Multi-Stage Hydrocarbon Accumulation and Formation Pressure Evolution in Sinian Dengying FormationCambrian Longwangmiao Formation, Gaoshiti-Moxi Structure, Sichuan Basin显示文摘Sichuan Basin is a typical superimposed basin, which experienced multi-phase tectonic movements, meanwhile Sinian–Cambrian underwent complex hydrocarbon accumulation processes, causing exploration difficulties in the past 60 years. Based on the microscopic evidence of fluid inclusions, combined with basin-modelling, this paper determines stages and time of hydrocarbon accumulation, reconstructs evolution of formation pressure and dynamic processes of hydrocarbon accumulation in Sinian Dengying Formation-Cambrian Longwangmiao Formation of Gaoshiti-Moxi structure. Three stages of inclusions are detected, including a stage of yellow-yellowgreen fluorescent oil inclusions, a stage of blue fluorescent oil-gas inclusions and a stage of non-fluorescent gas inclusions, reflecting the study area has experienced a series of complex hydrocarbon accumulation processes, such as formation of paleo-oil reservoirs, cracking of crude oil, formation of paleo-gas reservoirs and adjustment to present gas reservoirs, which occurred during 219–188, 192–146 and 168–0 Ma respectively. During the period of crude oil cracking, Dengying Formation-Longwangmiao Formation showed weak overpressure to overpressure characteristics, then after adjustment of paleo-gas reservoirs to present gas reservoirs, the pressure in Dengying Formation changed into overpressure but finally reduced to normal pressure system. However, due to excellent preservation conditions, the overpressure strength in Longwangmiao Formation only slightly decreased and was still kept to this day. | Juan Wu Shugen Liu Guozhi Wang Yihua Zhao Wei Sun Jinming Song Yanhong Tian | 2016 | Journal of Earth Science2016,27,5: | 8
Circular RNA circStag1 promotes bone regeneration by interacting with HuR显示文摘Postmenopausal osteoporosis is a common bone metabolic disorder characterized by deterioration of the bone microarchitecture, leading to an increased risk of fractures.Recently, circular RNAs(circ RNAs)have been demonstrated to play pivotal roles in regulating bone metabolism. However, the underlying functions of circ RNAs in bone metabolism in postmenopausal osteoporosis remain obscure. Here,we report that circ Stag1 is a critical osteoporosis-related circ RNA that shows significantly downregulated expression in osteoporotic bone marrow mesenchymal stem cells(BMSCs)and clinical bone tissue samples from patients with osteoporosis.Overexpression of circ Stag1 significantly promoted the osteogenic capability of BMSCs.Mechanistically,we found that circ Stag1 interacts with human antigen R(Hu R),an RNA-binding protein, and promotes the translocation of Hu R into the cytoplasm.A high cytoplasmic level of Hu R led to the activation of the Wnt signaling pathway by stabilizing and enhancing low-density lipoprotein receptor-related protein 5/6(Lrp5/6)andβ-catenin expression, thereby stimulating the osteogenic differentiation of BMSCs.Furthermore, overexpression of circ Stag1 in vivo by circ Stag1-loaded adeno-associated virus(circ Stag1-AAV)promoted new bone formation, thereby preventing bone loss in ovariectomized rats.Collectively,we show that circ Stag1 plays a pivotal role in promoting the regeneration of bone tissue via Hu R/Wnt signaling, which may provide new strategies to prevent bone metabolic disorders such as postmenopausal osteoporosis. | Gaoyang Chen Canling Long Shang Wang Zhenmin Wang Xin Chen Wanze Tang Xiaoqin He Zhiteng Bao Baoyu Tan Jin Zhao Yongheng Xie Zhizhong Li Dazhi Yang Guozhi Xiao Songlin Peng | 2022 | Bone Research2022,10,3: | 7
Effects of Fatty Acids on Low-Sulfur Diesel Lubricity:Experimental Investigation, DFT Calculation and MD Simulation显示文摘The continuous reduction in sulfur content of fuels would lead to diesel fuel with poor lubricity which could result in engine pump failure. In the present work, fatty acids were adopted as lubricity additives to low-sulfur diesel fuel. It was attempted to correlate the molecular structures of fatty acids, such as carbon chain length, degree of saturation and hydroxylation, to their lubricity enhancement, which was evaluated by the High-Frequency Reciprocating Rig (HFRR) method. The efficiency order was supported by the density functional theory (DFT) calculations and the molecular dynamics (MD) simulations. The lubricity enhancing properties of fatty acids are mainly determined by the cohesive energy of adsorbed films formed on iron surface. The greater the cohesive energy, the more efficiently the fatty acid would enhance the lubricity of low-sulfur diesel fuel. | Luo Hui Fan Weiyu Li Yang Zhao Pinhui Nan Guozhi | 2013 | China Petroleum Processing & Petrochemical Technology2013,15,2: | 6
