| 5 | Increased leptin by hypoxic-preconditioning promotes autophagy of mesenchymal stem cells and protects them from apoptosis显示文摘Autophagy is the basic catabolic progress involved in cell degradation of unnecessary or dysfunctional cellular components.It has been proven that autophagy could be utilized for cell survival under stresses.Hypoxic-preconditioning(HPC)could reduce apoptosis induced by ischemia and hypoxia/serum deprivation(H/SD)in bone marrow-derived mesenchymal stem cells(BMSCs).Previous studies have shown that both leptin signaling and autophagy activation were involved in the protection against apoptosis induced by various stress,including ischemia-reperfusion.However,it has never been fully understood how leptin was involved in the protective effects conferred by autophagy.In the present study,we demonstrated that HPC can induce autophagy in BMSCs by increased LC3-II/LC3-I ratio and autophagosome formation.Interestingly,similar effects were also observed when BMSCs were pretreated with rapamycin.The beneficial effects offered by HPC were absent when BMSCs were incubated with autophagy inhibitor,3-methyladenine(3-MA).In addition,down-regulated leptin expression by leptin-shRNA also attenuated HPC-induced autophagy in BMSCs,which in turn was associated with increased apoptosis after exposed to sustained H/SD.Furthermore,increased AMP-activated protein kinase phosphorylation and decreased mammalian target of rapamycin phosphorylation that were observed in HPC-treated BMSCs can also be attenuated by down-regulation of leptin expression.Our data suggests that leptin has impact on HPC-induced autophagy in BMSCs which confers protection against apoptosis under H/SD,possibly through modulating both AMPK and mTOR pathway. | WANG LiHan HU XinYang ZHU Wei JIANG Zhi ZHOU Yu CHEN PanPan WANG JianAn | 2014 | Science China(Life Sciences)2014,57,2: | 8 |
| 6 | The Glutamatergic Postrhinal Cortex–Ventrolateral Orbitofrontal Cortex Pathway Regulates Spatial Memory Retrieval显示文摘A deficit in spatial memory has been taken as an early predictor of Alzheimer’s disease(AD) or mild cognitive impairment(MCI). The uncinate fasciculus(UF) is a long-range white-matter tract that connects the anterior temporal lobe with the orbitofrontal cortex(OFC)in primates. Previous studies have shown that the UF impairment associated with spatial memory deficits may be an important pathological change in aging and AD, but its exact role in spatial memory is not well understood. The pathway arising from the postrhinal cortex(POR) and projecting to the ventrolateral orbitofrontal cortex(vlOFC)performs most of the functions of the UF in rodents.Although the literature suggests an association between spatial memory and the regions connected by the POR–vlOFC pathway, the function of the pathway in spatialmemory is relatively unknown. To further illuminate the function of the UF in spatial memory, we dissected the POR–vlOFC pathway in mice. We determined that the POR–vlOFC pathway is a glutamatergic structure, and that glutamatergic neurons in the POR regulate spatial memory retrieval. We also demonstrated that the POR–vlOFC pathway specifically transmits spatial information to participate in memory retrieval. | 7 | Effect of butanol on flotation separation of quartz from hematite with N-dodecyl ethylenediamine显示文摘In order to investigate the effect of butanol on quartz flotation when N-dodecyl ethylenediamine(ND)was used as collector, single mineral flotation and artificial mixed mineral(hematite and quartz were mixed at a mass ratio of 3:2) separation were conducted in the laboratory. Experimental results indicated that addition of butanol could improve the collecting performance of ND on quartz and enhance the floatability of quartz. Best flotation recovery of quartz was obtained when butanol was mixed with ND at a mass ratio of 1:1. Moreover, the molecular structure of alcohols had a significant effect on mineral recovery. Best separation efficiency could be obtained when tert-butanol was added as it had the largest cross-sectional area. Zeta potential measurements indicated that alcohols could strengthen electrostatic adsorption between quartz and collector. | 8 | Effect of hydration on the elasticity of mantle minerals and its geophysical implications显示文摘Recent studies have shown that major nominally anhydrous minerals in the Earth's mantle, such as olivine, pyroxene and garnet, can incorporate considerable amounts of water as structurally bound hydroxyl. Even a small amount of water is present in mantle minerals, it can strongly affect a