1The record of paleoclimatic change from stalagmites and the determination of termination II in the south of Guizhou Province, China显示文摘A high-resolution climate record from 163.00 kaBP to 113.80 kaBP has been obtained through TIMS-U series dating and carbon and oxygen isotope analysis of the three large stalagmites from two caves in the south of Guizhou Province, China. The record of the oxygen isotopes from the stalagmites reveals that the undulation characteristics between the cooling event of the glacial period and the warming event of the interglacial period in the research area can compare well to those of ice cores, lake sediments, loess and deep sea sediments on the scale of ten-thousand years or millennium time scale. The climate undulation provided by the record of the stalagmites has a coherence with the global changes and a tele-connection to the paleoclimate changes in the north polar region. Our results suggest that the direct dynamics of paleo-monsoon circulation changes reflected in the record of the stalagmites might be caused by changes of the global ice volume, and in turn related to various factors, including the solar radiation strength at the mid-latitudes in the Northern Hemisphere, the southern extension of the ice-rafted event in the North Atlantic, and changes of the equatorial Pacific sea surface temperature at the low-latitudes. Using δ18O values, we have calculated the temperatures and the results show that the temperature difference between the penultimate glacial period (with an average temperature of 8.1℃, and a minimum temperature range from 0.65℃ to -1.43℃ at stage 6 ) and the last interglacial period (with an average temperature of 18.24℃ at sub-stage 5e ) was about 10℃. This temperature difference from the record of the stalagmites corresponds in general to the record temperature variation (about 10℃) of measured ice cores. The climate records from the three stalagmites in the two caves have shown that the circulation strength of the Asian summer monsoon and the winter monsoon in the penultimate glacial period and the last interglacial period had a clear change. With the TIMS-U series method, termination II of the penultimate glacial period has been precisely dated at an age of (129.28±1.10) kaBP for the three stalagmites in the south of Guizhou Province, China. This borderline age represents the beginning of the last interglacial period or the boundary between the Middle Pleistocene and the Late Pleistocene, and corresponds to the beginning age of the last interglacial period shown by the ice cores and in the SPECMAP curve of the marine oxygen isotopes. 2Tumor-expressed B7-H3 mediates the inhibition of antitumor T-cell functions in ovarian cancer insensitive to PD-1 blockade therapy显示文摘Although PD-L1/PD-1 blockade therapy has been approved to treat many types of cancers,the majority of patients with solid tumors do not respond well,but the underlying reason remains unclear.Here,we studied ovarian cancer(OvCa),a tumor type generally resistant to current immunotherapies,to investigate PD-1-independent immunosuppression.We found that PD-L1 was not highly expressed in the tumor microenvironment(TME)of human OvCa.Instead,B7-H3,another checkpoint molecule,was highly expressed by both tumor cells and tumor-infiltrating antigen-presenting cells(APCs),which correlated with T-cell exhaustion in patients.Using ID8 OvCa mouse models,we found that B7-H3 expressed on tumor cells,but not host cells,had a dominant role in suppressing antitumor immunity.Therapeutically,B7-H3 blockade,but not PD-1 blockade,prolonged the survival of ID8 tumorbearing mice.Collectively,our results demonstrate that tumor-expressed B7-H3 inhibits the function of CD8^(+)T cells and suggest that B7-H3 may be a target in patients who are not responsive to PD-L1/PD-1 inhibition,particularly OvCa patients.Dongli Cai Jiaming Li Dingfeng Liu Shanjuan Hong Qin Qiao Qinli Sun Pingping Li Nanan Lyu Tiantian Sun Shan Xie Li Guo Ling Ni Liping Jin Chen Dong 2020Cellular & Molecular Immunology2020,17,3:8

4A fiber Bragg grating based earth and water pressures transducer with three-dimensional fused deposition modeling for soil mass显示文摘A novel fiber Bragg grating(FBG)sensor with three-dimensional(3D)fused deposition modeling(FDM)approach is proposed for effective stress measurement in soil mass.The three-diaphragm structure design is developed to measure earth and water pressures simultaneously.The proposed transducer has advantages of small size,high sensitivity,low cost,immunity to electromagnetic interference and rapid prototyping.The working principle,design parameters,and manufacturing details are discussed.The proposed transducer was calibrated for earth and water pressures measurement by using weights and a specially designed pressure chamber,respectively.The calibration results showed that the wavelength of the transducer was proportional to the applied pressure.The sensitivity coefficients of the earth and water pressures were 12.633 nm/MPa and 6.282 nm/MPa,respectively.Repeated tests and error analysis demonstrated the excellent stability and accuracy of the earth and water pressure measurements.The performance of the proposed transducer was further verified by a model experimental test and numerical analysis,which indicated that the proposed transducer has great potential for practical applications.Yue Qin Qiankun Wang Dongsheng Xu Jiaming Yan Shanshan Zhang 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,2:3

