| 1 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 3 | Associations between parental support for physical activity and moderate-to-vigorous physical activity among Chinese school children: A cross-sectional study显示文摘Purpose: The objective of this study was to examine the associations between parental support for physical activity(PA) and levels of moderate-to-vigorous physical activity(MVPA) among Chinese school children.Methods: Study participants were 81,857 school-aged children(aged 9–17 years; mean age = 13 years; 49.0% boys) and their parents or guardians who took part in the 2016 Physical Activity and Fitness in China—The Youth Study project. The independent variables included various forms of parental support for PA ascertained through a survey completed by parents or guardians. The dependent variable was the children's average MVPA in minutes per day by self-report. Structure equation modeling was used to examine the associations between parental support for PA and children's MVPA minutes.Results: The overall mean for MVPA participation time for the child participants was 48.82 ± 28.71 min/day. Significant associations between various forms of parental support and MVPA among children were found. Parental encouragement, accompanying, financial support, involvement,and role modeling for PA were positively associated with a high level of student MVPA participation. Parental PA knowledge-sharing was not associated with MVPA. | 5 | Prevalence and correlates of meeting the muscle-strengthening exercise recommendations among Chinese children and adolescents:Results from 2019 Physical Activity and Fitness in China—The Youth Study显示文摘Purpose This study aimed to describe the national prevalence of Chinese children and adolescents who met the World Health Organization muscle-strengthening exercise(MSE)recommendations and identify correlates of meeting the MSE recommendations.Methods Cross-sectional data from the 2019 Physical Activity and Fitness in China—The Youth Study,a nationally representative sample of Chinese children and adolescents(n=80,413;mean age=13.7 years;53.9%girls)and their parents,were analyzed.Children and adolescents who reported engaging≥3 days(up to 7 days)of MSE per week were classified as meeting the MSE recommendations.MSE,demographics,lifestyle behaviors(sport participation,moderate-to-vigorous physical activity,screen time,and sleep duration),exercise intention,peer and parental support,and parental MSE participation were assessed through self-reports.Logistic regression models were used to determine the correlates of meeting the MSE recommendations.The analyses were completed in 2020.Results Overall,39.3%of children and adolescents met the MSE recommendations.Girls,10th-12th graders,minorities,those from lower income households and those from families with lower parental education were less likely to meet the MSE recommendations.Children and adolescents who were proficient in≥2 sports were more likely to meet the MSE recommendations(adjusted odds ratio(aOR)=1.44,95%confidence interval(95%CI):1.26-1.65),as were those with more moderate-to-vigorous physical activity(aOR=1.57,95%CI:1.53-1.61).Additionally,children and adolescents with high exercise intention(aOR=1.60,95%CI:1.51-1.69),those whose parents met the adult MSE recommendations(aOR=1.46,95%CI:1.40-1.52),and those who received high peer(aOR=1.27,95%CI:1.20-1.34)and parental support(aOR=1.07,95%CI:1.04-1.12)were more likely to meet the MSE recommendations.Conclusion Less than two-fifths of Chinese children and adolescents met the World Health Organization MSE recommendations.The correlates identified in our study can help inform the development of school and community based strategies and policies to enhance participation in MSE and improve muscular fitness of all Chinese children and adolescents. | Fei Xin Zheng Zhu Sitong Chen Huan Chen Xiaoqing Hu Xiao Ma Kun Liang Yang Liu Lijuan Wang Yujun Cai Ang Chen Yan Tang | 2022 | Journal of Sport and Health Science2022,11,3: | 2 |

| 14 | Leakage detection of oil tank using terahertz spectroscopy显示文摘The necessity for safety in oil-gas storage and transportation has led to increasing technical requirements for on-line monitoring of damaged pores and oil leakage from tanks. In this study, the severity of damage of the oil tank at the micron level was detected by terahertz time-domain spectroscopy(THz-TDS), which is of great significance for the early detection and prevention of oil leakage. The THz amplitude(EP) was related to the severity of damaged according to the THz-TDS measurement of oil tanks with various degrees of damage, including intact, partially damaged, completely damaged, and seriously damaged points.Absorption and scattering effects co-occurred when the THz wave penetrated the tanks, with the collective tendencies being used to expressly monitor oil leakage from tanks. When the oil tank was damaged to form micron-level pores and the crude oil had not overflowed, the pore size was close to THz wavelength and the Mie scattering effect was obvious. After further destruction of the pores, the crude oil gradually spilled over and the scattering effect was gradually transformed from Mie scattering to Rayleigh scattering. In addition, the polar molecules in crude oil have strong resonance under the irradiation of THz wave, and the THz wave has strong attenuation. Eventually, surface tension of the oil flattens the surface of the tank, the scattering effect is gradually suppressed and replaced by the absorption effect. Absorption and scattering caused by THz waves passing through tanks coexist and have competing relationships. The change rule of EPcan successfully prove the phenomenon and can be considered as an important alternative for application to predict the degree of tank damage. | 18 | Identification of Expression Pattern and Clinical Significance of the Small Cajal Body-specific RNA SCARNA16 in Hepatocellular Carcinoma显示文摘Background and Aims:For high morbidity and mortality,hepatocellular carcinoma(HCC)becomes a major health issue worldwide.Nowadays,numerous non-coding RNAs(ncRNAs)are known to regulate the occurrence and patho-genesis of tumors.Some ncRNAs have also been developed as tumor biomarkers and therapeutic targets.However,the potential function of the small Cajal body-specific RNA(scaRNA)SCARNA16,a newly identified ncRNA,remains to be explored in HCC.Methods:In both HCC cell lines and specimens from 120 enrolled patients,the expression val-ues of SCARNA16 were detected.We divided patients into SCARNA16 high and low expression subgroups,and then analyzed the difference of various clinical characteristics and prognosis data between subgroups.Results:Compared to paired controls,SCARNA16 was significantly down-regulated in HCC cell lines and clinical specimens(p<0.01).Besides,HCC patients with lower SCARNA16 expression commonly presented with larger and more tumor lesions,more ves-sel carcinoma emboli,more capsular invasion and higher TNM stages(p<0.05).Moreover,SCARNA16 expression was negatively correlated with postoperative prognosis of HCC patients in 5-year follow-up,including tumor-free survival(TFS)(median time of low vs.high subgroups:14 vs.48 months,p=0.006)and overall survival(OS)(median time of low vs.high subgroups:39 vs.52 months,p=0.001).Besides,SCARNA16 acted as an independent prognostic bio-marker in TFS(hazard ratio[HR]:0.578,95%CI:0.345-0.969,p=0.038)and OS(HR:0.366,95%CI:0.178-0.752,p=0.006).Conclusions:Low expression patterns of SCAR-NA16 remarkably associated with severe clinical status and poor survival of patients,suggesting that SCARNA16 pos-sesses potency as a novel biomarker for HCC. | Sitong Zhang Yuan Ding Zhongquan Sun Yao Ge Yanjie Li Xin Han Qianhui Xu Liuzhi Zhou Yining Chen Hao Xu Yang Bai Chang Xu Hao Ding Sheng Yan Weilin Wang | 2022 | Journal of Clinical and Translational Hepatology2022,10,1: | 0 |
