1Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China,has become a public health emergency of international concern.The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models.Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs,viral replication,chest X-ray,histopathological changes and immune response.Results:Viral replication of nasopharyngeal swabs,anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema,notably,old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection.Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin 2020Animal Models and Experimental Medicine2020,3,1:22

3Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
4Relative importance of climate factors and human activities in impacting vegetation dynamics during 2000–2015 in the Otindag Sandy Land,northern China显示文摘In recent years, there has been increasing research interests in differentiating the relative importance of climate factors and human activities in impacting vegetation dynamics. In this study, based on residual trend method, we used MOD13A3(MODIS vegetation index product), MCD12Q1(MODIS land cover product) and meteorological datasets to differentiate the relative importance of climate factors and human activities in impacting vegetation dynamics during 2000–2015 in the Otindag Sandy Land, northern China. Results show that during the study period(2000–2015), the overall vegetation condition had improved in the Otindag Sandy Land. The driving forces of vegetation dynamics differed spatially in the whole study area over the study period. The area with vegetation degradation solely resulted from human activities accounted for 8.23% of the study area, while the area with vegetation degradation resulted from others(including climate factors and combination of climate factors and human activities) occupied 1.53%. The area with vegetation recovery benefitted from human activities occurred over 26.02% of the study area; the area benefitted from climate factors accounted for 23.69%; and the area benefitted from both climate factors and human activities occupied 37.74%. 5Transcriptome profiling of the developing postnatal mouse testis using next-generation sequencing显示文摘Mammalian testis development is a complex and highly sophisticated process. To study the dynamic change of normal testis development at the transcriptional level, we investigated mouse testes at three postnatal ages: 6 days postnatal, 4 weeks old, and 10 weeks old, representing infant (PN1), juvenile (PN2), and adult (PN3) stages, respectively. Using ultra high-throughput RNA sequencing (RNA-seq) technology, we obtained 211 million reads with a length of 35 bp. We identified 18837 genes that were expressed in mouse testes, and found that genes expressed at the highest level were involved in spermatogenesis. The gene expression pattern in PN1 was distinct from that in PN2 and PN3, which indicates that spermatogenesis has commenced in PN2. We analyzed a large number of genes related to spermatogenesis and somatic development of the testis, which play important roles at different developmental stages. We also found that the MAPK, Hedgehog, and Wnt signaling pathways were significantly involved at different developmental stages. These findings further our understanding of the molecular mechanisms that regulate testis development. 6A Comprehensive Transcriptomic Analysis of Infant and Adult Mouse Ovary显示文摘Ovary development is a complex process involving numerous genes. A well-developed ovary is essential for females to keep fertility and reproduce offspring. In order to gain a better insight into the molecular mechanisms related to the process of mammalian ovary development,we performed a comparative transcriptomic analysis on ovaries isolated from infant and adult mice by using next-generation sequencing technology(SOLi D). We identified 15,454 and 16,646 transcriptionally active genes at the infant and adult stage, respectively. Among these genes, we also identified 7021 differentially expressed genes. Our analysis suggests that, in general, the adult ovary has a higher level of transcriptomic activity. However, it appears that genes related to primordial follicle development, such as those encoding Figla and Nobox, are more active in the infant ovary,whereas expression of genes vital for follicle development, such as Gdf9, Bmp4 and Bmp15, is upregulated in the adult. These data suggest a dynamic shift in gene expression during ovary development and it is apparent that these changes function to facilitate follicle maturation, when additional functional gene studies are considered. Furthermore, our investigation has also revealed several important functional pathways, such as apoptosis, MAPK and steroid biosynthesis, that appear to be much more active in the adult ovary compared to those of the infant. 