1SCM-198 protects endometrial stromal cells from oxidative damage through Bax/Bcl-2 and ERK signaling pathways显示文摘In creasi ng amounts of evidence dem on strated that accumulative reactive oxyge n species (ROS) and apoptosis of human endometrial stromal cells (ESCs) are closely associated with endometrial dysfunction induced by oxidative stress, which plays an important role in the pathological process of multiple gyn ecological and reproduction-related diseases. SCM-198, an alkaloid active component of Leonurus japonicas Houtt, has been reported to have anti-oxidative activity. However, the specific mechanisms of SCM-198 in the prevention of endometrial damage remain unknown. In the present study, we assessed the effect of SCM-198 on hydrogen peroxide (H2O2Hnduced oxidative injury in ESCs. ESCs were pretreated with SCM-198 for 4 h and then challenged with H2O2.Morphology ch a nges, apoptosis rate, and intracellular ROS producti on were measured to assess the level of oxidative injury. Flow cytometry and western blot analysis were performed to detect the expression levels of Bax, Bcl-2, active-caspase-3, and mitogen-activated protein kinases pathways. Classic inflammation cytokines were measured by real-time polymerase chain reactions. Our results showed that SCM-198 attenuated apoptosis and ROS generation of ESCs induced by H2O2. H2O2 induced the apparent apoptotic characteristics, including fragmentation of DNA, upregulation of Bax/Bcl2, activation of caspase-3, and secretion of inflammation cytokines, which were all ameliorated by SCM-198. Furthermore, H2O2-induced apoptosis-related ERK1/2 pathway activation was restrained by SCM-198 pretreatme nt. 2Cancer-associated fibroblasts in breast cancer:Challenges and opportunities显示文摘The tumor microenvironment is proposed to contribute substantially to the progression of cancers,including breast cancer.Cancer-associated fibroblasts(CAFs)are the most abundant components of the tumor microenvironment.Studies have revealed that CAFs in breast cancer originate from several types of cells and promote breast cancer malignancy by secreting factors,generating exosomes,releasing nutrients,reshaping the extracellular matrix,and suppressing the function of immune cells.CAFs are also becoming therapeutic targets for breast cancer due to their specific distribution in tumors and their unique biomarkers.Agents interrupting the effect of CAFs on surrounding cells have been developed and applied in clinical trials.Here,we reviewed studies examining the heterogeneity of CAFs in breast cancer and expression patterns of CAF markers in different subtypes of breast cancer.We hope that summarizing CAFrelated studies from a historical perspective will help to accelerate the development of CAF-targeted therapeutic strategies for breast cancer.Dengdi Hu Zhaoqing Li Bin Zheng Xixi Lin Yuehong Pan Peirong Gong Wenying Zhuo Yujie Hu Cong Chen Lini Chen Jichun Zhou Linbo Wang 2022Cancer Communications2022,42,5:9
3Multiphysics simulation method of Lamb wavepropagation with piezoelectric transducers under load condition显示文摘Lamb Wave(LW) simulation under time-varying conditions is an effective and low cost way to study the problem of the low reliability of the structural health monitoring methods based on the LW and Piezoelectric Transducer(PT). In this paper, a multiphysics simulation method of the LW propagation with the PTs under load condition is proposed. With this method, two key mechanisms of the load influence on the LW propagation are considered and coupled with each other. The first mechanism is the acoustoelastic effect which is the main reason of the LW velocity change. The second key mechanism is the load influence on piezoelectric materials, which results in a change of the amplitude. Based on the computational platform of the COMSOL Multiphysics, a multiphysics simulation model of the LW propagation with the PTs under load condition is established. The simulation model includes two physical phenomena. The first one is called solid mechanics, which is used to simulate the acoustoelastic effect being combined with the hyperelastic material properties of the structure in which the LW propagates. The second one is called electromechanical coupling, which considers the simulation of the piezoelectric effect of the PTs for the LW excitation and sensing. To simulate the load influence on piezoelectric materials, a non-linear numerical model of the relationship between the load and the piezoelectric coefficient d31 is established based on an experiment of the load influence on the LW. The simulation results under uniaxial tensile load condition are obtained and are compared with the data obtained from the experiment. 6Dissecting the epigenomic dynamics of human fetal germ cell development at single-cell resolution显示文摘Proper development of fetal germ cells(FGCs)is vital for the precise transmission of genetic and epigenetic information through generations.The transcriptional landscapes of human FGC development have been revealed;however,the epigenetic reprogramming process of FGCs remains elusive.Here,we profiled the genome-wide DNA methylation and chromatin accessibility of human FGCs at different phases as well as gonadal niche cells at single-cell resolution.First,we found that DNA methylation levels of FGCs changed in a temporal manner,whereas FGCs at different phases in the same embryo exhibited comparable DNA methylation levels and patterns.Second,we revealed the phase-specific chromatin accessibility signatures at the promoter regions of a large set of critical transcription factors and signaling pathway genes.We also identified potential distal regulatory elements including enhancers in FGCs.Third,compared with other hominid-specific retrotransposons,SVA_D might have a broad spectrum of binding capacity for transcription factors,including SOX15 and SOX17.Finally,using an in vitro culture system of human FGCs,we showed that the BMP signaling pathway promoted the cell proliferation of FGCs,and regulated the WNT signaling pathway by orchestrating the chromatin accessibility of its ligand genes.Our single-cell epigenomic atlas and functional assays provide valuable insights for understanding the strongly heterogeneous,unsynchronized,yet highly robust nature of human germ cell development.Li Li Lin Li Qingqing Li Xixi Liu Xinyi Ma Jun Yong Shuai Gao Xinglong Wu Yuan Wei Xiaoye Wang Wei Wang Rong Li Jie Yan Xiaohui Zhu Lu Wen Jie Qiao Liying Yan Fuchou Tang 2021Cell Research2021,31,4:1

