1Cancer-associated fibroblasts in breast cancer:Challenges and opportunities显示文摘The tumor microenvironment is proposed to contribute substantially to the progression of cancers,including breast cancer.Cancer-associated fibroblasts(CAFs)are the most abundant components of the tumor microenvironment.Studies have revealed that CAFs in breast cancer originate from several types of cells and promote breast cancer malignancy by secreting factors,generating exosomes,releasing nutrients,reshaping the extracellular matrix,and suppressing the function of immune cells.CAFs are also becoming therapeutic targets for breast cancer due to their specific distribution in tumors and their unique biomarkers.Agents interrupting the effect of CAFs on surrounding cells have been developed and applied in clinical trials.Here,we reviewed studies examining the heterogeneity of CAFs in breast cancer and expression patterns of CAF markers in different subtypes of breast cancer.We hope that summarizing CAFrelated studies from a historical perspective will help to accelerate the development of CAF-targeted therapeutic strategies for breast cancer.Dengdi Hu Zhaoqing Li Bin Zheng Xixi Lin Yuehong Pan Peirong Gong Wenying Zhuo Yujie Hu Cong Chen Lini Chen Jichun Zhou Linbo Wang 2022Cancer Communications2022,42,5:9
2Variation in cis-regulation of a NAC transcription factor contributes to drought tolerance in wheat显示文摘Drought is a major environmental factor limiting wheat production worldwide,and developing drought-tolerant cultivars is a central challenge for wheat breeders globally.Therefore,it is important to identify genetic components determining drought tolerance in wheat.In this study,we identified a wheat NAC gene(TaNAC071-A)that is tightly associated with drought tolerance by a genome-wide association study.Knockdown of TaNAC071-A in wheat attenuated plant drought tolerance,whereas its overexpression significantly enhanced drought tolerance through improved water-use efficiency and increased expression of stress-responsive genes.This heightened water-saving mechanism mitigated the yield loss caused by water deficit.Further candidate gene association analysis showed that a 108-bp insertion in the promoter of TaNAC071-A alters its expression level and contributes to variation in drought tolerance among wheat accessions.This insertion contains two MYB cis-regulatory elements(CREs)that can be directly bound by the MYB transcription activator,TaMYBL1,thereby leading to increased TaNAC071-A expression and plant drought tolerance.Importantly,introgression of this 108-bp insertion allele,TaNAC071-AIn-693,into drought-sensitive cultivars could improve their drought tolerance,demonstrating that it is a valuable genetic resource for wheat breeding.Taken together,our findings highlight a major breakthrough in determining the genetic basis underlying phenotypic variation in wheat drought tolerance and showcase the potential of exploiting CRE-containing indels for improving important agronomical traits.Hude Mao Shumin Li Bin Chen Chao Jian Fangming Mei Yifang Zhang Fangfang Li Nan Chen Tian Li Linying Du Li Ding Zhongxue Wang Xinxiu Cheng Xiaojing Wang Zhensheng Kang 2022Molecular Plant2022,15,2:9
3Electroacupuncture at Tianshu (ST 25) for diarrhea-predominant irritable bowel syndrome using positron emission tomography Changes in visceral sensation center显示文摘Previous studies have demonstrated that electroacupuncture therapy is effective in the treatment of irritable bowel syndrome. However, the precise mechanism of this therapy is unknown. The present study served to investigate the effects of electroacupuncture therapy on treatment of patients with diarrhea-predominant irritable bowel syndrome (IBS). We compared brain activation maps based on the changes of cerebral glucose metabolism obtained by 18-fluorodeoxyglucose positron emission tomography scanning under three conditions: resting, rectal balloon distension and rectal balloon distension plus electroacupuncture. Under the resting condition, compared with healthy controls, IBS patients displayed an increasing regional cerebral metabolic rate of glucose over a wide range: bilateral superior temporal gyrus, right middle occipital gyrus, superior frontal gyrus and bilateral middle frontal gyrus. However, there was no significant activity in the visceral pain center. Compared with the resting condition, under the rectal balloon distension condition, patients with IBS had a greater regional cerebral metabolic rate of glucose in the prefrontal cortex, left anterior cingulate cortex, postcentral gyrus, precentral gyrus and temporal gyrus. Under the rectal balloon distension plus electroacupuncture condition, stimulation by electroacupuncture at Tianshu (ST 25) manifested a decreased regional cerebral metabolic rate of glucose in the left cingulate gyrus, right insula, right caudate nucleus, fusiform gyrus and hippocampal gyrus. 4Four-year dynamic observation and study on standardized management of elderly patients with type 2 diabetes in Beijing Yongding Road Community显示文摘Purpose:This study aimed to understand the joint standard of blood glucose,blood li-pids,and blood pressure in elderly patients>60 years of age with type 2 diabetes in the Beijing Yongding Road Community.Methods:One hundred ninety-two elderly patients>60 years of age who were diagnosed with type 2 diabetes in Yongding Road Community served as the investigation subjects,under-went standard management,and the dynamic changes in blood glucose,blood lipids,and blood pressure were monitored for 48 months.Results:At the end of the observation period,the standard rates of fasting blood glucose and postprandial blood glucose were 72.55%and 80.00%,respectively,which were increased compared with 55.73%and 56.08%at baseline(P<0.01).The standard rate of HbA1c was 59.81%,which was increased compared with 53.44%at baseline(P>0.05).The standard rates of TG and LDL-C were 76.71%and 60.38%,respectively,which were increased compared with 54.69%and 34.74%at base-line,and the standard rate of HDL-C was 13.64%,which was decreased compared with 40.10%at baseline(P<0.01).The standard rate of BP was 