1Modeling study of regional severe hazes over mid-eastern China in January 2013 and its implications on pollution prevention and control显示文摘The Nested Air Quality Prediction Model System(NAQPMS)was used to investigate the temporal and spatial variations of PM2.5over tropospheric central eastern China in January 2013.The impact of regional transport and its implications on pollution prevention and control were also examined.Comparison between simulated and observed PM2.5showed NAQPMS was able to reproduce the evolution of PM2.5during heavy haze episodes.The results indicated that regional transport of PM2.5played an important role in regional haze episodes in the city cluster including Hebei,Beijing and Tianjin(HBT).The cross-city clusters transport outside HBT and transport among cities inside HBT contributed 20%–35%and 26%–35%of PM2.5as compared with local emission,in HBT respectively.To meet the Air Quality Standards for Grade II,90%,90%and65%of emissions would have to be cut down in Hebei,Tianjin and Beijing,if non-control strategy was taken in the surrounding city clusters of HBT.This implicated that control of emissions in one city cluster is not sufficient to reduce regional haze events,and joint efforts among city clusters are essential.Besides regional transports,two-way feedback between boundary-layer evolution and PM2.5also significantly contributed to the formation of heavy hazes,which contributed 30%of monthly average PM2.5concentration in HBT.WANG ZiFa LI Jie WANG Zhe YANG WenYi TANG Xiao GE BaoZhu YAN PinZhong ZHU LiLi CHEN XueShun CHEN HuanSheng WAND Wei LI JianJun LIU Bing WANG XiaoYan WAND Wei ZHAO YiLin LU Ning SU DeBin 2014Science China Earth Sciences2014,57,1:106
2Mesenchymal stem cells derived from human placenta suppress allogeneic umbilical cord blood lymphocyte proliferation显示文摘ABSTRACTHuman placenta-derived mononuclear cells (MNC) were isolated by a Percoll density gradient and cultured in mesen-chymal stem cell (MSC) maintenance medium. The homogenous layer of adherent cells exhibited a typical fibroblast-like morphology, a large expansive potential, and cell cycle characteristics including a subset of quiescent cells. In vitrodifferentiation assays showed the tripotential differentiation capacity of these cells toward adipogenic, osteogenic andchondrogenic lineages. Flow cytometry analyses and immunocytochemistry stain showed that placental MSC was ahomogeneous cell population devoid of hematopoietic cells, which uniformly expressed CD29, CD44, CD73, CD105,CD166, laminin, fibronectin and vimentin while being negative for expression of CD31, CD34, CD45 and α-smoothmuscle actin. Most importantly, immuno-phenotypic analyses demonstrated that these cells expressed class I majorhistocompatibility complex (MHC-I), but they did not express MHC-II molecules. Additionally these cells could sup-press umbilical cord blood (UCB) lymphocytes proliferation induced by cellular or nonspecific mitogenic stimuli. Thisstrongly implies that they may have potential application in allograft transplantation. 3SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
4Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. 5Diagnosis and treatment of hepatic angiomyolipoma in 26 cases显示文摘AIM: To summarize the experience of the diagnosis and treatment of hepatic angiomyolipoma (HAML).METHODS: The clinical, imaging and pathological features,and treatment strategies of 26 patients with HAML treated at the authors′institute between October 1998 and January 2003 were retrospectively analyzed. All the patients received liver resection and were followed up till the study.Immunohistochemical assays were performed with a panel of antibodies.RESULTS: There was an obvious female predominance (21:5), and most of the patients (18/26) had no symptoms.Heterogeneous high echo was found in ultrasonography and punctiform or filiform vascular distribution pattern was found in color Doppler-sonography in most of the lesions (21/26).All of the 5 lesions further enhanced with Levovist showed early and prolonged enhancement. At contrast-enhanced spiral CT, the soft-tissue components of 24 lesions were markedly enhanced in the arterial phase and 18 lesions remained enhanced in the portal venous phase. MRI was performed in 9 patients, and showed hypointensity or hyperintensity on T1-weighted images and heterogeneous hyperintensity on T2-weighted images. Histopathologically,all lesions were composed of adipose tissues, smooth muscle and blood vessels with different proportions. Most lesions showed positive immunohistochemical staining for HMB45(26/26), A103 (24/26) and SMA (24/26). All of the 26 patients showed a benign course with no sign of recurrence.CONCLUSION: Preoperative radiological diagnosis of HAML is possible. The demonstration of intratumoral fat and central vessels is helpful in the diagnosis. 