1Kinsenoside attenuates osteoarthritis by repolarizing macrophages through inactivating NF-κB/MAPK signaling and protecting chondrocytes显示文摘The objective was to investigate the effect of kinsenoside(Kin) treatments on macrophage polarity and evaluate the resulting protection of chondrocytes to attenuate osteoarthritis(OA) progression.RAW264.7 macrophages were polarized to M1/M2 subtypes then administered with different concentrations of Kin. The polarization transitions were evaluated with quantitative real-time polymerase chain reaction(q RT-PCR), confocal observation and flow cytometry analysis. The mechanism of Kin repolarizing M1 macrophages was evaluated by Western blot. Further, macrophage conditioned medium(CM) and IL-1β were administered to chondrocytes. Micro-CT scanning and histological observations were conducted in vivo on anterior cruciate ligament transection(ACLT) mice with or without Kin treatment. We found that Kin repolarized M1 macrophages to the M2 phenotype. Mechanistically, Kin inhibited the phosphorylation of IκBα, which further reduced the downstream phosphorylation of P65 in nuclear factor-κB(NF-κB) signaling. Moreover, Kin inhibited mitogen-activated protein kinases(MAPK) signaling molecules p-JNK, p-ERK and p-P38. Additionally, Kin attenuated macrophage CM and IL-1β-induced chondrocyte damage. In vivo, Kin reduced the infiltration of M1 macrophages,promoted M2 macrophages in the synovium, inhibited subchondral bone destruction and reduced articular cartilage damage induced by ACLT. 2Targeting ferroptosis suppresses osteocyte glucolipotoxicity and alleviates diabetic osteoporosis显示文摘Diabetic osteoporosis(DOP) is the leading complication continuously threatening the bone health of patients with diabetes. A key pathogenic factor in DOP is loss of osteocyte viability. However, the mechanism of osteocyte death remains unclear. Here, we identified ferroptosis, which is iron-dependent programmed cell death, as a critical mechanism of osteocyte death in murine models of DOP. The diabetic microenvironment significantly enhanced osteocyte ferroptosis in vitro, as shown by the substantial lipid peroxidation, iron overload, and aberrant activation of the ferroptosis pathway. RNA sequencing showed that heme oxygenase-1(HO-1) expression was notably upregulated in ferroptotic osteocytes. Further findings revealed that HO-1 was essential for osteocyte ferroptosis in DOP and that its promoter activity was controlled by the interaction between the upstream NRF2 and c-JUN transcription factors. Targeting ferroptosis or HO-1 efficiently rescued osteocyte death in DOP by disrupting the vicious cycle between lipid peroxidation and HO-1 activation, eventually ameliorating trabecular deterioration. 3A review on tribology of polymer composite coatings显示文摘Self-lubricating polymer composite coatings,with tailorable tribological and mechanical properties,have been widely employed on mechanical parts to reduce friction and wear,which saves energy and improves the overall performance for applications such as aerospace satellite parts,shafts,gears,and bushings.The addition of functional fillers can overcome the limitations of single-polymer coatings and extend the service life of the coatings by providing a combination of low friction,high wear resistance,high load bearing,high temperature resistance,and high adhesion.This paper compares the heat resistance,and the tribological and mechanical properties of common polymer matrices,as well as the categories of functional fillers that improve the coating performance.Applicable scopes,process parameters,advantages,and limitations of the preparation methods of polymer coatings are discussed in detail.The tribological properties of the composite coatings with different matrices and fillers are compared,and the lubrication mechanisms are analyzed.Fillers reduce friction by promoting the formation of transfer films or liquid shear films.Improvement of the mechanical properties of the composite coatings with fillers of different morphologies is described in terms of strengthening and toughening mechanisms,including a stress transfer mechanism,shear yielding,crack bridging,and interfacial debonding.The test and enhancement methods for the adhesion properties between the coating and substrate are discussed.The coating adhesion can be enhanced through mechanical treatment,chemical treatment,and energy treatment of the substrate.Finally,we propose the design strategies for high-performance polymer composite coating systems adapted to specific operating conditions,and the limitations of current polymer composite coating research are identified.Yilong REN Lin ZHANG Guoxin XIE Zhanbo LI Hao CHEN Hanjun GONG Wenhu XU Dan GUO Jianbin LUO 2021Friction2021,9,3:13

