1Protective effect of naringenin on acetic acid-induced ulcerative colitis in rats显示文摘AIM:To evaluate the ameliorative effect of naringenin(NG)during ulcerative colitis(UC)in rats.METHODS:Rats were treated with three different doses(25,50 and 100 mg/kg per day)of NG and a single dose of mesalazine(MES,300 mg/kg per day)for seven days prior to ulcerative colitis induction by4%acetic acid(AA).Twenty four hours after AA rectal administration,animals were scarified and the colonic tissues were dissected.Colonic mucus content was estimated using Alcian blue dye binding technique.In colon tissues,levels of total glutathione sulphadryls(T-GSH),non-protein sulphadryls(NP-SH)and thiobarbituric acid reactive substances(TBARS)were evaluated.The activities of the antioxidant enzymes,catalase(CAT)and superoxide dismutase(SOD)were measured.Concentrations of nucleic acids(DNA and RNA)and total protein were also estimated in colon tissues.Colonic levels of tumor necrosis factor-(TNF-),interleukin-1(IL-1),interleukin-6(IL-6),prostaglandin E2(PGE2)and nitric oxide(NO)were estimated.In cross section of colitis tissue the histopathological changes were observed.RESULTS:Colonic mucus content was decreased in AA compared to controls(587.09±65.59 mg/kg vs941.78±68.41 mg/kg,P<0.001).AA administration markedly reduced T-GSH(5.25±0.37 nmol/L vs 3.04±0.24 nmol/L,P<0.01),NP-SH(3.16±0.04 nmol/L vs 2.16±0.30 nmol/L,P<0.01),CAT(6.77±0.40 U/mg vs 3.04±0.2 U/mg,P<0.01)and SOD(3.10±0.11U/mg vs 1.77±0.18 U/mg,P<0.01)while TBARS,TNF-,IL-1,IL-6,PGE2 and NO levels(15.09±3.84nmol/L vs 59.90±16.34 nmol/L,P<0.01;113.56±1.91 pg/mg vs 134.24±4.77 pg/mg,P<0.01;209.20±36.38 pg/mg vs 422.19±31.47 pg/mg,P<0.01;250.83±25.09 pg/mg vs 638.58±115.9 pg/mg,P<0.01;248.19±36.98 pg/mg vs 541.74±58.34 pg/mg,P<0.01 and 81.26±2.98 mmol/g vs 101.90±10.73 mmol/g,P<0.001)were increased in colon of rats with UC compared controls respectively.Naringenin supplementation,significantly and dose dependently increased the colonic mucus content.The elevated TBARS levels were significantly decreased(39.35±5.86nmol/L,P<0.05;26.74±3.17 nmol/L,P<0.01 nmol/L and 17.74±2.69 nmol/L,P<0.01)compared to AA(59.90±16.34 nmol/L)group while the decreased levels of T-GSH and NP-SH and activities of CAT and SOD found increased by NG treatments in dose dependent manner.The decreased values of nucleic acids and total protein in AA group were also significantly(P<0.01)increased in all three NG supplemented groupsrespectively.NG pretreatment inhibited the TNF-levels(123.76±3.76 pg/mg,122.62±3.41 pg/mg and121.51±2.61 pg/mg vs 134.24±4.78 pg/mg,P<0.05)compared to AA group,respectively.Interleukins,IL-1 and IL-6 levels were also decreased in NG50+AA(314.37±16.31 pg/mg and 292.58±23.68 pg/mg,P<0.05)and NG100+AA(416.72±49.62 pg/mg and 407.96±43.87 pg/mg,P<0.05)when compared to AA(352.46±8.58 pg/mg and 638.58±115.98pg/mg)group.Similar decrease(P<0.05)was seen in PGE2and NO values when compared to AA group.The group pretreated with MES,as a reference drug,showed significant(P<0.01)protection against the changes induced in colon tissue by AA administration respectively.CONCLUSION:In present study,NG produced antioxidant and anti-inflammatory effects demonstrating protective effect in inflammatory bowel disease.Salim S Al-Rejaie Hatem M Abuohashish Maher M Al-Enazi Abdullah H Al-Assaf Mihir Y Parmar Mohammed M Ahmed 2013World Journal of Gastroenterology2013,19,34:20
2Sensitive and rapid determination of amantadine without derivatization in human plasma by LC–MS/MS for a bioequivalence study显示文摘A highly sensitive, rapid and rugged liquid chromatography-tandem mass spectrometry(LC-ESI-MS/MS)method was developed for reliable estimation of amantadine(AMD), an antiviral drug in human plasma.The analyte and internal standard(IS), amantadine-d6(AMD-d6), were extracted from 200 m L plasma by solid phase extraction on Phenomenex Strata-X-C 33 m cartridges. Chromatography was performed on Synergi? Hydro-RP C18(150 mm ? 4.6 mm, 4 mm) analytical column using a mixture of acetonitrile and10 m M ammonium formate, p H 3.0(80:20, v/v) as the mobile phase. Detection and quantitation was done by multiple reaction monitoring in the positive ionization mode for AMD(m/z 152.1-135.1) and IS(m/z 158.0-141.1) on a triple quadrupole mass spectrometer. The assay was linear in the concentration range of 0.50–500 ng/m L with correlation coefficient(r2) Z 0.9969. The limit of detection of the method was 0.18 ng/m L. The intra-batch and inter-batch precisions were r 5.42% and the accuracy varied from98.47% to 105.72%. The extraction recovery of amantadine was precise and quantitative in the range of97.89%–100.28%. IS-normalized matrix factors for amantadine varied from 0.981 to 1.012. The stability of AMD in whole blood and plasma was evaluated under different conditions. The developed method was successfully applied for a bioequivalence study with 100 mg of AMD in 32 healthy volunteers. 