1Differentiating Crohn's disease from intestinal tuberculosis显示文摘Differentiating Crohn's disease(CD) and intestinal tuberculosis(ITB) has remained a dilemma for most of the clinicians in the developing world, which are endemic for ITB, and where the disease burden of inflammatory bowel disease is on the rise. Although, there are certain clinical(diarrhea/hematochezia/perianal disease common in CD; fever/night sweats common in ITB), endoscopic(longitudinal/aphthous ulcers common in CD; transverse ulcers/patulous ileocaecal valve common in ITB), histologic(caseating/confluent/large granuloma common in ITB; microgranuloma common in CD), microbiologic(positive stain/culture for acid fast-bacillus in ITB), radiologic(long segment involvement/comb sign/skip lesions common in CD; necrotic lymph node/contiguous ileocaecal involvement common in ITB), and serologic differences between CD and ITB, the only exclusive features are caseation necrosis on biopsy, positive smear for acid-fast bacillus(AFB) and/or AFB culture, and necrotic lymph node on cross-sectional imaging in ITB. However,these exclusive features are limited by poor sensitivity, and this has led to the development of multiple multi-parametric predictive models. These models are also limited by complex formulae, small sample size and lack of validation across other populations. Several new parameters have come up including the latest Bayesian meta-analysis, enumeration of peripheral blood T-regulatory cells, and updated computed tomography based predictive score. However, therapeutic anti-tubercular therapy(ATT) trial, and subsequent clinical and endoscopic response to ATT is still required in a significant proportion of patients to establish the diagnosis. 2Spectroscopic analysis on the binding interaction of biologically active pyrimidine derivative with bovine serum albumin显示文摘A biologically active antibacterial reagent, 2–amino-6-hydroxy–4–(4-N, N-dimethylaminophenyl)-pyrimidine-5-carbonitrile(AHDMAPPC), was synthesized. It was employed to investigate the binding interaction with the bovine serum albumin(BSA) in detail using different spectroscopic methods. It exhibited antibacterial activity against Escherichia coli and Staphylococcus aureus which are common food poisoning bacteria. The experimental results showed that the fluorescence quenching of model carrier protein BSA by AHDMAPPC was due to static quenching. The site binding constants and number of binding sites(n E 1) were determined at three different temperatures based on fluorescence quenching results. The thermodynamic parameters, enthalpy change(ΔH), free energy(ΔG) and entropy change(ΔS) for the reaction were calculated to be 15.15 k J/mol, –36.11 k J/mol and 51.26 J/mol K according to van't Hoff equation, respectively. The results indicated that the reaction was an endothermic and spontaneous process, and hydrophobic interactions played a major role in the binding between drug and BSA. The distance between donor and acceptor is 2.79 nm according to F?rster's theory. The alterations of the BSA secondary structure in the presence of AHDMAPPC were confirmed by UV–visible, synchronous fluorescence, circular dichroism(CD) and three-dimensional fluorescence spectra. All these results indicated that AHDMAPPC can bind to BSA and be effectively transported and eliminated in the body. It can be a useful guideline for further drug design.Vishwas D. 4Intestinal permeability and its association with the patient and disease characteristics in Crohn's disease显示文摘AIM:To assess the intestinal permeability (IP) in patients with Crohn's disease (CD) and study the association of IP with the patient and disease characteristics. METHODS: One hundred and twenty five consecutive patients of CD (Males: 66) were diagnosed on the basis of a combination of standard clinical, endoscopic, imaging and histological features. CD activity index (CDAI) was used to calculate the activity of the disease while the behavior of the disease was assessed by the modified Montreal classification. IP was measured by the ratio of the percentage excretion of ingested doses of lactulose and mannitol in urine (LMR). The upper limit of normality of LMR (0.037) was derived from 22 healthy controls. RESULTS: Thirty six percent of patients with CD had increased IP. There was no significant difference in mannitol excretion (patients vs controls = 12.5% vs 14.2%, P = 0.4652), but lactulose excretion was significantly higher in patients compared to healthy controls (patients vs controls = 0.326% vs 0.293%, P = 0.0391). The mean LMR was also significantly higher in the patients as compared to healthy controls [0.027 (0.0029-0.278) vs 0.0164 (0.0018-0.0548), P = 0.0044]. Male patients had a higher LMR compared to females [0.036 (95% CI 0.029, 0.046) vs 0.022 (95% CI 0.0178, 0.028) (P = 0.0024), though there was no difference