1Oral berberine improves brain dopa/dopamine levels to ameliorate Parkinson’s disease by regulating gut microbiota显示文摘The phenylalanine-tyrosine-dopa-dopamine pathway provides dopamine to the brain.Iin this process,tyrosine hydroxylase(TH)isthe rate-limiting enzyme that hydroxylates tyrosine and generates levodopa(L-dopa)with tetranydrobiopterin(BH_(4))as a coenzyme.Here,we show that oral berberine(BBR)might supply H^(·) through dihydroberberine(reduced BBR produced by bacterial nitroreductase)and promote the production of BHl from dihydrobiopterin;the increased BH,enhances TH activity,which accelerates the production of L-dopa by the gut bacteria.Oral BBR acts in a way similar to vitamins.The L-dopa produced by theintestinal bacteria enters the brain through the circulation and is transformed to dopamine.To verify the gut-brain dialog activatedby BBR's effect,Enterococcus foecalis or Enterococcus faecium was transplanted into Parkinson's disease(PD)mice.The bacteriasignificantly increased brain dopamine and ameliorated PD manifestation in mice;additionally,combination of BBR with bacteriashowed better therapeutic effect than that with bacteria alone.Moreover,2,4,6-trimethy-pyranylium tetrafluoroborate(TMP-TFB)-derivatized matrix-assisted laser desorption mass spectrometry(MALDI-MS)imaging of dopamine identihed elevated striataldopamine levels in mouse brains with oral Enterococcus,and BBR strengthened the imaging intensity of brain dopamine.Theseresults demonstrated that BBR was an agonist of TH in Enterococcus and could lead to the production of L-dopa in the gut.Furthermore,a study of 28 patients with hyperlipidemia conhrmed that oral BBR increased bloodfecal L-dopa by the intestinalbacteria.Hence,BBR might improve the brain function by upregulating the biosynthesis of-dopa in the gut microbiota through avitamin-like effect.Yan Wang Qian Tong Shu-Rong Ma Zhen-Xiong Zhao Li-Bin Pan Lin Cong Pei Han Ran Peng Hang Yu Yuan Lin Tian-Le Gao Jia-Wen Shou Xiao-Yang Li Xian-Feng Zhang Zheng-Wei Zhang Jie Fu Bao-Ying Wen Jin-Bo Yu Xuetao Cao Jian-Dong Jiang 2021Signal Transduction and Targeted Therapy2021,6,3:16
2Establishment of a novel corneal endothelial cell line from domestic rabbit,Oryctolagus curiculus显示文摘To develop a rabbit corneal endothelial (RCE) cell line, in vitro culture of RCE cells was initiated from Oryctolagus curiculus corneas and a novel RCE cell line was established in this study. To initiate the primary culture of RCE cells, corneas from rabbit eyes were sliced and attached into glutin-coated wells with endothelial cell surface down. After being cultured at a time-gradient interval from 48 to 6 h, the corneal slices were detached and reattached into new wells, respectively. Cells in the wells containing only a pure population of RCE cells were collected and cultured in 20% FBS-DMEM/F12 medium con- taining chondroitin sulfate, ocular extract, epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), carboxymethyl-chitosan, N-acetylglucosamine hydrochloride, glucosamine hydrochloride, culture medium of rabbit corneal stromal cells and oxidation-degradation products of chondroitin sul- fate at 37℃, 5% CO2. The cultured RCE cells, in quadrangle and polygonal shapes, proliferated to con- fluence 3 weeks later. During the subsequent subculture, the shape of RCE cells changed gradually from polygonal to more fibroblastic. A novel RCE cell line, growing at a steady rate, with a population doubling time of 53.8 h, has been established and subcultured to passage 67. Chromosome analysis showed that the RCE cells exhibited chromosomal aneuploidy with the modal chromosome number of 44. The results of immuno-cytochemical staining with neuron specific enolase (NSE) confirmed that the RCE cells were in neuroectodermal origin. Combined with the results of vascular endothelial growth factor (VEGF) treatment and endothelial cell morphology recovery, it can be concluded that the cell line established here is an RCE cell line. 4Strategies to minimize hypertrophy in cartilage engineering and regeneration显示文摘Due to a blood supply shortage,articular cartilage has a limited capacity for selfhealing once damaged.Articular chondrocytes,cartilage progenitor cells,embryonic stem cells,and mesenchymal stem cells are candidate cells for cartilage regeneration.Significant current attention is paid to improving chondrogenic differentiation capacity;unfortunately,the potential chondrogenic hypertrophy of differentiated cells is largely overlooked.Consequently,the engineered tissue is actually a transient cartilage rather than a permanent one.The development of hypertrophic cartilage ends with the onset of endochondral bone formation which has inferior mechanical properties.In this review,current strategies for inhibition of chondrogenic hypertrophy are comprehensively summarized;the impact of cell source options is discussed;and potential mechanisms underlying these strategies are also categorized.This paper aims to provide guidelines for the prevention of hypertrophy in the regeneration of cartilage tissue.This knowledge may also facilitate the retardation of osteophytes in the treatment of osteoarthritis.Song Chen Peiliang Fu Ruijun Cong HaiShan Wu Ming Pei 2015Genes & Diseases2015,2,1:11