number of physical properties, including density, sound velocity, melting temperature, and electrical conductivities. The presence of water can also influence the dynamic behavior, lead to lateral velocity heterogeneities, and affect the material circulation of the Earth's deep interior. In particular, seismic studies have reported the existence of low-velocity zones in various locations of the Earth's upper mantle and transition zone, which has been expected to be associated with the presence of water in the region. In the past two decades, the effect of water on the elasticity and sound velocities of minerals at relevant pressure-temperature(P-T) conditions of the Earth's mantle attracted extensive interests. Combining the high P-T experimental and theoretical mineralogical results with seismic observations provides crucial constraints on the distribution of water in the Earth's mantle. | 14 | Microstructure Evolution and Pore Formation Mechanism of Porous TiAl3 Intermetallics via Reactive Sintering显示文摘Porous TiAl_3 intermetallics were fabricated through vacuum reactive sintering from Ti–75 Al at.% elemental powder mixture. The phase compositions, expansion behaviors, pore characteristics and microstructure evolution of TiAl_3 intermetallics were investigated, and the pore formation mechanism was also proposed. It was found that the actual temperature of compacts showed an acute climb from 668 to 1244 °C in 166 s, while the furnace temperature maintained the linear growth of 5 °C/min, which indicated that an obvious thermal explosion(TE) reaction occurred during sintering,and only single-phase TiAl_3 intermetallic was synthesized in TE products. The open porosity increased from 22.2(green compact) to 32.8% after reactive diffusion sintering at 600 °C and rised to 58.7% after TE, then decreased to 51.2% after high-temperature homogenization at 1100 °C. Therefore, TE reaction is the dominated pore formation mechanism of porous TiAl_3 intermetallics. The pore evolution in porous TiAl_3 intermetallics occurred by the following mechanisms:certain intergranular pores remained among powder particles of green compact, then low-temperature sintering resulted in a further increase in porosity due to the Kirkendall effect. | 16 | Effect of Plaque Composition on Biomechanical Performance of a Carotid Stent: Computational Study显示文摘Clinical application of bare metal stents is constrained by the occurrence of instent restenosis,mainly due to the complex biomechanical environment in the body.Numerical simulation method was used to evaluate the effect of plaque composition on stent performance in a carotid artery.CT angiography(CTA)data were used as a reference,and zero-load state of the carotid artery was used to establish a 3D stenotic artery model.Different plaque compositions,calcified and hypo-cellular were defined in Model 1 and Model 2,respectively.Interactions between the stents and arterial tissues within the stent crimping-expansion process were analyzed to explore the effects of plaque composition on the mechanical parameters of carotid stents.Goodman diagram and fatigue safety factor(FSF)were analyzed to explore the effects of plaque composition on fatigue performance of a carotid stent in the stent service process.In the stent crimping-expansion process,the von Mises stress in the stent and the dog-boning ratio in Model 1 were higher than that in Model 2.The calcified plaque prevented the stent from expanding the stenotic vessel to a pre-set diameter.Thus,the risk of rupture in the calcified plaque was higher than that in the hypo-cellular plaque.Plaque also affected the stress/strain in the vessel wall,which was observed to be lower in Model 1 than in Model 2.This indicated that calcified plaque could decrease the stress-induced injury of arterial tissues.Within the stent service process,the stents used in these two models were predicted to not fail under fatigue rupture as calculated by the Goodman diagram.Additionally,the points closer to the fatigue limit were generally observed at the inner bend of the stent crowns.The FSF of the stent in Model 1 was lower than that in Model 2.The stent operating in the presence of calcified plaques suffered high risk of fractures.Reliability and fatigue performance of the stent were found to be associated with plaque composition.Hence,this study may provide stent designers an approach toward enhancing the mechanical reliability of a stent. | Xinyang Cui Qingshuai Ren Zihao Li Kun Peng Gaoyang Li ZhaoyongGu Aike Qiao | 2018 | Computer Modeling in Engineering & Sciences2018,,9: | 3 |
