1Aquaporin 5 is degraded by autophagy in diabetic submandibular gland显示文摘Autophagy is a catabolic process which is involved in the development of many diseases including diabetes mellitus and its complications. Hyposalivation is a common complication of diabetes mellitus, whereas its mechanism remains unclear. Here, we observed that the stimulated salivary flow rate of SMG was significantly decreased in db/db mice, a diabetic mice model. The expressions of aquaporin 5(AQP5), a water channel protein, were decreased, whereas the m RNA level of AQP5 was increased in SMGs of both diabetic patients and mice. Under transmission electron microcope, more autophagosomes were detected in diabetic SMGs. Expressions of autophagy related proteins LC3 II, Beclin-1 and ATG5 were increased, meanwhile autophagy substrate p62 was decreased in SMGs of diabetic patients and mice, indicating that autophagy was activated in diabetic SMG.Double immunofluorescence staining showed that the colocalization of AQP5 and LC3 was increased in SMGs of diabetic mice.In cultured SMG-C6 cells, high glucose(HG), but not high osmotic pressure, reduced AQP5 protein expression and induced autophagy. Moreover, inhibition of autophagy by 3-methyladenin, an autophagy inhibitor, or by autophagy-related gene 5 siRNA, decreased HG-induced AQP5 reduction in SMG-C6 cells. Additionally, the expression of p-p85, p-Akt and p-mTOR were decreased in HG-treated SMG-C6 cells. Pretreatment with 740 Y-P, a PI3 K agonist, significantly suppressed HG-induced autophagy and AQP5 degradation. Taken together, these results indicate that autophagy plays a crucial role in AQP5 degradation in diabetic SMG via PI3 K/Akt/mTOR signaling pathway, which contributes to the dysfunction of diabetic SMG. 4Observation of a bi-critical point between antiferromagnetic and superconducting phases in pressurized single crystal Ca0.73La0.27FeAs2显示文摘One of the most strikingly universal features of the high-temperature superconductors is that the superconducting phase emerges in the close proximity of the antiferromagnetic phase, and the interplay between these two phases poses a long-standing challenge. It is commonly believed that, as the antiferromagnetic transition temperature is continuously suppressed to zero, there appears a quantum critical point, around which the existence of antiferromagnetic fluctuation is responsible for the development of the superconductivity. In contrast to this scenario, we report the observation of a bi-critical point identified at 2.88 GPa and 26.02 K in the pressurized high-quality single crystal Ca_(0.73)La_(0.27)FeAs_2 by complementary in-situ high pressure measurements. 9Interleukin-13 promotes cellular senescence through inducing mitochondrial dysfunction in IgG4-related sialadenitis显示文摘Immunoglobulin G4-related sialadenitis(IgG4-RS)is an immune-mediated fibro-inflammatory disease and the pathogenesis is still not fully understood.The aim of this study was to explore the role and mechanism of interleukin-13(IL-13)in the cellular senescence during the progress of IgG4-RS.We found that the expression of IL-13 and IL-13 receptorα1(IL-13Rα1)as well as the number of senescent cells were significantly higher in the submandibular glands(SMGs)of IgG4-RS patients.IL-13 directly induced senescence as shown by the elevated activity of senescence-associatedβ-galactosidase(SA-β-gal),the decreased cell proliferation,and the upregulation of senescence markers(p53 and p16)and senescence-associated secretory phenotype(SASP)factors(IL-1βand IL-6)in SMG-C6 cells.Mechanistically,IL-13 increased the level of phosphorylated signal transducer and activator of transcription 6(p-STAT6)and mitochondrial-reactive oxygen species(mt ROS),while decreased the mitochondrial membrane potential,ATP level,and the expression and activity of superoxide dismutase 2(SOD2).Notably,the IL-13-induced cellular senescence and mitochondrial dysfunction could be inhibited by pretreatment with either STAT6 inhibitor AS1517499 or mitochondria-targeted ROS scavenger Mito TEMPO.Moreover,IL-13 increased the interaction between p-STAT6 and c AMP-response element binding protein(CREB)-binding protein(CBP)and decreased the transcriptional activity of CREB on SOD2.Taken together,our findings revealed a critical role of IL-13 in the induction of salivary gland epithelial cell senescence through the elevated mitochondrial oxidative stress in a STAT6–CREB–SOD2-dependent pathway in IgG4-RS.Mengqi Zhu Sainan Min Xiangdi Mao Yuan Zhou Yan Zhang Wei Li Li Li Liling Wu Xin Cong Guangyan Yu 2022International Journal of Oral Science2022,14,3:1

