1Drought Stress and Preharvest Aflatoxin Contamination in Agricultural Commodity: Genetics, Genomics and Proteomics显示文摘Throughout the world, aflatoxin contamination is considered one of the most serious food safety issues concerning health. Chronic problems with preharvest aflatoxin contamination occur in the southern US, and are particularly troublesome in corn, peanut, cottonseed, and tree nuts. Drought stress is a major factor to contribute to preharvest aflatoxin contamination. Recent studies have demonstrated higher concentration of defense or stress-related proteins in corn kernels of resistant genotypes compared with susceptible genotypes, suggesting that preharvest field condition (drought or not drought) influences gene expression differently in different genotypes resulting in different levels of 'end products': PR(pathogenesis-related) proteins in the mature kernels. Because of the complexity of Aspergillus-plant interactions, better understanding of the mechanisms of genetic resistance will be needed using genomics and proteomics for crop improvement. Genetic improvement of crop resistance to drought stress is one component and will provide a good perspective on the efficacy of control strategy. Proteomic comparisons of corn kernel proteins between resistant or susceptible genotypes to Aspergillus flavus infection have identified stress-related proteins along with antifungal proteins as associated with kernel resistance. Gene expression studies in developing corn kernels are in agreement with the proteomic studies that defense-related genes could be upregulated or downregulated by abiotic stresses.Baozhu Guo Zhi-Yuan Chen R. 2Monitoring the Expression of Maize Genes in Developing Kernels under Droug ht Stress usingOligo-microarray显示文摘Preharvest aflatoxin contamination of grain grown on the US southeastern Coast Plain is provoked and aggravated by abiotic stress. The primary abiotic stress is drought along with high temperatures. The objectives of the present study were to monitor gene expression in developing kernels in response to drought stress and to identify drought-responsive genes for possible use in germplasm assessment. The maize breeding line Tex6 was used, and gene expression profiles were analyzed in developing kernels under drought stress verses well-watered conditions at the stages of 25, 30, 35, 40, 45 d after pollination (DAP) using the 70 mer maize oligo-arrays. A total of 9 573 positive array spots were detected with unique gene IDs, and 7 988 were common in both stressed and well-watered samples. Expression patterns of some genes in several stress response-associated pathways, including abscisic acid, jasmonic acid and phenylalanine ammonia-lyase, were examined, and these specific genes were responsive to drought stress positively. Real-time quantitative polymerase chain reaction validated microarray expression data. The comparison between Tex6 and B73 revealed that there were significant differences in specific gene expression, patterns and levels. 6Characterization of novel Brown midrib 6 mutations affecting lignin biosynthesis in sorghum显示文摘The presence of lignin reduces the quality of lignocellulosic biomass for forage materials and feedstock for biofuels. In C4 grasses,the brown midrib phenotype has been linked to mutations to genes in the monolignol biosynthesis pathway. For example,the Bmr6 gene in sorghum(Sorghum bicolor) has been previously shown to encode cinnamyl alcohol dehydrogenase(CAD),which catalyzes the final step of the monolignol biosynthesis pathway. Mutations in this gene have been shown to reduce the abundance of lignin,enhance digestibility,and improve saccharification efficiencies and ethanol yields. Nine sorghum lines harboring five different bmr6 alleles were identified in an EMS-mutagenized TILLING population. DNA sequencing of Bmr6 revealed that the majority of the mutations impacted evolutionarily conserved amino acids while three-dimensional structural modeling predicted that all of these alleles interfered with the enzyme's ability to bind with its NADPH cofactor. All of the new alleles reduced in vitro CAD activity levels and enhanced glucose yields following saccharification. Further,many of these lines were associated with higher reductions in acid detergent lignin compared to lines harboring the previously characterized bmr6-ref allele. 7Utility of current thrombophilia screening in young patients with stroke and TIA显示文摘Introduction Approximately 40%of strokes in young adults are cryptogenic.The diagnostic yield of thrombophilia screening remains controversial.We aimed to determine utility of current thrombophilia testing for young patients with stroke and transient ischaemic attack(TIA).Methods We present a retrospective review of all patients with stroke and TIA≤60 years presenting to University College London Hospital stroke unit and daily TIA clinic from 1 January 2015 to 1 August 2016.Consecutive clinical records and thrombophilia tests,including factor V Leiden(FVL),prothrombin G20210A mutation(PGM),antiphospholipid antibody(APA),and protein S,C and antithrombin(AT)levels,were reviewed.results The mean age of 628 patients with stroke and TIA was 49.1 years(SD 9.2).Thrombophilia testing was performed in 360(57%)patients,including 171 with stroke and 189 with TIA.Positive tests were found in 50(14%)patients,of whom 24 patients were<50 years.Positive results were found in 36(10%)with acute ischaemic stroke,4(1%)with haemorrhagic stroke and 10(3%)with TIA.Thirteen patients(4%)had homozygous/heterozygous FVL or PGM,and 27(7.5%)had positive APA(anticardiolipin antibody,anti-β2 glycoprotein antibody or lupus anticoagulant).Of 27(7.5%)patients with protein C,S or AT deficiency,10(2.8%)had primary deficiency,presumed hereditary with other secondary causes excluded.9%of patients with protein C,S or AT and 27%with APA were followed by confirmatory testing.Conclusion Thrombophilia testing was positive in only 14%of cases overall.Thrombophilia mutations and protein C,S or AT abnormalities were found rarely and were very uncommon in patients with TIA.Follow-up of abnormal results was generally poor for all groups,which further limited the impact of the thrombophilia testing policy.Vafa Alakbarzade Alice Taylor Marie Scully Robert Simister Arvind Chandratheva 2018Stroke & Vascular Neurology2018,3,4:3
