1Chemokine expression in hepatocellular carcinoma versus colorectal liver metastases显示文摘AIM: To evaluate and compare the expression profiles of CXCL12 (SDF-1), CCL19 (MIP-3β), CCL20 (MIP-3α) and CCL21 (6Ckine, Exodus2) and their receptors on RNA and protein levels in hepatocellular carcinoma (HCC) versus colorectal liver metastases (CRLM) and to elucidate their impact on the carcinogenesis and progression of malignant liver diseases. METHODS: Chemokine expression was analyzed by RT-PCR and ELISA in 11 cases of HCC specimens and in 23 cases of CRLM and corresponding adjacent non- tumorous liver tissues, respectively. Expressions of their receptors CXCR4, CCR6 and CCR7 were analyzed by RT- PCR and Western blot analysis in the same cases of HCC and CRLM. RESULTS: Significant up-regulation for CCL20/CCR6 was detected in both cancer types. Moreover, CCL20 demonstrated significant overexpression in CRLM in relation to the HCC tissues. Being significantly up- regulated only in CRLM, CXCR4 displayed an aberrant expression pattern with respect to the HCC tissues. CONCLUSION: Correlation of CXCR4 expression with CRLM suggests CXCR4 as a potential predictive factor for CRLM. 21961-2010年黄河流域蒸发皿蒸发量变化及影响因子分析显示文摘利用黄河流域内61个气象站逐月观测资料,使用Mann-Kendall非参数检验方法对1961—2010年黄河流域小型蒸发皿蒸发量变化趋势进行了分析,并用SVD和多元回归方法检测影响蒸发量变化的因子。结果表明,黄河流域年蒸发皿蒸发量在1961—2010年显著下降,四季中夏季的下降趋势最显著,年和春、夏季蒸发量均在1979年发生突变。上、中、下游的年蒸发量变化率分别为-2.38 mm/a、-2.35 mm/a、-8.35 mm/a,下游下降幅度较大。空间变化分布上,年和春、夏季蒸发皿蒸发量均在黄河流域河源地区、河套地区西部及北部、河南北部有显著下降趋势,在河套地区东部呈显著上升趋势。利用SVD分析发现蒸发量的空间变化与不同因子作用有着显著关联,通过对不同区域内各影响因子的多元回归分析发现流域内蒸发量上升的地区主要是由气温上升所引起,而下降的地区则与风速减小有关。柳春 王守荣 梁有叶 Ruby L.Leung 2013气候变化研究进展2013,9,5:19

4黄河流域未来气候-水文变化的模拟研究显示文摘将大尺度半分布式水文模型VIC应用到黄河上中游流域(花园口水文断面以上),并利用区域气候模式RegCM4.0单向嵌套全球气候模式BCC_CSM1.1,动力降尺度到黄河流域的模拟结果驱动VIC模型,开展在新的典型浓度路径下(RCP4.5和RCP8.5)黄河流域未来气候和水文变化的离线模拟。模拟结果显示,在RCP4.5和RCP8.5排放情景下,黄河流域21世纪平均地表气温相对于1971—2000年均呈显著上升趋势,2019—2048年上升1.2—1.5℃,2069—2098年上升2.19—3.9℃。未来年平均降水量有微弱的增大,2019—2048年增幅为6%左右,2069—2098年增幅为1.4%—5.6%。未来蒸发量增大明显,2069—2098年年平均蒸发量最大可增加9.6%。2019—2048年花园口水文站的年平均径流量增大3.4%—7.4%,2069—2098年年平均径流量转为减少,减幅为3.3%—5.3%。黄河上游地区未来气候和水文变化趋势与黄河流域基本一致,但未来年径流量变幅低于黄河流域,相对比较稳定。康丽莉 LEUNG L Ruby 柳春 王守荣 2015气象学报2015,73,2:13
5Correlation of IL-8 with induction, progression and metastatic potential of colorectal cancer显示文摘AIM: To investigate the expression profile of IL-8 in inflammatory and malignant colorectal diseases to evaluate its potential role in the regulation of colorectal cancer (CRC) and the development of colorectal liver metastases (CRLM). METHODS: IL-8 expression was assessed by quantitative real-time PCR (Q-RT-PCR) and the enzyme-linked immunosorbent assay (ELISA) in resected specimens from patients with ulcerative colitis (UC, n = 6) colorectal adenomas (CRA, n = 8), different stages of colorectal cancer (n = 48) as well as synchronous and metachronous CRLM along with their corresponding primary colorectal tumors (n = 16). RESULTS: IL-8 mRNA and protein expression was significantly up-regulated in all pathological colorectal entities investigated compared with the corresponding neighboring tissues. However, in the CRC specimens IL-8 revealed a significantly more pronounced overexpression in relation to the CRA and UC tissues with an average 30-fold IL-8 protein up-regulation in the CRC specimens in comparison to the CRA tissues. Moreover, IL-8 expression revealed a close correlation with tumor grading. Most interestingly, IL-8 up-regulation was most enhanced in synchronous and metachronous CRLM, if compared with the corresponding primary CRC tissues. Herein, an up to 80-fold IL-8 overexpression in individual metachronous metastases compared to normal tumor neighbor tissues was found. 9chemokine/chemokine receptor pair CC L20/CC R6 in humancolorectal malignancy:An overview显示文摘Chemokines belong to a superfamily of small, cytokinelike proteins, which induce multiple physiological functions, particularly cytoskeletal rearrangement and compartment-specific migration through their interaction with G-protein-coupled receptors. Chemokines and their receptors have been widely acknowledged as essential and selective mediators in leukocyte migration in inflammatory response. It is now established that the chemokine/chemokine receptor system is also used by cancer cells to direct lymphatic and haematogenous spreading and additionally has an impact on the site of metastatic growth of different tumours. In recent years an increasing number of studies have drawn attention to CC-chemokine cysteine motif chemokine ligand 20(CCL20) and its physiological sole receptor CCR6 to play a role in the onset, development and metastatic spread of various gastrointestinal cancer entities. Among various cancer types CCR6 was also demonstrated to be significantly overexpressed in colorectal cancer(CRC) and stimulation by its physiological