1Resveratrol provides neuroprotection by regulating the JAK2/STAT3/PI3K/AKT/mTOR pathway after stroke in rats显示文摘Ischemic stroke is a common disease with high mortality and morbidity worldwide.One of the important pathophysiological effects of ischemic stroke is apoptosis.A neuroprotective effect is defined as the inhibition of neuronal apoptosis to rescue or delay the infarction in the surviving ischemic penumbra.Resveratrol is a natural polyphenol that reportedly prevents cerebral ischemia injury by regulating the expression of PI3K/AKT/mTOR.Therefore,this study aimed to elucidate the neuroprotective effect of resveratrol on cerebral ischemia/reperfusion injury and to investigate the signaling pathways and mechanisms through which resveratrol regulates apoptosis in the ischemic penumbra.Rats were subjected to middle cerebral artery occlusion for 2 h followed by 24 h reperfusion.Cerebral infarct volume was measured using 2%TTC staining.TUNEL staining was conducted to evaluate neuronal apoptosis.Western blotting and immunohistochemistry were used to detect the proteins involved in the JAK2/STAT3/PI3K/AKT/mTOR pathway.The results suggested that resveratrol significantly improved neurological function,reduced cerebral infarct volume,decreased neuronal damage,and markedly attenuated neuronal apoptosis;these effects were attenuated by the inhibition of PI3K/AKT with LY294002 and JAK2/STAT3 with AG490.We also found that resveratrol significantly upregulated the expression of p-JAK2,p-STAT3,p-AKT,p-mTOR,and BCL-2 and downregulated expression of cleaved caspase-3 and BAX,which was partially reversed by LY294002 and AG490.These results suggested that resveratrol provides a neuroprotective effect against cerebral ischemia/reperfusion injury,which is partially mediated by the activation of JAK2/STAT3 and PI3K/AKT/mTOR.Resveratrol may indirectly upregulate the PI3K/AKT/mTOR pathway by activating JAK2/STAT3.Yongying Hou Ke Wang Weijun Wan Yue Cheng Xia Pu Xiufeng Ye 2018Genes & Diseases2018,5,3:60
2Isolation and Characterization of an Algicidal Bacterium Indigenous to Lake Taihu with a Red Pigment able to Lyse Microcystis aeruginosa显示文摘Objective To isolate and characterize indigenous algicidal bacteria and their algae-lysing compounds active against Microcystis aeruginosa, strains TH1, TH2, and FACHB 905. Methods The bacteria were identified using the Biolog automated microbial identification system and 16S rDNA sequence analysis. The algae-lysing compounds were isolated and purified by silica gel column chromatography and reverse-phase high performance liquid chromatography. Their structures were confirmed by Nuclear Magnetic Resonance (NMR) and Fourier Transform Infrared (FT-IR) spectroscopy. Algae-lysing activity was observed using microscopy. Results The algae-lysing bacterium LTH-2 isolated from Lake Taihu was identified as Serratia marcescens. Strain LTH-2 secreted a red pigment identified as prodigiosin (C 20 H 25 N 3 O), which showed strong lytic activity with algal strains M. aeruginosa TH1, TH2, and FACHB 905 in a concentration-dependent manner. The 50% inhibitory concentration (IC 50 ) of prodigiosin with the algal strains was 4.8 (±0.4)×10 -2 μg/mL, 8.9 (±1.1)×10-2 μg/mL, and 1.7 (±0.1)×10 -1 μg/mL in 24 h, respectively. Conclusion The bacterium LTH-2 and its pigment had strong Microcystis-lysing activity probably related to damage of cell membranes. 4Concurrent intracranial and extracranial artery stenosis and the prognosis of transient ischaemic symptoms or imaging-negative ischaemic stroke显示文摘Background and purpose Transient ischaemic attack(TIA),transient symptoms with infarction(TSI)and diffusion-weighted imaging(DWI)-negative acute ischaemic stroke(AIS)share similar aetiologies but are considered to have a rather benign prognosis.We intended to investigate the association between intracranial atherosclerotic stenosis(ICAS),extracranial atherosclerotic stenosis(ECAS)and the prognosis of patients with TIA,TSI and DWI-negative AIS.Methods Clinical and imaging data of eligible participants were derived from the Chinese Intracranial Atherosclerosis study,according to symptom duration,acute infarction on DWI and discharge diagnosis.Based on the severity and location of arterial atherosclerosis,we categorised the study population into four groups:no or<50% ICAS and no ECAS;≥50% ICAS but no ECAS;no or<50%ICAS with ECAS;and concurrent≥50% ICAS and ECAS.Using multivariable Cox regression models,we analysed the relationship between the