2Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke.Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:8
3Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 5Prediction of radiosensitivity in primary central nervous system germ cell tumors using dynamic contrast-enhanced magnetic resonance imaging显示文摘Objective: To evaluate the feasibility of dynamic contrast-enhanced magnetic resonance imaging(DCEMRI) for predicting tumor response to radiotherapy in patients with suspected primary central nervous system(CNS) germ cell tumors(GCTs).Methods: DCE-MRI parameters of 35 patients with suspected primary CNS GCTs were obtained prior to diagnostic radiation, using the Tofts and Kermode model. Radiosensitivity was determined in tumors diagnosed 2 weeks after radiation by observing changes in tumor size and markers as a response to MRI. Taking radiosensitivity as the gold standard, the cut-off value of DCE-MRI parameters was measured by receiver operating characteristic(ROC) curve. Diagnostic accuracy of DCE-MRI parameters for predicting radiosensitivity was evaluated by ROC curve.Results: A significant elevation in transfer constant(Ktrans) and extravascular extracellular space(Ve)(P=0.000), as well as a significant reduction in rate constant(Kep)(P=0.000) was observed in tumors. Ktrans, relative Ktrans, and relative Kep of the responsive group were significantly higher than non-responsive groups. No significant difference was found in Kep, Ve, and relative Ve between the two groups. Relative Ktrans showed the best diagnostic value in predicting radiosensitivity with a sensitivity of 100%, specificity of 91.7%, positive predictive value(PPV) of 95.8%, and negative predictive value(NPV) of 100%.Conclusions: Relative Ktrans appeared promising in predicting tumor response to radiation therapy(RT). 6Understanding the Percolation Effect in Triboelectric Nanogenerator with Conductive Intermediate Layer显示文摘Introducing the conductive intermediate layer into a triboelectric nanogenerator(TENG)has been proved as an efficient way to enhance the surface charge density that is attributed to the enhancement of the dielectric permittivity.However,far too little attention has been paid to the companion percolation,another key element to affect the output.Here,the TENG with MXeneembedded polyvinylidene fluoride(PVDF)composite film is fabricated,and the dependence of the output capability on the MXene loading is investigated experimentally and theoretically.Specifically,the surface charge density mainly depends on the dielectric permittivity at lower MXene loadings,and in contrast,the percolation becomes the degrading factor with the further increase of the conductive loadings.At the balance between the dielectric and percolation properties,the surface charge density of the MXene-modified TENG obtained 350%enhancement compared to that with the pure PVDF.This work shed new light on understanding the dielectric and percolation effect in TENG,which renders a universal strategy for the high-performance triboelectronics.Binbin Zhang Guo Tian Da Xiong Tao Yang Fengjun Chun Shen Zhong Zhiming Lin Wen Li Weiqing Yang 2021Research2021,,1:2

8Revealing the linguistic and geographical disparities of public awareness to Covid-19 outbreak through social media显示文摘The Covid-19 has presented an unprecedented challenge to public health worldwide.However,residents in different countries showed diverse levels of Covid-19 awareness during the outbreak and suffered from uneven health impacts.This study analyzed the global Twitter data from January 1st to June 30th,2020,to answer two research questions.What are the linguistic and geographical disparities of public awareness in the Covid-19 outbreak period reflected on social media?Does significant association exist between the changing Covid-19 awareness and the pandemic outbreak?We established a Twitter data mining framework calculating the Ratio index to quantify and track awareness.The lag correlations between awareness and health impacts were examined at global and country levels.Results show that users presenting the highest Covid-19 awareness were mainly those tweeting in the