11Autonomic reinnervation and functional regeneration in autologous transplanted submandibular glands in patients with severe keratoconjunctivitis sicca显示文摘Autologous submandibular gland(SMG) transplantation has been proved to ameliorate the discomforts in patients with severe keratoconjunctivitis sicca. The transplanted glands underwent a hypofunctional period and then restored secretion spontaneously.This study aims to investigate whether autonomic nerves reinnervate the grafts and contribute to the functional recovery, and further determine the origin of these nerves. Parts of the transplanted SMGs were collected from the epiphora patients, and a rabbit SMG transplantation model was established to fulfill the serial observation on the transplanted glands with time. The results showed that autonomic nerves distributed in the transplanted SMGs and parasympathetic ganglionic cells were observed in the stroma of the glands. Low-dense and unevenly distributed cholinergic axons, severe acinar atrophy and fibrosis were visible in the patients' glands 4–6 months post-transplantation, whereas the cholinergic axon density and acinar area were increased with time. The acinar area or the secretory flow rate of the transplanted glands was statistically correlated with the cholinergic axon density in the rabbit model, respectively. Meanwhile, large cholinergic nerve trunks were found to locate in the temporal fascia lower to the gland, and sympathetic plexus concomitant with the arteries was observed both in the adjacent fascia and in the stroma of the glands. In summary, the transplanted SMGs are reinnervated by autonomic nerves and the cholinergic nerves play a role in the morphological and functional restoration of the glands. 12Surface coatings of two-dimensional metal-organic framework nanosheets enable stable zinc anodes显示文摘Aqueous zinc-ion batteries (ZIBs) have been considered as safe and scalable energy storage solutions,but the dendrite and corrosion issues of Zn anodes have hindered their further application.Herein,we demonstrate that two-dimensional metalorganic framework (MOF) nanosheets can act as protective coatings to prevent dendrite formation and hydrogen evolution of Zn anodes.The morphology of MOFs was tuned from octahedral nanoparticles (UiO-67-3D) to nanosheets (UiO-67-2D),leading to significantly enhanced protective performance.UiO-67-2D nanosheets-coated Zn anodes displayed smaller polarization,longer cycling lifetime and lower H_(2) evolution than those of UiO-67-3D nanoparticles in symmetrical cells,which has been attributed to the higher concentration of surface Zr-OH/H_(2)O to induce uniform Zn deposition and one-dimensional (1D) channels perpendicular to the Zn surface to regulate Zn^(2+) diffusion.The assembled UiO-67-2D@Zn||Mn_(2)O_(3)/C full cell shows a high capacity of240 m Ah g^(-1)at 1 A g^(-1) and excellent cycling stability.Liling Lei Feifan Chen Yulun Wu Jian Shen Xue-Jun Wu Shishan Wu Shuai Yuan 2022Science China Chemistry2022,65,11:0
13Mycorrhizal Networks Interacting with Litter Improves Nutrients and Growth for One Plant through the Vary of N/P Ratio under Karst Soil显示文摘Arbuscular mycorrhizae(AM)fungi affect nutrient uptake for host plants,while it is unclear how AM fungi interacting with soil litter affect plant growth and nutrient utilization through mycorrhizal networks in karst soil of deficient nutrients beyond the rhizosphere.An experiment was conducted in a microcosm composed of a planting compartment for Cinnamomum camphora seedlings with or without Glomus mosseae fungus(M+vs.M−)and an adjacent litter compartment containing or not containing additional litter material of Arthraxon hispidus(L+vs.L−),where the compartments are connected either by nylon mesh of 20μm or 0.45μm which either allow available mycorrhizal networks within the litter compartment or prevent mycelium entering into the litter compartment(N+vs.N−).Plant biomass and nutrients were measured.The results showed that the addition of litter changed the symbiotic process in mycorrhizal colonization,spore,and hyphal density,which when in association with the host plant then affected the biomass,and accumulations of N(nitrogen)and P(phosphorus)in the individual plant as well as root,stem,and leaf respectively.AM fungi increased N and P accumulations and N/P ratio in individual plants and plant tissues.A decrease of the N/P ratio of the individual plant was observed when AM fungus interacted significantly with litter through mycorrhizal networks in the litter compartment.The results indicate that the C.camphora seedlings benefited from litter in nutrient utilization of N and P through the vary of N/P ratio when accessing mycorrhizal networks.These findings suggest that mycorrhizal networks interacting with litter improve growth and nutrients of N and P for plants through the vary of N/P ratio in order to alleviate nutrient limitation under karst soil.Liling Kang Yuejun He Lipeng Zang Jianpeng Si Ying Yang Kaiping Shen Tingting Xia Qiyu Tan Bangli Wu Yun Guo Wei Wang Qin Liang 2021Phyton-International Journal of Experimental Botany2021,90,3:0
