| 3 | A Duplex Real-Time PCR Assay for the Simultaneous Detection of Porcine Circovirus 2 and Circovirus 3显示文摘Porcine circoviruses(PCV) include PCV1, PCV2, and the new-emerging PCV3. PCV2 is pathogenic to pigs, but the pathogenicity of PCV3 in pigs is debatable. Recently, there have been frequent reports of PCV2 and PCV3 co-infections in clinical samples. Thus, it would be practical to develop a duplex PCR method to detect PCV2 and PCV3 simultaneously. In this study, specific primers and probes were designed to target PCV2 cap and PCV3 rep genes. A duplex real-time PCR method was then developed to detect the two viruses. The assay was found to be highly specific, sensitive, and reproducible for PCV2/3 without cross-reactions with other swine pathogens. The sensitivity of this assay was 2.9 copies for the PCV2 plasmid and 22.5 copies for the PCV3 plasmid. The established assay was then used to detect PCV2/3 infection in 340 clinical samples collected in the first half of 2017. The results showed that the co-infection rate of PCV2/3 in the samples was 27.6%. | 5 | Evaluation of soil washing process with carboxymethyl-β-cyclodextrin and carboxymethyl chitosan for recovery of PAHs/heavy metals/fluorine from metallurgic plant site显示文摘Polycyclic aromatic hydrocarbons(PAHs)/heavy metals/fluorine(F) mixed-contaminated sites caused by abandoned metallurgic plants are receiving wide attention. To address the associated environmental problems,this study was initiated to investigate the feasibility of using carboxymethyl-β-cyclodextrin(CMCD) and carboxymethyl chitosan(CMC) solution to enhance ex situ soil washing for extracting mixed contaminants. Further,Tenax extraction method was combined with a first-three-compartment model to evaluate the environmental risk of residual PAHs in washed soil. In addition,the redistribution of heavy metals/F after decontamination was also estimated using a sequential extraction procedure. Three successive washing cycles using50 g/L CMCD and 5 g/L CMC solution were effective to remove 94.3% of total PAHs,93.2% of Pb,85.8% of Cd,93.4% of Cr,83.2% of Ni and 97.3% of F simultaneously. After the 3rd washing,the residual PAHs mainly existed as very slowly desorbing fractions,which were in the form of well-aged,well-sequestered compounds; while the remaining Pb,Cd,Cr,Ni and F mainly existed as Fe–Mn oxide and residual fractions,which were always present in stable mineral forms or bound to non-labile soil fractions. | 6 | High-performance Zn battery with transition metal ions co-regulated electrolytic MnO_(2)显示文摘Electrolytic MnO_(2)/Zn batteries have attracted extensive attention for use in large-scale energy storage applications due to their low cost,high output voltage,safety,and environmental friendliness.However,the poor electrical conductivity of MnO_(2)limits its deposition and dissolution at large capacities,which leads to sluggish reaction kinetics and drastic capacity decay.Here,we report a theory-guided design principle for an electrolytic MnO_(2)/Zn battery co-regulated with transition metal ions that has improved electrochemical performance in terms of deposition and stripping chemistries.We start with first-principles calculations to predict the electrolytic effects of regulating transition metal ions in the deposition/stripping chemistry of the MnO_(2)cathode.The results indicate that with the simultaneous incorporation of strongly electronegative Co and Ni,the MnO_(2)cathode tends to possess more active electron states,faster charge-transfer kinetics,and better electrical conductivity than either MnO_(2)regulated with Co or Ni on their own,or pristine MnO_(2);hence,this co-regulation is beneficial for the cathode solid/liquid MnO_(2)/Mn2t reactions.We then fabricate and demonstrate a novel Co2t and Ni2t coregulated MnO_(2)/Zn(Co-Ni-MnO_(2)/Zn)battery that yields significantly better electrochemical performance,finding that the synergistic regulation of Co and Ni on MnO_(2)can significantly increase its intrinsic conductivity and achieve high rates and Coulombic efficiencies at large capacities.The aqueous Co-Ni-MnO_(2)/Zn battery exhibits a high rate(10C,100 mA cm^(-2)),high Coulombic efficiency(91.89%),and excellent cycling stability(600 cycles without decay)at a large areal capacity of 10 mAh cm^(-2).Our proposed strategy of co-regulation with transition metal ions offers a versatile approach for improving the electrochemical performance of aqueous electrolytic MnO_(2)/Zn batteries in large-scale energy storage applications. | Mingyan Chuai Jinlong Yang Mingming Wang Yuan Yuan Zaichun Liu Yan Xu Yichen Yin Jifei Sun Xinhua Zheng Na Chen Wei Chen | 2021 | eScience2021,1,2: | 8 |

