee Dae Yul Kim 2020Neural Regeneration Research2020,15,8:11

6Reservoir Characteristics and Preservation Conditions of Longmaxi Shale in the Upper Yangtze Block, South China显示文摘The Upper Ordovician-Lower Silurian Longmaxi Shale in the Upper Yangtze block represents one of the most important shale gas plays in China. The shale composition,porosity,organic thermal maturity,and methane sorption were investigated at the Qilongcun section in the Dingshan area,southeastern Sichuan Basin. The results show that the Upper Ordovician-Lower Silurian Longmaxi Shale contains:(1) sapropelic I organic matter;(2) a 40-m thick bedded sequence where total organic carbon(TOC) content is > 2%;(3) a 30-m thick layer at the base of the Longmaxi Shale with a brittle mineral content higher than 50%; and(4) a mean methane adsorption capacity of 1.80 cm^3/g(7 MPa pressure). A positive correlation between TOC and sorbed gas indicates that organic matter content exerts an important control on methane storage capacity. Based on the analysis of the shale reservoir characteristics,the lower member of the Longmaxi Shale can thus be considered a favorable stratum for shale gas exploration and exploitation. It has similar reservoir characteristics with the Longmaxi Shale in the Jiaoshiba area tested with a high-yield industrial gas flow. However,based on tectonic analysis,differences in the level of industrial gas flow between the low-yield study area and the high-yield Jiaoshiba area may result from different tectonic preservation conditions. 7Association between socioeconomic status and chronic obstructive pulmonary disease in Jiangsu province, China: a population-based study显示文摘Background:Chronic obstructive pulmonary disease(COPD)is a common public health problem worldwide.Recent studies have reported that socioeconomic status(SES)is related to the incidence of COPD.This study aimed to investigate the association between SES and COPD among adults in Jiangsu province,China,and to determine the possible direct and indirect effects of SES on the morbidity of COPD.Methods:A cross-sectional study was conducted among adults aged 40 years and above between May and December of 2015 in Jiangsu province,China.Participants were selected using a multistage sampling approach.COPD,the outcome variable,was diagnosed by physicians based on spirometry,respiratory symptoms,and risk factors.Education,occupation,and monthly family average income(FAI)were used to separately indicate SES as the explanatory variable.Mixed-effects logistic regression models were introduced to calculate odds ratios(ORs)and 95%confidence intervals(CIs)for examining the SES-COPD relationship.A pathway analysis was conducted to further explore the pulmonary function impairment of patients with different SES.Results:The mean age of the 2421 participants was 56.63±9.62 years.The prevalence of COPD was 11.8%(95%CI:10.5%–13.1%)among the overall sample population.After adjustment for age,gender,residence,outdoor and indoor air pollution,body weight status,cigarette smoking,and potential study area-level clustering effects,educational attainment was negatively associated with COPD prevalence in men;white collars were at lower risk(OR:0.60,95%CI:0.43–0.83)of experiencing COPD than blue collars;compared with those within the lower FAI subgroup,participants in the upper(OR:0.68,95%CI:0.49–0.97)tertiles were less likely to experience COPD.Such negative associations between all these three SES indicators and COPD were significant among men only.Education,FAI,and occupation had direct or indirect effects on pulmonary function including post-bronchodilator forced expiratory volume in 1 s/forced vital capacity(FEV1/FVC),FEV1,FVC,and FEV1 percentage of predicted.Education,FAI,and occupation had indirect effects on pulmonary function indices of all participants mainly through smoking status,indoor air pollution,and outdoor air pollution.We also found that occupation could affect post-bronchodilator FEV1/FVC through body mass index.Conclusions:Education,occupation,and FAI had an adverse relationship with COPD prevalence in Jiangsu province,China.SES has both direct and indirect associations with pulmonary function impairment.SES is of great significance for COPD morbidity.It is important that population-based COPD prevention strategies should be tailored for people with different SES.Dan-Dan Zhang Jian-Nan Liu Qing Ye Zi Chen Ling Wu Xue-Qing Peng Gan Lu Jin-Yi Zhou Ran Tao Zhen Ding Fei Xu Linfu Zhou 2021Chinese Medical Journal2021,,13:5
