| 2 | RNA interference-mediated silencing of vATPase subunits A and E affect survival and development of the 28-spotted ladybeetle,Henosepilachna vigintioctopunctata显示文摘RNA interference(RNAi)has emerged as a powerful tool for developing novel management strategies for controlling insect pests.The 28-spotted ladybeetle,Henosepilachna vigintioctopunctata is one of the most important pests attacking solanaceous plants in Asia.In this study,the potential of dietary RNAi to manage H.vigintioctopunctata was investigated using both in vitro synthesized and bacterially expressed double-stranded RNAs(dsRNAs)of HvvATPase A and HvvATPase E.The expression levels of HvvATPase A and HvvATPase E were higher in Malpighian tubules than in other tissue types.The silencing of HvvATPase A and HvvATPase E led to significant mortality in H.vigintioctopunctata larvae.In addition,the ingestion of HvvATPase A and HvvATPase E significantly deterred feeding behavior and subsequently arrested the development of H.vigintioctopunctata.Notably,the bacterially expressed dsRNAs consistently caused higher mortality in larvae and adults.Finally,the nontarget effects of the dsRNAs of H.vigintioctopunctata on the predatory ladybeetle Propylaea japonica were evaluated.P.japonica 1st instar larvae were administered vATPase A and vATPase E dsRNAs from H.vigintioctopunctata and P.japonica under the worst-case scenario,in which dsGFP served as negative control.There were significant effects of dsHvvATPase A on P.japonica at the transcriptional level but not at the organismal level,whereas dsHvvATPase E did not effect P.japonica at either the transcriptional or the organismal level.Collectively,the results of the study suggest that HvvATPase A and HvvATPase E can act as novel molecular targets for the control of H.vigintioctopunctata. | Wei Guo Mujuan Guo Chunxiao Yang Zhuoqi Liu Shimin Chen Jing Lu Baoli Qiu Youjun Zhang Xuguo Zhou Huipeng Pan | 2021 | Insect Science2021,28,6: | 3 |
| 3 | The molecular mechanism of efficient transmission of plant viruses in variable virus-vector-plant interactions显示文摘Plant viruses are mainly transmitted by insect vectors in the non-persistent,semi-persistent,or persistent modes.In the non-persistent mode,plant viruses are retained in the stylets of their insect vectors.In the semi-persistent mode,plant viruses are carried to vector foreguts or salivary glands,but they cannot spread to salivary glands.In the persistent mode,plant viruses are retained in vector guts and can spread to salivary glands.In the non-persistent and semi-persistent modes,plant viruses are retained for a short time and cannot enter the hemolymph of insect vectors,whereas in the persistent mode,plant viruses are retained for a relatively long time and can be found in the hemolymph.Here,we reviewed recent studies that uncovered molecular mechanisms of how plant viruses manipulate host traits for efficient transmission by insect vectors.Normally,plants that are infected with viruses,regardless of the transmission mode,tend to release more attractive volatiles to vectors.However,plant defensive systems are regulated differently by viruses in these three modes.In the non-persistent mode,virus infections significantly induce plant defense responses,which probably trigger vectors(e.g.,winged aphids)to disperse and transmit viruses in a short time.In the semi-persistent mode,virus infections frequently suppress plant defense responses,resulting in an increase of vector population and facilitating viral transmissions during vector outbreaks.In the persistent mode,virus infections reduce plant defense responses and manipulate plant traits to become suitable feeding sites in a relatively long period of time.Understanding the underlying mechanisms of virus–vector–plant interactions will lay a foundation for preventing virus transmission. | Xiaobin Shi Zhuo Zhang Chi Zhang Xuguo Zhou Deyong Zhang Yong Liu | 2021 | Horticultural Plant Journal2021,7,6: | 3 |

| 7 | Defining honeybee subspecies in an evolutionary context warrants strategized conservation显示文摘Despite the urgent need for conservation consideration,strategic action plans for the preservation of the Asian honeybee,Apis cerana Fabricius,1793,remain lacking.Both the convergent and divergent adaptations of this widespread insect have led to confusing phenotypical traits and inconsistent infraspecific taxonomy.Unclear subspecies boundaries pose a significant challenge to honeybee conservation efforts,as it is difficult to effectively prioritize conservation targets without a clear understanding of subspecies identities.Here,we investigated genome variations in 362 worker bees representing almost all populations of mainland A.cerana to understand how evolution has shaped its population structure.Whole-genome single nucleotide polymorphisms(SNPs)based on nuclear sequences revealed eight putative subspecies,with all seven peripheral subspecies exhibiting mutually exclusive monophyly and distinct genetic divergence from the widespread central subspecies.Our results demonstrated that most classic morphological traits,including body size,were related to the climatic variables of the local habitats and did not reflect the true evolutionary history of the organism.Thus,such morphological traits were not suitable for subspecific delineation.Conversely,wing vein characters showed relative independence to the environment and supported the subspecies boundaries inferred from nuclear genomes.Mitochondrial phylogeny further indicated that the present subspecies structure was a result of multiple waves of population divergence from a common ancestor.Based on our findings,we propose that criteria for subspecies delineation should be based on evolutionary independence,trait distinction,and geographic isolation.We formally defined and described eight subspecies of mainland A.cerana.Elucidation of the evolutionary history and subspecies boundaries enables a customized conservation strategy for both widespread and endemic honeybee conservation units,guiding colony introduction and breeding. | Lifei Qiu Jiangxing Dong Xingan Li Sajad HParey Ken Tan Michael Orr Aquib Majeed Xue Zhang Shiqi Luo Xuguo Zhou Chaodong Zhu Ting Ji Qingsheng Niu Shanlin Liu Xin Zhou | 2023 | Zoological Research2023,44,3: | 1 |

| 20 | Inferring decelerated land subsidence and groundwater storage dynamics in Tianjin–Langfang using Sentinel-1 InSAR显示文摘To meet the growing demand for socioeconomic development,a large amount of groundwater is extracted from confined aquifers worldwide.The North China Plain has experienced considerable groundwater depletion and subsidence during the past six decades.In this study,we use Sentinel-1A/B SAR images from 2015 to 2020 to map the ground subsidence of the Tianjin–Langfang area.Three subsiding zones centered at Guangyang,Wuqing–Bazhou,and Jinghai are identified with maximum subsidence rates of 98.1,121.8,and 104.7 mm/yr.Seasonal and long-term signals are separated from time series subsidence and hydraulic measurements using continuous wavelet transform to retrieve aquifer parameters.The long-term subsidence,which fits well with an exponential decaying model,remarkably slows down in our study area.The elastic skeletal storage coefficients range between 0.52×10−3 and 9.66×10−3.We then retrieve the spatial–temporal variations of total groundwater storage,recoverable groundwater storage,and irreversible groundwater storage.Groundwater storage depletion rates are apparently reducing,which benefits from the operation of the South-to-North Water Transfer Project and local groundwater management practices. | Xuguo Shi Tongtong Zhu Wei Tang Mi Jiang Houjun Jiang Chen Yang Wei Zhan Zutao Ming Shaocheng Zhang | 2022 | International Journal of Digital Earth2022,15,1: | 0 |