| 11 | Stability influence factors analysis and construction of a deep beam anchorage structure in roadway roof显示文摘Deep beam anchorage structures based on spatial distribution analysis of the cable prestressed field have been proposed for roadway roof support. Stability and other factors that influence deep beam structures are studied in this paper using mechanical calculations, numerical analysis and field measurements. A mechanical model of deep beam structure subjected to multiple loading is established, including analysis of roof support in the return airway of S1203 working face in the Yuwu coal mine, China. The expression of maximum shear stress in the deep beam structure is deduced according to the stress superposition criterion. It is found that the primary factors affecting deep beam structure stability are deep beam thickness, cable pre-tension and cable spacing. The variation of maximum shear stress distribution and prestressed field diffusion effects according to various factors are analyzed using Matlabòand FLAC3D^(TM) software, and practical support parameters of the S1203 return airway roof are determined.According to the observations of rock pressure, there is no evidence of roof separation, and the maximum values of roof subsidence and convergence of wall rock are 72 and 48 mm, respectively. | 14 | Feasibility of Tea Saponin-Enhanced Soil Washing in a Soybean Oil-Water Solvent System to Extract PAHs/Cd/Ni Efficiently from a Coking Plant Site显示文摘Mixed contaminated brownfield sites have brought serious risks to human health and environmental safety. With the purpose of removing polycyclic aromatic hydrocarbons(PAHs) and heavy metals from a coking plant site, an innovative technology for ex-situ washing was developed in the present work. The combination of 15.0 mL L^(-1) soybean oil and 7.5 g L^(-1) tea saponin proved an effective method to extract co-pollutants from soil. After two consecutive washing cycles, the efficiency rates of removal for 3-, 4-,5(+6)-ring, and total PAHs, Cd, and Ni were approximately 98.2%, 96.4%, 92.3%, 96.3%, 94.1%, and 89.4%, respectively. Meanwhile,as evaluated by Tenax extraction method and metal stability indices, the residual PAHs and heavy metals after consecutive washing mainly existed in the form with extremely low bioaccessibility in the soil. Thus, in the soil after two washing cycles, there appeared limited environmental transfer risk of co-pollutants. Moreover, a subsequent precipitation method with alkaline solution and PAHdegrading strain Sphingobium sp. PHE9 inoculation effectively removed 84.6%–100% of Cd, 82.5%–91.7% of Ni, and 92.6%–98.4% of PAHs from the first and second washing solvents. The recovered solvents also exhibited a high recycling effectiveness. This problem is regarded as a pursuit-evasion game since satellites in both sides will try their best to capture or escape. In this game, the distance of these two players is small enough so that the highly nonlinear earth-centered gravitational dynamics can be reduced to the linear Clohessy-Wiltshire(CW) equations. The system is then simplified by introducing the zero effort miss variables. Saddle solution is formulated for the pursuit-evasion game and time-to-go is estimated similarly as that for the exoatmospheric interception. | 17 | Genetic improvement of heat tolerance in wheat:Recent progress in understanding the underlying molecular mechanisms显示文摘As a cool season crop, wheat(Triticum aestivum L.) has an optimal daytime