| 2 | Unbiased subgenome evolution following a recent whole-genome duplication in pear(Pyrus bretschneideri Rehd.)显示文摘Genome fractionation(also known as diploidization)frequently occurs following paleopolyploidization events.Biased fractionation between subgenomes has been found in some paleo-allopolyploids,while this phenomenon is absent in paleo-autopolyploids.Pear(Pyrus bretschneideri Rehd.)experienced a recent whole-genome duplication(WGD,~30 million years ago);however,the evolutionary fate of the two subgenomes derived from this WGD event is not clear.In this study,we identified the two paleo-subgenomes in pear using peach(Prunus persica)as an outgroup and investigated differences in the gene loss rate,evolutionary rate,gene expression level,and DNA methylation level between these two subgenomes.Fractionation bias was not found between the two pear subgenomes,which evolved at similar evolutionary rates.The DNA methylation level of the two subgenomes showed little bias,and we found no expression dominance between the subgenomes.However,we found that singleton genes and homeologous genes within each subgenome showed divergent evolutionary patterns of selective constraints,expression and epigenetic modification.These results provide insights into subgenome evolution following paleopolyploidization in pear. | Qionghou Li Xin Qiao Hao Yin Yuhang Zhou Huizhen Dong Kaijie Qi Leiting Li Shaoling Zhang | 2019 | Horticulture Research2019,6,1: | 4 |

| 4 | The Variation of Stone Cell Content in 236 Germplasms of Sand Pear(Pyrus pyrifolia)and Identification of Related Candidate Genes显示文摘Stone cells have been described to substantially influence pear fruit quality,as lignin and cellulose are the main components of stone cells.However,there are limited studies on the relationship between the variation and molecular basis of stone cells,lignin and cellulose content among different pear varieties.Here,to reveal the variation of stone cell content within different cultivated species,we collected 236 germplasms of sand pear(Pyrus pyrifolia)at 50 days after flower blooming(DAFB),the key stage of stone cell formation.In our results,we measured the content of stone cells,lignin and cellulose and found that these contents ranged from2.82%to 29.00%,8.84%to 55.30%and 11.52%to 30.55%,respectively.Further analysis showed that the variation coefficient of stone cell,lignin and cellulose content was 39.10%,28.03%and 16.71%,respectively.Additionally,a significant correlation between stone cell,lignin and cellulose content were detected,and the correlation coefficient between the contents of stone cell and lignin(0.912)was higher than between the contents of stone cell and cellulose(0.796).Moreover,the average lignin content(29.73%)was higher than the average cellulose content(18.03%)in stone cells in pear fruits,indicating that lignin is the main component of stone cell in pears.Finally,on the basic of the transcriptome data,we identified 10 transcription factors belonging to bHLH,ERF,MYB,and NAC transcript families,which might be involved in lignin formation in stone cells.qRT-PCR experiments verified coincident trends between expression of candidate genes and stone cell content.This research laid foundation for future studies on genetic variation of stone cells in pear fruits and provided important gene resources for stone cell regulation. | Jiaying Zhang Jiaming Li Cheng Xue Runze Wang Mingyue Zhang Kaijie Qi Jing Fan Hongju Hu Shaoling Zhang Jun Wu | 2021 | Horticultural Plant Journal2021,7,2: | 3 |
| 5 | The involvement of PybZIPa in light-induced anthocyanin accumulation via the activation of PyUFGT through binding to tandem G-boxes in its promoter显示文摘To gain insight into how anthocyanin biosynthesis is controlled by light in fruit,transcriptome and metabolome analyses were performed in the Chinese sand pear cultivar“Mantianhong”(Pyrus pyrifolia)after bagging and bag removal.We investigated transcriptional and metabolic changes and gene-metabolite correlation networks.Correlation tests of anthocyanin content and transcriptional changes revealed that 1,530 transcripts were strongly correlated with 15 anthocyanin derivatives(R2>0.9,P-value<0.05),with the top 130 transcripts categorized as being associated with flavonoid metabolism,transcriptional regulation,and light signaling.The connection network revealed a new photosensitive transcription factor,PybZIPa,that might play an important role during light-induced anthocyanin accumulation.The overexpression of PybZIPa promoted anthocyanin accumulation in pear and strawberry fruit as well as tobacco leaves.Dual luciferase and Y1H assays further verified that PybZIPa directly activated the expression of PyUFGT by binding to tandem G-box motifs in the promoter,which was key to differential anthocyanin accumulation in debagged pear skin,and the number of G-box motifs affected the transcriptional activation of PyUFGT by PybZIPa.The results indicate that the light-induced anthocyanin biosynthesis regulatory mechanism in pear differs from that described in previous reports