| 1 | In vitro assessment of thyroid hormone receptor activity of four organophosphate esters显示文摘Previous animal experiments have implied that organophosphate esters(OPEs) have a disruption effect on the thyroid endocrine system. However, knowledge of the toxicological mechanism remains limited. In this study, the activities of four OPEs have been characterized against the thyroid hormone(TH) nuclear receptor(TR) using two in vitro models, with the aim of evaluating their toxicity mechanisms towards the TR. The results of a TH-dependent cell proliferation assay showed that tris(2-chloro-1-(chloromethyl)ethyl)phosphate(TDCPP) could induce cell growth, while the other three OPEs had no effect. The results of a luciferase reporter gene assay revealed that all four of the OPEs tested in the current study showed agonistic activity towards TRβ, with TDCPP being the most potent one. Moreover, molecular docking revealed that all the tested OPEs could fit into the ligand binding pocket of TRβ, with TDCPP binding more effectively than the other three OPEs. | 3 | Single-cell transcriptomic analysis reveals a systemic immune dysregulation in COVID-19-associated pediatric encephalopathy显示文摘Unraveling the molecular mechanisms for COVID-19-associated encephalopathy and its immunopathology is crucial for developing effective treatments.Here,we utilized single-cell transcriptomic analysis and integrated clinical observations and laboratory examination to dissect the host immune responses and reveal pathological mechanisms in COVID-19-associated pediatric encephalopathy.We found that lymphopenia was a prominent characteristic of immune perturbation in COVID-19 patients with encephalopathy,especially those with acute necrotizing encephalopathy(AE).This was characterized a marked reduction of various lymphocytes(e.g.,CD8^(+)T and CD4^(+)T cells)and significant increases in other inflammatory cells(e.g.,monocytes).Further analysis revealed activation of multiple cell apoptosis pathways(e.g.,granzyme/perforin-,FAS-and TNF-induced apoptosis)may be responsible for lymphopenia.A systemic S100A12 upregulation,primarily from classical monocytes,may have contributed to cytokine storms in patients with AE.A dysregulated type I interferon(IFN)response was observed which may have further exacerbated the S100A12-driven inflammation in patients with AE.In COVID-19 patients with AE,myeloid cells(e.g.,monocytic myeloid-derived suppressor cells)were the likely contributors to immune paralysis.Finally,the immune landscape in COVID-19 patients with encephalopathy,especially for AE,were also characterized by NK and T cells with widespread exhaustion,higher cytotoxic scores and inflammatory response as well as a dysregulated B cell-mediated humoral immune response.Taken together,this comprehensive data provides a detailed resource for elucidating immunopathogenesis and will aid development of effective COVID-19-associated pediatric encephalopathy treatments,especially for those with AE. | Yi Wang Laurence Don Wai Luu Shuang Liu Xiong Zhu Siyuan Huang Fang Li Xiaolan Huang Linying Guo Jin Zhang Haiyan Ge Yuanyuan Sun Yi Hui Yanning Qu Huicong Wang Xiaoxia Wang Weilan Na Juan Zhou Dong Qu Jun Tai | 2023 | Signal Transduction and Targeted Therapy2023,8,11: | 0 |
| 4 | Comparison between pinning control of different chaotic complex dynamical networks显示文摘The stabilization properties of various typical complex dynamical networks composed of chaotic nodes are theoretically investigated and numerically simulated in detail. Some local stability properties of such pinned networks are derived and the valid stability regions are estimated based on eigenvalue analysis. Numerical simulations of such networks are given to explain why significantly less local controllers are needed by the specifically pinning scheme, which pins the most highly connected nodes in scale-free networks, than that required by the randomly pinning scheme. | 6 | Impact of Novel Coronavirus Vaccination on the Clinical Characteristics and Parental Care Needs of Infected Children During a High-Intensity Epidemic Period显示文摘Objective:This study aimed to investigate the impact of COVID-19 vaccination on the clinical characteristics and care needs of infected children during a high-intensity pandemic and to measure the potential effects of vaccination on children's health status and healthcare utilization.Methods:A sample of 903 children who had recently been infected with the novel coronavirus and came to our hospital's emergency department from January 11 to January 30,2023,was used as the research object in this retrospective study.The questionnaire on unknown coronavirus infection was filled out anonymously,and SPSS20.0 software was used for statistical data analysis.Results:The baseline conditions of 903 subjects were studied,including 505 males,accounting for 55.92%,and 398 females,accounting for 44.08%.The age of infection was mainly concentrated in preschool children(26.02%)and school-age children(38.76%).Regarding vaccination,561 cases were vaccinated,accounting for 62.13%.Among them,37 children received one dose of vaccine,accounting for 4.1%,463 children received two doses,accounting for 51.27%,and 61 children received three doses,accounting for 6.76%.For previous allergic diseases,180 children had a history of allergic diseases,accounting for 19.93%.Regarding sources of infection,the most common source was someone at home,accounting for 82.61%.Vaccination can effectively reduce the fever of children infected with the new coronavirus,the number of outpatient and emergency doctor visits,and the risk of hospitalization.In addition,regarding treatment,children in the vaccinated group were more likely to be treated at home,and the types of drugs used also differed from those in the non-vaccinated group.However,the vaccination group has a relatively high incidence of symptoms such as sore throat,cough,abnormal sense of smell and taste,muscle soreness,and headache.However,these are mild clinical symptoms and do not affect children's physical health and development.Parental care needs for children infected with COVID-19 are disparate,and parents of vaccinated children have lower proportions of needs for child care.Conclusion:Vaccination positively impacts the clinical outcomes of children infected with COVID-19 and their parents'well-being. | Hui Wang Yanyu Lyu Linying Guo | 2023 | Journal of Clinical and Nursing Research2023,7,5: | 0 |