growing temperature of 15 ℃ during the reproductive stage. With global climate change, heat stress is becoming an increasingly severe constraint on wheat production. In this review, we summarize recent progress in understanding the molecular mechanisms of heat tolerance in wheat. We firstly describe the impact of heat tolerance on morphology and physiology and its potential effect on agronomic traits. We then review recent discoveries in determining the genetic and molecular factors affecting heat tolerance, including the effects of phytohormone signaling and epigenetic regulation. | 18 | Ablation of Akt2 and AMPKα2 rescues high fat diet-induced obesity and hepatic steatosis through Parkin-mediated mitophagy显示文摘Given the opposing effects of Akt and AMP-activated protein kinase(AMPK)on metabolic homeostasis,this study examined the effects of deletion of Akt2 and AMPKα2 on fat diet-induced hepatic steatosis.Akt2–Ampkα2 double knockout(DKO)mice were placed on high fat diet for 5 months.Glucose metabolism,energy homeostasis,cardiac function,lipid accumulation,and hepatic steatosis were examined.DKO mice were lean without anthropometric defects.High fat intake led to adiposity and decreased respiratory exchange ratio(RER)in wild-type(WT)mice,which were ablated in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.High fat intake increased blood and hepatic triglycerides and cholesterol,promoted hepatic steatosis and injury in WT mice.These effects were eliminated in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.Fat diet promoted fat accumulation,and enlarged adipocyte size,the effect was negated in DKO mice.Fat intake elevated fatty acid synthase(FAS),carbohydrate-responsive element-binding protein(CHREBP),sterol regulatory element-binding protein 1(SREBP1),peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α),peroxisome proliferator-activated receptor-α(PPARα),PPARγ,stearoyl-CoA desaturase 1(SCD-1),phosphoenolpyruvate carboxykinase(PEPCK),glucose 6-phosphatase(G6Pase),and diglyceride O-acyltransferase 1(DGAT1),the effect was absent in DKO but not Akt2^(-/-) and Ampkα2^(-/-) mice.Fat diet dampened mitophagy,promoted inflammation and phosphorylation of forkhead box protein O1(FoxO1)and AMPKα1(Ser^(485)),the effects were eradicated by DKO.Deletion of Parkin effectively nullified DKO-induced metabolic benefits against high fat intake.Liver samples from obese humans displayed lowered microtubule-associated proteins 1A/1B light chain 3B(LC3B),Pink1,Parkin,as well as enhanced phosphorylation of Akt,AMPK(Ser^(485)),and FoxO1,which were consolidated by RNA sequencing(RNAseq)and mass spectrometry analyses from rodent and human livers.These data suggest that concurrent deletion of Akt2 and AMPKα2 offers resilience to fat diet-induced obesity and hepatic steatosis,possibly through preservation of Parkin-mediated mitophagy and lipid metabolism. | Shuyi Wang Jun Tao Huaguo Chen Machender R.Kandadi Mingming Sun Haixia Xu Gary D.Lopaschuk Yan Lu Junmeng Zheng Hu Peng Jun Ren | 2021 | Acta Pharmaceutica Sinica B2021,11,11: | 4 |

| 20 | Identification of stably expressed QTL for resistance to black shank disease in tobacco(Nicotiana tabacum L.) line Beinhart 1000-1显示文摘Cigar line Beinhart 1000-1 has effective durable resistance to black shank(BS) and is considered one of the most resistant sources in tobacco(Nicotiana tabacum L.). To investigate the inheritance and identification of stable quantitative trait loci(QTL) for BS response, F2,BC1 F2 individuals and BC1 F2:3 lines were produced from a cross between Beinhart 1000-1 and Xiaohuangjin 1025. Two major quantitative trait loci(M-QTL) named qBS7 and qBS17 were repeatedly detected under different conditions. QTL qBS7 was mapped to the region between PT30174 and PT60621 and explained 17.40%–25.60% of the phenotypic variance under different conditions. The other QTL qBS17 in interval PT61564–PT61538 of linkage group 17 was detected in a BC1 F2 population in the field and in BC1 F2:3 in both the field and at the seedling stage, explaining 6.90% to 11.60% of the phenotypic variance.
