1Kinsenoside attenuates osteoarthritis by repolarizing macrophages through inactivating NF-κB/MAPK signaling and protecting chondrocytes显示文摘The objective was to investigate the effect of kinsenoside(Kin) treatments on macrophage polarity and evaluate the resulting protection of chondrocytes to attenuate osteoarthritis(OA) progression.RAW264.7 macrophages were polarized to M1/M2 subtypes then administered with different concentrations of Kin. The polarization transitions were evaluated with quantitative real-time polymerase chain reaction(q RT-PCR), confocal observation and flow cytometry analysis. The mechanism of Kin repolarizing M1 macrophages was evaluated by Western blot. Further, macrophage conditioned medium(CM) and IL-1β were administered to chondrocytes. Micro-CT scanning and histological observations were conducted in vivo on anterior cruciate ligament transection(ACLT) mice with or without Kin treatment. We found that Kin repolarized M1 macrophages to the M2 phenotype. Mechanistically, Kin inhibited the phosphorylation of IκBα, which further reduced the downstream phosphorylation of P65 in nuclear factor-κB(NF-κB) signaling. Moreover, Kin inhibited mitogen-activated protein kinases(MAPK) signaling molecules p-JNK, p-ERK and p-P38. Additionally, Kin attenuated macrophage CM and IL-1β-induced chondrocyte damage. In vivo, Kin reduced the infiltration of M1 macrophages,promoted M2 macrophages in the synovium, inhibited subchondral bone destruction and reduced articular cartilage damage induced by ACLT. 2Targeting ferroptosis suppresses osteocyte glucolipotoxicity and alleviates diabetic osteoporosis显示文摘Diabetic osteoporosis(DOP) is the leading complication continuously threatening the bone health of patients with diabetes. A key pathogenic factor in DOP is loss of osteocyte viability. However, the mechanism of osteocyte death remains unclear. Here, we identified ferroptosis, which is iron-dependent programmed cell death, as a critical mechanism of osteocyte death in murine models of DOP. The diabetic microenvironment significantly enhanced osteocyte ferroptosis in vitro, as shown by the substantial lipid peroxidation, iron overload, and aberrant activation of the ferroptosis pathway. RNA sequencing showed that heme oxygenase-1(HO-1) expression was notably upregulated in ferroptotic osteocytes. Further findings revealed that HO-1 was essential for osteocyte ferroptosis in DOP and that its promoter activity was controlled by the interaction between the upstream NRF2 and c-JUN transcription factors. Targeting ferroptosis or HO-1 efficiently rescued osteocyte death in DOP by disrupting the vicious cycle between lipid peroxidation and HO-1 activation, eventually ameliorating trabecular deterioration. 3QTG-Seq Accelerates QTL Fine Mapping through QTL Partitioning and Whole-Genome Sequencing of Bulked Segregant Samples显示文摘Deciphering the genetic mechanisms underlying agronomic traits is of great importance for crop improvement. Most of these traits are controlled by multiple quantitative trait loci (QTLs), and identifying the underlying genes by conventional QTL fine-mapping is time-consuming and labor-intensive. Here, we devised a new method, named quantitative trait gene sequencing (QTG-seq), to accelerate QTL fine-mapping. QTGseq combines QTL partitioning to convert a quantitative trait into a near-qualitative trait, sequencing of bulked segregant pools from a large segregating population, and the use of a robust new algorithm for identifying candidate genes. Using QTG-seq, we fine-mapped a plant-height QTL in maize (Zea mays L.), qPH7, to a 300-kb genomic interval and verified that a gene encoding an NF-YC transcription factor was the functional gene. Functional analysis suggested that qPH7-encoding protein might influence plant height by interacting with a CO-like protein and an AP2 domain-containing protein. Selection footprint ana卜 ysis indicated that qPH7 was subject to strong selection during maize improvement. 