severity and distribution of large artery atherosclerosis and the prognosis of TIA,TSI and DWI-negative AIS.Results A total of 806 patients were included,67.3% of whom were male.The median age of the study participants was 63 years.Patients in the concurrent≥50% ICAS and ECAS subgroup had both a significantly higher 1-year recurrence rate(adjusted HR 3.4(95%CI 1.15 to 10.04),p=0.027)and a higher risk of composite vascular events(adjusted HR 3.82(95%CI 1.50 to 9.72),p=0.005).Conclusions Concurrent ICAS and ECAS is associated with a higher possibility of 1-year recurrent stroke or composite vascular events.Large artery evaluation is necessary to assess patients with transient ischaemic symptoms or DWI-negative AIS.Progress in shortening the time interval between symptom onset and large vessel evaluation is needed.Yue Suo Jing Jing Yuesong Pan Weiqi Chen Hongyu Zhou Hao Li Yuehua Pu Liping Liu Xingquan Zhao Yilong Wang Xia Meng Yongjun Wang 2021Stroke & Vascular Neurology2021,6,1:10

6miR-93-5p Transferred by Exosomes Promotes the Proliferation of Esophageal Cancer Cells via Intercellular Communication by Targeting PTEN显示文摘Objective To investigate the relationship between plasma miR-93-5p and the risk of esophageal cancer, as well as the influence of miR-93-5p on the biological function of esophageal cancer cells,exerted through exosomes.Methods The expression of plasma miR-93-5p in esophageal cancer patients and healthy controls was analysed by real-time quantitative PCR. The influence of miR-93-5p on the risk and prognosis of esophageal carcinoma was analyzed by conditional logistic regression and survival analysis. The effect of miR-93-5p on the biological function of recipient cells was investigated by establishing an in vitro donor cell co-culture model. 7A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 11Tumor-derived neomorphic mutations in ASXL1 impairs the BAP1-ASXL1-FOXK1/K2 transcription network显示文摘Additional sex combs-like 1(ASXL1)interacts with BRCA1-associated protein 1(BAP1)deubiquitinase to oppose the polycomb repressive complex 1(PRC1)-mediated histone H2A ubiquitylation.Germline BAP1 mutations are found in a spectrum of human malignancies,while ASXL1 mutations recurrently occur in myeloid neoplasm and are associated with poor prognosis.Nearly all ASXL1 mutations are heterozygous frameshift or nonsense mutations in the middle or to a less extent the C-terminal region,resulting in the production of C-terminally truncated mutant ASXL1 proteins.How ASXL1 regulates specific target genes and how the C-terminal truncation of ASXL1 promotes leukemogen-esis are unclear.Here,we report that ASXL1 interacts with forkhead transcription factors FOXK1 and FOXK2 to regulate a subset of FOXK1/K2 target genes.We show that the C-terminally truncated mutant ASXL1 proteins are expressed at much higher levels than the wild-type protein in ASXL1 heterozygous leukemia cells,and lose the ability to interact with FOXK1/K2.Specific deletion of the mutant allele eliminates the expression of C-termi-nally truncated ASXL1 and increases the association of wild-type ASXL1 with BAP1,thereby restoring the expression of BAP1-ASXL1-FOXK1/K2 target genes,particularly those involved in glucose metabolism,oxygen sensing,and JAK-STAT3 signaling pathways.In addition to FOXK1/K2,we also identify other DNA-bind-ing transcription regulators including transcription factors(TFs)which interact with wild-type ASXL1,but not C-terminally truncated mutant.Our results suggest that ASXL1 mutations result in neomorphic alleles that contribute to leukemogenesis at least in part through dominantly inhibiting the wild-type ASXL1 from interacting with BAP1 and thereby impairing the function of ASXL1-BAP1-TF in regulating target genes and leukemia cell growth.Yu-Kun Xia Yi-Rong Zeng Meng-Li Zhang Peng Liu Fang Liu Hao Zhang Chen-Xi He Yi-Ping Sun Jin-Ye Zhang Cheng Zhang Lei Song Chen Ding Yu-Jie Tang Zhen Yang Chen Yang Pu Wang Kun-Liang Guan Yue Xiong Dan Ye 2021Protein & Cell2021,12,7:6
