| 5 | Combination of mild therapeutic hypothermia and adipose-derived stem cells for ischemic brain injury显示文摘Mild therapeutic hypothermia has been shown to mitigate cerebral ischemia, reduce cerebral edema, and improve the prognosis of patients with cerebral ischemia. Adipose-derived stem cell-based therapy can decrease neuronal death and infiltration of inflammatory cells, exerting a neuroprotective effect. We hypothesized that the combination of mild therapeutic hypothermia and adipose-derived stem cells would be neuroprotective for treatment of stroke. A rat model of transient middle cerebral artery occlusion was established using the nylon monofilament method. Mild therapeutic hypothermia(33°C) was induced after 2 hours of ischemia. Adipose-derived stem cells were administered through the femoral vein during reperfusion. The severity of neurological dysfunction was measured by a modified Neurological Severity Score Scaling System. The area of the infarct lesion was determined by 2,3,5-triphenyltetrazolium chloride staining. Apoptotic neurons were detected by terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end labeling(TUNEL) staining. The regeneration of microvessels and changes in the glial scar were detected by immunofluorescence staining. The inflammatory responses after ischemic brain injury were evaluated by in situ staining using markers of inflammatory cells. The expression of inflammatory cytokines was measured by reverse transcription-polymerase chain reaction. Compared with mild therapeutic hypothermia or adipose-derived stem cell treatment alone, their combination substantially improved neurological deficits and decreased infarct size. They synergistically reduced the number of TUNEL-positive cells and glial fibrillary acidic protein expression, increased vascular endothelial growth factor levels, effectively reduced inflammatory cell infiltration and down-regulated the m RNA expression of the proinflammatory cytokines interleukin-1β, tumor necrosis factor-α and interleukin-6. | 6 | Clinical Application of Loewenstein Occupational Therapy Cognitive Assessment Battery-Second Edition in Evaluating of Cognitive Function of Chinese Patients with Post-stroke Aphasia显示文摘Objective To investigate the clinical application value of Loewenstein Occupational Therapy Cognitive Assessment(LOTCA)battery in Chinese patients with post-stroke aphasia.Methods Cognitive functions of 59 Chinese patients with aphasia following a stroke were assessed with the Chinese version of the second edition of LOTCA battery and their linguistic functions were tested with the Western Aphasia Battery(WAB)Scale,respectively.The results of LOTCA were analyzed and compared across different groups,in the light of gender,age,educational background,the length of illness,and the degree of aphasia.Results Neither the score of subtests of the LOTCA nor the overall scores of LOTCA of aphasia patients with different gender and educational background differed(all P>0.05).In different age groups,apart from thinking operation(F=3.373,P=0.016),visuomotor organization(F=3.124,P=0.022),attention(F=3.729,P=0.009)and the total score(F=2.683,P=0.041),there was no difference in terms of the other subtest scores of LOTCA(all P>0.05).In the groups of different length of time with illness,apart from orientation(F=2.982,P=0.039)and attention(F=3.485,P=0.022),the score of other subtests and the total score of LOTCA were not different(all P>0.05).In the groups of different degree of aphasia,apart from attention(F=2.061,P=0.074),both the score of other subtests and the total score of LOTCA differed(all P<0.05).Conclusion LOTCA might be suitable to assessing the cognitive ability of post-stroke Chinese patients with aphasia. | Zeng-zhi Yu Shu-jun Jiang Jun Li Sheng Bi Fei Li Tao Xie Rui Wang Xiao-tan Zhang | 2013 | Chinese Medical Sciences Journal2013,28,3: | 9 |
| 7 | Mitochondrial Modulation of Apoptosis Induced by Low-dose Radiation in Mouse Testicular Cells显示文摘Objective To investigate whether apoptosis induced by low-dose radiation(LDR) is regulated by mitochondrial pathways in testicular cells.Methods Male mice were exposed to whole-body LDR,and changes in mitochondrial function and in expression of apoptotic factors were analyzed in the testicular cells as follows.Total nitric-oxide synthase(T-NOS) and Na+/K+ATPase activities were biochemically assayed.Reactive oxygen species(ROS) and mitochondrial membrane potential(Δψm) were determined by flow cytometry using fluorescent probes.Levels of mRNAs encoding cytochrome c(Cyt c) and apoptosis-inducing factor(AIF) were quantified by real-time reverse-transcription PCR(RT-PCR).Expression of Cyt c,AIF,caspase-9,and caspase-3 at the proteinlevel was assessed by western blotting and immunohistochemistry.Results LDR induced an increase in T-NOS activity and ROS levels,and a decrease in Na+/K+ATPase activity and mitochondrial Δψm,in