8Optimal power allocation for secure directional modulation networks with a full-duplex UAV user显示文摘This paper makes an investigation of a secure unmanned aerial vehicle(UAV)-aided communication network based on directional modulation(DM). In this network, ground base station(GBS) acts as a control center to transmit confidential message and artificial noise(AN). The UAV user, moving along a linear flight trajectory, is intended to receive the useful information from GBS. At the same time, it also sends AN signals to further interference eavesdropper’s channel. Aiming at maximizing secrecy rate during the UAV flight process, a joint optimization problem is formulated with respect to power allocation(PA)factors, beamforming vector and AN projection matrices. For simplicity, maximum rate transmission, nullspace projection and the leakage-based method are applied to form the transmit beamforming vector, AN projection matrix at GBS, and AN projection vector at UAV user, respectively. Following this, the optimization problem reduces to a bivariate optimization programme with two PA factors. An alternating iterative algorithm(AIA) is proposed to optimize the two PA factors. 9Heavy mineral assemblage characteristics and the Cenozoic paleogeographic evolution in southwestern Qaidam Basin显示文摘Based on the analysis of heavy mineral assemblages in Cenozoic southwestern Qaidam Basin, we found that different areas have variable heavy mineral assemblage characteristics, which suggested that there were two source areas—the Altyn Mountains and the Qimen Tagh-East Kunlun Mountains. In Ganchaigou-Shizigou-Huatugou(Area A), which was mainly source from the Altyn Mountains, its heavy minerals were mainly composed of zircon, Ti-oxides, and wollastonite in the Paleoceneearly Eocene and mainly of unstable minerals, especially amphibole, in the middle Eocene-Oligene. Since the late OligoceneMiocene, the heavy minerals were still mainly unstable minerals, but the content of epidote increased and the content of amphibole decreased. In Qigequan-Hongliuquan(Area B), which was the mixed source from the Altyn Mountains and the Qimen Tagh-East Kunlun Mountains, its heavy minerals were mainly garnet, epidote, and amphibole. The source of LücaotanDongchaishan-Kunbei(Area C) was mainly from the Qimen Tagh-East Kunlun Mountains, heavy minerals in the sediments in Area C were mainly zircon and Ti-oxides in Paleogene and garnet, epidote, and amphibole in Neogene. In Yuejin-Youshashan(Area D), where the stable minerals and unstable minerals were present simultaneously, the heavy mineral assemblages was controlled by multi-direction source. The variation of heavy mineral assemblages in southwestern Qaidam Basin shows that Altyn Mountains was of low-lying topographic relief in Paleocene-early Eocene, and the rapid uplift of Altyn Mountains started from the middle Eocene. In Paleogene, the Altyn Tagh Fault had a slow strike-slip velocity, but the strike-slip velocity increased greatly since the late Oligocene, leading to a strike-slip displacement above 300 km since Neogene. 10Upregulation of annexin A5 affects the biological behaviors of lung squamous carcinoma cells in vitro显示文摘Annexin A5 is a Ca2?-dependent phospholipidbinding protein and protein kinase C inhibitory protein. It has a potential role in cellular signal transduction, inflammation, growth and differentiation. In this study, we evaluated the expression of this protein in lung tumor tissues and subsequently established a NCI-H520 cell line that stably expresses the wild-type ANXA5 gene to determine the effects of annexin A5 upregulation on the cell morphology, proliferation and metastasis potential in vitro.The effects of annexin A5 on NCI-H520 cells were tested by crystal violet staining, CCK-8 assay, scratch wound assay, and Transwell assay. The expressions of Akt,PCNA, vimentin, and E-cadherin were examined by Western blot assay. In this study, we demonstrated that annexin A5 is expressed at lower levels in tumor tissues compared with normal tissues. Additionally, the upregulation of this protein may inhibit the proliferation, migration, and invasion abilities of NCI-H520 cells in vitro. 13Application of Bis[2-(3,4-epoxycyclohexyl)ethyl]octamethyltetrasiloxane in the Preparation of a Photosensitive Resin for Stereolithography 3D Printing显示文摘Bis[2-(3,4-epoxycyclohexyl)ethyl]octamethyltetrasiloxane is also called diepoxycyclohexylethyl octamethyltetrasiloxane. In the present paper, diepoxycyclohexylethyl octamethyltetrasiloxane was synthesized, and the synthesized product was characterized by FTIR and 1 HMR. The synthesized product was compounded with some acrylates and an expoxide as well as photoinitiators to obtain a 3D printing stereolithography resin(3DSLR111). The properties of 3DSLR111 and its UV-cured samples were investigated by some instruments and equipments. The experimental results show that the critical exposure(Ec) of 3DSLR111 is 10.1 mJ/cm^2, its penetration depth(Dp) is 0.15 mm, and its viscosity at 30 ℃ is 319 mPa·s. Some samples were printed with 3DSLR111, and their linear shrinkage and warping factor were evaluated. 