| 1 | Risk factors and long-term health consequences of macrosomia:a prospective study in Jiangsu Province,China显示文摘We sought to determine risk factors associated with fetal macrosomia and to explore the long-term consequence of infant macrosomia at the age of 7 years.A prospective population based cohort study was designed to examine the associations between maternal and perinatal characteristics and the risk of macrosomia.A nested case-control study was conducted to explore the long-term health consequence of infant macrosomia.The mean maternal age of the macrosomia group was 24.74±3.32 years,which is slightly older than that in the control group(24.35±3.14 years,P = 0.000).The mean maternal body mass index(BMI) at early pregnancy was 22.75±2.81 kg/m 2,which was also higher than that in the control group(21.76±2.59 kg/m 2,P = 0.000).About 64.6% of macrosomic neonates were males,compared with 51.0% in the control group(P = 0.000).Compared with women with normal weight(BMI:18.5-23.9 kg/m 2),women who were overweight(BMI:24-27.9 kg/m 2) or obese(BMI ≥ 28 kg/m 2),respectively,had a 1.69-fold(P = 0.000) and a 1.49-fold(P = 0.000) increased risks of having a neonate with macrosomia,while light weight(BMI<18.5 kg/m 2) women had an approximately 50% reduction of the risk.Furthermore,macrosomia infant had a 1.52-fold and 1.50-fold risk,respectively,of developing overweight or obesity at the age of 7 years(P = 0.001 and P = 0.000).Older maternal age,higher maternal BMI at early pregnancy and male gender were independent risk factors of macrosomia.Macrosomic infant was associated with an increased predisposition to develop overweight or obesity at the beginning of their childhood. | Shouyong Gu Xiaofei An Liang Fang Xiaomin Zhang Chunyan Zhang Jingling Wang Qilan Liu Yanfang Zhang Yongyue Wei Zhibin Hu Feng Chen Hongbing Shen | 2012 | The Journal of Biomedical Research2012,26,4: | 14 |
| 2 | Establishing a human pancreatic stem cell line and transplanting induced pancreatic islets to reverse experimental diabetes in rats显示文摘The major obstacle in using pancreatic islet transplantation to cure type I and some type II diabetes is the shortage of the donors. One of ways to overcome such obstacle is to isolate and clone pancreatic stem cells as 'seed cells' and induce their differentiation into functional islets as an abundant trans-plantation source. In this study, a monoclonal human pancreatic stem cell (mhPSC) line was obtained from abortive fetal pancreatic tissues. Pancreatic tissues were taken from abortive fetus by sterile procedures, and digested into single cells and cell clusters with 0.1% type IV collagenase. Cultured in modified glucose-low DMEM with 10% fetal bovine serum (FBS), these single cells and cell clusters adhered to culture dishes, and then primary epidermal-like pancreatic stem cells started to clone. After digesting with 0.25% trypsin and 0.04% EDTA, fibroblasts and other cells were gradually eliminated and epithelioid pancreatic stem cells were gradually purified during generations. Using clone-ring selection, the mhPSCs were obtained. After addition of 10 ng/mL epidermal growth factor (EGF) in cell culture medium, the mhPSCs quickly grew and formed a gravelstone-like monolayer. Continuously proliferated, a mhPSC line, which was derived from a male abortive fetus of 4 months old, has been passed through 50 generations. More than 1×109 mhPSCs were cryo-preserved in liquid nitrogen. Karyotype analysis showed that the chromosome set of the mhPSC line was normal diploid. Immunocytochemistry results demonstrated that the mhPSC line was positive for the pdx1, glucagon, nestin and CK19, and negative for the insulin, CD34, CD44 and CD45 protein expression. RT-PCR revealed further that the mhPSCs expressed transcription factors of the pdx1, glucagon, nestin and CK19. Also, in vitro induced with β-mercaptoethanol, the mhPSCs differentiated into nerve cells that expressed the NF protein. Induced with nicotinamide, the mhPSCs differentiated into functional islet-like clusters, as identified by di-thizone staining, which expressed the transcription factor of the insulin and secreted the insulin and C-peptide. | 3 | Role of nitric oxide in the genotoxic response to chronic microcystin-LR exposure in