5Correlation of the temporal and spatial expression patterns of HQT with the biosynthesis and accumulation of chlorogenic acid in Lonicera japonica flowers显示文摘Hydroxycinnamoyl-CoA quinate transferase(HQT)is one of the key enzymes in the biosynthesis of chlorogenic acid(CGA)in the flowers of Lonicera japonica.However,the spatiotemporal expression patterns of HQT and its relationship to the dynamics of CGA biosynthesis,transport,and storage remain largely unknown.In this study,we collected L.japonica flower samples at different growth stages(S1–S6)and examined the spatiotemporal expression pattern of HQT and the dynamic accumulation patterns of CGA using a combination of molecular and cytological techniques.Our results suggest that the spatiotemporal expression pattern of HQT is directly correlated with dynamic changes in CGA accumulation and distribution in L.japonica flowers.We further show that CGA is synthesized primarily in the cytoplasm and chloroplasts.CGA synthesized in the cytoplasm first accumulates in specialized vesicles and is then transferred to large central vacuoles for storage by fusion of CGA-containing vesicles with vacuoles.Furthermore,CGA synthesized in the chloroplasts appears to be transferred into the vacuoles for storage by direct membrane fusion between the tonoplast and the disrupted chloroplast membranes.Collectively,our results suggest that CGA is synthesized in chloroplasts and cytoplasm and finally transferred to the vacuole for long-term storage.Yanqun Li Dexin Kong Mei Bai Hanjun He Haiyang Wang Hong Wu 2019Horticulture Research2019,6,1:7
6Fe5C2 nanoparticles as low-cost HER electrocatalyst:the importance of Co substitution显示文摘Constructing and understanding the doping effect of secondary metal in transition metal carbide(TMC)catalysts is pivotal for the design of low-cost hydrogen evolution reaction(HER) electrocatalysts. In this work, we developed a wet-chemistry strategy for synthesizing Co-modified Fe_5C_2 nanoparticles((Fe_(1-x)Cox)_5C_2 NPs) as highly active HER electrocatalysts in basic solution. The structure of(Fe_(1-x)Cox)_5C_2 NPs was characterized by X-ray diffraction(XRD), extended X-ray absorption fine structure spectra(EXAFS) and scanning/transmission electron microscopy(S/TEM), indicating that the isomorphous substitution of cobalt in the lattice of Fe_5C_2.(Fe_(0.75) Co_(0.25))_5C_2 exhibited the best HER activity(174 mV for j = -10 mA/cm^2). Computational calculation results indicate that Co provides the most active site for HER. X-ray adsorption spectra(XAS) studies further suggested that the electron transfer in Fe–C bonds are enhanced by the substitution of Co, which modulates the hydrogen adsorption on the adjacent electronic-enriched carbon, and therefore promotes HER activity. 7Effects of flexibility and strength training on peak hamstring musculotendinous strains during sprinting显示文摘Background:Hamstring injury is one of the most common injuries in sports involving sprinting.Hamstring flexibility and strength are often considered to be modifiable risk factors in hamstring injury.Understanding the effects of hamstring flexibility or strength training on the biomechanics of the hamstring muscles during sprinting could assist in improving prevention strategies and rehabilitation related to these injuries.The purpose of this study was to determine the effects of altering hamstring flexibility or strength on peak hamstring musculotendinous strain during sprinting.Methods:A total of 20 male college students(aged 18-24 years)participated and were randomly assigned to either a flexibility intervention group or a strength intervention group.Each participant executed exercise training 3 times a week for 8 weeks.Flexibility,sprinting,and isokinetic strength testing were performed before and after the 2 interventions.Paired t tests were performed to determine hamstring flexibility or strength intervention effects on optimal hamstring musculotendinous lengths and peak hamstring musculotendinous strains during sprinting.Results:Participants in the flexibility intervention group significantly increased the optimal musculotendinous lengths of the semimembranosus and biceps long head(p<0.026)and decreased peak musculotendinous strains in all 3 bi-articulate hamstring muscles(p<0.004).Participants in the strength-intervention group significantly increased the optimal musculotendinous lengths of all 3 hamstring muscles(p<0.041)and significantly decreased their peak musculotendinous strain during sprinting(p<0.017).Conclusion:Increasing hamstring flexibility or strength through exercise training may assist in reducing the risk of hamstring injury during sprinting for recreational male athletes.Xianglin Wan Shangxiao Li Thomas M.Best Hui Liu Hanjun Li Bing Yu 2021Journal of Sport and Health Science2021,10,2:6