58.33%,which was increased compared with 38.54%at baseline(P<0.01).The optimal control rate of blood glucose for 48 months(the standard times of the total measurement times in 48 months≥75%)was higher;the fasting blood glucose was 52.17%,the 2-h postprandial blood glucose was 60.22%,and the HbA1c was 46.45%.The optimal control rate of blood lipids was lower;LDL-C was 17.49%and HDL-C was 13.59%.The optimal control rate of BP was 9.13%.At the end of the observation period,the levels of fasting blood glucose and postprandial blood glucose were decreased by 0.7 mmol/L and 1.48 mmol/L,respectively,compared with the base-line(P<0.01).The level of HbA1c was decreased by 0.18%compared with the baseline(P<0.05).The levels of LDL-C and HDL-C were decreased by 0.4 mmol/L and 0.23 mmol/L,respectively,compared with the baseline(P<0.01).The levels of SBP and DBP were decreased by 4 mmHg compared with the baseline(P<0.01).At the end of the observation period,the joint standard rate of the three indices of HbA1c,LDL-C,and BP was 24.72%,which was increased compared with 6.25%at baseline(P<0.01).Conclusion:Standardized management of elderly patients with diabetes in the Community can improve the joint standard rate of blood glucose,blood lipids,and blood pressure.The op-timal control rate and joint standard rate are the important indices for evaluating the quality of diabetes management.Yue Li Dongyun Shen Wei Zhang Yimin Wang Hongmei Li Jie Geng Liying Yang Zhijun Xiao Yan Liu Zhigang Feng Linying Cao Yan Jin Lixia Shi 2014Family Medicine and Community Health2014,2,2:4

8Combination of a biopharmaceutic classification system and physiologically based pharmacokinetic models to predict absorption properties of baicalein in vitro and in vivo显示文摘Objective: To determine the in vitro and in vivo absorption properties of active ingredients of the Chinese medicine, baicalein, to enrich mechanistic understanding of oral drug absorption.Methods: The Biopharmaceutic Classification System(BCS) category was determined using equilibrium solubility, intrinsic dissolution rate, and intestinal permeability to evaluate intestinal absorption mechanisms of baicalein in rats in vitro. Physiologically based pharmacokinetic(PBPK) model commercial software GastroPlus~(TM) was used to predict oral absorption of baicalein in vivo.Results: Based on equilibrium solubility, intrinsic dissolution rate, and permeability values of main absorptive segments in the duodenum, jejunum, and ileum, baicalein was classified as a drug with low solubility and high permeability. Intestinal perfusion with venous sampling(IPVS) revealed that baicalein was extensively metabolized in the body, which corresponded to the low bioavailability predicted by the PBPK model. Further, the PBPK model predicted the key indicators of BCS, leading to reclassification as BCS-II. Predicted values of peak plasma concentration of the drug(C_(max)) and area under the curve(AUC)fell within two times of the error of the measured results, highlighting the superior prediction of absorption of baicalein in rats, beagles, and humans. The PBPK model supported in vitro and in vivo evidence and provided excellent prediction for this BCS class II drug.Conclusion: BCS and PBPK are complementary methods that enable comprehensive research of BCS parameters, intestinal absorption rate, metabolism, prediction of human absorption fraction and bioavailability, simulation of PK, and drug absorption in various intestinal segments across species. 16Single-cell transcriptomic analysis reveals a systemic immune dysregulation in COVID-19-associated pediatric encephalopathy显示文摘Unraveling the molecular mechanisms for COVID-19-associated encephalopathy and its immunopathology is crucial for developing effective treatments.Here,we utilized single-cell transcriptomic analysis and integrated clinical observations and laboratory examination to dissect the host immune responses and reveal pathological mechanisms in COVID-19-associated pediatric encephalopathy.We found that lymphopenia was a prominent characteristic of immune perturbation in COVID-19 patients with encephalopathy,especially those with acute necrotizing encephalopathy(AE).This was characterized a marked reduction of various lymphocytes(e.g.,CD8^(+)T and CD4^(+)T cells)and significant increases in other inflammatory cells(e.g.,monocytes).Further analysis revealed activation of multiple cell apoptosis pathways(e.g.,granzyme/perforin-,FAS-and TNF-induced apoptosis)may be responsible for lymphopenia.A systemic S100A12 upregulation,primarily from classical monocytes,may have contributed to cytokine storms in patients with AE.A dysregulated type I interferon(IFN)response was observed which may have further exacerbated the S100A12-driven inflammation in patients with AE.In COVID-19 patients with AE,myeloid cells(e.g.,monocytic myeloid-derived suppressor cells)were the likely contributors to immune paralysis.Finally,the immune landscape in COVID-19 patients with encephalopathy,especially for AE,were also characterized by NK and T cells with widespread exhaustion,higher cytotoxic scores and inflammatory response as well as a dysregulated B cell-mediated humoral immune response.Taken together,this comprehensive data provides a detailed resource for elucidating immunopathogenesis and will aid development of effective COVID-19-associated pediatric encephalopathy treatments,especially for those with AE.Yi Wang Laurence Don Wai Luu Shuang Liu Xiong Zhu Siyuan Huang Fang Li Xiaolan Huang Linying Guo Jin Zhang Haiyan Ge Yuanyuan Sun Yi Hui Yanning Qu Huicong Wang Xiaoxia Wang Weilan Na Juan Zhou Dong Qu Jun Tai 2023Signal Transduction and Targeted Therapy2023,8,11:0

Specifically,compartment B-to-A CSRs, enriched with hepatic genes, possessed a mosaic status of inactive chromatin and pre-existing and continuous accessibility in fibroblasts. Pre-existing accessibility enhanced the binding of inducible factor Foxa3, which triggered epigenetic activation and chromatin interaction as well as hepatic gene expression. Notably, these changes were restrained within B-to-A CSR boundaries that were defined by CTCF occupancy.