6Quantitation of tadalafil in human plasma using a sensitive and rapid LC-MS/MS method for a bioequivalence study显示文摘A highly sensitive liquid chromatography-tandem mass spectrometry(LC-MS/MS) method was developed for the determination of tadalafil(TAD) in human plasma. TAD and its deuterated internal standard(IS), tadalafil-d3, were extracted from 200 mL plasma using Phenomenex Strata-X-C 33 m extraction cartridges. Chromatographic analysis was carried out on Synergi? Hydro-RP C18(100 mm ? 4.6 mm, 4 mm)column with a mobile phase consisting of methanol and 10 m M ammonium formate, p H 4.0(90:10, v/v),delivered at a flow rate of 0.9 m L/min. Quantitation of the protonated analyte was done on a triple quadrupole mass spectrometer using multiple reaction monitoring via electrospray ionization. The precursor to product ions transitions monitored for TAD and TAD-d3 were m/z 390.3-268.2 and m/z393.1-271.2, respectively. The calibration curve was linear over the concentration range of 0.50–500 ng/m L with correlation coefficient, r2 Z 0.9994. Acceptable intra-batch and inter-batch precision(r3.7%) and accuracy(97.8% to 104.1%) were obtained at five concentration levels. The recovery of TAD from spiked plasma was highly precise and quantitative(98.95% to 100.61%). Further, the effect of endogenous matrix components was minimal. TAD was found to be stable under different storage conditions in human plasma and also in whole blood samples. The validated method was successfully used to determine TAD plasma concentration in a bioequivalence study with 20 mg TAD tablets in 24 healthy volunteers. 11Phylogeny and taxonomy of Afrocayratia,a new genus of Vitaceae from continental Africa and Madagascar显示文摘The genus Cayratia Juss.in the traditional sense(i.e.,Cayratia s.l.)of the grape family has been shown to be non-monophyletic.Previous studies supported the splitting of Cayratia s.l.into three genera,that is,Cayratia s.s.,Causonis Raf.,and a new genus representing the African Cayratia clade.However,the morphology of the African Cayratia clade has not been studied carefully and its phylogenetic position within Vitaceae remains unclear.Our study integrates molecular,distributional,and morphological data and supports the recognition of the new genus Afrocayratia from continental Africa and Madagascar.Phylogenetic analyses strongly support the monophyly of Afrocayratia and resolve it as a sister of Cayratia s.s.based on the chloroplast data,but it is placed sister to Cyphostemma based on the internal transcribed spacer dataset.Molecular dating suggests that Afrocayratia split with Cayratia s.s.during the Paleocene,but that the extant species of Afrocayratia did not diversify until the early Miocene.Afrocayratia differs from its allied genera in having short stigmas and seeds with subcircular ventral infold cavities in cross-section.Three clades are detected within Afrocayratia,with A.debilis(Baker)J.Wen&L.M.Lu as the first diverged lineage.The second diverged lineage includes A.delicatula(Willems)J.Wen&Z.D.Chen and A.gracilis(Guill.&Perr.)J.Wen&Z.D.Chen.The third diverged lineage includes A.imerinensis(Baker)J.Wen&L.M.Lu,A.longiflora(Desc.)J.Wen&Rabarijaona,and A.triternata(Baker)J.Wen&Rabarijaona from Madagascar,which form a monophyletic group that diverged from the second lineage in the middle Miocene.Combining the morphological and molecular evidence,we formally describe the new genus Afrocayratia,make seven new combinations,and provide a key to species of the genus.Romer Narindra Rabarijaona Viet-Cuong Dang Gaurav Parmar Bing Liu Jun Wen Zhi-Duan Chen Li-Min Lu 2020Journal of Systematics and Evolution2020,58,6:2

13Self nano-emulsifying drug delivery system for Embelin:Design,characterization and in-vitro studies显示文摘CThe objective of the present study was to prepare solid self-nanoemulsifying drug delivery system(S-SNEDDS)containing Capryol-90 as oil phase for the delivery of Embelin,a poorly water soluble herbal active ingredient.Box-Behnken experimental design was employed to optimise the formulation variables,X1(amount of oil;Capryol 90),X2(amount of surfactant;Acrysol EL 135)and X3(amount of co-surfactant;PEG 400).Systems were appraised for visual characteristics for self emulsifying time,globule size and drug release.Optimised liquid formulations were formulated into free flowing granules(S-SNEDDS)by adsorption on the porous materials like Aerosil 200 and Neusilin and thereby compressed into tablet.In vitro dissolution studies of SNEDDS revealed increased in the dissolution rate of the drug.FT-IR data revealed no physicochemical interaction between drug and excipients.Solid state characterization of S-SNEDDS by DSC and Powder XRD confirmed reduction in drug crystallinity which further supports the results of dissolution studies.TEM analysis exhibited spherical globules.Further,the accelerated stability studies for 6 months revealed that S-SNEDDS of Embelin are found to be stable without any significant change in physicochemical properties.Thus,the present studies demonstrated dissolution enhancement potential of porous carrier based S-SNEDDS for poorly water soluble herbal active ingredient,Embelin.Komal Parmar Jayvadan Patel Navin Sheth 2015Asian Journal of Pharmaceutical Sciences2015,10,5:2