12Prevalence of Overweight and Obesity among Students Aged 7-22 Years in Jiangsu Province,China显示文摘Objective To report the prevalence and trend of overweight and obesity among students aged 7-22 years in Jiangsu,2010 to 2013.Methods This cross-sectional study was carried out as part of students physical fitness and health survey in Jiangsu province.A total of 255,581 subjects(50.03% males and 49.97% females) enrolled in 82 school and 10 universities in Jiangsu.Weights and heights were obtained for each subject and its body mass index(BMI) was calculated using the Chinese Working Group on Obesity in China(CWGO).Results Anthropometric measurement including bodyweight,height,BMI and bust were significantly different between males in urban compared to females living rural areas(P<0.001).The total prevalence of overweight and obesity was 12.4% and 5.7%.Males had a significantly higher rate than in female's student.The prevalence of overweight and obesity by age groups was(14.5%,10.3%) at age 7-11 years,(11.2%,6.8%) at age 12-14 years,(11.7%,3.1%) at age 15-17 years,and(11.4%,2.3%) at age 18-22 years.By regions;the highest prevalence of overweight obesity reported in Taizhou(10.0%,14.2%),Xuzhou(9.4%,12.5%),and Nanjing(9.2%,15.6%),respectively.Conclusion The finding declares that overweight and obesity are important health problems among students in Jiangsu Province.Early intervention programme are needed to address this problems.Taha Hussein Musa WEI Li LI Xiao Shan PU Yue Pu WEI Ping Min 2016Biomedical and Environmental Sciences2016,29,10:5
13Artemisinin Biosynthesis in Non-glandular Trichome Cells of Artemisia annua显示文摘Artemisinin-based combination therapy(ACT)forms the first line of malaria treatment.However,the yield fluctuation of artemisinin has remained an unsolved problem in meeting the global demand for ACT.This problem is mainly caused by the glandular trichome(GT)-specific biosynthesis of artemisinin in all currently used Artemisia annua cultivars.Here,we report that non-GT cells of self-pollinated inbred A.annua plants can express the artemisinin biosynthetic pathway.Gene expression analysis demonstrated the transcription of six known pathway genes in GT-free leaves and calli of inbred A.annua plants.LC-qTOF-MS/MS analysis showed that these two types of GT-free materials produce artemisinin,artemisinic acid,and arteannuin B.Detailed IR-MALDESI image profiling revealed that these three metabolites and dihydroartemisinin are localized in non-GT cells of leaves of inbred A.annua plants.Moreover,we employed all the above approaches to examine artemisinin biosynthesis in the reported XL annua glandless(gl)mutant.The resulting data demonstrated that leaves of regenerated gl plantlets biosynthesize artemisinin.Codectively,these findings not only add new knowledge leading to a revision of the current dogma of artemisinin biosynthesis inannua but also may expedite innovation of novel metabolic engineering approaches for high and stable production of artemisinin in the future.Rika Judd M.Caleb Bagley Mingzhuo Li Yue Zhu Caiyan Lei Seyit Yuzuak Mans Ekelof Gaobin Pu Xiting Zhao David C.Muddiman De-Yu Xie 2019Molecular Plant2019,12,5:5

15miR-410 induces both epithelial–mesenchymal transition and radioresistance through activation of the PI3K/mTOR pathway in non-small cell lung cancer显示文摘Radiotherapy remains one of the major treatments for non-small cell lung cancer(NSCLC)patients;whereas intrinsic or acquired radioresistance limits its efficacy.Nevertheless,most studies so far have only focused on acquired resistance.The exact mechanisms of intrinsic radioresistance in NSCLC are still unclear.A few studies have suggested that epithelial–mesenchymal transition(EMT)is associated with radioresistance in NSCLC.However,little is known about whether the abnormal expression of specific microRNAs induces both EMT and radioresistance.We previously found that miR-410 has multiple roles as an oncomiRNA in NSCLC.In this study,we revealed that miR-410 overexpression promoted EMT and radioresistance,accompanied by enhanced DNA damage repair both in vitro and in vivo.Conversely,knockdown of miR-410 showed the opposite effects.We further demonstrated that PTEN was a direct target of miR-410 by using bioinformatic tools and dual-luciferase reporter assays,and the miR-410-induced EMT and radioresistance were reversed by PI3K,Akt,and mTOR inhibitors or by restoring the expression of PTEN in NSCLC cells.In addition,we preliminarily found that the expression of miR-410 was positively correlated with EMT and negatively associated with the expression of PTEN in NSCLC specimens.In summary,these results demonstrated that miR-410 is an important regulator on enhancing both NSCLC EMT and radioresistance by targeting the PTEN/PI3K/mTOR axis.The findings suggest that miR-410-induced EMT might significantly contribute to the enhanced radioresistance.Therefore,miR-410 may serve as a potential biomarker or therapeutic target for NSCLC radiotherapy.Yue Yuan Hu Liao Qiang Pu Xixian Ke Xueting Hu Yongfang Ma Xinmei Luo Qianqian Jiang Yi Gong Min Wu Lunxu Liu Wen Zhu 2020Signal Transduction and Targeted Therapy2020,5,1:5