6Matrine attenuates oxidative stress and cardiomyocyte apoptosis in doxorubicin-induced cardiotoxicity via maintaining AMPKα/UCP2 pathway显示文摘Oxidative stress and cardiomyocyte apoptosis are involved in the pathogenesis of doxorubicin(DOX)-induced cardiotoxicity. Matrine is well-known for its powerful anti-oxidant and anti-apoptotic capacities. Our present study aimed to investigate the effect of matrine on DOX-induced cardiotoxicity and try to unearth the underlying mechanisms. Mice were exposed with DOX to generate DOX-induced cardiotoxicity or normal saline as control. H9 C2 cells were used to verify the effect of matrine in vitro.DOX injection triggered increased generation of reactive oxygen species(ROS) and excessive cardiomyocyte apoptosis, which were significantly mitigated by matrine. Mechanistically, we found that matrine ameliorated DOX-induced uncoupling protein 2(UCP2) downregulation, and UCP2 inhibition bygenipin could blunt the protective effect of matrine on DOX-induced oxidative stress and cardiomyocyte apoptosis. Besides, 50-AMP-activated protein kinase α2(Ampkα2) deficiency inhibited matrine-mediated UCP2 preservation and abolished the beneficial effect of matrine in mice. Besides, we observed that matrine incubation alleviated DOX-induced H9 C2 cells apoptosis and oxidative stress level via activating AMPKα/UCP2, which were blunted by either AMPKα or UCP2 inhibition with genetic or pharmacological methods. 9Prevention of hepatocellular carcinoma in mice by IL-2 and B7-1 genes co-transfected liver cancer cell vaccines显示文摘AIM: To study the immunoprotective effect of liver cancer vaccine with co-transfected IL-2 and B7-1 genes on hepatocarcinogenesis in mice.METHODS: The murine liver cancer cell line Hepal-6 was transfected with IL-2 and/or B7-1 gene via recombinant adenoviral vectors and the liver cancer vaccines were prepared. C57BL/6 mice were immunized with these vaccines and challenged with the parental Hepal-6 cells afterwards.The immunoprotection was investigated and the reactive T cell line was assayed.RESULTS: The immunoprotection of the tumor vaccine was demonstrated. The effect of IL-2 and B7-1 genes cotransfected Hepal-6 liver cancer vaccine (Hep6-IL2/B7vaccine) on the onset of tumor formation was the strongest.When attacked with wild Hepal-6 cells, the median survival period of the mice immunized with Hep6-IL2/B7 vaccine was the longest (68 days, χ2=7.70-11.69, P<0.05) and the implanted tumor was the smallest (z =3.20-44.10, P<0.05).The effect of single IL-2 or B7-1 gene-transfected vaccine was next to the IL2/B7 gene co-transfected group, and the mean survival periods were 59 and 54 days, respectively.The mean survival periods of wild or enhanced green fluorescence protein gene modified vaccine immunized group were 51 and 48 days, respectively. The mice in control group all died within 38 days and the implanted tumor was the largest (z=3.20-40.21, P<0.05). The cellular immunofunction test and cytotoxicity study showed that the natural killer (NK) cell, lymphokine activated killer (LAK) cell and cytotoxic T lymphocyte (CTL) activities were significantly increased in mice immunized with the Hep6-IL2/B7 vaccine, (29.5±2.5%,65.0±2.9%, 83.1±1.5% respectively, compared with other groups, P<0.05).CONCLUSION: The Hep6-IL2/B7 liver cancer vaccines can induce the mice to produce activated and specific CTL against the parental tumor cells, and demonstrate stronger effect on the hepatocarcinogenesis than single gene modified or the regular tumor vaccine. 10Effect of endogenous insulin-like growth factor and stem cell factor on diabetic colonic dysmotility显示文摘AIM: To investigate whether the reduction of stem cell factor (SCF) is mediated by decreased endogenous insulin-like growth factor (IGF)-1 in diabetic rat colon smooth muscle. METHODS: Sixteen Sprague-Dawley rats were randomly divided into two groups: control group and streptozotocin-induced diabetic group. After 8 wk of streptozotocin administration, colonic motility function and contractility of circular muscle strips were measured. The expression of endogenous IGF-1 and SCF was tested in colonic tissues. Colonic smooth muscle cells were cultured from normal adult rats. IGF-1 siRNA transfection was used to investigate whether SCF expression was affected by endogenous IGF-1 expression in smooth muscle cells, and IGF-1 induced SCF expression effects were studied. The effect of high glucose on the expression of endogenous IGF-1 and SCF was also investigated. RESULTS: Diabetic rats showed prolonged colonic transit time (252 ± 16 min vs 168 ± 9 min, P < 0.01) and weakness of circular muscle contraction (0.81 ± 0.09 g vs 2.48 ± 0.23 g, P < 0.01) compared with the control group. Endogenous IGF-1 and SCF protein expression was significantly reduced in the diabetic colonic muscle tissues. IGF-1 and SCF mRNA expression also showed a paralleled reduction in diabetic