| 1 | Chromosome analysis of esophageal squamous cell carcinoma cell line KYSE 410-4 by repetitive multicolor fluorescence in situ hybridization显示文摘Chromosome aberrations are distinctive features of human malignant tumors. Analysis of chromosomal changes can illuminate the molecular mechanisms underlying the development and progression of cancer. To establish the technique of multicolor fluorescence in situ hybridization (M-FISH) for identifying chromosome aberrations in esophageal carcinoma cell line KYSE 410-4, four pools of 6-color whole-chromosome painting probes have been designed and hybridized on the same metaphase spread by four rounds of repetitive FISH. Repetitive 6-color M-FISH was successfully established and the cytogenetic abnormalities in KYSE 410-4 cells were characterized. Chromosome gains occurred at 2q, 3, 8, 17p, and X. An isochromosome 3q was visualized in the cell line, which might be one intermedi- ate mechanism leading to 3p losses and/or 3q gains. Furthermore, 16 structural arrangements were detected, including four derivative chromosomes. The rearrangement of the centromeric regions accounted for approximately 44% of all rearrangements. The results added a more complete and accurate information of the genetic alterations to the classical cytogenetic description of KYSE 410-4 and provided a detailed cytogenetic background data for appropriate use of the cell line. | 2 | Morphological, mechanical and thermal properties of cyanate ester/benzoxazine resin composites reinforced by silane treated natural hemp fibers显示文摘This present study deals with the reinforcement of thermosetting resin blends composed of cyanate ester(CE) and benzoxazine(BOZ) resins with natural hemp fibers(NHFs). These NHFs were initially treated by using a silane coupling agent(SCA) in order to chiefly enhance their distributions as well as adhesions within the CE/BOZ resin matrix,then incorporated with various weight amounts ranging from 5 wt% to 20 wt% with a regular interval of 5 wt%. The obtained results showed that at the maximum treated fiber loading(20 wt%), distinctive enhancements in the mechanical properties in terms of flexural strength and microhardness were obtained. Besides, the thermal stability and glass transition temperature(Tg) were appreciably enhanced and were higher than those of the pure CE/BOZ resin properties. With respect to the astonishing properties of the NHFs, these enhancements could be possibly due to the good dispersion and adhesion of the treated NHFs inside the CE/BOZ resin achieved upon using the SCA. | 3 | An Efficient and Reliable Assay for Investigating the Effects of Hypoxia/Anoxia on Drosophila显示文摘Stroke is a leading cause of death worldwide. Up to one thousand potential drugs or interventions have been developed to treat stroke, out of which ~160 have gone on to clinical trials. However, none of them has been successful. New insights into the molecular and cellular mechanisms of ischemia-induced injury are needed for discovering new therapeutic targets. Recently, Drosophila has been used to uncover new hypoxia-related genes. In this study, we describe an efficient and reliable assay with a sophisticated apparatus for studying the effects of oxygen deprivation on flies. Using this assay, wild-type flies were exposed to an anoxic environment for varying lengths of time, then the cumulative death rate and mobility recovery were systematically analyzed. We found that anoxia for over one hour caused lethality. The cumulative death rate on day 5 after anoxia was linearly and positively correlatedwith the duration of anoxia, and reached 50% when the duration was 2.5 h–3 h. | 5 | Segmental Translocation Contributed to the Origin of the Brassica S-locus显示文摘Self-incompatibility(SI),which has recurred during the evolution of plants,is one of the most important cross-pollination mating systems.Three S-loci have been reported in Brassicaceae,namely,Arabidopsis lyrata(Al),Brassica(Br),and Leavenworthia alabamica(La)S-loci.Here,through multi-genomic comparative analysis of 20 species,we revealed that the most ancient S-locus was formed prior to the divergence of Brassicaceae lineage I and II.Itwas retained and inherited by Arabidopsis,as the Al S-locus in Brassicaceae lineage I.Furthermore,we found that the Br S-locus,which has been widely used in the breeding of Brassica crops to generate hybrid seeds,was formed through segmental translocation(ST)in the hexaploid ancestor of Brassica in Brassicaceae lineage II.The Br S-locus was evolved through a ST from one of the triplicated ancestral S-locus paralogs in the Brassica hexaploidy ancestor,while the other two S-locus paralogs were lost.Together with the previous discovery that the La S-locus was formed through a secondary origin in Brassicaceae lineage I,we conclude the monophyletic origin of Al and Br S-loci and clarify the evolutionary route of S-loci in the Brassicaceae family.Our findings will contribute to evolutionary studies and breeding applications of the S-locus in Brassicaceae. | Yinan Cui Mu Zhuang Jian Wu Jisheng Liu Yiyue Zhang Lingkui Zhang Yile Huang Xu Cai Jianli Liang Kang Zhang Xiaowu Wang Feng Cheng | 2020 | Horticultural Plant Journal2020,6,3: | 2 |