human–hamster hybrid cells显示文摘Microcystin-LR(MC-LR) is the most abundant and toxic microcystin congener and has been classified as a potential human carcinogen(Group 2B) by the International Agency for Research on Cancer. However, the mechanisms underlying the genotoxic effects of MC-LR during chronic exposure are still poorly understood. In the present study, human–hamster hybrid(AL) cells were exposed to MC-LR for varying lengths of time to investigate the role of nitrogen radicals in MC-LR-induced genotoxicity. The mutagenic potential at the CD59 locus was more than 2-fold higher(p < 0.01) in ALcells exposed to a cytotoxic concentration(1 μmol/L) of MC-LR for 30 days than in untreated control cells, which was consistent with the formation of micronucleus. MC-LR caused a dose-dependent increase in nitric oxide(NO) production in treated cells. Moreover, this was blocked by concurrent treatment with the NO synthase inhibitor NG-methyl-L-arginine(L-NMMA), which suppressed MC-LRinduced mutations as well. The survival of mitochondrial DNA-depleted(ρ0) ALcells was markedly decreased by MC-LR treatment compared to that in ALcells, while the CD59 mutant fraction was unaltered. | 4 | Functional Characterization of the Apple RING E3 Ligase MdMIEL1 in Transgenic Arabidopsis显示文摘E3 ubiquitin ligases are involved in various physiological processes,and they play pivotal roles in growth and development.In this study,we identified a previously unknown gene in the apple fruit(Malus×domestica)and named it MdMIEL1.The MdMIEL1 gene encoded a protein that contained a zinc-finger domain at its N-terminus and a RING-finger motif at its C-terminus.To investigate MdMIEL1 functions,we generated transgenic Arabidopsis lines expressing the MdMIEL1 gene under the control of the Cauliflower mosaic virus 35S promoter.Interestingly,ectopic expression of MdMIEL1 in Arabidopsis produced multiple phenotypes,including early germination,early flowering and a lateral root number increase relative to wild-type plants.Further analysis indicated that MdMIEL1 regulated lateral root initiation by increasing auxin accumulation in the roots.In a word,these results suggest that,MdMIEL1 as a novel RING-finger ubiquitin ligase influences plant growth and development,and highlight that MdMIEL1 regulates lateral root growth. | AN Jianping LIU Xin SONG Laiqing YOU Chunxiang WANG Xiaofei HAO Yujin | 2017 | Horticultural Plant Journal2017,3,2: | 3 |
| 5 | The Growth-Promoting Mechanism of Brevibacillus laterosporus AMCC100017 on Apple Rootstock Malus robusta显示文摘Plant growth-promoting rhizobacteria(PGPR) located in rhizobacteria soil are beneficial to plant growth and development. A PGPR strain AMCC100017 of Brevibacillus laterosporus synthesizes the plant hormone IAA in a tryptophan-dependent manner. In this study, the AMCC100017 strain was used to treat Malus robusta, an excellent natural rootstock for apple production, and assess its ability to promote growth. The fresh weight, dry weight, plant height, and lateral root growth of M. robusta were significantly increased with treatment. The presence of the AMCC100017 strain increased IAA content in M. robusta and promoted root secretion of tryptophan. Colonization of the strain in the roots allowed continuous synthesis of IAA and promoted plant growth. In addition, the photosynthetic efficiency in leaves increased after microbial treatment, and the utilization of nitrogen, zinc, iron, copper and magnesium in leaves was increased, which was conducive to photosynthesis.Interestingly, the activities of CAT and SOD, as well as the contents of ROS in plants colonized by AMCC100017 were increased compared to control plants, but the activities of POD and MDA contents were decreased. AMCC100017 strain affected the antioxidant capacity and stress resistance of plants. AMCC100017 strain increased the content of soluble protein and soluble sugar in plants, improved plant metabolic activity and stress resistance. | 6 | Geochemical characteristics of hadal sediment in the northern Yap Trench显示文摘Two sediment cores were retrieved in the hadal zone of the Yap Trench,and their concentrations of six major elements Mg,Al,Ca,Ti,Mn and Fe and nine trace elements Sr,Ba,Pb,V,Cr,Co,Ni,Cu and Zn were determined in inductively