rats. In the IGF-1 siRNA transfected smooth muscle cells, SCF mRNA and protein expression was significantly decreased. IGF-1 could induce SCF expression in a concentration and time-dependent manner, mainly through the extracellular-signal-regulated kinase 1/2 signal pathway. High glucose inhibited endogenous IGF-1 and SCF expression and the addition of IGF-1 to the medium reversed the SCF expression. CONCLUSION: Myopathy may resolve in colonic motility dysfunction in diabetic rats. 11Effect of Ce doping into V_(2)O_(5)-WO_(3)/TiO_(2) catalysts on the selective catalytic reduction of NO_(x) by NH_(3)显示文摘In this work, the effectiveness of V_2O_5-WO_3/TiO_2 catalysts modified with different CeO_2 contents by impregnation and co-precipitation methods on the selective catalytic reduction of NOxby NH3 have been studied comparatively by various experimental techniques. The results showed that the NO conversion of V_2O_5-WO_3/CeO_2-TiO_2 catalysts modified by co-precipitation method obviously increased with the Ce doping contents in the studied range below 20%(All Ce contents are in mass fractions), but the NO conversion of V_2O_5-WO_3/CeO_2/TiO_2 catalysts modified by impregnation methods was lower than V_2O_5-WO_3/CeO_2-TiO_2 catalysts especially beyond 2.5% Ce doping contents. The V_2O_5-WO_3/CeO_2-TiO_2 catalysts showed better SCR activity, wider reaction window, and higher sulfur and water resistance. The characterization results elucidated that the modified catalysts by co-precipitation method exhibited higher specific surface area, much better dispersity of Ce component, more Ce^(3+)species and more Br?nsted acid sites than that by impregnation. The vacancies caused by more Ce^(3+)species were favorable for more NO oxidation to NO_2, and the interaction between Ce species and WOxspecies generated more Br?nsted acid sites. It could be supposed that dispersed Ce Oxspecies and WOxspecies offered more second active centers respectively to adsorb oxygen and activate ammonia as co-catalysis to the primary active center of V ions, thus facilitated the better SCR activity of modified V_2O_5-WO_3/CeO_2-TiO_2 catalysts by coprecipitation methods. 13Hot deformation behavior and constitutive model of TC18 alloy during compression显示文摘The hot deformation behavior of TC18 alloy at strain rates ranging from 1 9 10-4to 1 9 10-2s-1and temperatures ranging from 25 to 800 °C was studied using a WDW-300 electronic universal testing machine. The relationships between true stress and true strain show that flow stress decreases with the increase of temperature and increases as strain rate increases. The effect of strain rate on the flow stress becomes pronounced at higher temperatures. At room temperature, the river pattern characteristic of brittle fracture and the dimple pattern typical of ductile fracture are found to exist in different regions of fracture surfaces of the samples. An improved constitutive relationship is proposed to accurately describe the flow stress of TC18 by considering the effect of strain. And a microscopic model is also deduced which can link the physical mechanisms to the macroscopic experimental results. 16Effects of cryopreservation and phenylacetate on biological characters of adherent LAK cells from patients with hepatocellular carcinoma显示文摘AIM: To improve the preparation of adherent lymphokine-activated killer (A-LAK) cells and to study the effects ofcryopreservation and phenylacetate (PA) on biologicalcharacters of A-LAK cells.METHODS: A-LAK cells were obtained from peripheral bloodmononuclear cells (PBMCs) of the patients withhepatocellular caminoma (HCC) by using L-phenylalaninemethyl ester ( PME ) to deplete immunosuppressivermonocytes. Proliferative activity of SMMC7721 cell line aftertreatment with phenylacetate (PA) was observed. A-LAKcells were treated with the supernatant of SMMC7721 cellsthat had been pretreated with PA. The changes ofproliferation, cytotoxicity and phenotype of A-LAK cellswere investigated after cryopreservstion.RESULTS: The expansion of A-LAK cells (96.79± 69.10 foldson Day 14 ) was significantly higher then that of non-adherent LAK (NA-LAK) cells (22.77 ± 13.20) as well asconventional LAK cells (4. 64 ± 0. 91 ). PA significantlysuppressed the growth of SMMC7721 cells, and the inhibitorratio was 46 %. rite supernatant of cultured tumor cellsintensively suppressed the proliferation and cytotoxicity ofA-LAK cells, but the suppressive effect of the supernatantwas previously decreased after treatment with PA.Impairments in proliferation and cytotoxicity of A-LAK cellsimmediately after thawing of cryopreservation and recoveryafter reincubation with IL-2 were observed. The cytotoxicltyof thawed A-LAK cells on Day 5 was significantly higher thanthat of fresh A-LAK before freezing (54.8 ± 10.2 % vs 40.5 ±6. 