8A 3D-bioprinted scaffold with doxycycline-controlled BMP2-expressing cells for inducing bone regeneration and inhibiting bacterial infection显示文摘Large bone defects face a high risk of pathogen exposure due to open wounds,which leads to high infection rates and delayed bone union.To promote successful repair of infectious bone defects,fabrication of a scaffold with dual functions of osteo-induction and bacterial inhibition is required.This study describes creation of an engineered progenitor cell line(C3H10T1/2)capable of doxycycline(DOX)-mediated release of bone morphogenetic protein-2(BMP2).Three-dimensional bioprinting technology enabled creation of scaffolds,comprising polycaprolactone/mesoporous bioactive glass/DOX and bioink,containing these engineered cells.In vivo and in vitro experiments confirmed that the scaffold could actively secrete BMP2 to significantly promote osteoblast differentiation and induce ectopic bone formation.Additionally,the scaffold exhibited broad-spectrum antibacterial capacity,thereby ensuring the survival of embedded engineered cells when facing high risk of infection.These findings demonstrated the efficacy of this bioprinted scaffold to release BMP2 in a controlled manner and prevent the occurrence of infection;thus,showing its potential for repairing infectious bone defects.Minqi Wang Hanjun Li Yiqi Yang Kai Yuan Feng Zhou Haibei Liu Qinghui Zhou Shengbing Yang Tingting Tang 2021Bioactive Materials2021,6,5:3

10Bone marrow stromal cell versus neural stem cell transplantation in a C6 glioma rat model显示文摘BACKGROUND: Embryonic neural stem cells (NSCs) have provided positive effects for the treatment of glioma. However, the source for embryonic NSCs remains limited and high amplification conditions are required. Bone marrow stromal cells (BMSCs) have been proposed for the treatment of glioma. OBJECTIVE: To investigate biological changes in NSCs and BMSCs following transplantation into rat models of glioma. DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at the Embryonic Stem Cell Research Laboratory of Yunyang Medical College from February 2006 to August 2008. MATERIALS: The rat C6 glioma cell line was purchased from Institute of Biochemistry and Cell Biology, Shanghai Institutes for Biological Sciences, Chinese Academy of Sciences; mouse anti-bromodeoxyuridine (BrdU) monoclonal antibody and Cy3-labeled goat anti-mouse IgG antibody was purchased from Upstate, USA. METHODS: A total of 95 Sprague Dawley rats were randomly assigned to three groups: NSC (n = 35), transplanted with > 6 × 106 NSCs via left medial hind limb; BMSC (n = 35), transplanted with > 1 × 106 BMSCs via left medial hind limb; model group (n = 25), injected with the same volume of 0.1 mmol/L phosphate buffered saline. MAIN OUTCOME MEASURES: Gliomal growth and size were assessed by nuclear magnetic resonance, and glioma morphological features were observed following hematoxylin-eosin staining and BrdU immunohistochemistry 3 and 4 weeks following transplantation. RESULTS: The average survival of rats in the BMSC, NSC, and model groups was 4.03, 4.28, and 3.88 weeks. At 3 weeks, there was no significant difference in the average glioma diameter between the BMSC and model groups (P > 0.05). However, gliomal diameter was significantly decreased in the NSC group compared with the model group (P < 0.05). At 4 weeks, there was no statistical difference between the groups (P > 0.05). BrdU immunohistochemistry revealed that BMSCs and NSCs appeared to migrate to the gliomas. CONCLUSION: NSCs inhibited glioma cell growth and prolonged rat survival. 