Experimental and Molecular Dynamics Simulations for Investigating the Effect of FattyAcid and Its Derivatives on Low Sulfur Diesel Lubricity显示文摘In this work, fatty acid and its derivatives were adopted as lubricity additives for low sulfur diesel.Tribological evaluation obtained from the High-Frequency Reciprocating Rig(HFRR)apparatus showed that the lubricating performance of the additives increased in the following order:stearic acid>glycol monopalmitate>stearyl alcohol>ethyl palmitate>cetyl ethyl ether. The adsorption behavior of the additives on Fe(110)surface and Fe2O3(001)surface was investigated by molecular dynamics(MD)simulations to verify their lubricity performance. The results suggested that adsorption energies of the additives on Fe(110)surface are determined by the van der Waals forces, while adsorptions on Fe2O3(001)surface are significantly attributed to the electrostatic attractive forces.Higher values of adsorption energy of the additives on Fe2O3(001)surface indicate that the additive has more efficient lubricity enhancing properties. | Luo Hui Fan Weiyu Li Yang Zhao Pinhui Nan Guozhi | 2013 | China Petroleum Processing & Petrochemical Technology2013,15,3: | 4
Inhibition of Axinl in osteoblast precursor cells leads to defects in postnatal bone growth through suppressing osteoclast formation显示文摘Axinl is a negative regulator of β-catenin signaling and its role in osteoblast precursor cells remains undefined. In the present studies,we determined changes in postnatal bone growth by deletion of Axinl in osteoblast precursor cells and analyzed bone growth in newborn and postnatal Axin1 O5X mice and found that hypertrophic cartilage area was largely expanded in AxinlOSX KO mice.A larger number of chondrocytes and unabsorbed cartilage matrix were found in the bone marrow cavity of Axin1OSX KO mice.Osteoclast formation in metaphyseal and subchondral bone areas was significantly decreased, demonstrated by decreased TRAPpositive cell numbers, associated with reduction of MMP9-and cathepsin K-positive cell numbers in Axin1 O5X KO mice.OPG expression and the ratio of O p g to Rankl were significantly increased in osteoblasts of Axinl O5X KO mice.Osteoclast formation in primary bone marrow derived microphage(BMM)cells was significantly decreased when BMM cells were cultured with conditioned media(CM)collected from osteoblasts derived from Axin1OSX mice compared with BMM cells cultured with CM derived from WT mice. Thus, the loss of Axinl in osteoblast precursor cells caused increased OPG and the decrease in osteoclast formation, leading to delayed bone growth in postnatal Axin1°sx KO mice. | Bing Shu Yongjian Zhao Shitian Zhao Haobo Pan Rong Xie Dan Yi Ke Lu Junjie Yang Chunchun Xue Jian Huang Jing Wang Dongfeng Zhao Guozhi Xiao Yongjun Wang Di Chen | 2020 | Bone Research2020,8,3: | 3

Long noncoding RNA(lncRNA)H19:An essential developmental regulator with expanding roles in cancer, stem cell differentiation, and metabolic diseases显示文摘Recent advances in deep sequencing technologies have revealed that, while less than 2%of the human genome is transcribed into mRNA for protein synthesis, over 80%of the genome is transcribed, leading to the production of large amounts of noncoding RNAs(ncRNAs).It has been shown that ncRNAs,especially long non-coding RNAs(lncRNAs),may play crucial regulatory roles in gene expression. As one of the first isolated and reported lncRNAs,H19 has gained much attention due to its essential roles in regulating many physiological and/or pathological processes including embryogenesis,development,tumorigenesis,osteogen-esis,and metabolism.Mechanistically,H19 mediates diverse regulatory functions by serving as competing endogenous RNAs(CeRNAs),Igf2/H19 imprinted tandem gene, modular scaffold, cooperating with H19 antisense, and acting directly with other mRNAs or lncRNAs.Here,we summarized the current understanding of H19 in embryogenesis and development, cancer development and progression, mesenchymal stem cell lineage-specific differentiation, and metabolic diseases. We discussed the potential regulatory mechanisms underlying H19’s func-tions in those processes although more in-depth studies are warranted to delineate the exact molecular, cellular, epigenetic, and genomic regulatory mechanisms underlying the physiolog-ical and pathological roles of H19.Ultimately,these lines of investigation may lead to the development of novel therapeutics for human diseases by exploiting H19 functions. | Junyi Liao Bowen Chen Zhenglin Zhu Chengcheng Du Shengqiang Gao Guozhi Zhao Piao Zhao Yonghui Wang Annie Wang Zander Schwartz Lily Song Jeffrey Hong William Wagstaff Rex CHaydon Hue HLuu Jiaming Fan Russell RReid Tong-Chuan He Lewis Shi Ning Hu Wei Huang | 2023 | Genes & Diseases2023,10,4: | 1