in the number of patients with abnormal IP in boththe sexes. Patients with an ileo-colonic disease had a higher LMR than those with only colonic disease [0.045 (95% CI 0.033, 0.06) vs 0.021 (95% CI 0.017, 0.025) (P < 0.001)]. Of patients with ileo-colonic disease, 57.8% had an abnormal IP, compared to 26.7% with colonic and 15.6% with small intestinal disease. Patients with a stricturing disease had significantly higher LMR compared to non-fistulising non-stricturing disease [0.043 (95% CI 0.032, 0.058) vs 0.024 (95% CI 0.019, 0.029) (P = 0.0062)]. There was no correlation of IP with age, disease activity, duration of illness, D-xylose absorption, upper GI involvement, perianal disease, and extra- intestinal manifestations. On multiple regression analysis, male gender and ileo-colonic disease were independent factors associated with increased IP. Gender, location, behavior of the disease and upper GI involvement could explain up to 23% of variability in IP (R2 = 0.23). CONCLUSION: IP was increased in 36% of patients with CD. 5Recommendations to quantify villous atrophy in video capsule endoscopy images of celiac disease patients显示文摘AIM To quantify the presence of villous atrophy in endoscopic images for improved automation.METHODS There are two main categories of quantitative descriptors helpful to detect villous atrophy:(1) Statistical and(2) Syntactic. Statistical descriptors measure the small intestinal substrate in endoscope-acquired images based on mathematical methods. Texture is the most commonly used statistical descriptor to quantify villous atrophy. Syntactic descriptors comprise a syntax, or set of rules, for analyzing and parsing the substrate into a set of objects with boundaries. The syntax is designed to identify and distinguish three-dimensional structures based on their shape.RESULTS The variance texture statistical descriptor is useful to describe the average variability in image gray level representing villous atrophy, but does not determine the range in variability and the spatial relationships between regions. Improved textural descriptors will incorporate these factors, so that areas with variability gradients and regions that are orientation dependent can be distinguished. The protrusion syntactic descriptor is useful to detect three-dimensional architectural components, but is limited to identifying objects of a certain shape. 7Prediction of daily sediment discharge using a back propagation neural network training algorithm:A case study of the Narmada River,India显示文摘Most of the studies on Artificial Neural Network (ANN) models remain restricted to smaller rivers and catchments. In this paper, an attempt has been made to correlate variability of sediment loads with rainfall and runoff through the application of the Back Propagation Neural Network (BPNN) algorithm for a large tropical river. The algorithm and simulation are done through MATLAB environment. The methodology comprised of a collection of data on rainfall, water discharge, and sediment discharge for the Narmada River at various locations (along with time variables) and application to develop a threelayer BPNN model for the prediction of sediment discharges. For training and validation purposes a set of 549 data points for the monsoon (16 June-15 November) period of three consecutive years (1996–1998) was used. For testing purposes, the BPNN model was further trained using a set of 732 data points of monsoon season of four years (2006–07 to 2009–10) at nine stations. The model was tested by predicting daily sediment load for the monsoon season of the year 2010–11. To evaluate the performance of the BPNN model, errors were calculated by comparing the actual and predicted loads. The validation and testing results obtained at all these locations are tabulated and discussed. Results obtained from the model application are robust and encouraging not only for the sub-basins but also for the entire basin. These results suggest that the proposed model is capable of predicting the daily sediment load even at downstream locations, which show nonlinearity in the transportation process. 8Electrospun CNT embedded ZnO nanofiber based biosensor for electrochemical detection of Atrazine: a step closure to single molecule detection显示文摘In this study we have reported the design and development of a facile,sensitive,selective,and label-free electrochemical sensing platform for the detection of atrazine based on MWCNT-embedded ZnO nanofibers.Electrospun nanofibers were characterized using scanning electron microscope(SEM),transmission electron microscope(TEM),X-ray diffraction(XRD),X-ray photoelectron spectroscope(XPS),UV-Visible spectroscope(UV-VIS),and Fourier-transform infrared spectroscope(FTIR).Electrochemical properties of MWCNT-ZnO nanofiber-modified electrodes were assessed using electrochemical impedance spectroscopy(EIS)and cyclic voltammetry(CV).Binding event of atrazine to anti-atrazine antibody,which immobilized on nanofiber-modified electrode via EDC and NHS chemistry,was transduced with EIS.Due to high conductivity,surface area,and low bandgap of MWCNT-ZnO nanofibers,we have achieved the sensitivity and limit of detection(LoD)of sensor as 21.61(KΩμg^(−1) mL^(−1))cm^(−2) and 5.368zM for a wide detection range of 10 zM–1µM.The proposed immunosensing platform has good stability,selectivity,repeatability,and reproducibility,and are less prone to interference.Patta Supraja Vikrant Singh Siva Rama Krishna Vanjari Shiv Govind Singh 2020Microsystems & Nanoengineering2020,6,1:3