16Ultrafine Pt nanoparticles supported on double-shelled C/TiO2 hollow spheres material as highly efficient methanol oxidation catalysts显示文摘Catalyst support is extremely important for future fuel cell devices.In this work,we developed doubleshelled C/TiO2(DSCT)hollow spheres as an excellent catalyst support via a template-directed method.The combination of hollow structure,TiO2 shell and carbon layer results in excellent electron conductivity,electrocatalytic activity,and chemical stability.These uniformed DSCT hollow spheres are used as catalyst support to synthesize Pt/DSCT hollow spheres electrocatalyst.The resulting Pt/DSCT hollow spheres exhibited high catalytic performance with a current density of 462 mA mg^-1 for methanol oxidation reaction,which is 2.52 times higher than that of the commercial Pt/C.Furthermore,the increased tolerance to carbonaceous poisoning with a higher If/Ibratio and a better long-term stability in acid media suggests that the DSCT hollow sphere is a promising C/TiO2-based catalyst support for direct methanol fuel cells applications.Xiaoyu Yue Yuguang Pu Wen Zhang Ting Zhang Wei Gao 2020Journal of Energy Chemistry2020,29,10:4

19Urinary levels of nickel and chromium associated with dental restoration by nickel-chromium based alloys显示文摘This paper aims to investigate if the dental restoration of nickel-chromium based alloy (Ni-Cr) leads to the enhanced excretions of Ni and Cr in urine. Seven hundred and ninety-five patients in a dental hospital had single or multiple Ni-Cr alloy restoration recently and 198 controls were recruited to collect information on dental restoration by questionnaire and clinical examination. Urinary concentrations of Ni and Cr from each subject were measure by graphite furnace atomic absorption spectrometry. Compared to the control group, the urinary level of Ni was significantly higher in the patient group of 〈 1 month of the restoration duration, among which higher Ni excretions were found in those with either a higher number of teeth replaced by dental alloys or a higher index of metal crown not covered with the porcelain. Urinary levels of Cr were significantly higher in the three patient groups of 〈1, 1 to 〈3 and 3 to 〈6 months, especially in those with a higher metal crown exposure index. Linear curve estimations showed better relationships between urinary Ni and Cr in patients within 6-month groups. Our data suggested significant increased excretions of urinary Ni and Cr after dental restoration. 20Studies on blood enrichment and anti-tumor effects of combined Danggui Buxue Decoction,Fe and rhEPO based on colon cancer-related anemia model and gut microbiota modulation显示文摘Colon cancer-related anemia(CCRA)is mainly caused by systemic inflammation,intestinal bleeding,iron deficiency and chemotherapy-induced myelosuppression in colon cancer.However,the best therapeutic schedule and related mechanism on CCRA were still uncertain.Studies on blood enrichment and anti-tumor effects of combined Danggui Buxue Decoction(DBD),Fe and rhEPO based on CCRA and gut microbiota modulation were conducted in this paper.Here,CCRA model was successfully induced by subcutaneous inoculation of CT-26 and i.p.oxaliplatin,rhEPO+DBD high dosage+Fe(EDF)and rhEPO+DBD high dosage(ED)groups had the best blood enrichment effect.Attractively,EDF group also showed antitumor activity.The sequencing results of gut microbiota showed that compared to P group,the relative abundances of Lachnospiraceae and opportunistic pathogen(Odoribacter)in ED and EDF groups were decreased.Interestingly,EDF also decreased the relative abundances of cancer-related bacteria(Helicobacter,Lactococcus,Alloprevotella)and imbalance-inducing bacteria(Escherichia-Shigella and Parabacteroides)and increased the relative abundances of butyrate-producing bacteria(Ruminococcaceae_UCG-014),however,ED showed the opposite effects to EDF,this might be the reason of the smaller tumor volume in EDF group.Our findings proposed the best treatment combination of DBD,rhEPO and Fe in CCRA and provided theoretical basis and literature reference for CCRA-induced intestinal flora disorder and the regulatory mechanism of EDF.SHI Xu-Qin ZHU Zhen-Hua YUE Shi-Jun TANG Yu-Ping CHEN Yan-Yan PU Zong-Jin TAO Hui-Juan ZHOU Gui-Sheng DUAN Jin-Ao 2021Chinese Journal of Natural Medicines2021,19,6:4