8USP29 maintains the stability of cGAS and promotes cellular antiviral responses and autoimmunity显示文摘Cyclic GMP-AMP synthase(cGAS)is an essential sensor of cytosolic DNA and critically mediates innate immune responses and autoimmunity.Modulating the activity and stability of cGAS provides potential strategies for treating viral or autoimmune diseases.Here,we report that ubiquitin-specific protease 29(USP29)deubiquitinates and stabilizes cGAS and promotes cellular antiviral responses and autoimmunity.Knockdown or knockout of USP29 severely impairs Herpes simplex virus 1(HSV-1)-or cytosolic DNA-induced expression of type I interferons(IFNs)and proinflammatory cytokines.Consistently,Usp29m/m mice produce decreased type I IFNs and proinflammatory cytokines after HSV-1 infection and are hypersensitive to HSV-1 infection compared to the wild-type littermates.In addition,genetic ablation of USP29 in Trex1−/−mice eliminated the detectable pathological and molecular autoimmune phenotypes.Mechanistically,USP29 constitutively interacts with cGAS,deconjugates K48-linked polyubiquitin chains from cGAS and stabilizes cGAS in uninfected cells or after HSV-1 infection.Reconstitution of cGAS into Usp29−/−cells fully rescues type I IFN induction and cellular antiviral responses after HSV-1 infection.Our findings thus reveal a critical role of USP29 in the innate antiviral responses against DNA viruses and autoimmune diseases and provide insight into the regulation of cGAS.Qiang Zhang Zhen Tang Ran An Liya Ye Bo Zhong 2020Cell Research2020,30,10:5

12Identification of coastal water quality by multivariate statistical techniques in two typical bays of northern Zhejiang Province,East China Sea显示文摘The Hangzhou Bay(HZB) and Xiangshan Bay(XSB), in northern Zhejiang Province and connect to the East China Sea(ECS) were considerably affected by the consequence of water quality degradation. In this study, we analyzed physical and biogeochemical properties of water quality via multivariate statistical techniques. Hierarchical cluster analysis(HCA) grouped HZB and XSB into two subareas of different pollution sources based on similar physical and biogeochemical properties. Principal component analysis(PCA) identified three latent pollution sources in HZB and XSB respectively and emphasized the importance of terrestrial inputs, coastal industries as well as natural processes in determining the water quality of the two bays. 13Modeling unsaturated flow in fractured rocks with scaling relationships between hydraulic parameters显示文摘Modeling unsaturated flow in fractured rocks is essential in various subsurface engineering applications,but it remains a great challenge due to the difficulties in determining the unsaturated hydraulic properties of rocks that contain various scales of fractures.It is generally accepted that the van Genuchten(VG)model can be applied to fractured rocks,provided that the hydraulic parameters could be representatively determined.In this study,scaling relationships between the VG parameters(a and n)and hydraulic conductivity(K)across 8 orders of magnitude,from 10^(-10)m/s to 10^(-2)m/s,were proposed by statistical analysis of data obtained from 1416 soil samples.The correlations were then generalized to predict the upper bounds of VG parameters for fractured rocks from the K data that could be obtained more easily under field conditions,and were validated against a limited set of data from cores,fractures and fractured rocks available in the literature.The upper bound estimates significantly narrow the ranges of VG parameters,and the representative values of a and n for fractured rocks at the field scale can then be determined with confidence by inverse modeling using groundwater observations in saturated zones.The proposed methodology was applied to saturated-unsaturated flow modeling in the right-bank slope at the Baihetan dam site with a continuum approach,showing that most of the flow behaviors in fractured rocks in this complex hydrogeological condition could be properly reproduced.The proposed method overcomes difficulties in suction measurement in fractured rocks with strong heterogeneity,and provides a feasible way for modeling of saturated-unsaturated flow in fractured rocks with acceptable engineering accuracy.Yi-Feng Chen Yuke Ye Ran Hu Zhibing Yang Chuang-Bing Zhou 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,6:4
14Recovery from prolonged disorders of consciousness:A dual-center prospective cohort study in China显示文摘BACKGROUND Recent innovations in intensive care have improved the prognosis of patients with severe brain injuries and brought more patients with disorders of consciousness(DoC).Data are lacking regarding the long-term outcomes of those patients in China.It is necessary to study the long-term outcomes of patients with prolonged DoC in light of many factors likely to influence crucial decisions about their care and their life.AIM To present the preliminary results of a DoC cohort.METHODS This was a two-center prospective cohort study of inpatients with vegetative state(VS)/unresponsive wakefulness syndrome(UWS).The study outcomes were the recovery from VS/UWS to minimally conscious state(MCS)and the long-term status of patients