5Endothelial CDS2 deficiency causes VEGFA-mediated vascular regression and tumor inhibition显示文摘The response of endothelial cells to signaling stimulation is critical for vascular morphogenesis,homeostasis and function.Vascular endothelial growth factor-a(VEGFA)has been commonly recognized as a pro-angiogenic factor in vertebrate developmental,physiological and pathological conditions for decades.Here we report a novel finding that genetic ablation of CDP-diacylglycerol synthetase-2(CDS2),a metabolic enzyme that controls phosphoinositide recycling,switches the output of VEGFA signaling from promoting angiogenesis to unexpectedly inducing vessel regression.Live imaging analysis uncovered the presence of reverse migration of the angiogenic endothelium in cds2 mutant zebrafish upon VEGFA stimulation,and endothelium regression also occurred in postnatal retina and implanted tumor models in mice.In tumor models,CDS2 deficiency enhanced the level of tumorsecreted VEGFA,which in-turn trapped tumors into a VEGFA-induced vessel regression situation,leading to suppression of tumor growth.Mechanistically,VEGFA stimulation reduced phosphatidylinositol(4,5)-bisphosphate(PIP2)availability in the absence of CDS2-controlled-phosphoinositide metabolism,subsequently causing phosphatidylinositol(3,4,5)-triphosphate(PIP3)deficiency and F0X01 activation to trigger regression of CDS2-null endothelium.Thus,our data indicate that the effect of VEGFA on vasculature is context-dependent and can be converted from angiogenesis to vascular regression.Wencao Zhao Le Cao Hanru Ying Wenjuan Zhang Dantong Li Xiaolong Ziiu Wenzhi Xue Shuang Wu Mengye Cao Cong Fu Haonan Qi Yimei Hao Yun-Chi Tang Jun Qin Tao PZhong Xiaoxi Lin Luyang Yu Xuri Li Lin Li Dianqing Wu Weijun Pan 2019Cell Research2019,29,11:9
6Spatial Properties of Mismatch Negativity in Patients with Disorders of Consciousness显示文摘In recent decades, event-related potentials have been used for the clinical electrophysiological assessment of patients with disorders of consciousness(DOCs). In this paper, an oddball paradigm with two types of frequencydeviant stimulus(standard stimuli were pure tones of 1000 Hz; small deviant stimuli were pure tones of 1050 Hz; large deviant stimuli were pure tones of 1200 Hz) was applied to elicit mismatch negativity(MMN) in 30 patients with DOCs diagnosed using the JFK Coma Recovery ScaleRevised(CRS-R). The results showed that the peak amplitudes of MMN elicited by both large and small deviant stimuli were significantly different from baseline.In terms of the spatial properties of MMN, a significant interaction effect between conditions(small and large deviant stimuli) and electrode nodes was centered at the frontocentral area. Furthermore, correlation coefficients were calculated between MMN amplitudes and CRS-R scores for each electrode among all participants to generate topographic maps. Meanwhile, a significant negative correlation between the MMN amplitudes elicited by large deviant stimuli and the CRS-R scores was also found at the frontocentral area. 13Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 14Two case reports and literature review for hepatic epithelioid angiomyolipoma: Pitfall of misdiagnosis显示文摘BACKGROUND Hepatic epithelioid angiomyolipoma(HEAML) is a rare liver disease and is easily misdiagnosed. Enhanced recognition of HEAML is beneficial to the differential diagnosis of rare liver diseases.CASE SUMMARY We presented two cases of HEAML in Changzheng Hospital, Naval Medical University, and then collected and analyzed all reports about HEAML recorded in PubMed, MEDLINE, China Science Periodical Database, and VIP database from January 2000 to March 2018. A total of 409 cases of HEAML in 97 reports were collected, with a ratio of men to women of 1:4.84 and an age range from 12 years to 80 years(median 44 years). Among the patients with clinical symptoms mentioned, 61.93%(205/331) were asymptomatic, 34.74%(115/331) showed upper or right upper quadrant abdomen discomfort, while a few of them showed abdominal mass, gastrointestinal symptoms, low fever, or weight loss. The misdiagnosis rate of HEAML was as high as 40.34%(165/409) due to its nonspecific imaging findings. Most of the tumors were solitary and round in morphology, with clear boundaries. Ultrasound scan indicated low echo with internal nonuniformity and rich blood supply in most cases. Computer tomography/magnetic resonance imaging enhanced scan showed varied characteristics. The ratio of fast wash-in and fast wash-out, fast wash-in and slow wash-out, and delayed enhancement was roughly 4:5:1. A definite diagnosis of HEAML depended on the pathological findings of the epithelioid cells in lesions and the expression of human melanoma black 45, smooth muscle actin,melanoma antigen, and actin by immunohistochemical staining. HEAML had a relatively low malignant rate of 3.91%. 