1Variation in cis-regulation of a NAC transcription factor contributes to drought tolerance in wheat显示文摘Drought is a major environmental factor limiting wheat production worldwide,and developing drought-tolerant cultivars is a central challenge for wheat breeders globally.Therefore,it is important to identify genetic components determining drought tolerance in wheat.In this study,we identified a wheat NAC gene(TaNAC071-A)that is tightly associated with drought tolerance by a genome-wide association study.Knockdown of TaNAC071-A in wheat attenuated plant drought tolerance,whereas its overexpression significantly enhanced drought tolerance through improved water-use efficiency and increased expression of stress-responsive genes.This heightened water-saving mechanism mitigated the yield loss caused by water deficit.Further candidate gene association analysis showed that a 108-bp insertion in the promoter of TaNAC071-A alters its expression level and contributes to variation in drought tolerance among wheat accessions.This insertion contains two MYB cis-regulatory elements(CREs)that can be directly bound by the MYB transcription activator,TaMYBL1,thereby leading to increased TaNAC071-A expression and plant drought tolerance.Importantly,introgression of this 108-bp insertion allele,TaNAC071-AIn-693,into drought-sensitive cultivars could improve their drought tolerance,demonstrating that it is a valuable genetic resource for wheat breeding.Taken together,our findings highlight a major breakthrough in determining the genetic basis underlying phenotypic variation in wheat drought tolerance and showcase the potential of exploiting CRE-containing indels for improving important agronomical traits.Hude Mao Shumin Li Bin Chen Chao Jian Fangming Mei Yifang Zhang Fangfang Li Nan Chen Tian Li Linying Du Li Ding Zhongxue Wang Xinxiu Cheng Xiaojing Wang Zhensheng Kang 2022Molecular Plant2022,15,2:9
2Copper-ceria sheets catalysts: Effect of copper species on catalytic activity in CO oxidation reaction显示文摘Copper-ceria sheets catalysts with different loadings of copper(2 wt.%, 5 wt.% and 10 wt.%) supported on ceria nanosheets were synthesized via a depositioneprecipitation(DP) method. The prepared catalysts were systematically characterized with various structural and textural detections including X-ray diffraction(XRD), Raman spectra, transmission electron microscopy(TEM), X-ray absorption fine structure(XAFS), and temperature-programmed reduction by hydrogen(H_2-TPR), and tested for the CO oxidation reaction. Notably, the sample containing 5 wt.% of Cu exhibited the best catalytic performance as a result of the highest number of active CuO species on the catalyst surface. Further increase of copper content strongly affects the dispersion of copper and thus leads to the formation of less active bulk CuO phase, which was verified by XRD and H_2-TPR analysis. Moreover, on the basis of in-situ diffuse reflectance infrared Fourier transform spectroscopy(in-situ DRIFTS) results, the surface Cu^+ species, which are derived from the reduction of Cu^(2+), are likely to play a crucial role in the catalyzing CO oxidation.Consequently, the superior catalytic performance of the copper-ceria sheets is mainly attributed to the highly dispersed CuO_x cluster rather than Cu-[O_x]-Ce structure, while the bulk CuO phase is adverse to the catalytic activity of CO oxidation.Linying Du Weiwei Wang Han Yan Xu Wang Zhao Jin Qisheng Song Rui Si Chunjiang Jia 2017Journal of Rare Earths2017,35,12:5

4Wheat adaptation to environmental stresses under climate change:Molecular basis and genetic improvement显示文摘Wheat(Triticum aestivum)is a staple food for about 40%of the world’s population.As the global population has grown and living standards improved,high yield and improved nutritional quality have become the main targets for wheat breeding.However,wheat production has been compromised by global warming through the more frequent occurrence of extreme temperature events,which have increased water scarcity,aggravated soil salinization,caused plants to be more vulnerable to diseases,and directly reduced plant fertility and suppressed yield.One promising option to address these challenges is the genetic improvement of wheat for enhanced resistance to environmental stress.Several decades of progress in genomics and genetic engineering has tremendously advanced our understanding of the molecular and genetic mechanisms underlying abiotic and biotic stress responses in wheat.These advances have heralded what might be considered a“golden age”of functional genomics for the genetic improvement of wheat.Here,we summarize the current knowledge on the molecular and genetic basis of wheat resistance to abiotic and biotic stresses,including the QTLs/genes involved,their functional and regulatory mechanisms,and strategies for genetic modification of wheat for improved stress resistance.In addition,we also provide perspectives on some key challenges that need to be addressed.Hude Mao Cong Jiang Chunlei Tang Xiaojun Nie Linying Du Yuling Liu Peng Cheng Yunfeng Wu Huiquan Liu Zhensheng Kang Xiaojie Wang 2023Molecular Plant2023,16,10:0
5Robust Benchmark Structural Variant Calls of An Asian Using State-of-the-art Long-read Sequencing Technologies显示文摘The importance of structural variants(SVs)for human phenotypes and diseases is now recognized.Although a variety of SV detection platforms and strategies that vary in sensitivity and specificity have been developed,few benchmarking procedures are available to confidently assess their performances in biological and clinical research.To facilitate the validation and application of these SV detection approaches,we established an Asian reference material by characterizing the genome of an Epstein-Barr virus(EBV)-immortalized B lymphocyte line along with identified benchmark regions and high-confidence SV calls.We established a high-confidence SV callset with 8938 SVs by integrating four alignment-based SV callers,including 109×Pacific Bio sciences(PacBio)continuous long reads(CLRs),22×PacBio circular consensus sequencing(CCS)reads,104×Oxford Nanopore Technologies(ONT)long reads,and 114×Bionano optical mapping platform,and one de novo assembly-based SV caller using CCS reads.A total of 544 randomly selected SVs were validated by PCR amplification and Sanger sequencing,demonstrating the robustness of our SV calls.Combining trio-binning-based haplotype assemblies,we established an SV benchmark for identifying false negatives and false positives by constructing the continuous high-confidence regions(CHCRs),which covered 1.46 gigabase pairs(Gb)and 6882 SVs supported by at least one diploid haplotype assembly.Establishing high-confidence SV calls for a benchmark sample that has been characterized by multiple technologies provides a valuable resource for investigating SVs in human biology,disease,and clinical research.Xiao Du Lili Li Fan Liang Sanyang Liu Wenxin Zhang Shuai Sun Yuhui Sun Fei Fan Linying Wang Xinming Liang Weijin Qiu Guangyi Fan Ou Wang Weifei Yang Jiezhong Zhang Yuhui Xiao Yang Wang Depeng Wang Shoufang Qu Fang Chen Jie Huang 2022Genomics, Proteomics & Bioinformatics2022,20,1:0