11Identification of an immune classifier for predicting the prognosis and therapeutic response in triple-negative breast cancer显示文摘Triple-negative breast cancer(TNBC)poses a significant challenge due to the lack of reliable prognostic gene signatures and an understanding of its immune behavior.Methods:We analyzed clinical information and mRNA expression data from 162 TNBC patients in TCGA-BRCA and 320 patients in METABRIC-BRCA.Utilizing weighted gene coexpression network analysis,we pinpointed 34 TNBC immune genes linked to survival.The least absolute shrinkage and selection operator Cox regression method identified key TNBC immune candidates for prognosis prediction.We calculated chemotherapy sensitivity scores using the“pRRophetic”package in R software and assessed immunotherapy response using the Tumor Immune Dysfunction and Exclusion algorithm.Results:In this study,34 survival-related TNBC immune gene expression profiles were identified.A least absolute shrinkage and selection operator-Cox regression model was used and 15 candidates were prioritized,with a concomitant establishment of a robust risk immune classifier.The high-risk TNBC immune groups showed increased sensitivity to therapeutic agents like RO-3306,Tamoxifen,Sunitinib,JNK Inhibitor VIII,XMD11-85h,BX-912,and Tivozanib.An analysis of the Search Tool for Interaction of Chemicals database revealed the associations between the high-risk group and signaling pathways,such as those involving Rap1,Ras,and PI3K-Akt.The low-risk group showed a higher immunotherapy response rate,as observed through the tumor immune dysfunction and exclusion analysis in the TCGA-TNBC and METABRIC-TNBC cohorts.Conclusion:This study provides insights into the immune complexities of TNBC,paving the way for novel diagnostic approaches and precision treatment methods that exploit its immunological intricacies,thus offering hope for improved management and outcomes of this challenging disease.KUAILU LIN QIANYU GU XIXI LAI 2023BIOCELL2023,47,12:0
12Preparation and Properties of Sapphire by Edge-defined Film-fed Growth(EFG) Method with Different Growth Directions显示文摘Sapphire, belonging to hexagonal crystal system, is typically anisotropic which makes it direction-sensitive. To research the effects of growth directions on properties of sapphire, c-[0001] seed(c-sapphire) and a-[11-20] seed(a-sapphire) were used to prepare sapphire by edge-defined film-fed growth(EFG) method. The samples were analyzed through lattice integrity, dislocation and corrosion performance by double-crystal XRD, OM, AFM, SEM and EDX. It was shown that the lattice integrities of two growth-direction crystals were both well due to the small FWHM values. While the average densities of dislocation in c-sapphire and a-sapphire were 9.2×103 and 3.9×103 cm-2 respectively, the energy of dislocation in c-sapphire was lower than that in a-sapphire. During Strong Phosphoric Acid(SPA) etching, the surface of c-sapphire basically kept smooth but in a-sapphire there were many point-like corrosion pits where aluminum and oxygen atoms lost by 2:1. 13New perspective of skeletal stem cells显示文摘Tissue-resident stem cells are a group of stem cells distinguished by their capacity for self-renewal and multilineage differentiation capability with tissue specificity.Among these tissue-resident stem cells,skeletal stem cells(SSCs)were discovered in the growth plate region through a combination of cell surface markers and lineage tracing series.With the process of unravelling the anatomical variation of SSCs,researchers were also keen to investigate the developmental diversity outside the long bones,including in the sutures,craniofacial sites,and spinal regions.Recently,fluorescence-activated cell sorting,lineage tracing,and single-cell sequencing have been used to map lineage trajectories by studying SSCs with different spatiotemporal distributions.The SSC niche also plays a pivotal role in regulating SSC fate,such as cell-cell interactions mediated by multiple signalling pathways.This review focuses on discussing the spatial and temporal distribution of SSCs,and broadening our understanding of the diversity and plasticity of SSCs by summarizing the progress of research into SSCs in recent years.Guixin Yuan Zan Li Xixi Lin Na Li Ren Xu 2022Biomaterials Translational2022,3,4:0
14Inductive effect of hydroxyl safflower yellow-A on apoptosis in abnormal HUVEC via the mitochondrial pathway显示文摘Objective:To investigate the mechanism by which hydroxyl safflower yellow A,an active component of safflower (Carthamus tinctorius L.),promotes apoptosis in abnormal human umbilical vein endothelial cells (HUVECs).Methods:Supernatant of BGC-823 was used to stimulate HUVECs to establish a model of abnormal proliferation of HUVECs.After determining an ideal concentration of HSYA by MTT assay,apoptosis was detected with flow cytometry and TUNEL assay.Mechanism of apoptosis was assessed using quantitative real-time polymerase chain reaction,Western blot,and ELISA.Results:A range of concentrations of HSYA inhibited proliferation and promoted apoptosis of abnormal HUVECs.As the rate of apoptosis increased,mRNA expression of caspase-3 increased while expression of mutant p53 decreased.HSYA had no effect on Fas gene expression.Analogously,protein expression of Bax was increased while those of Bcl-2,Fas,and Fas-L were decreased.Conclusions:HSYA appears to induce apoptosis of HUVECs with the stimulation of the supematant of tumor cells.The mechanism of apoptosis by HSYA may involve activation of the mitochondrial apoptotic pathway and regulation of the expressions of Bcl-2,Bax,and p53.Na Si Jingjing Wang Yingying Xu Lintao Liu Xu Wang Hongmei Sun Zong Lin Xixi Wang Li Liu Qi Zhang Qian Zhang Cuiling Feng 2015Journal of Traditional Chinese Medical Sciences2015,2,1:0