4 %). No significant change in the percentage oflymphocyte subsets was identified in frozen A-LAK cells ascompared with that in the fresh control cells.CONCLUSION: A-LAK cells can he simply prepared by usingPME, and showed a synergistic anti-tumor effect with thecombination of PA. 17Progress of the technique of coal microwave desulfurization显示文摘With the advantages of its fast speed,effective and moderate controllable conditions,desulfurization of coal by microwave has become research focus in the field of clean coal technology.Coal is a homogeneous mixture which consists of various components with different dielectric properties,so their abilities to absorb microwaves are different,and the sulfur-containing components are better absorbers of microwave,which makes them can be selectively heated and reacted under microwave irradiation.There still remain controversies on the principle of microwave desulfurization at present,thermal effects or non-thermal effects.The point of thermal effects of microwave is mainly base on its characters of rapidly and selectly heating.While,in view of non-thermal effect,direct interactions between the microwave electromagnetic field and sulfur containing components are proposed.It is a fundamental problem to determine the dielectric properties of coal and the sulfur-containing components to reveal the interaction of microwave and sulfur-containing compounds.However,the test of dielectric property of coal is affected by many factors,which makes it difficult to measure dielectric properties accurately.In order to achieve better desulfurization effect,the researchers employ methods of adding chemical additives such as acid,alkali,oxidant,reductant,or changing the reaction atmosphere,or combining with other methods such as magnetic separation,ultrasonic and microorganism.Researchers in this field have also put forward several processes,and have obtained a number of patents.Obscurity of microwave desulfurization mechanism,uncertainties in qualitative and quantitative analysis of sulfur-containing functional groups in coal,and the lack of special microwave equipment have limited further development of microwave desulfurization technology.Xiuxiang Tao Ning Xu Maohua Xie Longfei Tang 2014International Journal of Coal Science & Technology2014,1,1:15
18Placement of prophylactic pancreatic stents to prevent post-endoscopic retrograde cholangiopancreatography pancreatitis in high-risk patients: A meta-analysis显示文摘AIM:To assess the effectiveness of pancreatic stents for preventing pancreatitis in high-risk patients after endoscopic retrograde cholangiopancreatography(ERCP).METHODS:PubMed,Embase,Science Citation Index,and Cochrane Controlled Trials Register were searched to identify relevant trials published in English.Inclu-sion and exclusion criteria were used to screen for suitable studies.Two reviewers independently judged the study eligibility while screening the citations.The methodological quality of the included trials was assessed using the Jadad scoring system.All results were expressed as OR and 95%CI.Data were analyzed using Stata12.0 software.RESULTS:Ten eligible randomized controlled trials were selected,including 1176 patients.A fixed-effects model in meta-analysis supported that pancreatic duct stents significantly decreased the incidence of postERCP pancreatitis(PEP)in high-risk patients(OR=0.25;95%CI:0.17-0.38;P<0.001).Pancreatic stents also alleviated the severity of PEP(mild pancreatitis after ERCP:OR=0.33;95%CI:0.21-0.54;P<0.001;moderate pancreatitis after ERCP:OR=0.30;95%CI:0.13-0.67;P=0.004).The result of severe pancreatitis after ERCP was handled more rigorously(OR=0.24;95%CI:0.05-1.16;P=0.077).Serum amylase levels were not different between patients with pancreatic stents and control patients(OR=1.08;95%CI:0.82-1.41;P=0.586).CONCLUSION:Placement of prophylactic pancreatic stents may lower the incidence of post-ERCP pancreatitis in high-risk patients and alleviate the severity of this condition.Qing-Qing Shi Xiao-Yi Ning Ling-Ling Zhan Guo-Du Tang Xiao-Ping Lv 2014World Journal of Gastroenterology2014,20,22:15
19DISTRIBUTION AND CHARACTERIZATION OF POLYCYCLIC AROMATIC HYDROCARBON COMPOUNDS IN AIRBORNE PARTICULATES OF EAST ASIA显示文摘A review is presented on the distribution and characterization of polycyclic aromatic hydrocarbons (PAHs) and their derivatives, including nitro-PAHs and hydro-PAHs, on atmospheric particulates of East Asia. Generally, PAH compounds with two or three aromatic rings are released mainly into the gas phase, while those containing three or more aromatic rings are associated with particulate matter (PM) emission. Particle-associated PAHs are primarily adsorbed on fine particles, and little associated with coarse particles. Investigation into the concentration level of PAHs in different areas can serve not only to reflect the pollutant status and sources but also to lead to the formulation of control strategies.