the testicular cells.The intensity of these effects increased with time after irradiation and with dose.The cells showed remarkable swelling and vacuolization of mitochondria,and displayed a time- and dose-dependent increase in the expression of Cyt c,AIF,procaspase-9,and procaspase-3.Activation of the two procaspases was confirmed by detection of the cleaved caspases.The changes in expression of the four apoptotic factors were mostly limited to spermatogonia and spermatocytes.Conclusion LDR can induce testicular cell apoptosis through mitochondrial signaling pathways. | FANG Fang GONG Ping Sheng ZHAO Hong Guang BI Yu Jing ZHAO Gang GONG Shou Liang WANG Zhi Cheng | 2013 | Biomedical and Environmental Sciences2013,26,10: | 9 |
| 8 | Blood Pressure Associated with Arsenic Methylation and Arsenic Metabolism Caused by Chronic Exposure to Arsenic in Tube Well Water显示文摘Objective The effects of arsenic exposure from drinking water, arsenic metabolism, and arsenic methylation on blood pressure(BP) were observed in this study. Methods The BP and arsenic species of 560 participants were determined. Logistic regression analysis was applied to estimate the odds ratios of BP associated with arsenic metabolites and arsenic methylation capability. Results BP was positively associated with cumulative arsenic exposure(CAE). Subjects with abnormal diastolic blood pressure(DBP), systolic blood pressure(SBP), and pulse pressure(PP) usually had higher urinary i As(inorganic arsenic), MMA(monomethylated arsenic), DMA(dimethylated arsenic), and TAs(total arsenic) than subjects with normal DBP, SBP, and PP. The i As%, MMA%, and DMA% differed slightly between subjects with abnormal BP and those with normal BP. The PMI and SMI were slightly higher in subjects with abnormal PP than in those with normal PP. Conclusion Our findings suggest that higher CAE may elevate BP. Males may have a higher risk of abnormal DBP, whereas females have a higher risk of abnormal SBP and PP. | 11 | Deletion analysis of SMN1 and NAIP genes in southern Chinese children with spinal muscular atrophy显示文摘Spinal muscular atrophy (SMA) is a disorder characterized by degeneration of lower motor neurons and occasionally bulbar motor neurons leading to progressive limb and trunk paralysis as well as muscular atrophy. Three types of SMA are rec- ognized depending on the age of onset, the maximum muscular activity achieved, and survivorship: SMA1, SMA2, and SMA3. The survival of motor neuron (SMN) gene has been identified as an SMA determining gene, whereas the neuronal apoptosis inhibitory protein (NAIP) gene is considered to be a modifying factor of the severity of SMA. The main objective of this study was to analyze the deletion of SMN1 and NAIP genes in southern Chinese children with SMA. Here, polymerase chain reaction (PCR) combined with restriction fragment length polymorphism (RFLP) was performed to detect the deletion of both exon 7 and exon 8 of SMN1 and exon 5 of NAIP in 62 southern Chinese children with strongly suspected clinical symptoms of SMA. All the 32 SMA1 patients and 76% (13/17) of SMA2 patients showed homozygous deletions for exon 7 and exon 8, and all the 13 SMA3 patients showed single deletion of SMN1 exon 7 along with 24% (4/17) of SMA2 patients. Eleven out of 32 (34%) SMA1 patients showed NAIP deletion, and none of SMA2 and SMA3 patients was found to have NAIP deletion. The findings of homozygous deletions of exon 7 and/or exon 8 of SMN1 gene confirmed the diagnosis of SMA, and suggested that the deletion of SMN1 exon 7 is a major cause of SMA in southern Chinese children, and that the NAIP gene may be a modifying factor for disease severity of SMA1. | 15 | Biodegradation behavior of magnesium and ZK60 alloy in artificial urine and rat models显示文摘In this work, the biodegradable and histocompatibility properties of pure Mg and ZK60 alloy wereinvestigated as new temporary implants for urinary applications. The corrosion mechanism in artificialurine was proposed using electrochemical impedance spectroscopy and potentiodynamic polarizationtests. The corrosion potential of pure magnesium and ZK60 alloy were -1820 and -1561 mV, respectively,and the corrosion current densities were 59.66 ± 6.41 and 41.94 ± 0.53 μA cm^-2, respectively. Thein vitro degradation rates for pure Mg and ZK60 alloy in artificial urine were 0.382 and 1.023 mm/y,respectively, determined from immersion tests. The ZK60 alloy degraded faster than the pure Mg in bothartificial urine and in rat bladders (the implants of both samples are ø 3 mm × 5 mm). Histocompatibilityevaluations showed good histocompatibility for the pure Mg and ZK60 alloy during the 3 weeks postimplantationin rat bladders, and no harm was observed in the bladder, liver and kidney tissues. | 17 | Expression and Purification of Recombinant Hepatitis Delta Virus(HDV) Antigen for Use in a Diagnostic ELISA for HDV Infection Using the