9Implementation of a polling protocol for predicting celiac disease in videocapsule analysis显示文摘AIM: To investigate the presence of small intestinal villous atrophy in celiac disease patients from quantitative analysis of videocapsule image sequences.METHODS: Nine celiac patient data with biopsy-proven villous atrophy and seven control patient data lacking villous atrophy were used for analysis. Celiacs had biopsy-proven disease with scores of Marsh Ⅱ-Ⅲ C except in the case of one hemophiliac patient. At four small intestinal levels (duodenal bulb, distal duodenum, jejunum, and ileum), video clips of length 200 frames (100 s) were analyzed. Twenty-four measurements were used for image characterization. These measurements were determined by quantitatively processing the videocapsule images via techniques for texture analysis, motility estimation, volumetric reconstruction using shape-from-shading principles, and image transformation. Each automated measurement method, or automaton, was polled as to whether or not villous atrophy was present in the small intestine, indicating celiac disease. Each automaton's vote was determined based upon an optimized parameter threshold level, with the threshold levels being determined from prior data. A prediction of villous atrophy was made if it received the majority of votes (≥ 13), while no prediction was made for tie votes (12-12). Thus each set of images was classified as being from either a celiac disease patient or from a control patient. RESULTS: Separated by intestinal level, the overall sensitivity of automata polling for predicting villous atrophy and hence celiac disease was 83.9%, while the specificity was 92.9%, and the overall accuracy of automata-based polling was 88.1%. The method of image transformation yielded the highest sensitivity at 93.8%, while the method of texture analysis using subbands had the highest specificity at 76.0%. Similar results of prediction were observed at all four small intestinal locations, but there were more tie votes at location 4 (ileum). Incorrect prediction which reduced sensitivity occurred for two celiac patients with Marsh type Ⅱ pattern, which is characterized by crypt hyperplasia, but normal villous architecture. Pooled from all levels, there was a mean of 14.31 ± 3.28 automaton votes for celiac vs 9.67 ± 3.31 automaton votes for control when celiac patient data was analyzed (P<0.001). Pooled from all levels, there was a mean of 9.71 ± 2.8128 automaton votes for celiac vs 14.32 ± 2.7931 automaton votes for control when control patient data was analyzed (P<0.001). CONCLUSION: Automata-based polling may be useful to indicate presence of mucosal atrophy, indicative of celiac disease, across the entire small bowel, though this must be confirmed in a larger patient set. Since the method is quantitative and automated, it can potentially eliminate observer bias and enable the detectionof subtle abnormality in patients lacking a clear diagnosis. 11Growth, photosynthesis and nitrogen metabolism in soybean varieties after exclusion of the UV-B and UV-A/B components of solar radiation显示文摘A field experiment was conducted to study the impact of the exclusion of the solar UV components on growth, photosynthesis and nitrogen metabolism in soybean(Glycine max)varieties PK-472, Pusa-24, JS 71-05, JS-335, NRC-7 and Kalitur. The plants were grown in specially designed UV exclusion chambers wrapped with filters to exclude UV-B or UV-A/B and transmitted all UV. Exclusion of UV significantly enhanced the growth of the aerial parts as well as the growth of the below ground parts in all of the six soybean varieties.Nitrate reductase activity(NRA) was significantly reduced, whereas leghemoglobin(Lb)content, total soluble protein, net photosynthesis(Pn) and α-tocopherol content were enhanced after UV exclusion. The exclusion of solar UV-A/B enhanced all parameters to a larger extent than the exclusion of solar UV-B in four of the six varieties of soybean except for NRC-7 and Kalitur. These two varieties responded more to UV-B exclusion compared to UV-A/B exclusion. A significant inverse correlation between the NRA and the number of nodules per plant was observed. The extent of response in all parameters was greater in PK-472 and JS71-05 than that in Kalitur and JS-335 after UV exclusion. The exclusion of UV augmented the growth of nodules, Lb content and α-tocopherol levels and conferred higher rates of Pnto support better growth of nodules. 