| 1 | Development of Mining Technology and Equipment for Seafloor Massive Sulfide Deposits显示文摘Seafloor massive sulfide(SMS) deposits which consist of Au, Ag, Cu, and other metal elements, have been a target of commercial mining in recent decades. The demand for established and reliable commercial mining system for SMS deposits is increasing within the marine mining industry. The current status and progress of mining technology and equipment for SMS deposits are introduced. First, the mining technology and other recent developments of SMS deposits are comprehensively explained and analyzed.The seafloor production tools manufactured by Nautilus Minerals and similar mining tools from Japan for SMS deposits are compared and discussed in turn. Second, SMS deposit mining technology research being conducted in China is described, and a new SMS deposits mining tool is designed according to the environmental requirement. Finally, some new trends of mining technology of SMS deposits are summarized and analyzed. | 2 | Comparative studies on histological and ultra-structure of the pituitary of different ploidy level fishes显示文摘The histological and ultra-structure of the pituitary in diploid red crucian carp(Carassius auratus red var.),triploid crucian carp and allotetraploid hybrids within and after the breeding season were comparatively studied.The result showed that there were six endocrine cell types in the pituitary of these three kinds of fishes,and there was an obvious difference in cell size among different ploidy level fishes.As for the same type of pituitary cells,the cell size was increased gradually with the in- creasing ploidy level.In the breeding season,the allotetraploid hybrids had higher proportion of go- nadotropin cells(GTH)than triploids,and the triploids had higher proportion of GTH than diploids.The results were related to the earlier sexual maturity of allotetraploid hybrids and sterility of triploid cru- cian carp.On the other hand,among the three kinds of fishes,the proportion of somatotropin(STH) cells in triploids crucian carp was the highest,whereas that in allotetraploid hybrids was the lowest. | 3 | The cloning of Dmc1 cDNAs and a comparative study of its expression in different ploidy cyprinid fishes显示文摘Dmc1 (disrupted meiotic cDNA) is a functionally specific gene, which was firstly discovered in yeast and then found to encode a protein required for homologous chromosome synapsis during the process of meiosis. In this investigation, we cloned the partial cDNAs of Dmc1 of diploid red crucian carp, Japanese crucian carp, common carp, triploid crucian carp and allotetraploid hybrids by using a pair of degenerate primers based on the conservative sequence of amino acids of the DMC1 protein in yeast, mouse and human. The full length cDNAs were then obtained by rapid amplification of cDNA ends (RACE). Our data showed that the full length cDNAs of Dmc1 in the three diploid fishes are all 1375 bp long, while it is 1383 bp long in triploids and 1379 bp long in allotetraploids. And despite of the variation in length, all the cDNAs encode a protein of 342 amino acids. A high homology of 97.3% of the DMC1 protein can be drawn by comparing the amino acid sequences in the three diploids, which is also of 86%, 86% and 95% similarity to human, mouse and zebrafish, respectively. A comparative study of the expression pattern of Dmc1 was carried out by RT-PCR using specific primers against the same se-quences of coding regions in different ploidy cyprinid fishes, from which it was showed that Dmc1 was expressed only in gonads of these five kinds of fishes. The expression pattern of Dmc1 in both ovaries and testes from different ploidy fishes within breeding season was also studied by Real-time PCR, and the results showed that the expression of this gene was greatly different among the three different ploidy fishes, which was the highest of triploid and lowest of allotetraploids. The histological sections data showed matured gonads of both diploid red crucian carp and allotetraploids in breeding season, although the latter demonstrated a higher maturation, and no gonadal maturation could be observed in triploids. In conclusion, we suggest that Dmc1 is specifically expressed in the period of meiosis in all the ploidy cyprinid fishes and directly related with the development of gonad in a manner of ploidy-independent way. | 4 | Rapid growth and sterility of growth hormone gene transgenic triploid carp显示文摘Triploid carp(100%)with 150(3n=150)chromosomes were obtained by crossing the females of improved tetraploid hybrids(♀, 4n=200)of red crucian carp(♀)×common carp(♂)with the males of diploid yellow river carp(♂,2n=100).The crosses yielded transgenic triploid carp(positive triploid fish,44.2%of the progeny)and non-transgenic triploid carp(negative triploid fish). | 5 | Studies on the evolution process of rockbursts in deep tunnels显示文摘This paper focuses on the evolution processes of different types of rockbursts occurring in deep tunnels. A series of laboratory tests and in-situ monitoring in deep tunnels excavated by tunnel boring machine (TBM) and drill-and-blast (D&B) method have been conducted to understand the mechanisms and processes of the evolution of different types of rockbursts, including strain rockburst, strain-structure slip rockburst, immediate rockburst and time-delayed rockburst. Three different risk assessment methods are proposed to evaluate the intensity and potential failure depth of rockbursts. These methods can be applied before excavation and the results can be updated according to the real-time information during excavation. Two micro-seismicity based real-time warning systems have been established for predicting various intensities of rockbursts, such as slight, moderate, intensive and extremely intensive rockbursts. Meanwhile, the probability and intensity of the rockburst are also given. The strategy for excavation and support design has been suggested for various intensities of rockbursts before excavation. The strategy for dynamic control of the rockburst evolution process is also proposed according to the monitoring results. The methodology has been successfully applied to rockburst risk reduction for deep tunnels at Jinping II hydropower project. | 7 | Toxicity detection of sodium nitrite,borax and aluminum potassium sulfate using electrochemical method显示文摘Based on the