official languages of India and Bangladesh.Asian countries showed more disparities in awareness than European countries,and awareness in Eastern Europe was higher than in central Europe.Finally,the Ratio index had high correlations with global mortality rate,global case fatality ratio,and country-level mortality rate,with 21-31,35-42,and 13–18 leading days,respectively.This study yields timely insights into social media use in understanding human behaviors for public health research.Binbin Lin Lei Zou Nick Duffield Ali Mostafavi Heng Cai Bing Zhou Jian Tao Mingzheng Yang Debayan Mandal Joynal Abedin 2022International Journal of Digital Earth2022,15,1:2
9Overexpression of Sirt6 ameliorates sleep deprivation induced-cognitive impairment by modulating glutamatergic neuron function显示文摘Sleep benefits the restoration of energy metabolism and thereby suppo rts neuronal plasticity and cognitive behaviors.Sirt6 is a NAD+-dependent protein deacetylase that has been recognized as an essential regulator of energy metabolism because it modulates various transcriptional regulators and metabolic enzymes.The aim of this study was to investigate the influence of Sirt6 on cerebral function after chronic sleep deprivation(CSD).We assigned C57BL/6J mice to control or two CSD groups and subjected them to AAV2/9-CMV-EGFP or AAV2/9-CMV-Sirt6-EGFP infection in the prelimbic cortex(PrL).We then assessed cerebral functional connectivity(FC) using resting-state functional MRI,neuron/astrocyte metabolism using a metabolic kinetics analysis;dendritic spine densities using sparse-labeling;and miniature excitato ry postsynaptic currents(mEPSCs) and action potential(AP) firing rates using whole-cell patchclamp recordings.In addition,we evaluated cognition via a comprehensive set of behavioral tests.Compared with controls,Sirt6 was significantly decreased(P<0.05) in the PrL after CSD,accompanied by cognitive deficits and decreased FC between the PrL and accumbens nucleus,piriform cortex,motor co rtex,somatosensory co rtex,olfactory tubercle,insular cortex,and cerebellum.Sirt6 ove rexpression reve rsed CSD-induced cognitive impairment and reduced FC.Our analysis of metabolic kinetics using [1-13C] glucose and [2-13C] acetate showed that CSD reduced neuronal Glu4and GABA2synthesis,which could be fully restored via forced Sirt6 expression.Furthermore,Sirt6 ove rexpression reversed CSD-induced decreases in AP firing rates as well as the frequency and amplitude of mEPSCs in PrL pyramidal neurons.These data indicate that Sirt6 can improve cognitive impairment after CSD by regulating the PrL-associated FC network,neuronal glucose metabolism,and glutamatergic neurotransmission.Thus,Sirt6 activation may have potential as a novel strategy for treating sleep disorder-related diseases.Jinpiao Zhu Chang Chen Zhen Li Xiaodong Liu Jingang He Ziyue Zhao Mengying He Binbin Nie Zili Liu Yingying Chen Kuanpin Su Xiang Li Juxiang Chen Hongbing Xiang Fuqiang Xu Kangguang Lin Zongze Zhang Jie Wang 2023Neural Regeneration Research2023,18,11:1

16Harmful algal bloom-forming dinofl agellate Prorocentrum donghaiense inhibits the growth and photosynthesis of seaweed Sargassum fusiformis embryos显示文摘Harmful algal bloom(HAB)is an ecological disaster to local mariculture.At present,its impact on macrophytes has not been well studied.In this study,we cultivated sexually propagated embryos of S argassum fusiformis-an edible seaweed-in Prorocentrum donghaiense suspensions at different cell densities(0,0.50×10^(5),0.75×10^(5),1.00×10^(5),and 1.50×10^(5) cells/mL)for 10 days,during which growth and photosynthetic activities of the embryos were determined,and a monocultivation was set up for comparison.Results show that the relative growth rate and photosynthetic activities of the embryos co-cultivated with P.donghaiense were inhibited mostly and significantly in the cell densities of 0.75×10^(5),1.00×10^(5),and 1.50×10^(5) cells/mL,and the inhibitory effects increased in overall with increased cell densities.The maximum relative electron transport rates(rETR max)and apparent photosynthetic efficiency(a)of co-cultivated embryos were all significantly lower than