1Screening test for anti-Helicobacter pylori activity of traditional Chinese herbal medicines显示文摘AIM:To evaluate the anti-Helicobacter pylori (H.pylori) activity of 50 traditional Chinese herbal medicines in order to provide the primary evidence for their use in clinical practice.METHODS:A susceptibility test of water extract from 50 selected traditional Chinese herbal medicines for in vitro H.pylori Sydney strain 1 was performed with broth dilution method.Anti-H.pylori activity of the selected Chinese herbal medicines was evaluated according to their minimum inhibitory concentration (MIC).RESULTS:The water extract from Rhizoma Coptidis,Radix Scutellariae and Radix isatidis could significantly inhibit the H.pylori activity with their MIC less than 7.8 mg/mL,suggesting that traditional Chinese herbal medicines have anti-inflammatory and antibacterial effects and can thus be used in treatment of H.pylori infection.CONCLUSION:Rhizoma Coptidis,Radix Scutellariae and Radix isatidis are the potential sources for the synthesis of new drugs against H.pylori.Feng Ma Ye Chen Jing Li He-Ping Qing Ji-De Wang YaLi Zhang Yang Bai Bei-Guo Long 2010World Journal of Gastroenterology2010,16,44:34
2Ablation of gut microbiota alleviates obesity-induced hepatic steatosis and glucose intolerance by modulating bile acid metabolism in hamsters显示文摘Since metabolic process differs between humans and mice, studies were performed in hamsters, which are generally considered to be a more appropriate animal model for studies of obesityrelated metabolic disorders. The modulation of gut microbiota, bile acids and the farnesoid X receptor(FXR) axis is correlated with obesity-induced insulin resistance and hepatic steatosis in mice. However,the interactions among the gut microbiota, bile acids and FXR in metabolic disorders remained largely unexplored in hamsters. In the current study, hamsters fed a 60% high-fat diet(HFD) were administeredvehicle or an antibiotic cocktail by gavage twice a week for four weeks. Antibiotic treatment alleviated HFD-induced glucose intolerance, hepatic steatosis and inflammation accompanied with decreased hepatic lipogenesis and elevated thermogenesis in subcutaneous white adipose tissue(sWAT). In the livers of antibiotic-treated hamsters, cytochrome P450 family 7 subfamily B member 1(CYP7 B1) in the alternative bile acid synthesis pathway was upregulated, contributing to a more hydrophilic bile acid profile with increased tauro-β-muricholic acid(TβMCA). The intestinal FXR signaling was suppressed but remained unchanged in the liver. 3Epidemiology of Diabetes in Adults Aged 35 and Older from Shanghai,China显示文摘Objective This study aimed to investigate the epidemiology of diabetes in adults from Shanghai, aged 35 and older, in 2013. Methods We estimated the diabetes prevalence in a representative sample of 18,736 adults who were selected through a multistage stratified cluster sampling process. A standard questionnaire containing questions about demographic characteristics and lifestyle factors was distributed. After an overnight fast of at least 10 hours, a venous blood sample was collected from each participant. For each patient without a history of diabetes, another blood sample was drawn 120 min after an oral glucose tolerance test to identify undiagnosed diabetes and prediabetic condition. Results Among Shanghai residents aged 35 and above, the overall weighted prevalence of diabetes was 17.6% [95% confidence interval(CI): 16.4%-18.8%]. The prevalences were 19.3% in men and 15.8% in women as well as 19.1%, 15.4%, and 16.1% in urban, suburban, and rural residents, respectively. In addition, the weighted prevalence of prediabetes was 16.5%(95% CI: 15.3%-17.8%), with the prevalences of 16.5% in men, 16.6% in women, 15.2% in urban residents, 18.0% in suburban residents, and 18.5% in rural residents. Among all patients with diabetes, 68.1%(95% CI: 64.3%-71.6%) were aware of their status, 63.5%(95% CI: 60.0%-66.9%) received diabetes treatment, but only 35.1%(95% CI: 32.4%-37.8%) had adequate glycemic control. Conclusion In Shanghai, diabetes and prediabetes are highly prevalent. 