1Structural basis of SARS-CoV-2 3CL^pro and anti-COVID-19 drug discovery from medicinal plants显示文摘The recent pandemic of coronavirus disease 2019(COVID-19)caused by SARS-CoV-2 has raised global health concerns.The viral 3-chymotrypsin-like cysteine protease(3CL^pro)enzyme controls coronavirus replication and is essential for its life cycle.3CL^pro is a proven drug discovery target in the case of severe acute respiratory syndrome coronavirus(SARS-CoV)and Middle East respiratory syndrome coronavirus(MERS-CoV).Recent studies revealed that the genome sequence of SARS-CoV-2 is very similar to that of SARS-CoV.Therefore,herein,we analysed the 3CL^pro sequence,constructed its 3D homology model,and screened it against a medicinal plant library containing 32,297 potential anti-viral phytochemicals/traditional Chinese medicinal compounds.Our analyses revealed that the top nine hits might serve as potential anti-SARS-CoV-2 lead molecules for further optimisation and drug development process to combat COVID-19.Muhammad Tahir ul Qamar Safar MAlqahtani Mubarak AAlamri Ling-Ling Chen 2020Journal of Pharmaceutical Analysis2020,10,4:29
2游离胫后动脉皮瓣修复80例口腔颌面部软组织缺损效果评价显示文摘目的:探讨胫后动脉皮瓣游离移植修复口腔颌面部软组织缺损的临床设计及效果。方法:2017年11月-2018年8月,应用胫后动脉皮瓣修复中山大学孙逸仙纪念医院收治的80例口腔颌面部肿瘤术后缺损。术前应用计算机体层血管成像(CTA)对双侧下肢血管进行评估,明确胫后动、静脉走向及穿支数量,完成穿支体表定位。术中根据受区部位、缺损大小、形态以设计、制备皮瓣,并转移至受区进行修复。对皮瓣大小、血管蒂长度、血管直径、穿支数量、穿支长度、存活率、外形、供区和受区术后并发症等进行总结分析。结果:80例患者中,男56例,女24例;年龄24~90(58.17±1.43)岁;皮瓣大小5 cm×6 cm^7 cm×13 cm,皮瓣厚度0.30~1.00(0.53±0.2) cm,血管蒂长度8~14(10.03±1.4) cm;供区动脉直径1.50~3.00(2.36±0.4) mm,供区静脉直径1.50~4.0(2.99±0.6) mm;穿支数量1~5(2.61±0.9)支(集中于小腿内侧中下1/3),穿支长度0.3~6(1.70±0.7) cm。皮瓣存活率为100%,伤口愈合良好。覆盖于小腿供区缺损处的皮片2例(2.5%)完全坏死,7例(8.7%)部分坏死,但经过换药处理后恢复满意。患者对外形及功能恢复满意。结论:应用胫后动脉皮瓣修复口腔颌面部缺损是一种较好的修复方式。胫后动脉皮瓣可携带近心端筋膜及皮下脂肪组织,用以丰满舌体形态、充填口底缺损、保护血管蒂。麦潋曦 Mubarak Mashrah 严凌健 万全 林钊宇 潘朝斌 2019中国口腔颌面外科杂志2019,17,3:10
3Zonisamide,an antiepileptic drug,alleviates diabetic cardiomyopathy by inhibiting endoplasmic reticulum stress显示文摘Endoplasmic reticulum stress(ER stress)plays a key role in the development of cardiac hypertrophy and diabetic cardiomyopathy(DCM).Zonisamide(ZNS)was originally developed as an antiepileptic drug.Studies have shown that ZNS suppresses ER stress-induced neuronal cell damage in the experimental models of Parkinson’s disease.Herein,we investigated whether ZNS improved DCM by attenuating ER stress-induced apoptosis.C57BL/6J mice were fed with high-fat diet(HFD)and intraperitoneally injected with low-dose streptozotocin(STZ)to induce type 2 diabetes mellitus(T2DM),and then treated with ZNS(40 mg·kg^(−1)·d^(−1),i.g.)for 16 weeks.We showed that ZNS administration slightly ameliorated the blood glucose levels,but significantly alleviated diabetes-induced cardiac dysfunction and hypertrophy.Furthermore,ZNS administration significantly inhibited the Bax and caspase-3 activity,upregulated Bcl-2 activity,and decreased the proportion of TUNEL-positive cells in heart tissues.We analyzed the hallmarks of ER stress in heart tissues,and revealed that ZNS administration significantly decreased the protein levels of GRP78,XBP-1s,ATF6,PERK,ATF4,and CHOP,and elevated Hrd1 protein.In high glucose(HG)-treated primary cardiomyocytes,application of ZNS(3μM)significantly alleviated HG-induced cardiomyocyte hypertrophy and apoptosis.ZNS application also suppressed activated ER stress in HG-treated cardiomyocytes.Moreover,preapplication of the specific ER stress inducer tunicamycin(10 ng/mL)eliminated the protective effects of ZNS against HG-induced cardiac hypertrophy and ER stress-mediated apoptosis.Our findings suggest that ZNS improves the cardiac diastolic function in diabetic mice and prevents T2DM-induced cardiac hypertrophy by attenuating ER stress-mediated apoptosis.Jia-hui Tian Qian Wu Yong-xiang He Qi-ying Shen Mubarak Rekep Gui-ping Zhang Jian-dong Luo Qin Xue Ying-hua Liu 2021Acta Pharmacologica Sinica2021,42,3:8