7Sub-nanometer ultrathin epitaxy of AlGaN and its application in efficient doping显示文摘Solving the doping asymmetry issue in wide gap semiconductors is a key dificulty and long-standing challenge for device applications.Here,a desorption-tailoring strategy is proposed to juggle the carrier concentration and transport.Specific to the p-doping issue in Al-rich AlGaN,self-assembled p-AlGaN superlattices with an average Al composition of over 50%are prepared by adopting this approach.The hole concentration as high as 8.1×10^(18)cm^(-3)is thus realized at room temperature,which is attributed to the signifcant reduction of effective Mg activation energy to 17.5 meV through modulating the activating path,as well as the highlighted Mg surface-incorporation by an intentional interruption for desorption.More importantly,benefting from the constant ultrathin barrier thickness of only three monolayers via this approach,vertical miniband transport of holes is verifed in the p-AlGaN superlattices,greatly satisfying the demand of hole injection in device application.280 nm deep-ultraviolet light emitting diodes are then fabricated as a demo with the desorption-tailored Al-rich p-AlGaN superlattices,which exhibit a great improvement of the carrier injection efficiency and light extraction efficiency,thus leading to a 55.7%increase of the light output power.This study provides a solution for p-type doping of Al-rich AlGaN,and also sheds light on solving the doping asymmetry issue in general for wide-gap semiconductors.Jiaming Wang Mingxing Wang Fujun Xu Baiyin Liu Jing Lang Na Zhang Xiangning Kang Zhixin Qin Xuelin Yang Xinqiang Wang Weikun Ge Bo Shen 2022Light(Science & Applications)2022,11,4:1

12Genomic selection of eight fruit traits in pear显示文摘Genomic selection (GS) has the potential to improve selection efficiency and shorten the breeding cycle in fruit tree breeding. In this study,we evaluated the effect of prediction methods, marker density and the training population (TP) size on pear GS for improving its performance and reducing cost. We evaluated GS under two scenarios:(1) five-fold cross-validation in an interspecific pear family;(2) independent validation. Based on the cross-validation scheme, the prediction accuracy (PA) of eight fruit traits varied between 0.33 (fruit core vertical diameter)and 0.65 (stone cell content). Except for single fruit weight, a slightly better prediction accuracy (PA) was observed for the five parametrical methods compared with the two non-parametrical methods. In our TP of 310 individuals, 2 000 single nucleotide polymorphism (SNP) markers were sufficient to make reasonably accurate predictions. PAs for different traits increased by 18.21%-46.98%when the TP size increased from 50to 100, but the increment was smaller (-4.13%-33.91%) when the TP size increased from 200 to 250. For independent validation, the PAs ranged from 0.11 to 0.45 using rrBLUP method. In summary, our results showed that the TP size and SNP numbers had a greater impact on the PA than prediction methods. 13SiRNA-mediated ankyrin-G silence modulates the expression of voltage-gated Na channels in murine hippocampal HT22 cells显示文摘Background:Voltage-gated sodium channels are the targets of many commonly used antiepileptic drugs.NaV1.6,encoded by Scn8a,increased in chronic mesial temporal epilepsy animal models and co-localized with Ankyrin-G,encoded by Ank3.We hypothesized that inhibition of Ank3 transcription by siRNA decrease the expression of NaV1.6.Results:We characterized expression of the target genes in hippocampal neuron HT22 cells by Real time-PCR.The melt peak in the resolution curve of Scn1a,Scn8a and Ank3 were all unique.Ank3 transcription was interfered and the relative Ank3 mRNA levels of the three interfered groups compared to GAPDH were 0.89±0.13,0.52±0.07 and 0.26±0.05 while that of the negative control group was 1.01±0.08(P<0.05).When Ank3 transcription was inhibited by siRNA,the relative mRNA levels of Scn8a decreased in the three groups(0.91±0.09,0.33±0.06 and 0.25±0.05),compared to the negative control group(1.10±0.09).Tested by Western blotting,protein levels of ankyrinG and Nav1.6 decreased after ank3-siRNA.Ankyrin-G in negative control group,group1,group2 and group1+2 were 0.813±0.051,0.744±0.041,0.477±0.055 and 0.351±0.190 respectively(P<0.01)while Nav1.6 were 0.934±0.036,0.867±0.078,0.498±0.070 and 0.586±0.180(P<0.01).The quantity analysis of immunofluorescence showed significant decrease of ankyrin-G and Nav1.6(Student’s test,P=0.046 and 0.016 respectively).Conclusion:We therefore concluded that in HT22 cells the expression of Nav1.6 was down-regulated by Ank3 RNA interference.Guanzhong Ni Xiaoting Hao Xiaodong Cai Jiaming Qin Liemin Zhou Patrick Kwan Ziyi Chen 2019Acta Epileptologica2019,1,1:0