ligand CCL20 has been reported to promote CRC cell proliferation and migration in vitro. Further, the CCL20/CCR6 system apparently plays a role in the organ-selective liver metastasis of CRC. 10Effect of preoperative FOLFOX chemotherapy on CCL20/CCR6 expression in colorectal liver metastases显示文摘AIM:To evaluate the influence of preoperative FOLFOX chemotherapy on CCL20/CCR6 expression in liver metastases of stage Ⅳ colorectal cancer(CRC) patients.METHODS:Using Real Time-PCR,enzyme-linked immunosorbent assay,Western Blots and immunohistochemistry,we have analyzed the expression of CCL20,CCR6 and proliferation marker Ki-67 in colorectal liver metastasis(CRLM) specimens from stage Ⅳ CRC patients who received preoperative FOLFOX chemotherapy(n = 53) and in patients who did not receive FOLFOX chemotherapy prior to liver surgery(n = 29).RESULTS:Of the 53 patients who received FOLFOX,time to liver surgery was ≤ 1 mo in 14 patients,≤ 1 year in 22 patients and > 1 year in 17 patients,respectively.In addition,we investigated the proliferation rate of CRC cells in liver metastases in the different patient groups.Both CCL20 and CCR6 mRNA and protein expression levels were significantly increased in patients who received preoperative FOLFOX chemotherapy ≤ 12 mo before liver surgery(P < 0.001) in comparison to patients who did not undergo FOLFOX treatment.Further,proliferation of CRLM cells as measured by Ki-67 was increased in patients who underwent FOLFOX treatment.CCL20 and CCR6 expression levels were significantly increased in CRLM patients who had undergone preoperative FOLFOX chemotherapy.CONCLUSION:This chemokine/receptor up-regulation could lead to increased proliferation/migration through an autocrine mechanism which might be used by surviving metastatic cells to escape cell death caused by FOLFOX.Claudia Rubie Vilma Oliveira Frick Pirus Ghadjar Mathias Wagner Christoph Justinger Stefan Graeber Jens Sperling Otto Kollmar Martin K Schilling 2011World Journal of Gastroenterology2011,17,26:5

13Projected changes in mean and interannual variability of surface water over continental China显示文摘Five General Circulation Model(GCM) climate projections under the RCP8.5 emission scenario were used to drive the Variable Infiltration Capacity(VIC) hydrologic model to investigate the impacts of climate change on hydrologic cycle over continental China in the 21 st century. The bias-corrected climatic variables were generated for the Fifth Assessment Report of the Intergovernmental Panel on Climate Change(IPCC AR5) by the Inter-Sectoral Impact Model Intercomparison Project(ISIMIP). Results showed much larger fractional changes of annual mean Evapotranspiration(ET) per unit warming than the corresponding fractional changes of Precipitation(P) per unit warming across the country, especially for South China, which led to a notable decrease of surface water variability(P-E). Specifically, negative trends for annual mean runoff up to -0.33%/ year and soil moisture trends varying between -0.02% to -0.13%/year were found for most river basins across China. Coincidentally, interannual variability for both runoff and soil moisture exhibited significant positive trends for almost all river basins across China, implying an increase in extremes relative to the mean conditions. Noticeably, the largest positive trends for runoff variability and soil moisture variability, which were up to 0.41%/year and 0.90%/year, both occurred in Southwest China. In addition to the regional contrast, intra-seasonal variation was also large for the runoff mean and runoff variability changes, but small for the soil moisture mean and variability changes. Our results suggest that future climate change could further exacerbate existing water-related risks(e.g., floods and droughts) across China as indicated by the marked decrease of surface water amounts combined with a steady increase of interannual variability throughout the 21 st century. 14Loss of the vitamin D receptor in human breast and prostate cancers strongly induces cell apoptosis through downregulation of Wnt/β-catenin signaling显示文摘Vitamin D co-regulates cell proliferation, differentiation and apoptosis in numerous tissues, including cancers. The known anti-proliferative and pro-apoptotic actions of the active metabolite of vitamin D,1,25-dihydroxy-vitamin D [1,25(OH)2 D] are mediated through binding to the vitamin D receptor(VDR). Here,we report on the unexpected finding that stable knockdown of VDR expression in the human breast and prostate cancer cell lines, MDA-MB-231 and PC3, strongly induces cell apoptosis and inhibits cell proliferation in vitro. Implantation of these VDR knockdown cells into the mammary fat pad(MDA-MB-231),subcutaneously(PC3) or intra-tibially(both cell lines) in immune-incompetent nude mice