4Zinc alloy-based bone internal fixation screw with antibacterial and anti-osteolytic properties显示文摘There is no targeted effective treatment for patients undergoing internal fixation surgery/two-stage total joint revision surgery with a high risk of postoperative infection and osteolysis,while the rate of reoperation due to infection and osteolysis remains high.In this study,we report a pioneering application of implants made of biodegradable Zn-Ag alloy with active antibacterial and anti-osteolytic properties in three classical animal models,illustrating antibacterial,anti-osteolysis,and internal fixation for fractures.The antibacterial activity of the Zn-2Ag alloy was verified in a rat femur osteomyelitis prevention model,while the anti-osteolytic properties were evaluated using a mouse cranial osteolysis model.Moreover,the Zn-2Ag based screws showed similar performance in bone fracture fixation compared to the Ti-6Al-4V counterpart.The fracture healed completely after 3 months in the rabbit femoral condyle fracture model.Furthermore,the underlying antibacterial mechanism may include inhibition of biofilm formation,autolysis-related pathways,and antibiotic resistance pathways.Osseointegration mechanisms may include inhibition of osteoclast-associated protein expression,no effect on osteogenic protein expression,and no activation of related inflammatory protein expression.The empirical findings here reveal the great potential of Zn-Ag-based alloys for degradable biomaterials in internal fixation surgery/two-stage total joint revision surgery for patients with a high risk of postoperative infection and osteolysis.Xinhua Qu Hongtao Yang Bo Jia Minqi Wang Bing Yue Yufeng Zheng Kerong Dai 2021Bioactive Materials2021,6,12:4
5A 3D-bioprinted scaffold with doxycycline-controlled BMP2-expressing cells for inducing bone regeneration and inhibiting bacterial infection显示文摘Large bone defects face a high risk of pathogen exposure due to open wounds,which leads to high infection rates and delayed bone union.To promote successful repair of infectious bone defects,fabrication of a scaffold with dual functions of osteo-induction and bacterial inhibition is required.This study describes creation of an engineered progenitor cell line(C3H10T1/2)capable of doxycycline(DOX)-mediated release of bone morphogenetic protein-2(BMP2).Three-dimensional bioprinting technology enabled creation of scaffolds,comprising polycaprolactone/mesoporous bioactive glass/DOX and bioink,containing these engineered cells.In vivo and in vitro experiments confirmed that the scaffold could actively secrete BMP2 to significantly promote osteoblast differentiation and induce ectopic bone formation.Additionally,the scaffold exhibited broad-spectrum antibacterial capacity,thereby ensuring the survival of embedded engineered cells when facing high risk of infection.These findings demonstrated the efficacy of this bioprinted scaffold to release BMP2 in a controlled manner and prevent the occurrence of infection;thus,showing its potential for repairing infectious bone defects.Minqi Wang Hanjun Li Yiqi Yang Kai Yuan Feng Zhou Haibei Liu Qinghui Zhou Shengbing Yang Tingting Tang 2021Bioactive Materials2021,6,5:3

7近年中国北方草地变绿受降水增加的驱动显示文摘中国北方的暖湿化是近期的热点话题,然而其对植被生长的影响仍不清楚。本研究基于长时间序列(1982–2018年)数据,研究了归一化植被指数(NDVI)的时间动态及其气候驱动因子之间的关系,以探索近年来气候的暖湿化是否会导致该区植被变绿。我们采用分段回归探测了NDVI的变化趋势是否存在转变点,用Pearson相关分析描述了植被指数与气候因子的关系。最后,采用逐步多元回归方法研究了气候因子对NDVI时间变化的贡献率。研究结果表明,NDVI时间变化趋势的转变点出现在2008年,GIMMS NDVI在1982–2008年略有增加,上升速率为0.00022 yr?1,在2008–2015年上升速率达到0.002 yr?1,MODIS NDVI在2008–2018年上升速率为0.0018 yr?1。降水是NDVI变化的主要驱动因子,气温和饱和水汽压差(VPD)对NDVI的变化影响较小。总体看来,NDVI时间序列变化趋势存在转变点,并且近期气候的暖湿化主导了中国北方草地植被变绿,这为今后更好地预测该地区气候变化下的植被覆盖变化提供了依据。Kai Di Zhongmin Hu Mei Wang Ruochen Cao Minqi Liang Genan Wu Ruru Chen Guangcun Hao Yaolong Zhao 2021Journal of Plant Ecology2021,14,5:2
8The Influence of Ultrasonic Frequency on the Properties of Ni-Co Coatings Prepared by Ultrasound-assisted Electrodeposition显示文摘In this paper,Ni-Co coatings were electrodeposited onto carbon steel substrates with the aid of ultrasonic agitation.The coatings were analyzed by energy dispersive X-ray analysis(EDX),X-ray difraction analysis(XRD)and scanning electron microscopy(SEM).The efects of the ultrasonic frequency on the roughness,hardness and corrosion resistance of the Ni-Co coatings were also investigated.The results indicated that the increase of the ultrasonic frequency from 20 to 120 kHz reduced the Ni content and the grain size of Ni-Co coatings.Moreover,the phase structure of the electrodeposited coatings was influenced by the ultrasonic frequency.Under 55 kHz ultrasonic agitation,the Ni-Co coating was single fcc phase and showed the finest roughness and the strongest corrosion resistance in 5 wt.%NaCl solution at the ambient temperature.Under ultrasonic agitation with frequency of 90 kHz,the coating was a mixture of fcc and hcp structure and showed the maximal hardness of about 420 HV.Therefore,ultrasonic agitation helped decrease the roughness,and enhance hardness and corrosion resistance of Ni-Co coatings.Minqi SHENG Chenkai LV Lan HONG Mingwang SHAO Kang WAN Fan LV 2013Acta Metallurgica Sinica(English Letters)2013,26,6:2