monocultivation ones on the 10 th day.Furthermore,the photosynthetic activity detected by chlorophyll-a fluorescence transient(i.e.,OJIP),the electron transport among electron transfer accepters of PSII(photosystem II)and that from PSII to PSI(photosystem I)was restricted,which is probably responsible for the decreases of rETR max andain the co-cultivated embryos.In addition,parts of the photosynthetic reaction centers of PSII in the co-cultivated embryos were inactivated.Therefore,P.donghaiense bloom could restrain the development and photosynthetic activities of S.fusiformis embryos,reduce the seedlings stock,and eventually hinder the development of S.fusiformis production industry.Caixia WANG Min WANG Binbin CHEN Wenli QIN Lidong LIN Chuanjun DAI Hengguo YU Renhui LI Min ZHAO Zengling MA 2021Journal of Oceanology and Limnology2021,39,6:1
17Myokine interleukin-15 expression profile is different in suckling and weaning piglets显示文摘Interleukin-15(IL-15) is a cytokine highly expressed in skeletal muscle. The objective of the present study was to investigate the development of muscle IL-15 expression in suckling piglets and in early weaning piglets(day 14) at each level, that is, mRNA, protein, and secretion. Eight litters(eight piglets per litter) of newborn healthy piglets(Large × White × Landrace) with a similar initial weight(1618.0 ± 140.1 g) were chosen and divided into two groups, Group one used suckling piglets that were killed, respectively, at days 1,7,14,21, and group two used early(day 14) weaning piglets that were killed respectively, at days 15, 17, 19, 21. In group one, IL-15 gene expression levels increased significantly(P < 0.05) along with increased body weight over time. IL-15 protein expression levels in piglets at day 21 of age were higher(P < 0.05) than those in piglets at other ages, and there was no difference(P > 0.05)among piglets at other ages. These findings indicated that increased IL-15 mRNA expression did not result in a corresponding increase of its protein expression. In group two, which used early weaning piglets from days 15-19, IL-15 mRNA and protein expression levels increased constantly(P < 0.05) and were higher(P < 0.05) than those in suckling piglets. Moreover, there was no gain of body weight(P > 0.05) compared with suckling piglets at day 14 of age. However, IL-15 protein expression levels in early weaning piglets at day 21 of age dropped significantly(P < 0.05) to the levels as suckling piglets at day 21 of age, while body weight increased(P < 0.05) markedly to the levels as suckling piglets at day 21 of age. In both groups, the serum IL-15 levels of piglets decreased significantly(P < 0.01) over time. Taken together, our results indicate that IL-15 expression differs in suckling piglets and in weaning piglets. 19Air Temperature Estimation with MODIS Data over the Northern Tibetan Plateau显示文摘Time series of MODIS land surface temperature(T_s) and normalized difference vegetation index(NDVI) products,combined with digital elevation model(DEM) and meteorological data from 2001 to 2012,were used to map the spatial distribution of monthly mean air temperature over the Northern Tibetan Plateau(NTP). A time series analysis and a regression analysis of monthly mean land surface temperature(T_s) and air temperature(T_a) were conducted using ordinary linear regression(OLR) and geographical weighted regression(GWR). The analyses showed that GWR,which considers MODIS T_s,NDVI and elevation as independent variables,yielded much better results [R_(Adj)~2> 0.79; root-mean-square error(RMSE) =0.51℃–1.12℃] associated with estimating T_a compared to those from OLR(R_(Adj)~2= 0.40-0.78; RMSE = 1.60℃–4.38℃).In addition,some characteristics of the spatial distribution of monthly T_a and the difference between the surface and air temperature(T_d) are as follows. According to the analysis of the 0℃ and 10℃ isothermals,T_a values over the NTP at elevations of 4000–5000 m were greater than 10℃ in the summer(from May to October),and T_a values at an elevation of3200 m dropped below 0℃ in the winter(from November to April). T_a exhibited an increasing trend from northwest to southeast.