High-Density Fermentation Strategy in Escherichia coli显示文摘Objective Hepatitis Delt a Virus(HDV) antigen is widely used as a capture antigen in ELISAs for the identification of HDV infection; large amounts of recombinant HDV antigen with active antigenicity are required for this purpose. Methods Reconstruct the gene of HDV antigen based on the bias code of Escherichia coli, the recombinant protein expresses by high-density fermentation with fed-batch feeding strategy, and purify by immobilized metal chromatography. The sensitivity and specificity of this antigen detect by ELISA method. Results The expression of HDV antigen can reach 20% of the total cell mass in the soluble form. The recombinant HDV antigen can be conveniently purified(98%) by immobilized metal ion affinity chromatography(IMAC) using the interaction between a His-tag and nickel ions. Production of recombinant HDV antigen can reach 0.5 g/L under conditions of high-density cell fermentation. Applied to the diagnostic ELISA method, the recombinant HDV antigen shows excellent sensitivity(97% for IgM and 100% for IgG) and specificity(100% for IgG and IgM) for the detection of anti-HDV antibodies. Conclusion Expression and purification the recombinant HDV antigen as a candidate protein for application in a diagnostic ELISA for HDV infection. | 19 | Estimation of tropical cyclone parameters and wind fields from SAR images显示文摘The traditional method of Synthetic Aperture Radar(SAR)wind field retrieval is based on an empirical relation between the near surface winds and the normalized radar backscatter cross section to estimate wind speeds,where this relation is called the geophysical model function(GMF).However,the accuracy rapidly decreases due to the impact of rainfall on the measurement of SAR and the saturation of backscattered intensity under the condition of tropical cyclone.Because of no available instrument synchronously monitoring rain rate on the satellite platform of SAR,we have to derive the precipitation of the SAR observation time from non-simultaneous passive microwave observations of rain in combination with geostationary IR images,and then use the model of rain correction to remove the impact of rain on SAR wind field measurements.For the saturation of radar backscatter cross section in high wind speed conditions,we develop an approach to estimate tropical cyclone parameters and wind fields based on the improved Holland model and the SAR image features of tropical cyclone.To retrieve the low-to-moderate wind speed,the wind direction of tropical cyclone is estimated from the SAR image using wavelet analysis.And then the maximum wind speed and the central pressure of tropical cyclone are calculated by a least square minimization of the difference between the improved Holland model and the low-to-moderate wind speed retrieved from SAR.In addition,wind fields are estimated from the improved Holland model using the above-mentioned parameters of tropical cyclone as input.To evaluate the accuracy of our approach,the SAR images of typhoon Aere,typhoon Khanun,and hurricane Ophelia are used to estimate tropical cyclone parameters and wind fields,which are compared with the best track data and reanalyzed wind fields of the Joint Typhoon Warning Center(JTWC)and the Hurricane Research Division(HRD).The results indicate that the tropical cyclone center,maximum wind speed,and central pressure are generally consistent with the best track data,and wind fields agree well with reanalyzed data from HRD. | ZHOU Xuan YANG XiaoFeng LI ZiWei YU Yang BI HaiBo MA Sheng LI XiaoFeng | 2013 | Science China Earth Sciences2013,56,11: | 4 |
| 20 | Hepatitis B Immunoprophylactic Failure and Characteristics of the Hepatitis B Virus Gene in Mother-Infant Pairs in Parts of China显示文摘Objective To determine the hepatitis B immunoprophylactic failure rate in infants born to hepatitis B virus(HBV) infected mothers and to characterize HBV genes. Methods HBV-serological testing was conducted for pregnant women and infants. The complete genomes of 30 HBV isolates were sequenced, and genetic characteristics were analyzed using MEGA 5 software. Results The immunoprophylactic failure rate for infants who had completed the scheduled hepatitis B vaccination program was 5.76%(32/556). High sequence homology(99.8%-100%) was observed in 8 of the 10 mother-infant pairs. We identified 19 subgenotype C2 strains, 9 subgenotype B2 strains, and 2 subgenotype C1 strains. Three serotypes were detected: adr(19/30), adw(9/30), and ayw(2/30). The frequency of amino acid mutation of the ‘a' determinant region was 16.67%(5/30), including that of Q129 H, F134 Y, S136 Y, and G145 E. We detected 67 amino acid mutations in the basal core promoter, precore, and core regions of the genome. Conclusion The immunoprophylactic failure rate in infants born to HBV-infected mothers is low in the regions of China examined during this study.