Geochemistry, U-Pb, Hf isotopic characteristics and geological significance of Zhalaxiageyong trachydacite in Tuotuohe area, Qinghai显示文摘The Zhalaxiageyong lead-zinc-copper polymetallic deposit is a typical porphyry deposit of the Tuotuohe area. Whole-rock geochemical analyses,Zircon U-Pb dating and Hf isotope analysis are undertaken for the ore host trachydacite with the aim of constraining its petrogenesis, magma source and regional tectonic setting.LA-ICP-MS zircon U-Pb dating indicates that the trachydacite was formed in 32. 68 ± 0. 50 Ma( MSWD =1. 6),i. e.,Oligocene. The trachydacite is rich in potassium and poor in Mg#( 5. 10-9. 70),belonging to the peraluminous shoshonite series. The rocks are enriched in LILE( large ion lithophile elements) Rb,Ba,K and LREE,depleted in HFSE( high field strength elements) Nb,Ta,P,Ti,with high Sr and low Y and Yb,having the characteristics of the C type adakite. It is calculated that the initial εHf( t) of the zircons range from-0. 92 to 2. 07 and their two-stage Hf model ages T_(DM2) range from 978 Ma to 1 169 Ma. The magma source should be mainly the partially melt mafic rocks of the thickened Middle Neoproterozoic lower crust of the Northern Qiangtang massif with the addition of ancient aluminosilica material in the melting process. The rocks formed in the tectonic setting of delamination of lithosphere and extension of the thickened crust. During the period of 40-32 Ma,large-scale potassium rich alkaline magmatism occurred in this area. The porphyry metallogenesis is related to the magmatic activities in this period. | ZHANG Junyi LI Bile ZHAO Guoquan NING Chuanqi SUN Jing WANG Guozhi | 2017 | Global Geology2017,20,3: | 1
SV40 large T antigen-induced immortalization reprograms mouse cardiomyocyte progenitors with mesenchymal stem cell characteristics and osteogenic potential显示文摘While progenitor cell-based cardiomyocyte regeneration holds great promise of repairing an injured heart, primary cardiomyogenic progenitors(CPs)have a limited life span in culture, hampering the use of CPs for in vitro and in vivo studies. We previously isolated primary CPs from mouse E15.5 fetal heart, and reversibly immortalized them with SV40 large T antigen(SV40 LTA),resulting in immortalized CPs(iCPs),which maintain long-term proliferation and ex-press cardiomyogenic markers and retain differentiation potential under appropriate differentiation conditions. | Yichun Yu Jiamin Zhong Connie Chen Yannian Gou Guozhi Zhao Piao Zhao Yonghui Wang Wei Zeng Annie Wang William D.Wagstaff Jr Rex C.Haydon Tong-Chuan He Russell R.Reid Michael J.Lee Hue H.Luu Jiaming Fan | 2023 | Genes & Diseases2023,10,4: | 0

Niclosamide(NA)overcomes cisplatin resistance in human ovarian cancer显示文摘Ovarian cancer(OC)is one of the most lethal malignancies of the female reproduc-tive system.OC patients are usually diagnosed at advanced stages due to the lack of early diag-nosis.The standard treatment for OC includes a combination of debulking surgery and platinum-taxane chemotherapy, while several targeted therapies have recently been approved for maintenance treatment. The vast majority of OC patients relapse with chemoresistant tu-mors after an initial response. Thus, there is an unmet clinical need to develop new therapeu-tic agents to overcome the chemoresistance of OC.The anti-parasite agent niclosamide(NA)has been repurposed as an anti-cancer agent and exerts potent anti-cancer activities in human cancers including OC.Here,we investigated whether NA could be repurposed as a therapeutic agent to overcome cisplatin-resistant(CR)in human OC cells. To this end,we first established two CR lines SKOV3CR and OVCAR8CR that exhibit the essential biological characteristics of cisplatin resistance in human cancer. We showed that NA inhibited cell proliferation,sup-pressed cell migration, and induced cell apoptosis in both CR lines at a low micromole range.Mechanistically,NA inhibited multiple cancer-related pathways including AP1,ELK/SRF,HIF1,and TCF/LEF,in SKOV3CR and OVCAR8CR cells.NA was further shown to effectively inhibit xenograft tumor growth of SKOV3CR cells.Collectively, our findings strongly suggest that NA may be repurposed as an efficacious agent to combat cisplatin resistance in chemoresistant hu-man OC,and further clinical trials are highly warranted. | Linjuan Huang Jing Zhang Youling Deng Hao Wang Piao Zhao Guozhi Zhao Wei Zeng Yonghui Wang Connie Chen William Wagstaff Rex C.Haydon Russell R.Reid Tong-Chuan He Le Shen Hue H.Luu Ling Zhao | 2023 | Genes & Diseases2023,10,4: | 0