9Computing rarity on uncertain data显示文摘The essence of uncertain data management has been well adopted since data uncertainty widely exists in lots of applications,such as Web,sensor networks,etc.Most of the uncertain data models are based on the possible world semantics.Because the number of the possible worlds will blowup exponentially with the growth of the data set,it is much more challenging to handle uncertain data than deterministic data.In this paper,we take the first attempt to study the rarity,an important statistic that describes the proportion of items with the same frequency,upon uncertain data.We have proposed three novel solutions,including an exact method and an approximate method to compute the rarity of a given frequency respectively,and a method to find the frequency of the maximum rarity.Analysis in theorem and extensive experimental results demonstrate the effectiveness and efficiency of the proposed solutions.JIN CheQing ZHOU MinQi ZHOU AoYing 2011Science China(Information Sciences)2011,54,10:1

14Polyurethane grouting materials with different compositions for the treatment of mud pumping in ballastless track subgrade beds:properties and application effect显示文摘Mud pumping in subgrade beds under ballastless tracks will deteriorate the dynamic performance of infrastructure under railway lines,reduce the smoothness of the railway lines,and seriously affect the comfort and safety of the trains.Due to their good mechanical properties,twocomponent polyurethane materials can be used for grouting to treat the fouling problems caused by ballastless track mud pumping.To develop a polyurethane formula suitable for the treatment of ballastless track mud pumping,we first performed indoor experiments to investigate the mechanical properties and gelation time of polyurethane elastomers synthesized with different raw material composition ratios,to determine an optimal composition ratio of the raw materials.Then,we conducted a dynamic field test to verify the remediation effect of the polyurethane material fabricated according to the design ratio.The results showed that polyurethane grouting material with the selected design ratios improved the contact characteristics between the surface layer of the subgrade bed and the base plate in the area,coordinating the dynamic response between the track structure and the subgrade bed.Thus,the obtained polyurethane grouting material could be used to renovate mud pumping areas of ballastless tracks with a good treatment effect.Zhichao Huang Qian Su Junjie Huang Minqi Dong Dong Li Ting Liu 2022Railway Engineering Science2022,30,2:1

16Carbon nanofibers membrane bridged with graphene nanosheet and hyperbranched polymer for high-performance osmotic energy harvesting显示文摘Reverse electrodialysis(RED),based on ion-selective membranes,is one of the most promising technologies for capturing osmotic energy.As key elements of the RED system,ion-selective membranes must meet the crucial demands of mechanical stability,anti-fouling characteristics,easy fabrication,and high power density;however,this still remains a challenge.In this study,we demonstrated a large-scale,mechanically stable,and high-porosity membrane obtained by combining carbon nanomaterials and hyperbranched polyethyleneimine(h-PEI),thereby achieving a high power density of 5.0 W·m−2 with seawater and river water.Carbon nanofibers(CNFs)were subsequently bridged with graphene and h-PEI to strengthen the interaction between the CNFs,reduce the channel size and increase the space charge density,mechanical strength,and toughness.The large-scale and mechanically stable membrane fabricated using the modified CNFs exhibited anion selectivity and high ionic conductivity,thereby achieving a high-performance osmotic energy conversion.Furthermore,the anti-fouling property of the membrane was confirmed by the stability of the osmotic energy conversion in a solution with algae,which can be attributed to the high porosity of carbon nanomaterials.This economic and convenient method for the ion-selective membrane preparation is believed to be promising for large-scale osmotic energy harvesting.Weijie Wang Jinlin Hao Qian Sun Minqi Zhao Huiyi Liu Chao Li Xin Sui 2023Nano Research2023,16,1:1