15Berberine treats atherosclerosis via a vitamine-like effect downregulating Choline-TMA-TMAO production pathway in gut microbiota显示文摘Trimethylamine-N-oxide(TMAO)derived from the gut microbiota is an atherogenic metabolite.This study investigates whether or not berberine(BBR)could reduce TMAO production in the gut microbiota and treat atherosclerosis.Effects of BBR on TMAO production in the gut microbiota,as well as on plaque development in atherosclerosis were investigated in the culture of animal intestinal bacterial,HFD-fed animals and atherosclerotic patients,respectively.We found that oral BBR in animals lowers TMAO biosynthesis in intestine through interacting with the enzyme/co-enzyme of choline-trimethylamine lyase(CutC)and flavincontaining monooxygenase(FMO)in the gut microbiota.This action was performed by BBR’s metabolite dihydroberberine(a reductive BBR by nitroreductase in the gut microbiota),via a vitamine-like effect down-regulating Choline-TMA-TMAO production pathway.Oral BBR decreased TMAO production in animal intestine,lowered blood TMAO and interrupted plaque formation in blood vessels in the HFD-fed hamsters.Moreover,21 patients with atherosclerosis exhibited the average decrease of plaque score by 3.2%after oral BBR(0.5 g,bid)for 4 months(^(*)P<0.05,n=21);whereas the plaque score in patients treated with rosuvastatin plus aspirin,or clopidogrel sulfate or ticagrelor(4 months,n=12)increased by 1.9%.TMA and TMAO in patients decreased by 38 and 29%in faeces(^(*)P<0.05;^(*)P<0.05),and 37 and 35%in plasma(^(***)P<0.001;^(*)P<0.05),after 4 months on BBR.BBR might treat atherosclerotic plaque at least partially through decreasing TMAO in a mode of action similar to that of vitamins.Shu-Rong Ma Qian Tong Yuan Lin Li-Bin Pan Jie Fu Ran Peng Xian-Feng Zhang Zhen-Xiong Zhao Yang Li Jin-Bo Yu Lin Cong Pei Han Zheng-Wei Zhang Hang Yu Yan Wang Jian-Dong Jiang 2022Signal Transduction and Targeted Therapy2022,7,8:6

19Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 20Response of Soil Moisture to Rainfall Event in Black Locust Plantations at Different Stages of Restoration in Hilly-gully Area of the Loess Plateau, China显示文摘Precipitation plays an important role in the water supplies that support ecological restoration by sustaining large-scale artificial plantations in arid and semiarid regions, especially black locust(Robinia pseudoacacia) plantations(RP plantations), which are widely planted due to R. pseudoacacia being an excellent pioneer species. Characterizing the response of soil moisture to rainfall events at different stages of restoration is important for assessing the sustainability of restoration in RP plantations. In this study, we quantified the response of soil moisture to rainfall events at different years of restoration(15, 20 and 30 yr) representing different restoration stages in RP plantations in a typical hilly-gully area, i.e., the Yangjuangou Catchment, of the Loess Plateau, China. Over the growing season(June to September) of 2017, smart probes were placed at nine depths(10, 20, 40, 60, 80, 100, 120, 150, and 180 cm below the soil surface) to obtain volumetric soil water information at 30-min intervals in the three RP plantations. The advance of the wetting front was depicted, and the total cumulative water infiltration was measured. Soil moisture was mainly replenished by eight heavy rainfall events(mean rainfall amount = 46.3 mm), accounting for 88.7% of the rainfall during the growing season. The mean soil moisture content profiles of RP plantations at the three restoration stages were ordered as 30-yr(14.07%) > 20-yr(10.12%) > 15-yr(8.03%), and this relationship displayed temporal stability. Soil moisture was primarily replenished by rainfall at the 0-60 cm soil depth, and soil moisture remained stable below the 100-cm soil depth. The rainfall regime influenced the advancement of the wetting front. Here, a single rainfall event of 30 mm was the rainfall threshold for infiltration into the 60-cm soil layer. The total infiltration time ranged from 310.5-322.0 h, but no significant differences were found among RP plantations at different restoration stages. Young and old RP plantations had more total infiltration(more than 228.2 mm) and deeper infiltration depths(80-100 cm) than middle-aged plantations. The RP plantation at the intermediate restoration stage exhibited minimal total infiltration(174.2 mm) and a shallow infiltration depth(60 cm) due to the soil physical structure of the plot, which may have limited rain infiltration.