14Thermosensitive injectable hydrogel loaded with hypoxia-induced exosomes maintains chondrocyte phenotype through NDRG3-mediated hypoxic response显示文摘Current clinical treatments cannot effectively delay the progression of osteoarthritis(OA).Consequently,joint replacement surgery is required for late-stage OA when patients cannot tolerate pain and joint dysfunction.Therefore,the prevention of OA progression in the early and middle stages is an urgent clinical problem.In a previous study,we demonstrated that NDRG3-mediated hypoxic response might be closely related to the development and progression of OA.In this study,an injectable thermosensitive hydrogel was established by cross-linking Pluronic F-127 and hyaluronic acid(HA)for the sustained release of hypoxia-induced exosomes(HExos)derived from adipose-derived mesenchymal stem cells.We demonstrated that for OA at the early and middle stages,the HExos-loaded HP hydrogel could maintain the chondrocyte phenotype by enhancing chondrocyte autophagy,reducing chondrocyte apoptosis,and promoting chondrocyte activity and proliferation through the NDRG3-mediated hypoxic response.This novel composite hydrogel,which could activate the NDRG3-mediated hypoxic response,may provide new ideas and a theoretical basis for the treatment of early-and mid-stage OA.Xiongbo Song Liling Gu Qiming Yang Jiarui Wu Junrong Chen Xiaobin Tian Li Sun Long Chen 2023Chinese Chemical Letters2023,34,8:0

16Pyk2 suppresses contextual fear memory in an autophosphorylation-independent manner显示文摘Clustered protocadherins(Pcdhs)are a large family of cadherin-like cell adhesion proteins that are central for neurite selfavoidance and neuronal connectivity in the brain.Their downstream nonreceptor tyrosine kinase Pyk2(proline-rich tyrosine kinase 2,also known as Ptk2b,Cakb,Raftk,Fak2,and Cadtk)is predominantly expressed in the hippocampus.We constructed Pyk2-null mouse lines and found that these mutant mice showed enhancement in contextual fear memory,without significant change in auditory-cued and spatial-referenced learning and memory.In addition,by preparing Y402F mutant mice,we observed that Pyk2 suppressed contextual fear memory in an autophosphorylation-independent manner.Moreover,using high-throughput RNA sequencing,we found that immediate early genes,such as Npas4,cFos,Zif268/Egr1,Arc,and Nr4a1,were enhanced in Pyk2-null mice.We further showed that Pyk2 disruption affected pyramidal neuronal complexity and spine dynamics.Thus,we demonstrated that Pyk2 is a novel fear memory suppressor molecule and Pyk2-null mice provide a model for understanding fearrelated disorders.These findings have interesting implications regarding dysregulation of the Pcdh–Pyk2 axis in neuropsychiatric disorders.Jin Zheng Lun Suo Yuxiao Zhou Liling Jia Jingwei Li Yanping Kuang Donghong Cui Xuehong Zhang Qiang Wu 2021Journal of Molecular Cell Biology2021,13,11:0
17No evidence of superconductivity in a compressed sample prepared from lutetium foil and H_(2)/N_(2) gas mixture显示文摘A material described as lutetium–hydrogen–nitrogen(Lu-H-N in short)was recently claimed to have“near-ambient superconductivity”[Dasenbrock-Gammon et al.,Nature 615,244–250(2023)].If this result could be reproduced by other teams,it would be a major scientific breakthrough.Here,we report our results of transport and structure measurements on a material prepared using the same method as reported by Dasenbrock-Gammon et al.Our x-ray diffraction measurements indicate that the obtained sample contains three substances:the facecentered-cubic(FCC)-1 phase(Fm-3m)with lattice parameter a=5.03Å,the FCC-2 phase(Fm-3m)with a lattice parameter a=4.755Å,and Lu metal.The two FCC phases are identical to the those reported in the so-called near-ambient superconductor.However,we find from our resistance measurements in the temperature range from 300 K down to 4 K and the pressure range 0.9–3.4 GPa and our magnetic susceptibility measurements in the pressure range 0.8–3.3 GPa and the temperature range down to 100 K that the samples show no evidence of superconductivity.We also use a laser heating technique to heat a sample to 1800 XC and find no superconductivity in the produced dark blue material below 6.5 GPa.In addition,both samples remain dark blue in color in the pressure range investigated.Shu Cai Jing Guo Haiyun Shu Liuxiang Yang Pengyu Wang Yazhou Zhou Jinyu Zhao Jinyu Han Qi Wu Wenge Yang Tao Xiang Ho-kwang Mao Liling Sun 2023Matter and Radiation at Extremes2023,8,4:0

20Quantum phase transition and destruction of Kondo effect in pressurized SmB6显示文摘SmB_6 has been a well-known Kondo insulator for decades, but recently attracts extensive new attention as a candidate topological system. Studying SmB_6 under pressure provides an opportunity to acquire the much-needed understanding about the effect of electron correlations on both the metallic surface state and bulk insulating state. Here we do so by studying the evolution of two transport gaps(low temperature gap E_l and high temperature gap E_h) associated with the Kondo effect by measuring the electrical resistivity under high pressure and low temperature(0.3 K) conditions. We associate the gaps with the bulk Kondo hybridization, and from their evolution with pressure we demonstrate an insulator-tometal transition at ~4 GPa. At the transition pressure, a large change in the Hall number and a divergence tendency of the electron-electron scattering coefficient provide evidence for a destruction of the Kondo entanglement in the ground state.