5Isolation and characterization of dengue virus serotype 2 from the large dengue outbreak in Guangdong, China in 2014显示文摘Dengue has been well recognized as a global public health threat,but only sporadic epidemics and imported cases were reported in recent decades in China.Since July 2014,an unexpected large dengue outbreak has occurred in Guangdong province,China,resulting in more than 40000 patients including six deaths.To clarify and characterize the causative agent of this outbreak,the acute phase serum from a patient diagnosed with severe dengue was subjected to virus isolation and high-throughput sequencing(HTS).Traditional real-time RT-PCR and HTS with Ion Torrent PGM detected the presence of dengue virus serotype 2(DENV-2).A clinical DENV-2 isolate GZ05/2014 was obtained by culturing the patient serum in mosquito C6/36 cells.The complete genome of GZ05/2014 was determined and deposited in Gen Bank under the access number KP012546.Phylogenetic analysis based on the complete envelope gene showed that the newly DENV-2 isolate belonged to Cosmopolitan genotype and clustered closely with other Guangdong strains isolated in the past decade.No amino acid mutations that are obviously known to increase virulence or replication were identified throughout the genome of GZ05/2014.The high homology of Guangdong DENV-2 strains indicated the possibility of establishment of local DENV-2 circulation in Guangdong,China.These results help clarify the origin of this epidemic and predict the future status of dengue in China.ZHAO Hui ZHAO Ling Zhai JIANG Tao LI Xiao Feng FAN Hang HONG Wen Xin ZHANG Yu ZHU Qin YE Qing TONG Yi Gang CAO Wu Chun ZHANG Fu Chun QIN Cheng Feng 2014Science China(Life Sciences)2014,57,12:21
6Contribution of diffusion,perfusion and functional MRI to the disconnection hypothesis in subcortical vascular cognitive impairment显示文摘Vascular cognitive impairment(VCI)describes all forms of cognitive impairment caused by any type of cerebrovascular disease.Early identification of VCI is quite difficult due to the lack of both sensitive and specific biomarkers.Extensive damage to the white matter tracts,which connect the cortical and subcortical regions,has been shown in subcortical VCI(SVCI),the most common subtype of VCI that is caused by small vessel disease.Two specific MRI sequences,including diffusion tensor imaging(DTI)and functional MRI(fMRI),have emerged as useful tools for identifying subtle white matter changes and the intrinsic connectivity between distinct cortical regions.This review describes the advantages of these two modalities in SVCI research and the current DTI and fMRI findings on SVCI.Using DTI technique,a variety of studies found that white matter microstructural damages in the anterior and superior areas are more specific to SVCI.Similarly,functional brain abnormalities detected by fMRI have also been mainly shown in anterior brain areas in SVCI.The characteristic distribution of brain abnormalities in SVCI interrupts the prefrontal-subcortical loop that results in cognitive impairments in particular domains,which further confirms the‘disconnection syndrome’hypothesis.In addition,another MRI technique,arterial spin labelling(ASL),has been used to describe the disconnection patterns in a variety of conditions by measuring cerebral blood flow.The role of the ASL technique in SVCI research is also assessed.Finally,the review proposes the application of multimodality fusion in the investigation of SVCI pathogenesis.Qing Ye Feng Bai 2018Stroke & Vascular Neurology2018,3,3:20
7Echinacoside Protects Against MPP+-Induced Neuronal Apoptosis via ROS/ATF3/CHOP Pathway Regulation显示文摘Echinacoside(ECH) is protective in a mouse model of Parkinson's disease(PD) induced by 1-methyl-4-phenylpyridinium ion(MPP^+). To investigate the mechanisms involved, SH-SY5Y neuroblastoma cells were treated with MPP^+or a combination of MPP^+and ECH,and the expression of ATF3(activating transcription factor3), CHOP(C/EBP-homologous protein), SCNA(synuclein alpha), and GDNF(glial cell line-derived neurotrophic factor) was assessed. The results showed that ECH significantly improved cell survival by inhibiting the generation of MPP^+-induced reactive oxygen species(ROS). In addition, ECH suppressed the ROS and MPP^+-induced expression of apoptotic genes(ATF3, CHOP, and SCNA). ECH markedly decreased the MPP^+-induced caspase-3 activity in a dose-dependent manner. ATF3-knockdown also decreased the CHOP and cleaved caspase-3 levels and inhibited the apoptosis induced by MPP^+.Interestingly, ECH