4Rapid adsorption of toxic Pb(Ⅱ) ions from aqueous solution using multiwall carbon nanotubes synthesized by microwave chemical vapor deposition technique显示文摘Multiwall carbon nanotubes(MWCNTs) were synthesized using a tubular microwave chemical vapor deposition technique, using acetylene and hydrogen as the precursor gases and ferrocene as catalyst. The novel MWCNT samples were tested for their performance in terms of Pb(Ⅱ)binding. The synthesized MWCNT samples were characterized using Fourier Transform Infrared(FT-IR), Brunauer, Emmett and Teller(BET), Field Emission Scanning Electron Microscopy(FESEM) analysis, and the adsorption of Pb(Ⅱ) was studied as a function of p H,initial Pb(Ⅱ) concentration, MWCNT dosage, agitation speed, and adsorption time, and process parameters were optimized. The adsorption data followed both Freundlich and Langmuir isotherms. On the basis of the Langmuir model, Qmaxwas calculated to be 104.2 mg/g for the microwave-synthesized MWCNTs. In order to investigate the dynamic behavior of MWCNTs as an adsorbent, the kinetic data were modeled using pseudo first-order and pseudo second-order equations. Different thermodynamic parameters, viz., ΔH0, ΔS0and ΔG0were evaluated and it was found that the adsorption was feasible, spontaneous and endothermic in nature. The statistical analysis revealed that the optimum conditions for the highest removal(99.9%) of Pb(Ⅱ) are at p H 5, MWCNT dosage 0.1 g, agitation speed 160 r/min and time of 22.5 min with the initial concentration of 10 mg/L. 6利用瞬态瑞雷面波法探测浅层玄武岩三维分布——以沙特Sabkhah Ad Dumathah地区为例显示文摘沙特Sabkhah Ad Dumathah地区浅层玄武岩的分布影响着该区钙结岩型铀矿的发育空间和分布范围,然而常规钻探、(电)磁法、重磁法等技术手段,对于该区浅层(约30 m以浅)玄武岩,存在探测难度.本文提出了利用瞬态瑞雷面波法开展浅层玄武岩探查试验研究,首先通过面波数据采集技术的系统试验研究,获取了较适用的面波数据采集方法,其次经过面波资料处理得到用于解释的面波速度映像图,最后基于该处理的三维面波速度映像图,解释了浅层玄武岩的三维分布范围和形态特征,通过与测区内已知钻孔资料进行对比,探测结果与地质认识相吻合.可见本文使用的面波勘探法能够较好地探测浅层玄武岩的三维空间分布规律特征,为今后类似工作提供重要参考和借鉴.吴曲波 潘自强 陈金勇 Mubarak Hani 2019地球物理学进展2019,34,5:5
71,2,3-Triazole incorporated coumarin derivatives as potential antifungal and antioxidant agents显示文摘A series of novel ethyl-7-((1-(benzyl)-1H-1,2,3-triazol-4-yl)methoxy)-2-oxo-2H-chromene-3-carboxylates 8a-h as potential antifungal agents were synthesized via click chemistry. The antifungal activity was evaluated against five human pathogenic fungal strains, such as Candida albicans, Fusarium oxysporum, Aspergillus flavus, Aspergillus niger and Cryptococcus neoformans. Compound 8c, 8d, 8e and 8h were found to be equipotent against C. albicans when compared with miconazole and compound 8f was found to be two-fold more active compared with miconazole and equipotent to fluconazole against C.albicans. The coumarin-based triazole derivatives were also evaluated for antioxidant activity and compound 8a was found to be potent antioxidant when compared with standard drug. Furthermore,molecular docking study of the newly synthesized compounds was performed and results showed good binding mode in the active site of fungal C. albicans enzyme P450 cytochrome lanosterol 14 ademethylase. 