31p NMR study of magnetic phase transitions of MnP single crystal under 2 GPa pressure显示文摘Superconductivity on the border of the long-range magnetic order has been discovered in Mn P under high pressures. In order to investigate the nature of the magnetic properties adjacent to the superconducting state, we performed zero-field 31 P NMR for Mn P single crystal under ambient and hydrostatic pressure of 2 GPa, respectively. Radio frequency power level was used to determine whether NMR signal originates from a helical state or not. When 2 GPa pressure was applied, the signal from helical state exists even above 160 K, while that from the ferromagnetic phase was not observed. Our NMR results indicate that the magnetic phase which is adjacent to the superconducting state is in a helical magnetic structure. | GuoZhi Fan Bo Zhao Wei Wu Ping Zheng JianLin Luo | 2016 | Science China(Physics,Mechanics & Astronomy)2016,59,5: | 0
Study on time-resolved fluorescence dynamics of cyanine dye sensitizing AgBr显示文摘The fluorescence spectra of three different dyes adsorbed on the tabular and cubic AgBr microcrystals are obtained by the picosecond time-resolved streak camera technique. The dependence of the ultrafast electron transferring from dye-aggre-gates to the conduction band of AgBr and the efficiency of spectral sensitization on different kinds of dyes with different concentrations is analyzed. Further more, the microcosmic mechanism of the sensitization process is discussed. It is found that the fluorescence decay curves are fitted very well by the double exponential func-tion,consisting of a slow component and a fast one with large amplitude. We con-sider this fast one mainly attributable to the electron transfer from dye J-aggre-gates to the conduction band of AgBr. | YANG ShaoPeng FAN ShanShan LI ChunLei FAN GuoZhi MENG Tao LI XiaoWei FU GuangSheng AI XiCheng ZHAO XiaoHui YE JianPing WANG LingXuan | 2008 | Science China(Physics,Mechanics & Astronomy)2008,51,3: | 0
Engineered nucleus-free mesenchymal stem cells (MSCs) for the targeted delivery of therapeutics to disease site显示文摘Specialized therapeutic delivery, or use of pharmaceuticals and other biomaterials to target specific parts of the body or diseased tissue, has long been sought as an ideal way of treating human diseases. A recent article published in Nature Biomedical Engineering revealed an innovative strategy to engineer nucleus-free human mesenchymal stem cells (MSCs) for targeted delivery of therapeutics to disease site.1 MSCs have emerged as promising vehicles of therapeutic delivery.2,3 MSCs are undifferentiated pluripotent stem cells derived from areas such as bone marrow and adipose tissue.4,5 MSCs are sought after for their chemotaxis, or ability to home towards a chemical stimulus, and capacity for modification with elements such as chemoattractant receptors and adhesion molecules.1 These properties allow for site-specific and minimally-invasive therapeutic administration and treatment. | Zander Schwartz Piao Zhao Annie Wang Guozhi Zhao Wei Zeng Yonghui Wang Hue H. | 18 | miR-204 ameliorates osteoarthritis pain by inhibiting SP1-LRP1 signaling and blocking neuro-cartilage interaction显示文摘Osteoarthritis(OA)is a painful degenerative joint disease and is the leading cause of chronic disability among elderly individuals. To improve the quality of life for patients with OA,the primary goal for OA treatment is to relieve the pain.During OA progression, nerve ingrowth was observed in synovial tissue and articular cartilage. These abnormal neonatal nerves act as nociceptors to detect OA pain signals. The molecular mechanisms for transmitting OA pain in the joint tissues to the central nerve system(CNS)is currently unknown.MicroRNA miR-204 has been demonstrated to maintain the homeostasis of joint tissues and have chondro-protective effect on OA pathogenesis. However, the role of miR-204 in OA pain has not been determined. In this study,we investigated interactions between chondrocytes and neural cells and evaluated the effect and mechanism of miR-204 delivered by exosome in the treatment of OA pain in an experimental OA mouse model. Our findings demonstrated that miR-204 could protect OA pain by inhibition of SP1-LDL Receptor Related Protein 1(LRP1)signaling and blocking neuro-cartilage interaction in the joint. Our studies defined novel molecular targets for the treatment of OA pain. | Ke Lu Qingyun Wang Liuzhi Hao Guizheng Wei Tingyu Wang William WLu Guozhi Xiao Liping Tong Xiaoli Zhao Di Chen | 2023 | Bioactive Materials2023,,8: | 0
