| 1 | Sonic hedgehog signaling in kidney fibrosis: a master communicator显示文摘The hedgehog signaling cascade is an evolutionarily conserved pathway that regulates multiple aspects of embryonic development and plays a decisive role in tissue homeostasis. As the best studied member of three hedgehog ligands, sonic hedgehog(Shh) is known to be associated with kidney development and tissue repair after various insults. Recent studies uncover an intrinsic link between dysregulated Shh signaling and renal fibrogenesis. In various types of chronic kidney disease(CKD), Shh is upregulated specifically in renal tubular epithelium but targets interstitial fibroblasts, thereby mediating a dynamic epithelialmesenchymal communication(EMC). Tubule-derived Shh acts as a growth factor for interstitial fibroblasts and controls a hierarchy of fibrosis-related genes, which lead to the excessive deposition of extracellular matrix in renal interstitium. In this review, we recapitulate the principle of Shh signaling, its activation and regulation in a variety of kidney diseases. We also discuss the potential mechanisms by which Shh promotes renal fibrosis and assess the efficacy of blocking this signaling in preclinical settings. | 2 | Current and future therapies for inherited cholestatic liver diseases显示文摘Familial intrahepatic cholestasis(FIC) comprises a group of rare cholestatic liver diseases associated with canalicular transport defects resulting predominantly from mutations in ATP8B1, ABCB11 and ABCB4. Phe-notypes range from benign recurrent intrahepatic cholestasis(BRIC), associated with recurrent cholestatic attacks, to progressive FIC(PFIC). Patients often suffer from severe pruritus and eventually progressive cholestasis results in liver failure. Currently, first-line treatment includes ursodeoxycholic acid in patients with ABCB4 deficiency(PFIC3) and partial biliary diversion in patients with ATP8B1 or ABCB11 deficiency(PFIC1 and PFIC2). When treatment fails, liver transplantation is needed which is associated with complications like rejection, post-transplant hepatic steatosis and recurrence of disease. Therefore, the need for more and better therapies for this group of chronic diseases remains. Here, we discuss new symptomatic treatment options like total biliary diversion, pharmacological diversion of bile acids and hepatocyte transplantation. | 5 | Invariant and energy analysis of an axially retracting beam显示文摘The mechanism of a retracting cantilevered beam has been investigated by the invariant and energy-based analysis. The time-varying parameter partial differential equation governing the transverse vibrations of a beam with retracting motion is derived based on the momentum theorem.The assumed-mode method is used to truncate the governing partial differential equation into a set of ordinary differential equations(ODEs) with time-dependent coefficients. It is found that if the order of truncation is not less than the order of the initial conditions, the assumed-mode method can yield accurate results. The energy transfers among assumed modes are discussed during retraction. The total energy varying with time has been investigated by numerical and analytical methods,and the results have good agreement with each other. | 6 | Modeling large reversible electric-field-induced strain in ferroelectric materials using 90° orientation switching显示文摘Reversible large electric-field-induced strain caused by reversible orientation switchings in BaTiO3 is modeled using the Landau's theory of phase transition. A triple well free energy function is constructed. Each of its minima is associated with one of the polarization orientations involved. Nonlinear constitu- tive laws accounting for reversible orientation switchings and electrostriction effects are obtained by using thermodynamic equilibrium conditions. Hysteretic dynamics of one-dimensional structures is described by coupled nonlinear differential equations. Double hysteretic loops in the electric and me- chanic fields are both successfully modeled. Giant reversible electrostriction is modeled as a conse-quence of reversible orientation switchings via electro-mechanical couplings. | 18 | The Appearance of Ulva laetevirens(Ulvophyceae, Chlorophyta) in the Northeast Coast of the United States of America显示文摘Introduced species may outcompete or hybridize with native species, resulting in the loss of native biodiversity or even alteration of ecosystem processes. In this study, we reported an alien distromatic Ulva species, which was found in an embayment(Holly Pond) connected with Long Island Sound, USA. The morphological and anatomical observations in combination with molecular data were used for its identification to species. Anatomy of collected specimens showed that the cell shape in rhizoidal and basal regions was round and the marginal teeth along the basal and median region were not found. These characteristics were primarily identical to the diagnostic characteristics of Ulva laetevirens Areschoug(Chlorophyta). The plastid-encoding tufA and nucleus-encoding ITS1 were used for its molecular identification. Phylogenetic analysis for the tufA gene placed the specimens from Holly Pond in a well-supported clade along with published sequences of U. laetevirens identified early without any sequence divergence. In ITS tree, the sample also formed well-supported clades with the sequences of U. laetevirens with an estimated sequence divergence among the taxa in these clades as low as 1%. These findings confirmed the morpho-anatomical conclusion. Native to Australia, this species was reported in several countries along the Mediterranean coast after the late of 1990 s.