inhibition effect on the respiratory chain activity of microorganisms by toxicants,an electrochemical method has been developed to measure the current variation of a mediator in the presence of microorganisms contacted with a toxicant.Microelectrode arrays were adopted in this study,which can accelerate the mass transfer rate of an analyte to the electrode and also increase the total current signal,resulting in an improvement in detection sensitivity.We selected Escherichia coli as the testee and the standard glucoseglutamic acid as an exogenous material.Under oxygen restriction,the experiments in the presence of toxicant were performed at optimum conditions(solution pH 7.0,37°C and reaction for 3 hr).The resulting solution was then separated from the suspended microorganisms and was measured by an electrochemical method,using ferricyanide as a mediator.The current signal obtained represents the reoxidation of ferrocyanide,which was transformed to inhibiting efficiency,IC 50,as a quantitative measure of toxicity.The IC 50 values measured were 410,570 and 830mg/L for sodium nitrite,borax and aluminum potassium sulfate,respectively.The results show that the toxicity sequence for these three food additives is consistent with the value reported by other methods.Furthermore,the order of damage degree to the microorganism was also observed to be:sodium nitrite > borax > aluminum potassium sulfate > blank,according to the atomic force microscopy images of E.coli after being incubated for 3 hr with the toxic compound in buffer solutions.The electrochemical method is expected to be a sensitive and simple alternative to toxicity screening for chemical food additives. | Dengbin Yu Daming Yong Shaojun Dong | 2013 | Journal of Environmental Sciences2013,25,4: | 9 |
| 8 | Authigenic sulfide minerals and their sulfur isotopes in sediments of the northern continental slope of the South China Sea and their implications for methane flux and gas hydrate formation显示文摘This is a report of the study of the authigenic sulfide minerals and their sulfur isotopes in a sediment core (NH-1) collected on the northern continental slope of the South China Sea, where other geo-physical and geochemical evidence seems to suggest gas hydrate formation in the sediments. The study has led to the findings: (1) the pyrite content in sediments was relatively high and its grain size relatively large compared with that in normal pelagic or hemipelagic sediments; (2) the shallowest depth of the acid volatile sulfide (AVS) content maximum was at 437.5 cm (>2 μmol/g), which was deeper than that of the authigenic pyrite content maximum (at 141.5-380.5 cm); (3) δ 34S of authigenic pyrite was positive (maximum: +15‰) at depth interval of 250-380 cm; (4) the positive δ 34S coincided with pyrite enrichment. Compared with the results obtained from the Black Sea sediments by Jorgen-sen and coworkers, these observations indicated that at the NH-1 site, the depth of the sulfate-methane interface (SMI) would be or once was at about 437.5-547.5 cm and the relatively shallow SMI depth suggested high upward methane fluxes. This was in good agreement with the results obtained from pore water sulfate gradients and core head-space methane concentrations in sediment cores collected in the area. | 13 | Evaluating the emission status of light-duty gasoline vehicles and motorcycles in Macao with real-world remote sensing measurement显示文摘Roadside remote sensing measurement was used to explore the real-world emission status of light duty gasoline vehicles(LDGVs) and motorcycles in Macao. Both fuel-based and distance-based emission factors were derived using the mass balance method. The emission concentration profile of LDGVs illustrated the benefits of tightening emission standards at the source country or region of import. The distance-based emission factors for CO, HC and NOx of LDGVs registered before 2000 were 8.00, 1.04 and 1.36 g/km,respectively. The distance-based emission factors for CO, HC and NOx of LDGVs registered in or after 2000 were 1.16, 0.15 and 0.18 g/km, respectively. The fuel-based CO emission factors of light duty motorcycles(LDMCs) and heavy duty motorcycles(HDMCs) registered before 2000 were about 10 times higher than those of LDGVs of the same age group. As the emissions of LDGVs decreased more quickly after 2000, the gap widens for newer vehicles.The distance-based HC emission factors of LDMCs and HDMCs registered before 2000 were4.81 and 2.91 g/km, respectively. The distance-based HC emission factors of LDMCs and HDMCs registered in or after 2000 were 3.52 and 0.93 g/km, respectively. The poor emission performance of motorcycles and their larger share in the traffic flow will cause them to be the major contributor to traffic CO and HC emissions. | 17 | Efficacy of Cigu Xiaozhi pill(慈菇消脂丸) on non-alcoholic steatohepatitis-associated lipoapoptosis through stress-activated c-Jun N-terminal kinase signalling pathway显示文摘OBJECTIVE: To investigate the efficacy of Cigu Xiaozhi pill(慈菇消脂丸, CGXZ) on non-alcoholic steatohepatitis(NASH)-associated lipoapoptosis through the stress-activated c-Jun N-terminal kinase(JNK)/stress-activated protein kinase signalling pathway.METHODS: Sixty male Sprague-Dawley rats were randomly divided into the following groups(10 rats each): blank control, model, low-dose CGXZ,medium-dose CGXZ, high-dose CGXZ, and positive control(treated with SP600125, a JNK inhibitor).The NASH model was established and the histomor-phological characteristics of haematoxylin and eosin-stained liver tissues were examined under a light microscope. Cell apoptosis in liver tissues was assessed via terminal deoxynucleotidyl transferase d UTP nick-end labelling assay. In addition, the m RNA and protein expression levels of p-JNK,p-c-Jun, caspase-8, Fas, and Fas-L were determined via fluorescence-based quantitative real-time PCR,immunohistochemical and Western blot assays.RESULTS: Histopathological examination of the liver showed that the model rats had moderate-to-severe steatosis with infiltration of inflammatory cells as well as significantly higher expression levels of the p-JNK, p-c-Jun, caspase-8, Fas, and Fas-L proteins, compared with those in the blank control group(P < 0.01). Hepatic lobules of the rats in the treatment groups showed significantly reduced vacuolar degeneration and steatosis as well as alleviated inflammatory cell infiltration.