partially restored the GDNF expression that was down-regulated by MPP^+. ECH also improved dopaminergic neuron survival during MPP^+treatment and protected these neurons against the apoptosis induced by MPTP. 9Quantification and timing of porosity evolution in tight sand gas reservoirs:an example from the Middle Jurassic Shaximiao Formation,western Sichuan,China显示文摘The diagenesis and porosity evolution of the Middle Jurassic Shaximiao sandstones were analyzed based on petrographic observations.X-ray diffractometry.scanning electron microscopy observations,carbon and oxygen stable isotope geochemistry,fluid inclusion microthermometry,and thermal and burial history modeling results.The point count data show that secondary pores(av.5.5%) are more abundant than primary pores(av.3.7%) and are thus the dominant pore type in the Shaximiao sandstones.Analysis of porosity evolution indicates that alteration of sandstones mainly occurred during two paragenetic stages.Mechanical compaction and cementation by early chlorite,calcite,and quartz typically decrease the depositional porosity(40.9%) by an average of37.2%,leaving porosity of 3.7%after stage Ⅰ(<85 ℃,175-145 Ma).The original intergranular porosity loss due to compaction is calculated to be 29.3%,suggesting that mechanical compaction is the most significant diagenetic process in primary porosity destruction.Stage Ⅱ can be further divided into two sub-stages(Stage Ⅱ_a and StageⅡ_b).Stage Ⅱ_a(85-120 ℃,145-125 Ma) is characterized by late dissolution,which enhanced porosity by 8.8%,and the porosity increased from 3.7%to 12.5%.During stageⅡ_b(>120 ℃,125-0 Ma),the precipitation of late chlorite,calcite,quartz,and kaolinite destroyed 3.3%porosity,leaving porosity of 9.2%in the rock today.Zheng-Xiang Lü Su-Juan Ye Xiang Yang Rong Li Yuan-Hua Qing 2015Petroleum Science2015,12,2:13
10Dynamic alterations in the gut microbiota and metabolome during the development of methionine-choline-deficient diet-induced nonalcoholic steatohepatitis显示文摘AIM To investigate changes in gut microbiota and metabolism during nonalcoholic steatohepatitis(NASH) development in mice fed a methionine-choline-deficient(MCD) diet. METHODS Twenty-four male C57 BL/6 J mice were equally divided into four groups and fed a methionine-choline-sufficient diet for 2 wk(Control 2 w group,n = 6) or 4 wk(Control 4 w group,n = 6) or the MCD diet for 2 wk(MCD 2 w group,n = 6) or 4 wk(MCD 4 w group,n = 6). Liver injury,fibrosis,and intestinal barrier function were evaluated after 2 and 4 wk of feeding. The fecal microbiome and metabolome were studied using 16 s r RNA deep sequencing and gas chromatography-mass spectrometry. RESULTS The mice fed the MCD diet presented with simple hepatic steatosis and slight intestinal barrier deterioration after 2 wk. After 4 wk of feeding with the MCD diet,however,the mice developed prominent NASH with liver fibrosis,and the intestinal barrier was more impaired. Compared with the control diet,the MCD diet induced gradual gut microbiota dysbiosis,as evidenced by a marked decrease in the abundance of Alistipes and the(Eubacterium) coprostanoligenes group(P < 0.001 and P < 0.05,respectively) and a significant increase in Ruminococcaceae UCG 014 abundance(P < 0.05) after 2 wk. At 4 wk,the MCD diet significantly reduced the promising probiotic Bifidobacterium levels and markedly promoted Bacteroides abundance(P < 0.05,and P < 0.01,respectively). The fecal metabolomic profile was also substantially altered by the MCD diet: At 2 wk,arachidic acid,hexadecane,palmitic acid,and tetracosane were selected as potential biomarkers that were significantly different in the corresponding control group,and at 4 wk,cholic acid,cholesterol,arachidic acid,tetracosane,and stearic acid were selected. 