12Low dose oral curcumin is not effective in induction of remission in mild to moderate ulcerative colitis: Results from a randomized double blind placebo controlled trial显示文摘AIM To evaluate the role of oral curcumin in inducing clinical remission in patients with mild to moderate ulcerative colitis(UC).METHODS A prospective randomized double-blind placebo-controlled trial comparing the remission inducing effect of oral curcumin and mesalamine 2.4 g with placebo and mesalamine 2.4 g in patients of ulcerative colitis with mild to moderate severity was conducted from January 2003 to March 2005. The included patients received 1 capsule thrice a day of placebo or curcumin(150 mg) for 8 wk. Patients were evaluated clinically and endoscopically at 0,4 and 8 wk. The primary outcome was clinical remission at 8 wk and secondary outcomes were clinical response, mucosal healing and treatment failure at 8 wk. The primary analysis was intention to treat worst case scenario(ITT-WCS).RESULTS Of 300 patients with UC, 62 patients(curcumin: 29, placebo: 33) fulfilled the inclusion criteria and were randomized at baseline. Of these, 21 patients did not complete the trial, 41 patients(curcumin: 16, placebo: 25) finally completed 8 wk. 13Artificial neural network simulation for prediction of suspended sediment concentration in the River Ramganga,Ganges Basin,India显示文摘The relation between the water discharge (Q) and suspended sediment concentration (SSC) of the River Ramganga at Bareilly, Uttar Pradesh, in the Himalayas, has been modeled using Artificial Neural Networks (ANNs). The current study validates the practical capability and usefulness of this tool for simulating complex nonlinear, real world, river system processes in the Himalayan scenario. The modeling approach is based on the time series data collected from January to December (2008-2010) for Q and SSC. Three ANNs (T1-T3) with different network configurations have been developed and trained using the Levenberg Marquardt Back Propagation Algorithm in the Matlab routines. Networks were optimized using the enumeration technique, and, finally, the best network is used to predict the SSC values for the year 2011. The values thus obtained through the ANN model are compared with the observed values of SSC. The coefficient of determination (R2), for the optimal network was found to be 0.99. The study not only provides insight into ANN modeling in the Himalayan river scenario, but it also focuses on the importance of understanding a river basin and the factors that affect the SSC, before attempting to model it. 16Preslaughter diet management in sheep and goats:effects on physiological responses and microbial loads on skin and carcass显示文摘Sixteen crossbred buck goats(Kiko x Spanish;BW = 32.8 kg) and wether sheep(Dorset x Suffolk;BW = 39.9 kg) were used to determine the effect of preslaughter diet and feed deprivation time(FDT) on physiological responses and microbial loads on skin and carcasses.Experimental animals were fed either a concentrate(CD) or a hay diet(HD)for 4 d and then deprived of feed for either 12-h or 24-h before slaughter.Blood samples were collected for plasma Cortisol and blood metabolite analyses.Longisimus muscle(LM) pH was measured.Skin and carcass swabs were obtained to assess microbial loads.Plasma creatine kinase activity(863.9 and 571.7 ± 95.21 IU) and non-esterified fatty acid concentrations(1,056.1 and 589.8 + 105.01 mEq/L) were different(P< 0.05) between sheep and goats.Species and diet treatments had significant effects on the ultimate pH of LM.Pre-holding total coliform(TCC) and aerobic plate counts(APC) of skin were significantly different between species.Goats had lower(P < 0.05) TCC(2.1 vs.3.0 log_(10) CFU/cm^2) and APC(8.2 vs.8.5 log_(10) CFU/cm^2) counts in the skin compared to sheep.Preslaughter skin E.coli counts and TCC were different(P < 0.05) between species.Goats had lower(P < 0.05) counts of E.coli(2.2 vs.2.9 log_(10) CFU/cm^2) and TCC(2.3 vs.3.0 log_(10) CFU/cm^2) in the skin compared with those in sheep.Diet,species,and FDT had no effect(P> 0.05) on E.coli and TCC in carcass swab samples.The APC of carcass swab samples were only affected(P < 0.05) by the FDT.The results indicated that preslaughter dietary management had no significant changes on hormone and blood metabolite concentrations and sheep might be more prone for fecal contamination than goats in the holding pens at abattoir.Govind Kannan Venkat R Gutta Jung Hoon Lee Brou Kouakou Will R Getz George W McCommon 2015Journal of Animal Science and Biotechnology2015,6,1:2
