| 7 | Green fluorescent protein (GFP) transgenic pig produced by somatic cell nuclear transfer显示文摘Transgenic somatic cell nuclear transfer is a very promising route for producing transgenic farm ani-mals. Research on GFP transgenic pigs can provide useful information for breeding transgenic pigs, human disease models and human organ xenotransplantation. In this study, a liposomal transfection system was screened and transgenic embryos were reconstructed by nuclear transfer of GFP positive cells into enucleated in vitro matured oocytes. The development of reconstructed embryos both in vitro and in vivo was observed, and GFP expression was determined. The results showed that porcine fe-tal-derived fibroblast cells cultured with 4.0 μL/mL liposome and 1.6 μg/mL plasmid DNA for 6 h re-sulted in the highest transfection rate (3.6%). The percentage of GFP reconstructed embryos that de-veloped in vitro to the blastocyst stage was 10%. Of those the GFP positive percentage was 48%. Re-constructed transgenic embryos were transferred to 10 recipients. 5 of them were pregnant, and 3 de-livered 6 cloned piglets in which 4 piglets were transgenic for the GFP as verified by both GFP protein expression and GFP DNA sequence analysis. The percentage of reconstructed embryos that resulted in cloned piglets was 1.0%; while the percentage of piglets that were transgenic was 0.7%. | 8 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 10 | Subseasonal features of the Asian summer monsoon in the NCEP climate forecast system显示文摘The operational climate forecast system(CFS)of the US National Centers for Environmental Prediction provides climate predictions over the world,and CFS products are becoming an important source of information for regional climate predictions in many Asian countries where monsoon climate dominates.Recent studies have shown that,on monthly-to-seasonal time-scales,the CFS is highly skillful in simulating and predicting the variability of the Asian monsoon.The higher-frequency variability of the Asian summer monsoon in the CFS is analyzed,using output from a version with a spectral triangular truncation of 126 waves in horizontal and 64 sigma layers in vertical,focusing on synoptic,quasi-biweekly,and intraseasonal time-scales.The onset processes of different regional monsoon components were investigated within Asia.Although the CFS generally overestimates variability of monsoon on these time-scales,it successfully captures many major features of the variance patterns,especially for the synoptic time-scale.The CFS also captures the timing of summer monsoon onsets over India and the Indo-China Peninsula.However,it encounters difficulties in simulating the onset of the South China Sea monsoon.The success and failure of the CFS in simulating the onset of monsoon precipitation can also be seen from the associated features of simulated atmospheric circulation processes.Overall,the CFS is capable of simulating the synoptic-to-intraseasonal variability of the Asian summer monsoon with skills.As for seasonal-to-interannual time-scales shown previously,the model is expected to possess a potential for skillful predictions of the high-frequency variability of the Asian monsoon. | Song YANG WEN Min R Wayne HIGGINS | 2008 | Acta Oceanologica Sinica2008,27,3: | 3 |

| 14 | Newly self-administered two-step tool for screening cognitive function in an ageing Chinese population:an exploratory cross-sectional study显示文摘Background Early screening of cognitive function is critical to dementia treatment and care.However,traditional tests require face-to-face administration and are often limited by implementation costs and biases.Aims This study aimed to assess whether the Thoven Cognitive Self-Assessment(TCSA),a novel,innovative two-step touchscreen-based cognition assessment tool,could identify early cognitive impairment due to dementia in older adults.Methods The TCSA was administered to 61 healthy controls(HCs),46 participants with mild cognitive impairment(MCI)and 44 participants diagnosed with dementia recruited from Shanghai.Two outcome measures were generated from the TCSA test:the TCSA_(primary task)score and the TCSA_(secondary task)score.Results The total average scores in the control group for the TCSA_(primary task)and TCSA_(secondary task)were significantly higher than those in the MCI and dementia groups(TCSA_(primary task):HCs vs MCI group vs dementia group,8.58±1.76 vs 5.40±2.67 vs 2.74±2.11,F=75.40,p<0.001;TCSA_(secondary task):HCs vs MCI group vs dementia group,23.02±3.31 vs 17.95±4.93 vs 11.93±5.50,F=76.46,p<0.001).Moreover,receiver operating characteristic analysis showed that a score below 7.5 for the TCSA_(primary task)and a score below 22.5 for the TCSA_(secondary task)were indicators of MCI.Conclusions The TCSA appears to be efficacious for the detection of cognitive impairment in older adults.It demonstrates the potential for large-scale cognition screening in community service settings. | Jing Nie Yang Yang Yining Gao Wenwen Jiang Aisikeer Aidina Fei Sun Lucas R Prieto Jie Yu Kang Ju Lisheng Song Xia Li | 2023 | General Psychiatry2023,36,1: | 2 |

| 18 | Geometry deviation effects of railway catenaries on pantograph– catenary interaction: a case study in Norwegian Railway System显示文摘This paper presents a non-contact measurement of the realistic catenary geometry deviation in the Norwegian railway network through a laser rangefinder.The random geometry deviation is included in the catenary model to investigate its effect on the pantograph–catenary interaction.The dispersion of the longitudinal deviation is assumed to follow a Gaussian distribution.A power spectrum density represents the vertical deviation in the contact wire.Based on the Monte Carlo method,several geometry deviation samples are generated and included in the catenary model.A lumped mass pantograph with flexible collectors is employed to reproduce the high-frequency behaviours.The stochastic analysis results indicate that the catenary geometry deviation causes a significant dispersion of the pantograph–catenary interaction response.The contact force standard deviations measured by the inspection vehicle are within the scope of the simulation results.A critical cut-off frequency that covers 1/16 of the dropper interval is suggested to fully describe the effect of the catenary geometry deviation on the contact force.The statistical minimum contact force is recommended to be modified according to the tolerant contact loss rate at high frequency.An unpleasant interaction performance of the pantograph–catenary can be expected at the catenary top speed when the random catenary geometry deviation is included. | Yang Song Tengjiao Jiang Petter Navik Anders Rønnquist | 2021 | Railway Engineering Science2021,29,4: | 2 |