| 6 | Clinical characteristics and prognostic values of 1p32.3 deletion detected through fluorescence in situ hybridization in patients with newly diagnosed multiple myeloma: a single-center study in China显示文摘This study aimed to investigate the prevalence,clinical characteristics,and prognostic impact of 1p32.3 deletion in patients with newly diagnosed multiple myeloma (MM).A retrospective analysis was conducted on 411 patients with newly diagnosed MM;among which,270 received bortezomib-based therapies,and 141 received thalidomide-based therapies.Fluorescence in situ hybridization (FISH) was performed to detect six cytogenetic abnormalities,namely,del(1p32.3),gain(1q21),del(17p13),del(13q14),t(4;14),and t(11;14).Results showed that 8.3% of patients with MM were detected with del(1p32.3) and had significantly more bone marrow plasma cells (P =0.025),higher β2-microglobulin levels (P =0.036),and higher lactate dehydrogenase levels (P =0.042) than those without del(1p32.3).Univariate analysis showed that patients with del(1p32.3) under thalidomide-based therapies were strongly associated with short progression-free survival (PFS) (median PFS 11.6 vs.31.2 months,P =0.002) and overall survival (OS) (median OS 16.8 vs.45.9 months,P < 0.001).Multivariate analysis revealed that del(1p32.3) remained a powerful independent factor with worse PFS (P =0.006) and OS (P =0.016) for patients under thalidomide-based treatments.Patients with del(1 p32.3) under bortezomib-based treatments tended to have short PFS and OS.In conclusion,del(1p32.3) is associated with short PFS and OS in patients with MM who received thalidomide-or bortezomib-based treatments. | Huanping Wang Haitao Meng Jinghan Wang Yinjun Lou Yile Zhou Peipei Lin Fenglin Li Lin Liu Huan Xu Min Yang Jie Jin | 2020 | Frontiers of Medicine2020,14,3: | 2 |
| 7 | FACS-iChip:a high-efficiency iChip system for microbial‘dark matter’mining显示文摘The isolation chip method(iChip)provides a novel approach for culturing previously uncultivable microorganisms;this method is currently limited by the user being unable to ensure single-cell loading within individual wells.To address this limitation,we integrated flow cytometry-based fluorescence-activated cell sorting with a modified iChip(FACS-iChip)to effectively mine microbial dark matter in soils.This method was used for paddy soils with the aim of mining uncultivable microorganisms and making preliminary comparisons between the cultured microorganisms and the bulk soil via 16S rRNA gene sequencing.Results showed that the FACS-iChip achieved a culture recovery rate of almost 40%and a culture retrieval rate of 25%.Although nearly 500 strains were cultured from 19 genera with 8 FACS-iChip plates,only six genera could be identified via 16S rRNA gene amplification.This result suggests that the FACS-iChip is capable of detecting strains in the currently dead spaces of PCR-based sequencing technology.We,therefore,conclude that the FACS-iChip system provides a highly efficient and readily available approach for microbial‘dark matter’mining. | Haoze Liu Ran Xue Yiling Wang Erinne Stirling Shudi Ye Jianming Xu Bin Ma | 2021 | Marine Life Science & Technology2021,3,2: | 1 |

| 9 | Influence of litter decomposition on iron and manganese in the sediments of wetlands for acid mine drainage treatments显示文摘Plant litter will influence the bioavailability of heavy metals in sediments of wetlands used to treat acid mine drainage. To investigate the effect of plant litter on sediments in wetlands and define the comprehensive and continuous role of plant litter, different mass ratios(0%,5%, 20%) of litter were added into the sediments to study the influence of litter decomposition on the overlying water and sediments. The changes in pH, EC, Eh, Fe, and Mn of the overlying water and the organic matter in the sediments and the forms of Fe and Mn after 1, 7, 14, 21, and 28 days of litter decomposition were studied. The results indicated that litter decomposition increased the pH, EC, and reduced Eh of the overlying water. Litter decomposition promoted the release of Fe and Mn from the sediments into the overlying water and with the continuous decomposition of litter, the concentration of Fe and Mn in the overlying water declined. Litter decomposition increased the content of the organic matter in the sediment, and the forms of Fe and Mn indicated that litter decomposition could significantly affect the transformation of the forms of Fe and Mn.Reducible Fe was the main form in the sediments. Litter decomposition promoted the transformation of reducible Fe, the main form found in the sediments, intoexchangeable and oxidizable Fe, but had no effect on residual form. In this paper, we introduce several well-known compression algorithms in the research area as well as the boosting industry standardization works. Specifically,based on various applications of this 3D data, we summarize the static and dynamic point cloud compression, both including irregular geometry and photometry information that represent the spatial structure information and corresponding attributes, respectively.