coupled plasma atomic emission spectrometry(ICP-AES).According to the vertical distribution profiles of the 15 elements,the correlation of their concentrations,the ratios of Ni/Co,V/Cr,Fe/Al,and Ti/Al,and morphological characteristics of the sediment samples,the implications of the depositional environment and the sediment provenance were analyzed.The results show that the ratio of Ni/Co in all depths of the two sediment cores were below 5,and the ratio of V/Cr were lower than 2,indicating that the depositional environment of the hadal zone in the trench was oxidative and might have inflow of the Antarctic bottom oxygen-rich water.The sediment samples on the eastern side of the trench were siliceous mud mainly composed of diatoms,radiolarian,and sponge needles from surface to deep layer.The vertical profile of the elements,the concentration of TOC and the fossil record indicated that the sediment sample from station Dive113 was well mixed from surface to bottom layer.Based on the correlation of concentrations of the elements,the morphological characteristics of the sediment,and the ratios of Fe/Al and Ti/Al,we inferred that the sediment in the hadal zone of the trench had terrestrial,volcanic,biological,and authigenic sources.Major source of the sediment in the eastern side of the trench were terrestrial;whereas the sediment in the western side of the trench received more volcaniclastic input. | HUANG Yuhuan SUN Chengjun YANG Guipeng YUE Xin’an JIANG Fenghua CAO Wei YIN Xiaofei GUO Chaonan NIU Jiaohong DING Haibing | 2020 | Journal of Oceanology and Limnology2020,38,3: | 3 |

| 9 | Structural and functional characteristics of microbiota in oropharynx of sub-healthy children with gastrointestinal heat retention syndrome differentiated by traditional Chinese medicine显示文摘Objective:To explore the structural and functional characteristics of microbiota in oropharynx of subhealthy children with gastrointestinal heat retention syndrome(GHRS)differentiated by traditional Chinese medicine,and screen the biometric operational taxonomic units(OTUs)to assist the clinical diagnosis.Methods:We recruited children according to the“GHRS diagnostic scale”,collected their oropharyngeal swabs,and sequenced the 16 SrDNA V4 region.We described the bacterial structure with alpha-indexes,beta-distances,and relative abundances;moreover,we screened the differential genera/OTUs with Wilcoxon rank-sum test,Metagenome Seq analysis,and linear discriminant analysis effect size(LEf Se)analysis,in which biometric OTUs were selected to construct the receiver operating characteristic curve to verify the diagnostic value.The bacterial function was predicted with Kyoto Encyclopedia of Genes and Genomes pathways according to 16S rDNA gene by using Phylogenetic Investigation of Communities by Reconstruction of Unobserved States.Results:The study population was composed of 10 children with GHRS and 10 healthy control children.GHRS children were more likely to overeat(gluttony,P=.033).Alpha-indexes,such as Sobs,abundancebased coverage estimator,Bootstrap,and Qstat,were significantly higher in the GHRS group,while betadistances did not exhibit any significant intergroup differences.There were 9 differently distributed nonpredominant genera between the groups in Wilcoxon rank-sum test,as well as 13 non-predominant genera in Metagenome Seq analysis and 3 non-predominant OTUs in LEfSe analysis.OTU44 and OTU196 were used to construct the receiver operating characteristic curve,and the area under curve was 0.92.Predicted functions showed that pathways related to oxidative phosphorylation and carbon metabolism were enriched in healthy control samples,while the pathway related to renin secretion was remarkably enriched in GHRS samples.Conclusion:Unique oropharyngeal microbial structure and function were identified in GHRS children.OTU44 and OTU196 were specific OTUs,which could be used as biomarkers of GHRS to assist clinical diagnosis. | Jianhua Zhen He Yu Xiaofei Li Fei Dong Zi'an Zheng Xueyan Ma Yuxiang Wan Tiegang Liu Lu Fan Xiaohong Gu | 2022 | Journal of Traditional Chinese Medical Sciences2022,9,3: | 1 |
