| 1 | APC and K-ras gene mutation in aberrant crypt foci of human colon显示文摘AIM To study the genetic alteration in ACF andto define the possibility that ACF may be a veryearly morphological lesion with molecularchanges, and to explore the relationshipbetween ACF and colorectal adenoma evencarcinoma.METHODS DNA from 35 CRC, 15 adenomas, 34ACF and 10 normal mucus was isolated by meansof microdissection. Direct gene sequencing of K-ras gene including codon 12, 13 and 61 as well asthe mutation cluster region (MCR) of APC genewas performed.RESULTS K-ras gene mutation frequency inACF, adenoma and carcinoma was 17.6% (6/34), 13.3% (2/ 15), and 14.3% (5/ 35)respectively, showing no difference ( P > 0.05)in K-fas gene mutation among three pathologicprocedures. The K-ras gene mutation inadenoma, carcinoma and 4 ACF restricted incodon 12 (GGT→GAT), but the other 2 mutationsfrom ACF located in codon 13 (GGC→GAC). K-res gene mutation was found more frequently inolder patients and patients with polypoidcancer. No mutation in codon 61 was found in thethree tissue types. Mutation rate of APO gene inadenoma and carcinoma was 22.9% (8/35) and26.7% (4/ 15), which was higher than ACF(2.9%) (P < 0.05). APC gene mutation incarcinoma was not correlated with age ofpatients, location, size and differentiation oftumor.CONCLUSION ACF might be a very earlymorphological lesion in the tumorogenesis ofcolorectal tumor. The morphological feature andgene mutation status was different in ACF andadenoma. ACF is possibly putative'microadenoma' that might be the precursor ofadenoma. | 2 | Effect of heparin on apoptosis in human nasopharyngeal carcinoma CNE2 cells显示文摘In order to study the mechanism of the effect of heparin on apoptosis in carcinoma cells, the nasopharyngeal carcinoma cell line CNE2 was used to identify the effect of heparin on apoptosis associated with the expression of c-myc, bax, bcl-2 proteins by use of Hoechst 33258 staining, terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL), agarose gel electrophoresis, and flow cytometry, as well as Western blot analysis. The results showed that heparin induced apoptosis of CNE2 cells including the morphologic changes such as reduction in the volume, and the nuclear chromatin condensation, as well as the “ladder pattern” revealed by agarose gel electrophoresis of DNA in a concentration-dependent manner.The number of TUNEL-positive cells was dramatically increased to 33.6±1.2% from 2.8±0.3% by treat ment with heparin in different concentrations (10~40 kU/L). The apoptotic index was increased to 32.5% from 3.5% by detecting SubG1 peaks on flow cytometry. | 4 | Exogenous hydrogen sulfide inhibits apoptosis by regulating endoplasmic reticulum stress-autophagy axis and improves myocardial reconstruction after acute myocardial infarction显示文摘During acute myocardial infarction,endoplasm ic reticulum(ER)stress-induced autophagy and apoptosis have been shown as im portant pathogeneses of myocardial reconstruction.Im portantly,hydrogen sulfide(H2S),as a third endogenous gas signaling molecule,exerts strong cytoprotective effect on anti-ER stress,autophagy regulation and antiapoptosis.Here,we showed that H2 S treatm ent inhibits apoptosis by regulating ER stress-autophagy axis and im proves myocardial reconstruction after acute myocardial infarction.We found that H2S intervention im proved left ventricle function,reduced glycogen deposition in myocardial tissue mesenchyme,and inhibited apoptosis.Moreover,the expressions of fibrosis indicators(Col3a1 and Col1a2),ER stress-related proteins(CHOP and BIP/ERP78),autophagy-related proteins(Beclin and ATG5),apoptosis protein(Bax),as well as fibrosis protein Col4a3bp were all decreased after treatm ent w ith H2 S.H2 S adm inistration also m aintained MMP/TIMP balance.Mechanistically,H2S activated the PI3K/AKT signaling pathway.In addition,H2S treatm ent also reduced the expressions of ER stress-related proteins,autophagy-related proteins,and apoptins in in wtro experiments.Interestingly,activation of ER stress-autophagy axis could reverse the inhibitory effect of H2S on myocardial apoptosis.Altogether,these results suggested that exogenous H2S suppresses myocardial apoptosis by blocking ER stress-autophagy axis,which in turn reverses cardiac rem odeling after myocardial infarction. | Yaling Li Maojun Liu Jiali Yi Xiong Song Xia Zheng Da Liu Sen Wang Chun Chu Jun Yang | 2020 | Acta Biochimica et Biophysica Sinica2020，52，12： | 5 |