8Sugar beet yield and industrial sugar contents improved by potassium fertilization under scarce and adequate moisture conditions显示文摘Sugar beet(Beta vulgaris L.) is an industrial crop, grown worldwide for sugar production. In Pakistan, sugar is mostly extracted from sugarcane, soil and environmental conditions are equally favorable for sugar beet cultivation. Beet sugar contents are higher than sugarcane sugar contents, which can be further increased by potassium(K) fertilization. Total K concentration is higher in Pakistani soils developed from mica minerals, but it does not represent plant available K for sustainable plant growth. A pot experiment was conducted in the wire-house of Institute of Soil and Environmental Sciences at University of Agriculture Faisalabad, Pakistan. K treatments were the following: no K(K_0), K application at 148 kg ha^(–1)(K_1) and 296 kg ha^(–1)(K_2). Irrigation levels were used as water sufficient at 60% water holding capacity and water deficient at 40% water holding capacity. The growth, yield and beet quality data were analyzed statistically using LSD. The results revealed that increase in the level of K fertilization at water sufficient level significantly increased plant growth, beet yield and industrial beet sugar content. The response of K fertilization under water deficient condition was also similar, however overall sugar production was less than that in water sufficient conditions. 9High-dose-rate intraluminal brachytherapy during preoperative chemoradiation for locally advanced rectal cancers显示文摘AIM:To determine the feasibility and safety of high dose rate intraluminal brachytherapy(HDR-ILBT) boost during preoperative chemoradiation for rectal cancer.METHODS:Between 2008 and 2009,thirty-six patients with locally advanced rectal cancer(≥ T3 or N+),were treated initially with concurrent capecitabine(825 mg/m2 oral twice daily) and pelvic external beam radiotherapy(EBRT)(45 Gy in 25 fractions),then were randomized to group A;HDR-ILBT group(n = 17) to receive 5.5-7 Gy×2 to gross tumor volume(GTV) and group B;EBRT group(n = 19) to receive 5.4 Gy×3 fractions to GTV with EBRT.All patients underwent total mesorectal excision.RESULTS:Grade 3 acute toxicities were registered in 12 patients(70.6%) in group A and in 8(42.1%) in group B.Complete pathologic response of T stage(ypT0) in group A was registered in 10 patients(58.8%) and in group B,3 patients(15.8%) had ypT0(P < 0.0001).Sphincter preservation was reported in 6/9 patients(66.7%) in group A and in 5/10 patients(50%) in group B(P < 0.01).Overall radiological response was 68.15% and 66.04% in Group A and B,respectively.During a median follow up of 18 mo,late grade 1 and 2 sequelae were registered in 3 patients(17.6%) and 4 patients(21.1%) in the groups A and B,respectively.CONCLUSION:HDR-ILBT was found to be effective dose escalation technique in preoperative chemoradiation for rectal cancers,with higher response rates,downstaging and with manageable acute toxicities.Mutahir Ali Tunio Mansoor Rafi Altaf Hashmi Rehan Mohsin Abdul Qayyum Mujahid Hasan Amjad Sattar Muhammad Mubarak 2010World Journal of Gastroenterology2010,16,35:3