12Effect of roadway turnings on gas explosion propagation characteristics in coal mines显示文摘In order to reveal the effect of turnings on explosion propagation, experiments were performed in three different pipes (single bend, U-shaped pipe and Z-shaped pipe). Flame and pressure transducers were used to track the velocity at the explosion front. When the pipes were filled with methane, the explosion strength was significantly enhanced due to the turbulence induced by increasing the number of turnings, while the flame speed (S f) and peak overpressure (DP max) increased dramatically. In addition, the strength of the explosion increased in violence as a function of the number of turnings. However, when the bend was without methane, the turnings weakened the strength of the explosion compared with the ordinary pipe, shown by the decrease in the values of DP max and S f . In addition, the propagation characteristics in a U-shaped pipe were similar to those in a Z-shaped pipe and the values of DP max and S f were also close. The results show that the explosion propagation characteristics largely depend on gas distribution in the pipes and the number of turnings. 13Diurnal characteristics of geoelectric fields and their changes associated with the Alxa Zuoqi M_S5.8 earthquake on 15 April 2015(Inner Mongolia)显示文摘In China, efforts are being made to monitor geoelectric fields through a large network of stations deployed and managed by the China Earthquake Administration. The diurnal variations in the geoelectric field waveforms were similar in the quiet magnetic periods when K<5(generally, K<3 indicates a quiet time). The arrival time points of the maxima in the geoelectric field waveforms exhibited differences in local time related to geographic longitude. The amplitude of diurnal variation was several to 16.6 mV/km and decreased with increasing latitude. Further,the amplitude of diurnal variation, which was related to seasonal changes, was larger in summer and autumn than in spring and winter. The periods of diurnal changes during quiet days were 24, 12, 8, 6, 4 hours and several minutes over large areas. 14Altered intrinsic functional connectivity of the primary visual cortex in youth patients with comitant exotropia: a resting state fMRI study显示文摘AIM: To evaluate the differences in the functional connectivity(FC) of the primary visual cortex(V1) between the youth comitant exotropia(CE) patients and health subjects using resting functional magnetic resonance imaging(f MRI) data.METHODS: Totally, 32 CEs(25 males and 7 females) and 32 healthy control subjects(HCs)(25 males and 7 females) were enrolled in the study and underwent the MRI scanning. Two-sample t-test was used to examine differences in FC maps between the CE patients and HCs. RESULTS: The CE patients showed significantly less FC between the left brodmann area(BA17) and left lingual gyrus/cerebellum posterior lobe, right middle occipital gyrus, left precentral gyrus/postcentral gyrus and right inferior parietal lobule/postcentral gyrus. 16Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11

18Effect of Morinda officinalis capsule on osteoporosis in ovariectomized rats显示文摘AIM:To explore the therapeutic effects of Morinda officinalis capsules(MOP) on osteoporosis in ovariectomized rats.METHOD:Six-month-old female Sprague–Dawley rats were induced for postmenopausal osteoporosis(PMOP) by bilateral ovariectomy and divided into seven groups as follows:sham-operated group,ovariectomized(OVX) control group,OVX treated with xianlinggubao(XLGB)(270 mg·kg-1·d-1),OVX treated with alendronate sodium(ALN)(3 mg·kg-1·d-1),and OVX treated with Morinda officinalis capsule(MOP) of graded doses(90,270 and 810 mg·kg-1·d-1) groups.Oral treatments were administered daily on the 4th week after ovariectomy and lasted for 12 weeks.The bone mineral density was evaluated by dual-energy X-ray absorptiometry.The tartrate-resistant acid phosphatase(TRAP),alkaline phosphatase(AKP),and osteocalcin(OC) levels in the serum and plasma were determined by standard colorimetric and enzyme immunoassays methods.Bone biomechanical properties and morphological parameters were analyzed by three-point bending test and histomorphometry respectively.RESULTS:Morinda officinalis capsules at all doses were able to significantly prevent the OVX-induced loss of bone mass due to diminishing serum AKP and TRAP levels while elevating OC level in the plasma.Morinda officinalis capsules also enhanced the bone strength and prevented the deterioration of trabecular microarchitecture.CONCLUSION:Morinda officinalis capsules possess potent anti-osteoporotic activity in OVX rats which could be an effective treatment for postmenopausal osteoporosis.LI Ye Lü Shan-Shan TANG Gui-Ying HOU Min TANG Qing ZHANG Xiao-Na CHEN Wei-Hai CHEN Gang XUE Qiang ZHANG Cong-Cong ZHANG Ji-Fen CHEN Yi XU Xiao-Yu 2014Chinese Journal of Natural Medicines2014,12,3:10