with prolonged DoC considered in VS/UWS or MCS for up to 6 years.The patients were evaluated using the Glasgow coma scale,coma recovery scale-revised,and Glasgow outcome scale.The endpoint of follow-up was recovery of full consciousness or death.The changes in the primary clinical outcome improvement in clinical diagnosis were evaluated at 12 mo compared with baseline.RESULTS The study population included 93 patients(62 VS/UWS and 31 MCS).The postinjury interval range was 28-634 d.Median follow-up was 20 mo(interquartile range,12-37 mo).At the endpoint,33 transitioned to an emergence from MCS or full consciousness,eight had a locked-in syndrome,and there were 35 patients remaining in a VS/UWS and 11 in an MCS.Seven(including one locked-in syndrome)patients(7.5%)died within 12 mo of injury.Compared with the unresponsive group(n=52)at 12 mo,the responsive group(n=41)had a higher proportion of males(87.8%vs 63.5%,P=0.008),shorter time from injury(median,40.0 d vs 65.5 d,P=0.006),higher frequency of vascular etiology(68.3%vs 38.5%,P=0.007),higher Glasgow coma scale score at admission(median,9 vs 6,P<0.001),higher coma recovery scale-revised score at admission(median,9 vs 2.5,P<0.001),at 1 mo(median,14 vs 5,P<0.001),and at 3 mo(median,20 vs 6,P<0.001),lower frequency of VS/UWS(36.6%vs 90.0%,P<0.001),and more favorable Glasgow outcome scale outcome(P<0.001).CONCLUSION Patients with severe DoC,despite having strong predictors of poor prognosis,might recover consciousness after a prolonged time of rehabilitation.An accurate initial diagnosis of patients with DoC is critical for predicting outcome and a long-term regular follow-up is also important.Wei-Guan Chen Ran Li Ye Zhang Jian-Hui Hao Ju-Bao Du Ai-Song Guo Wei-Qun Song 2020World Journal of Clinical Cases2020,8,12:4
15Instability waves and low-frequency noise radiation in the subsonic chevron jet显示文摘Spatial instability waves associated with lowfrequency noise radiation at shallow polar angles in the chevron jet are investigated and are compared to the round counterpart. The Reynolds-averaged Navier–Stokes equations are solved to obtain the mean flow fields, which serve as the baseflow for linear stability analysis. The chevron jet has more complicated instability waves than the round jet, where three types of instability modes are identified in the vicinity of the nozzle, corresponding to radial shear, azimuthal shear,and their integrated effect of the baseflow, respectively. The most unstable frequency of all chevron modes and round modes in both jets decrease as the axial location moves downstream. Besides, the azimuthal shear effect related modes are more unstable than radial shear effect related modes at low frequencies. Compared to a round jet, a chevron jet reduces the growth rate of the most unstable modes at downstream locations. Moreover, linearized Euler equations are employed to obtain the beam pattern of pressure generated by spatially evolving instability waves at a dominant low frequency St = 0.3, and the acoustic efficiencies of these linear wavepackets are evaluated for both jets. 16Genesis of the Changba–Lijiagou Giant Pb-Zn Deposit, West Qinling, Central China: Constraints from S-Pb-C-O isotopes显示文摘The extensive Changba-Lijiagou Pb-Zn deposit is located in the north of the Xihe–Chengxian ore cluster in West Qinling. The ore bodies are mainly hosted in the marble, dolomitic marble and biotite-calcite-quartz schist of the Middle Devonian Anjiacha Formation, and are structurally controlled by the fault and anticline. The ore-forming process can be divided into three main stages, based on field geological features and mineral assemblages. The mineral assemblages of hydrothermal stage I are pale-yellow coarse grain, low Fe sphalerite, pyrite with pits, barite and biotite. The mineral assemblages of hydrothermal stage II are black-brown cryptocrystalline, high Fe shalerite, pyrite without pits, marcasite or arsenopyrite replace the pyrite with pits, K-feldspar. The features of hydrothermal stage III are calcite-quartz-sulfide vein cutting the laminated, banded ore body. Forty-two sulfur isotope analyses, twenty-five lead isotope analyses and nineteen carbon and oxygen isotope analyses were determined on sphalerite, pyrite, galena and calcite. The δ34 S values of stage I(20.3 to 29.0‰) are consistent with the δ34 S of sulfate(barite) in the stratum. Combined with geological feature, inclusion characteristics and EPMA data, we propose that TSR has played a key role in the formation of the sulfides in stage I. The δ34 S values of stage II sphalerite and pyrite(15.1 to 23.0‰) are between sulfides in the host rock, magmatic sulfur and the sulfate(barite) in the stratum. This result suggests that multiple S reservoirs were the sources for S2-in stage II. The δ34 S values of stage III(13.1 to 22‰) combined with the structure of the geological