| 10 | Assessment of the cytotoxic and mutagenic potential of the Jialu River and adjacent groundwater using human-hamster hybrid cells显示文摘The Jialu River in China has been seriously polluted by the direct discharge of industrial and domestic wastewater. The predominant contaminants of the Jialu River and its adjacent groundwater were recently investigated. However, the potential genotoxic impact of polluted water on human health remains to be clarified. Here, we used human–hamster hybrid(A_L) cells, which are sensitive for detecting environmental mutagens. We found that the cytotoxicity and mutagenicity of the groundwater in the Jialu River basin were influenced by the infiltration of the Jialu River. Hydrological periods significantly affected the cytotoxicity, but not the mutagenic potential, of surface and groundwater. Further, the mutagenic potential of groundwater samples located < 1 km from the Jialu River(S_(M-2) water samples) was detected earlier than that of groundwater samples located approximately 20 km from the Jialu River(S_N water samples). Because of high cytotoxicity, the mutagenic potential of water samples from the Jialu River(S_(M-1) water samples) was not significantly enhanced compared with that of untreated controls. To further assess the mutagenic dispersion potential, an artificial neural network model was adopted. The results showed that the highest mutagenic potential of groundwater was observed approximately 10 km from the Jialu River. For such a system, we propose a mobile-service based max-min fairness resource scheduling scheme, where service amount which is more suitable for high mobility scenarios is adopted to characterize the information transmission capacity of the links and the max-min criteria is adopted to meet the fairness requirement of the moving vehicles. Simulation results demonstrate the effectiveness of our proposed scheme. | 13 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment. | 15 | A study on wheeze models显示文摘In this paper, the finite differenee schemes for the linear and nonlinear models of wheezes are given. The waveform of wheczes is analysed. It is shown that the numerical results of the lincar model of wheezes and Grotberg’s theorehcal results coincide well. The numerical results of the nonlincar model of wheczes are in qualita tive agreement with our expcrimental observations. It is suggested that the nonlinear elastance coefficient (β) causes damping, and is closely related to the flow limitation and the airway collapse. | 16 | Regional characteristics of ultrasonic structural scattering in liver显示文摘In this paper, the ultrasonic spectra of liver’s structural scattering wave obtained by autoregressive model and the spectral characteristics were analyzed statisticaJly The experimellt and analysis results show that a liver can be divided into three distingulshed scattering regions. The top layer region shows approkimate Rayleigh scattering characteristic, the central region shows diffosion scattering, and the region between the top layer and centre shows random scattering. These distribution characteristics correlate with the distribution of the microstructure characteristics of liver. | 17 | THE ANTHOCYANIN BIOSYNTHETIC REGULATOR MDMYB1 POSITIVELY REGULATES ASCORBIC ACID BIOSYNTHESIS IN APPLE显示文摘Ascorbic acid(AsA,vitamin C)is involved in the regulation of many aspects of plant growth and development.It is an essential micronutrient for humans and can prevent scurvy,maintain the health of gums and blood vessels,reduce the level of plasma cholesterol and enhance the immune systen.Apple cultivars Orin and Guanghui were crossed to obtain a group of hybrid offspring with and without red flesh in the course of assessing apple germplasm resources.Unexpectedly,the red-flesh apples had higher AsA contents than other apples.Further studies showed that the anthocyanin biosynthetic regulator MdMYBl directly activates the expression of dehydroascorbate reductase gene MdDHAR,thus promoting the activity of the DHAR enzyme and the accumulation of AsA.This finding reveals the mechanism leading to high AsA levels in red-flesh apples and suggests a new idea to cultivate red-flesh apples with high AsA contents and produce AsA efficiently and without pollution. | Jianping AN Xiaofei WANG Chunxiang YOU Yujin HAO | 2021 | Frontiers of Agricultural Science and Engineering2021,8,2: | 0 |