14Molecular characterization and antibiotic resistance of Salmonella enterica serovar 1,4,[5],12:i:-environmental isolates from poultry farms显示文摘Salmonella enterica serovar 1,4,[5],12:i:-(S.1,4,[5],12:i:-)has been recognized as an emerging foodborne pathogen in recent years.It can cause human salmonellosis predominated by the contamination of animal-derived foods such as raw poultry and pork.This study aimed to characterize the genetic diversity,plasmid replicon types,and antibiotic resistance of 15 S.1,4,[5],12:i:-environmental isolates collected from two poultry farms using pulsed-field gel electrophoresis(PFGE),multilocus sequence typing(MLST),polymerase chain reaction-based replicon typing,and minimum inhibitory concentration approach.Ten different PFGE genotypes were detected,indicating a high diversity among these S.1,4,[5],12:i:-isolates.Three sequence types(ST19,ST1544,ST34)were identified by MLST.Among them,ST1544 was first detected in S.1,4,[5],12:i:-environmental isolates from poultry farms.All isolates were resistant to cefazolin,cefotetan,tobramycin,amikacin,and gentamicin,but susceptible to piperacillin-tazobactam,aztreonam,ceftazidime,cefepime,and ertapenem.Five incompatibility groups(Inc)of plasmids were identified,including IncFIIs(66.7%),IncHI2(20%),IncI1(6.7%),IncN(6.7%),and IncQ(6.7%).Among these isolates,80%carried at least one plasmid replicon type,and 20%carried multiple plasmid replicon types.Interestingly,the multidrug-resistant isolate 263 carried numerous resistance genes(i.e.qnrS,aac(6ʹ)-Ib-cr,bla_(TEM),bla_(CTX-M-9),bla_(OXA-1),sul1,sul2,sul3,floR,and mcr-1)and class I integronase gene intI1,which possessed both IncHI2 and IncQ plasmids,suggesting that resistance genes may be horizontally transferred by the combination of IncHI2 and IncQ plasmids.Collectively,antibiotic-resistant S.1,4,[5],12:i:-isolates were first found in poultry farm environments in China,and surveillance should be strengthened to prevent their further spread from poultry farms to foods.Xiaojie Qin Linlin Xiao Jiaming Li Mingzhe Yang Changying Yang Qingli Dong 2022Food Quality and Safety2022,6,4:0

19Canonical and noncanonical Wnt signaling: Multilayered mediators, signaling mechanisms and major signaling crosstalk显示文摘Wnt signaling plays a major role in regulating cell proliferation and differentiation.The Wnt ligands are a family of 19 secreted glycoproteins that mediate their signaling effects via binding to Frizzled receptors and LRP5/6 coreceptors and transducing the signal either throughβ-catenin in the canonical pathway or through a series of other proteins in the nonca-nonical pathway.Many of the individual components of both canonical and noncanonical Wnt signaling have additional functions throughout the body,establishing the complex interplay between Wnt signaling and other signaling pathways.This crosstalk between Wnt signaling and other pathways gives Wnt signaling a vital role in many cellular and organ processes.Dys-regulation of this system has been implicated in many diseases affecting a wide array of organ systems,including cancer and embryological defects,and can even cause embryonic lethality.The complexity of this system and its interacting proteins have made Wnt signaling a target for many therapeutic treatments.However,both stimulatory and inhibitory treatments come with potential risks that need to be addressed.This review synthesized much of the current knowl-edge on the Wnt signaling pathway,beginning with the history of Wnt signaling.It thoroughly described the different variants of Wnt signaling,including canonical,noncanonical Wnt/PCP,and the noncanonical Wnt/Ca2+pathway.Further description involved each of its components and their involvement in other cellular processes.Finally,this review explained the various other pathways and processes that crosstalk with Wnt signaling.Kevin Qin Michael Yu Jiaming Fan Hongwei Wang Piao Zhao Guozhi Zhaoo Wei Zeng Connie Chen Yonghui Wang Annie Wang Zander Schwartz Jeffrey Hong Lily Song William Wagstaff Rex C.Haydon Hue H.Luu Sherwin H.Ho Jason Strelzow Russell R.Reid Tong-Chuan He Lewis L.Shi 2024Genes & Diseases2024,11,1:0