10Treatment of perforated giant gastric ulcer in an emergency setting显示文摘AIM:To study and assess clinical outcomes of various modes of treatment for perforated giant gastric ulcer in an emergency setting.METHODS:From May 2010 to February 2013,20cases of perforated giant gastric ulcer(>2 cm)were operated on in an emergency setting.All the patients presented with features of peritonitis and were resuscitated aggressively before taking for surgery.In the first4 cases,primary closure was done after taking a biopsy and among these,the 3rdcase also underwent partial distal gastrectomy and gastrojejunostomy and the 4th case underwent a radical subtotal gastrectomy with D2 lymphadenectomy and gastrojejunostomy for malignancy.All the remaining 16 cases underwent partial distal gastrectomy and gastrojejunostomy.RESULTS:Among the first 4 cases,2 had an uneventful recovery and were discharged on the 6thpostoperative day.The 3rdand 4thpatients developed gastricfistula,leading to prolonged hospitalization.For the 3rd patient,conservative management was tried for 1 wk,followed by partial distal gastrectomy and gastrojejunostomy,and he was discharged on the 20thday after admission,while the 4thpatient underwent a radical subtotal gastrectomy with D2 lymphadenectomy and gastrojejunostomy.Postoperatively,he developed adult respiratory distress syndrome,multiorgan dysfunction syndrome and expired on the 3rdpostoperative day of the second surgery.All the remaining 16 patients underwent partial distal gastrectomy and gastrojejunostomy and recovered well.Among these,4 of them were malignant and the remaining were benign ulcers.All had an uneventful recovery.The percentage of malignancy in our series was 30%(6 out of 20 cases).In our study,86%had an uneventful recovery,complications were seen in about 10%,and mortality was about 5%.CONCLUSION:In giant gastric ulcer,the chances of malignancy and leak after primary closure are high.So,we feel that partial distal gastrectomy and gastrojejunostomy is better.Pradeep Kumar Hosni Mubarak Khan Safarulla Hasanrabba 2014World Journal of Gastrointestinal Surgery2014,6,1:3
11Polyethylene glycol functionalized carbon nanotubes/gelatin-chitosan nanocomposite: An approach for significant drug release显示文摘This research work blooms the new idea of developing a safe and controlled drug releasing matrix using multi-walled carbon nanotubes(MWCNTs).In aqueous solution,uniform and highly stable dispersion of MWCNTs was obtained after secondary functionalization with polyethylene glycol(PEG)which was studied by Fourier transmission infrared spectroscopy(FTIR)and thermogravimetric analysis(TGA).Solution casting method was used to prepare MWCNTs/gelatin-chitosan nanocomposite films and the effect of MWCNTs on physico-mechanical,thermal and water uptake properties of the nanocomposites were evaluated.Incorporation of MWCNTs into the porous gelatin-chitosan matrix showed interesting stiffness and dampness along with developed microfibrillar structures within the pore walls intended at being used in tissue engineering of bone or cartilage.A common antibiotic drug,ciprofloxacin was incorporated into nanocomposite matrix.The evaluation of the effect of MWCNTs on drug release rate by dissolution test and antimicrobial susceptibility test was performed.Sharp release of the drug was found at early stages(~1 h),but the rate was reduced afterwards,showing a sustained release.It was observed that for all microorganisms,the antibacterial activities of drug loaded MWCNTs/gelatin-chitosan nanocomposites were higher than that of drug loaded gelatin-chitosan composite films containing no MWCNTs.Comparative statistical studies by ANOVA techniques also showed remarkable difference between the antibacterial activities,exhibited by MWCNTs-incorporated and non-incorporated composite films.Sadia Sharmeen A.F.M.Mustafizur Rahman Mostakima M.Lubna Kh Samaher Salem Rafiqul Islam Mubarak A.Khan 2018Bioactive Materials2018,3,3:3