13Modulating Pd e_(g) orbital occupancy in Pd-Au metallic aerogels for efficient carbon dioxide reduction显示文摘The electronic structure of electrocatalysts plays a critical role in energy conversion,whereas for an efficient catalyst,it is challenging to modulate the orbitals.Herein,we present a new strategy to modulate the e_(g) orbital occupancy of Pd by constructing composition-controllable Pd-Au metallic aerogels(MAs),optimizing the d-band center of Pd to achieve excellent performance for electrochemical carbon dioxide reduction reaction(CO_(2)RR).Specifically,Pd_(1)Au_(2) MAs achieve almost 100% Faraday efficiency(FE) of CO in the range of-0.40 to-0.80 V vs.reversible hydrogen electrode(RHE),as well as the long-term stability,being one of the best Pd-based materials for CO_(2)RR.The X-ray photoelectron spectroscopy(XPS) results and density functional theory(DFT) calculations demonstrate that the introduction of Au modulates the Pd e_(g) orbital occupancy,which significantly weakens *CO adsorption on Pd,reduces the CO_(2)RR energy barrier and consequently improves the electrocatalytic activity and stability for long-term applications.Our work highlights a new strategy for designing efficient electrocatalysts for CO_(2)RR and beyond.Yao Chen Juan Wang Tingjie Mao Cun Chen Hanjun Li Honggang Huang Hui Fu Feili Lai Jiadong Chen Nan Zhang Tianxi Liu 2023Journal of Energy Chemistry2023,,12:0

15The Soft-Strain Effect Enabled High-Performance Flexible Pressure Sensor and Its Application in Monitoring Pulse Waves显示文摘Flexible and wearable pressure sensors attached to human skin are effective and convenient in accurate and real-time tracking of various physiological signals for disease diagnosis and health assessment.Conventional flexible pressure sensors are constructed using compressible dielectric or conductive layers,which are electrically sensitive to external mechanical stimulation.However,saturated deformation under large compression significantly restrains the detection range and sensitivity of such sensors.Here,we report a novel type of flexible pressure sensor to overcome the compression saturation of the sensing layer by softstrain ffect,enabling an utra-high sensitivity of~636 kPa^(-1) and a wide detection range from 0.1 kPa to 56 kPa.In addition,the cyclic loading-unloading test reveals the excellent stability of the sensor,which maintains its signal detection after 10.000 cycles of 10 kPa compression.The sensor is capable of monitoring arterial pulse waves from both deep tissue and distal parts,such as digital arteries and dorsal pedal arteries,which can be used for blood pressure estimation by pulse transit time at the same artery branch.Yue Li Yuan Wei Yabao Yang Lu heng Lei Luo Jiuwei Gao Hanjun Jiang Juncai Song Manzhang Xu Xuewen Wang Wei Huang 2023Research2023,,1:0

17d-d Orbital coupling induced by crystal-phase engineering assists acetonitrile electroreduction to ethylamine显示文摘The d-d orbital coupling induced by crystal-phase engineering can effectively adjust the electronic structure of electrocatalysts,thus showing significant catalytic performance,while it has been rarely explored in electrochemical acetonitrile reduction reaction(ARR)to date.Herein,we successfully realize the structural transformation of Pd Cu metallic aerogels(MAs)from face-centered cubic(FCC)to body-centered cubic(BCC)through annealing treatment.Specifically,the BCC Pd Cu MAs exhibit excellent ARR performance with high ethylamine selectivity of 90.91%,Faradaic efficiency of 88.60%,yield rate of 316.0 mmol h^(-1)g^(-1)_(Pd+Cu)and long-term stability for consecutive electrolysis within 20 h at-0.55 V vs.reversible hydrogen electrode,outperforming than those of FCC Pd Cu MAs.Under the membrane electrode assembly system,BCC Pd Cu MAs also demonstrate excellent ethylamine yield rate of 389.5 mmol h^(-1)g^(-1)_(Pd+Cu).Density functional theory calculation reveals that the d-d orbital coupling in BCC Pd Cu MAs results in an evident correlation effect for the interaction of Pd and Cu sites,which boosts up the Cu sites electronic activities to enhance ARR performance.Our work opens a new route to develop efficient ARR electrocatalysts from the perspective of crystalline structure transformation.Honggang Huang Yao Chen Hui Fu Cun Chen Hanjun Li Zhe Zhang Feili Lai Shuxing Bai Nan Zhang Tianxi Liu 2024Journal of Energy Chemistry2024,89,2:0