and mineral features suggest a magmatic hydrothermal origin of the mineralization. The lead isotope compositions of the sulfides have 206 Pb/204 Pb ranging from 17.9480 to 17.9782, 207 Pb/204 Pb ranging from 15.611 to 15.622, and 208 Pb/204 Pb ranging from 38.1368 to 38.1691 in the three ore-forming stages. The narrow and symmetric distributions of the lead isotope values reflect homogenization of granite and mantle sources before the Pb-Zn mineralization. The δ13 CPDB and δ18 OSMOW values of stage I range from-0.1 to 2.4‰ and from 18.8 to 21.7‰. The values and inclusion data indicate that the source of fluids in stage I was the dissolution of marine carbonate. The δ13 CPDB and δ18 OSMOW values of stage II range from-4 to 1‰ and from 12.3 to 20.3‰, suggesting multiple C-O reservoirs in the Changba deposit and the addition of mantle-source fluid to the system. The values in stage III are-3.1‰ and 19.7‰, respectively. We infer that the process of mineralization involved evaporitic salt and sedimentary organic-bearing units interacting through thermochemical sulfate reduction through the isotopic, mineralogy and inclusion evidences. Subsequently, the geology feature, mineral assemblages, EPMA data and isotopic values support the conclusion that the ore-forming hydrothermal fluids were mixed with magmatic hydrothermal fluids and forming the massive dark sphalerite, then yielding the calcite-quartz-sulfide vein ore type at the last stage. 19Pulmonary toxicity induced by three forms of titanium dioxide nanoparticles via intra-tracheal instillation in rats显示文摘Titanium dioxide (TiO2) nanoparticles are in wide commercial use worldwide. To evaluate if acute pulmonary toxicity can be induced by nano-TiO2 particles, rats were intra-tracheally instilled with 0.5, 5, or 50 mg/kg of 5, 21, and 50 nm TiO2 primary particles. Toxic effects were determined with the coefficients of lung tissues to body weight, histopathology, biochemical parameters of blood, activity of lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and acid phosphatase (ACP) in tissues, and the phagocytotic ability of alveolar macrophages (AMs). All the indicators were observed in sacrificed rats one week post-exposure. There was a significant difference of coefficients of pulmonary tissues between the high-dose group and the low-or moderate-dose groups with an exposure of 5 nm TiO2. At the same time, 5 nm TiO2 primary particles increased the activity of LDH and ALP when exposure dose was >5 mg/kg. A significant difference in LDH and ALP activity was observed between the 50 mg/kg group and 0.5 or 5 mg/kg group with exposure of 5 nm TiO2. Lung tissues showed increased ALP activity only if treated with 5 and 50 mg/kg of 21 nm TiO2 particles. There was no significant difference in LDH and ALP activity in the 50 nm TiO2 group and control group. Histo-pathologic examination of lung tissues indicated that the pulmonary response to exposure to TiO2 particles in rats manifested as dose-dependent inflammatory lesions, which mainly consisted of inflltration of inflammatory cells and interstitial thickening. Analysis of uptake of neutral red dye showed that 50 nm TiO2 particles significantly increased phagocytotic ability of AMs compared with controls (P<0.05), whereas exposure with 5 nm TiO2 reduced the phagocytotic ability of AMs when the exposure dose was 50 mg/kg. These results suggest that particle size and exposure dose may have important roles in pulmonary toxicity. The toxic effect of TiO2 nanopar-ticles in lung tissue exhibited a dose-response relationship. After exposure with TiO2 particles of >5.0 mg/kg, 5 and 21 nm TiO2 particles induced standing pulmonary lesions; and 5 nm TiO2 particles may suppress the phagocytotic ability of AMs if exposure dose was P50 mg/kg. 20Hybrid Precoder and Combiner Design with Finite Resolution PSs for mmWave MIMO Systems显示文摘Hybrid precoding and combining have been considered as a promising technology, which can provide a compromise between hardware complexity and system performance in millimeter wave multiple-input multiple-output systems. However, most existing hybrid precoder and combiner designs generally assume that infinite resolution phase shifters(PSs) are used to produce the analog beamformers. In a practical scene, the design with accurate PSs can lead to high hardware cost and power consumption. In this paper, we investigate the hybrid precoder and combiner design with finite resolution PSs in millimeter wave systems. We employ alternate optimization as the main strategy to jointly design analog precoder and combiner. In addition, we propose a low complexity algorithm, where the analog beamformers are implemented only by finite resolution PSs to maximize spectral efficiency. Then, the digital precoder and combiner are designed based on the obtained analog beamformers to improve the spectral efficiency.