The evolving roles of Wnt signaling in stem cell proliferation and differentiation, the development of human diseases, and therapeutic opportunities显示文摘The evolutionarily conserved Wnt signaling pathway plays a central role in develop-ment and adult tissue homeostasis across species.Wnt proteins are secreted,lipid-modified signaling molecules that activate the canonical(β-catenin dependent)and non-canonical(β-catenin independent)Wnt signaling pathways.Cellular behaviors such as proliferation,differ-entiation,maturation,and proper body-axis specification are carried out by the canonical pathway, which is the best characterized of the known Wnt signaling paths.Wnt signaling has emerged as an important factor in stem cell biology and is known to affect the self-renewal of stem cells in various tissues. This includes but is not limited to embryonic, hematopoietic, mesenchymal, gut, neural, and epidermal stem cells.Wnt signaling has also been implicated in tumor cells that exhibit stem cell-like properties.Wnt signaling is crucial for bone formation and presents a potential target for the development of therapeutics for bone disorders. Not surprisingly, aberrant Wnt signaling is also associated with a wide variety of diseases, including cancer.Mutations of Wnt pathway members in cancer can lead to unchecked cell proliferation,epithelial-mesenchymal transition, and metastasis.Altogether, advances in the understand-ing of dysregulated Wnt signaling in disease have paved the way for the development of novel therapeutics that target components of the Wnt pathway.Beginning with a brief overview of the mechanisms of canonical and non-canonical Wnt,this review aims to summarize the cur-rent knowledge of Wnt signaling in stem cells, aberrations to the Wnt pathway associated with diseases, and novel therapeutics targeting the Wnt pathway in preclinical and clinical studies. | Michael Yu Kevin Qin Jiaming Fan Guozhi Zhao Piao Zhao Wei Zeng iconnie Chen Annie Wang Yonghui Wang jiamin Zhong Yi Zhu William Wagstaff Rex C.Haydon Hue H.Luu Sherwin Ho Michael J.Lee Jason Strelzow Russell R.Reid Tong-Chuan He | 2024 | Genes & Diseases2024,11,3: | 0
Canonical and noncanonical Wnt signaling: Multilayered mediators, signaling mechanisms and major signaling crosstalk显示文摘Wnt signaling plays a major role in regulating cell proliferation and differentiation. The Wnt ligands are a family of 19 secreted glycoproteins that mediate their signaling effects via binding to Frizzled receptors and LRP5/6 coreceptors and transducing the signal either throughβ-catenin in the canonical pathway or through a series of other proteins in the nonca-nonical pathway. Many of the individual components of both canonical and noncanonical Wnt signaling have additional functions throughout the body, establishing the complex interplay between Wnt signaling and other signaling pathways. This crosstalk between Wnt signaling and other pathways gives Wnt signaling a vital role in many cellular and organ processes.Dys-regulation of this system has been implicated in many diseases affecting a wide array of organ systems, including cancer and embryological defects, and can even cause embryonic lethality. The complexity of this system and its interacting proteins have made Wnt signaling a target for many therapeutic treatments. However, both stimulatory and inhibitory treatments come with potential risks that need to be addressed. This review synthesized much of the current knowl-edge on the Wnt signaling pathway, beginning with the history of Wnt signaling. It thoroughly described the different variants of Wnt signaling, including canonical, noncanonical Wnt/PCP,and the noncanonical Wnt/Ca2+pathway.Further description involved each of its components and their involvement in other cellular processes. Finally, this review explained the various other pathways and processes that crosstalk with Wnt signaling. | Kevin Qin Michael Yu Jiaming Fan Hongwei Wang Piao Zhao Guozhi Zhaoo Wei Zeng Connie Chen Yonghui Wang Annie Wang Zander Schwartz Jeffrey Hong Lily Song William Wagstaff Rex C.Haydon Hue H.Luu Sherwin H.Ho Jason Strelzow Russell R.Reid Tong-Chuan He Lewis L.Shi | 2024 | Genes & Diseases2024,11,1: | 0