suggesting that a bZIP family member co-regulates anthocyanin biosynthesis with other transcription factors in apple and Arabidopsis.It was found that,in response to light,PybZIPa promoted anthocyanin biosynthesis by regulating important transcription factors(PyMYB114,PyMYB10,and PyBBX22)as well as structural genes(PyUFGT)via binding to G-boxes within promoters.This activation was amplified by the self-binding of PybZIPa to activate its own promoter.Overall,we demonstrate the utility of a multiomics integrative approach for discovering new functional genes and pathways underlying light-induced anthocyanin biosynthesis. | Hainan Liu Jun Su Yangfan Zhu Gaifang Yao Andrew C.Allan Charles Ampomah-Dwamena Qun Shu Kui Lin-Wang Shaoling Zhang Jun Wu | 2019 | Horticulture Research2019,6,1: | 3 |

| 7 | Treatment of PC-3 cells with ultrasound combined with microbubbles induces distinct alterations in the expression of Bcl-2 and Bax显示文摘The objective of the present study was to investigate whether ultrasound combined with microbubbles induces apoptotic cell death in androgen-independent prostate cancer cells and to identify the probable mechanism.We used ultrasound in continuous wave mode with a frequency of 21 kHz and a spatial-average temporal-average intensity of 46 mW/cm2.Ultrasound combined with microbubbles(200μL SonoVue)was used to treat androgen-independent human prostate cancer PC-3 cells for 30 s.PC-3 cells were divided into three groups:the control group,the ultrasound group and the ultrasound combined with microbubbles group.Immediately after treatment,trypan blue exclusion was used to assess cell viability.Cell apoptosis at 24 h after treatment was measured using transmission electron microscopy and flow cytometry.Western blotting was used to evaluate the expression of the apoptosis-related proteins,Bcl-2 and Bax.Ultrasound combined with microbubbles had a minimal effect on the viability of PC-3cells and induced minimal levels of cell lysis.The level of apoptosis in PC-3 cells induced by this modality was significantly higher than in controls(12.77±0.31%vs.2.56±0.22%,P<0.01).Treatment with ultrasound combined with microbubbles increased the expression of Bax,a pro-apoptotic protein,and decreased the expression of Bcl-2,an anti-apoptotic protein.It was concluded that ultrasound combined with microbubbles induces apoptotic cell death in human prostate cancer PC-3 cells through down-regulation of Bcl-2 and up-regulation of Bax. | BAI WenKun YANG ShaoLing SHEN E ZHANG JiZhen SHEN ZhiYong HU Bing | 2013 | Chinese Science Bulletin2013,58,28: | 2 |
| 8 | REMOTE SENSING OF ALGAL BLOOMS USING A TURBIDITY-FREE FUNCTION FOR NEAR-INFRARED ANDRED SIGNALS显示文摘This article presents a method for real- time mapping of algal blooms in turbid coastal waters using the remote sensing reflectance of red band (Channel 1, 580-680 nm) and near-infrared band (Channel 2, 720-1100 nm) of the AVHRR sensor on the NOAA series satellites. A turbidity-free function for near-infrared and red signals, α0 = (bb(1)/bb(2))(a(2)/ a(1)) based on the first order bb/(a+bb) model deduc-ing equation Rrs(2)?1 = α0 Rrs(1) ?1+ g?1 (1?α0), were se-lected as a chlorophyll-a related index for detecting algal blooms, and the algal blooms with chlorophyll-a concentration of 64-256 mg/L could be defined by window of 1.6 < α0 < 5.2 and 0.01< Rrs(2)/g < 0.2. Such turbidity-free two-band method is supported by both sea-truth data and remote sensing experiment for an algal blooms event on the near-shore water off the Minjiang estuary of southeastern China during early June of 2003. | 18 | Phylogenetic and Expression Analyses of With-No-Lysine Kinase Genes Reveal Novel Gene Family Diversity in Fruit Trees显示文摘With-No-Lysine kinases(WNK) have been reported to be associated with plant growth regulation in Arabidopsis, soybean, and rice, but little is known of their roles in fruits. In this study, a total of 114 WNK genes were identified from 8 fruit trees species, and these WNK genes belonged to 2 classes(I and II) that respectively contain 4(A, B, C, D) and 2 groups(E, F). The WNK genes had variable exon-intron structures and were randomly distributed among most chromosomes of each genus. The expression levels of six of the 18 WNK genes in peach were almost undetectable in fruits, suggesting that they may not be associated with fruit development and ripening. Of the other 12 WNK genes, PpWNK.B1 exhibited stronger stability of expression levels than the best reference genes reported previously in fruits from two peach cultivars. These results suggested that PpWNK.B1 could be a reliable reference gene for gene expression studies of peach fruit. Moreover, qRT-PCR assays yielded evidence that when using single reference gene as internal control, mistakes were more possible, indicating that the use of three reference genes is necessary for gene expression studies. Based on the gene expression profiles validated by qRT-PCR, PpWNK.A1 is predicted to likely be involved in fruit ripening, while PpWNK.A2 and PpWNK.E3.1 are likely associated with early fruit development.