15Vasculitis with renal involvement in essential mixed cryoglobulinemia:Case report and mini-review显示文摘The discovery of a strong association between hepatitis C virus(HCV) infection and mixed cryoglobulinemia(MC) has led to an increasingly rare diagnosis of idiopathic essential MC(EMC).The incidence of EMC is high in regions where there is a comparatively low HCV infection burden and low in areas of high infection prevalence,including HCV.The diagnosis of EMC requires an extensive laboratory investigation to exclude all possible causes of cryoglobulin formation.In addition,although cryoglobulin testing is simple,improper testing conditions will result in false negative results.Here,we present a 46-year-old female patient with a case of EMC with dermatological and renal manifestations,highlighting the importance of extensive investigation to reacha proper diagnosis.We review the need for appropriate laboratory testing,which is often neglected in clinical practice and which can result in false negative results.This review also emphasizes the significance of an extended testing repertoire necessary for better patient management.Despite a strong association of MC with HCV infection and other causes that lead to cryoglobulin formation,EMC remains a separate entity.Correct diagnosis requires proper temperature regulation during sample handling,as well as characterization and quantification of the cryoprecipitate.Inclusion of rheumatoid factor activity and complement levels in the cryoglobulin test-panel promotes better patient management and monitoring.Consensus guidelines should be developed and implemented for cryoglobulin detection and the diagnosis of cryoglobulinemic syndrome,which will reduce variability in inter-laboratory reporting.Sabiha Anis Khawar Abbas Mohammad Mubarak Ejaz Ahmed Sajid Bhatti Rana Muzaffar 2014World Journal of Clinical Cases2014,2,5:2

18Effect of methanolic extract of Tetrapleura tetraptera(Schum and Thonn)Taub leaves on hyperglycemia and indices of diabetic complications in alloxan-induced diabetic rats显示文摘Objective:To investigate the ameliorative role of Tetrapleura tetraplera(Schum and Thonn)Taub(T.tetraplera)leaf in hyperglycemia with associated conditions like oxidative stress,kidney damage and disorders in lipid metabolism.Methods:Five groups of five rats each intraperitoneally received the following treatment schedules for 7 d:untreated normal control,untreated alloxan-diabetic control,diabetic treated with glibenclamide,normal rats treated with extract(50 mg/kg)and diabetic rats treated with the extract.Evaluations were made for fasting blood sugar,body weight changes,malondialdehyde,aspartate aminotransferase,alanine aminotransferase,bilirubin,superoxide dismutase,catalase,lipid profile,packed cell volume,hemoglobin,urea and creatinine in all the rats.Results:Whereas the untreated diabetic rats showed a significant decrease(P<0.05)in packed cell volume,superoxide dismutase,catalase and high-density lipoprotein-cholesterol with a concomitant increase in the levels of malondialdehyde,fasting blood sugar,aspartate aminotransferase,alanine aminotransferase,bilirubin,urea and creatinine,administration of methanolic extract of T.tetraplera leaf or glibenclamide alleviated these altered parameters in the treated rats.Conclusions:Methanolic extract of T.tetraplera leaves possesses a potent capacity for treatment of diabetes and the accompanying complications,including oxidative stress and hyperlipidemia.Sunday Ene-Ojo Atawodi Ojochenemi Ejeh Yakubu Mubarak Labaran Liman Dorothy Uju iliemene 2014Asian Pacific Journal of Tropical Biomedicine2014,4,4:1