resulted in reduced tumor growth associated with increased apoptosis and reduced cell proliferation compared with controls.These growth-retarding effects of VDR knockdown occur in the presence and absence of vitamin D and are independent of whether cells were grown in bone or soft tissues. Transcriptome analysis of VDR knockdown and non-target control cell lines demonstrated that loss of the VDR was associated with significant attenuation in the Wnt/β-catenin signaling pathway. In particular, cytoplasmic and nuclear β-catenin protein levels were reduced with a corresponding downregulation of downstream genes such as Axin2, Cyclin D1,interleukin-6(IL-6), and IL-8. Stabilization of β-catenin using the GSK-3β inhibitor BIO partly reversed the growth-retarding effects of VDR knockdown. Our results indicate that the unliganded VDR possesses hitherto unknown functions to promote breast and prostate cancer growth, which appear to be operational not only within but also outside the bone environment. 15Effect of Long-Term Soil Management on the Mutual Interaction Among Soil Organic Matter, Microbial Activity and Aggregate Stability in a Vineyard显示文摘Vineyard management practices to enhance soil conservation principally focus on increasing carbon(C)input,whereas mitigating impacts of disturbance through reduced tillage has been rarely considered.Furthermore,information is lacking on the effects of soil management practices adopted in the under-vine zone on soil conservation.In this work,we evaluated the long-term effects(22 years)of alley with a sown cover crop and no-tillage(S+NT),alley with a sown cover crop and tillage(S+T),and under-vine zone with no vegetation and tillage(UV)on soil organic matter(SOM),microbial activity,aggregate stability,and their mutual interactions in a California vineyard in USA.Vegetation biomass,microbial biomass and activity,organic C and nitrogen(N)pools,and SOM size fractionation and aggregate stability were analysed.Soil characteristics only partially reflected the differences in vegetation biomass input.Organic C and N pools and microbial biomass/activity in S+NT were higher than those in S+T,while the values in UV were intermediate between the other two treatments.Furthermore,S+NT also exhibited higher particulate organic matter C in soil.No differences were found in POM C between S+T and UV,but the POM fraction in S+T was characterized by fresher material.Aggregate stability was decreased in the order:S+NT>UV>S+T.Tillage,even if shallow and performed infrequently,had a negative effect on organic C and N pools and aggregate stability.Consequently,the combination of a sown cover crop and reduced tillage still limited SOM accumulation and reduced aggregate stability in the surface soil layer of vineyards,suggesting relatively lower resistance of soils to erosion compared to no-till systems.Sergio A.BELMONTE Luisella CELI Ruby J.STAHEL Eleonora BONIFACIO Vittorino NOVELLO Ermanno ZANINI Kerri L.STEENWERTH 2018Pedosphere2018,28,2:3
16The microbial content of unexpired pasteurized milk from selected supermarkets in a developing country显示文摘Objective:To determine the presence and levels of microbes in unexpired pasteurized milk from randomly selected supermarkets in Kingston,Jamaica.Methods:The quantitative study used a stratified random sampling technique in the selection of the 20 representative milk samples from six(6) supermarkets.Microbiological tests such as methylene blue reduction,standard plate count(SPC),coliform plate count(CPC),purity plate culture,gram staining and biochemical tests were performed to examine the microbes in purchased unexpired pasteurized milk.Results:One sample(BCr016) had a pH of 4.0.a rancid odour and curdled appearance.It decolourized within one hour during the methylene blue reduction test and was classified as class 4 milk.Seven of the samples were sterile with no microbe growth on the plate count agar and violet red bile salt agar(VRBA).The milk samples that appeared to be safe for consumption were all 10,11,12 and 13 days before expiration.The VRBA sample BCr016,had a colony count of 13 400 CFU/ mL.There was the presence of Escherichia coli in sample LCr021 which had a standard plate count of 1 580 SPC/mL and a coliform count of 500 CFU/mL.Enterobacter sp.was present in colonies from BCr016 and all the other milk samples.Conclusions:Unacceptable levels of Entembacter spp.and Escherichia coli were found in most of the samples.Effective measures to ensure safe milk for human consumption such as the phosphatase test and methylene blue reduction test should be routinely performed on each batch of milk processed by dairy plants.Melisa Anderson Patrice Hinds Stacyann Hurditt Princena Miller Donovan McGrowder Ruby Alexander-Lindo 2011Asian Pacific Journal of Tropical Biomedicine2011,1,3:3