14Transcriptomic analysis reveals key regulators of mammogenesis and the pregnancy-lactation cycle显示文摘An organ unique to mammals,the mammary gland develops 90%of its mass after birth and experiences the pregnancy-lactation-involution cycle(PL cycle)during reproduction.To understand mammogenesis at the transcriptomic level and using a ribo-minus RNA-seq protocol,we acquired greater than 50 million reads each for the mouse mammary gland during pregnancy(day 12 of pregnancy),lactation(day 14 of lactation),and involution(day 7 of involution).The pregnancy-,lactation-and involution-related sequencing reads were assembled into 17344,10160,and 13739 protein-coding transcripts and1803,828,and 1288 non-coding RNAs(ncRNAs),respectively.Differentially expressed genes(DEGs)were defined in the three samples,which comprised 4843 DEGs(749 up-regulated and 4094 down-regulated)from pregnancy to lactation and4926 DEGs(4706 up-regulated and 220 down-regulated)from lactation to involution.Besides the obvious and substantive upand down-regulation of the DEGs,we observe that lysosomal enzymes were highly expressed and that their expression coincided with milk secretion.Further analysis of transcription factors such as Trps1,Gtf2i,Tcf7l2,Nupr1,Vdr,Rb1,and Aebp1,and ncRNAs such as mir-125b,Let7,mir-146a,and mir-15 has enabled us to identify key regulators in mammary gland development and the PL cycle.ZHOU YuanYuan GONG Wei XIAO JingFa WU JiaYan PAN LinLin LI XiaoNuan WANG XuMin WANG WeiWei HU SongNian YU Jun 2014Science China(Life Sciences)2014,57,3:4
15Macrobenthic assemblage characteristics under stressed waters and ecological health assessment using AMBI and M-AMBI:a case study at the Xin'an River Estuary,Yantai,China显示文摘To understand the ecological status and macrobenthic assemblages of the Xin'an River Estuary and its adjacent waters, a survey was conducted for environmental variables and macrobenthic assemblage structure in September 2012(Yantai, China). Several methods are adopted in the data analysis process: dominance index,diversity indices, cluster analysis, non-metric multi-dimentional scaling ordination, AMBI and M-AMBI. The dissolved inorganic nitrogen and soluble reactive phosphorus of six out of eight sampling stations were in a good condition with low concentration. The average value of DO((2.89±0.60) mg/L) and p H(4.28±0.43) indicated that the research area faced with the risk of ocean acidification and underlying hypoxia. A total of 62 species were identified, of which the dominant species group was polychaetes. The average abundance and biomass was577.50 ind./m2 and 6.01 g/m2, respectively. Compared with historical data, the macrobenthic assemblage structure at waters around the Xin'an River Estuary was in a relatively stable status from 2009 to 2012.Contaminant indicator species Capitella capitata appeared at Sta. Y1, indicating the animals here suffered from hypoxia and acidification. AMBI and M-AMBI results showed that most sampling stations were slightly disturbed,which were coincided with the abiotic measurement on evaluating the health conditions. 16Preparation of a Novel Cationic Photosensitive Resin(3D-SLR01) for Stereolithography 3D Printing and Determination of Its Some Properties显示文摘A novel cationic photosensitive resin(3 DSLR-01) for stereolithography 3 D printing was prepared with 3,4-epoxycyclohexylmethyl-3’,4’-epoxycyclohexane carboxylate(2021 P),1,4-cyclohexanedimethanol glycidyl ether(JX-026), diglycidyl 4,5-epoxycyclohexane-1,2-dicarboxylate(S-186), polycaprolactone polyol(Polyol-0305), novolac epoxy resin(F-51), bis(3-ethyl-3-oxetanylmethyl) ether(S-221) and a mixture of triarylsulfonium hexafluoroantimonate salts solution(UVI-6976). The properties of the photosensitive resin and its UV-cured films were investigated by some instruments and equipment.The experimental results show that the critical exposure(Ec) of the photosensitive resin is 16.3 mJ/cm^2, the penetration depth(Dp) is 0.14 mm, and the optical property of the photosensitive resin is excellent. 17Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. However, the role of the neuroinflammatory response in regulating secondary impairment in the visual cortex after ON injury remains unclear. The NOD-like receptor family pyrin domain containing 3(NLRP3) is an important regulator of neuroinflammation. In this study, we established a mouse model of unilateral ON crush(ONC) and showed that the expression of NLRP3 was significantly increased in the primary visual cortex(V1) as a response to ONC and that the NLRP3 inflammasome was activated in the contralateral V1 1 days–14 days after ONC. Ablation of the NLRP3 gene significantly decreased the trans-neuronal degeneration within 14 days. Visual electrophysiological function was improved in NLRP3-/- mice. 19Sensitivity of SARS-CoV-2 to different temperatures显示文摘This study was designed to investigate the sensitivity of SARS-CoV-2 to different temperatures,to provide basic data and a scientific basis for the control of COVID-19 epidemic.The virus was dispersed in 1 mL basal DMEM medium at a final concentration of 103.2 TCID 50/mL and then incubated at 4,22,30,35,37,38,39 and 40°C for up to 5 days.The infectivity of residual virus was titrated using the Vero E6 cell line.The results showed that the virus remained viable for 5 days at 4°C,and for 1 day only at 22 and 30°C.We found that the infectivity of the virus was completely lost after less than 12 hours at 37,38 and 39°C,while at 40°C,the inactivation time of the virus was rapidly reduced to 6 hours.We show that SARS-CoV-2 is sensitive to heat,is more stable at lower temperatures than higher temperature,remains viable for longer at lower temperatures,and loses viability rapidly at higher temperatures.Qi Lv Mingya Liu Feifei Qi Shuran Gong Shasha Zhou Shisheng Zhan Linlin Bao 2020Animal Models and Experimental Medicine2020,3,4:3