19Multi-omics analysis based on 3D-bioprinted models innovates therapeutic target discovery of osteosarcoma显示文摘Current in vitro models for osteosarcoma investigation and drug screening,including two-dimensional(2D)cell culture and tumour spheroids(i.e.cancer stem-like cells),lack extracellular matrix(ECM).Therefore,results from traditional models may not reflect real pathological processes in genuine osteosarcoma histological structures.Here,we report a three-dimensional(3D)bioprinted osteosarcoma model(3DBPO)that contains osteosarcoma cells and shrouding ECM analogue in a 3D frame.Photo-crosslinkable bioinks composed of gelatine methacrylamide and hyaluronic acid methacrylate mimicked tumour ECM.We performed multi-omics analysis,including transcriptomics and DNA methylomics,to determine differences between the 3DBPO model and traditional models.Compared with 2D models and tumour spheroids,our 3DBPO model showed significant changes in cell cycle,metabolism,adherens junctions,and other pathways associated with epigenetic regulation.The 3DBPO model was more sensitive to therapies targeted to the autophagy pathway.We showed that simulating ECM yielded different osteosarcoma cell metabolic characteristics and drug sensitivity in the 3DBPO model compared with classical models.We suggest 3D printed osteosarcoma models can be used in osteosarcoma fundamental and translational research,which may contribute to novel therapeutic strategy discovery.Yixuan Lin Yiqi Yang Kai Yuan Shengbing Yang Shuhong Zhang Hanjun Li Tingting Tang 2022Bioactive Materials2022,7,12:0
20Irisin Attenuates Osteoarthritis by Inhibiting Apoptosis of Osteocytes Through Activating Erk Signaling Pathway显示文摘Osteoarthritis(OA)is an inflammatory disease involving the joints that is prevalent in the global aging population.The purpose of this study is to determine whether irisin can attenuate osteoarthritis(OA)progression in anterior cruciate ligament transection(ACLT)mice models and the mechanism of irisin therapy effect on OA by increase the resistance of apoptosis in MLO-Y4 cells induced by mechanical stretch in vitro.Methods For in vivo study,3-month-old male C57BL/6 J mice were randomized to three groups,sham-operated,anterior cruciate ligament transection(ACLT)-operated treated with vehicle,and ACLT-operated treated with irisin by intraperitoneal injection once a week.Cartilage erosion was observed by HE staining.Osteoarthritis Research Society International(OARSI)scores were evaluated according to the safranin O stai-ning.The microstructure of tibia cortical bone,trabecular bone,and subchondral bone was analyzed by micro-CT and the bone histomorphometry has been administrated including mineral apposition rate(MAR).Edu staining and cck-8 were used for the detection of the proliferation of MLO-Y4 cells.For mechanical stress,cells were seeded on the collagen-I coated chamber subjected with a peak biaxial stretch of 20%at 1 Hz for 16 hours to induce apoptosis.Flow cytometry was used for the detection of apoptosis and cell cycle.TUNNEL was used for staining the apoptotic cells and rt-PCR was applied for quantifying the expression of mRNA such as Bax,Bcl-2,SOST,c-myc,Opg.Western blot was utilized to confirm the mechanism of how irisin decrease the osteocyte apoptosis.Results In vivo,irisin can attenuate articular cartilage degeneration.Irisin maintains the proportion of hyaline cartilage and calcified cartilage and keep fewer cartilage erosions in ACLT-operated mice.For immunohistochemical(IHC)staining,irisin reduced the expression of caspase3,Bax and matrix metalloproteinase-13 in both cartilage and subchondral bone.Irisin-treated ACLT group shows higher Trabecular number(Tb.N)and bone volume fraction(BV/TV)compared to the vehicle-treated ACLT group.In vitro, irisin significantly increased the proliferation of MLO-Y4 cells detected by Edu and Ki67 staining,and irisin can protect the cells from both mechanical stretchinduced apoptosis detected by FITC-PI flow cytometry and maintain the cell activity by regulating the expression of Bax,Bcl-2,and c-myc.Transcriptome sequencing shows that irisin significantly activates the MAPK signaling pathway and we confirm the result by western blot:irisin effectively activates the Erk signaling pathway through phosphorylation and has a certain activation effect on p38 signaling pathway,no activation was observed for FAK signaling pathway.Conclusions Irisin can attenuate the progression of OA by decrease the apoptosis of osteocyte,which can improve the microarchitecture of subchondral bone.Erk pathway activation plays an important role in reducing the apoptosis of osteocyte.Zihao He Hanjun Li Feng Zhou Jingke Du Shuhong Zhang Tingting Tang Zhifeng Yu 2019医用生物力学2019,34,A01:0
