1TRIM35 mediates protection against influenza infection by activating TRAF3 and degrading viral PB2显示文摘Tripartite motif(TRIM)family proteins are important effectors of innate immunity against viral infections.Here we identified TRIM35 as a regulator of TRAF3 activation.Deficiency in or inhibition of TRIM35 suppressed the production of type I interferon(IFN)in response to viral infection.777m35-deficient mice were more susceptible to influenza A virus(IAV)infection than were wild-type mice.TRIM35 promoted the RIG-Imediated signaling by catalyzing Lys63-linked polyubiquitination of TRAF3 and the subsequent formation of a signaling complex with VISA and TBK1.IAV PB2 polymerase countered the innate antiviral immune response by impeding the Lys63-linked polyubiquitination and activation of TRAF3.TRIM35 mediated Lys48-linked polyubiquitination and proteasomal degradation of IAV PB2,thereby antagonizing its suppression of TRAF3 activation.Our in vitro and in vivo findings thus reveal novel roles of TRIM35,through catalyzing Lys63-or Lys48-linked polyubiquitination,in RIG-I antiviral immunity and mechanism of defense against IAV infection.Nan Sun Li Jiang Miaomiao Ye Yihan Wang Guangwen Wang Xiaopeng Wan Yuhui Zhao Xia Wen Libin Liang Shujie Ma Liling Liu Zhigao Bu Hualan Chen Chengjun Li 2020Protein & Cell2020,11,12:4
2Effects of gravity on the microstructure of Zr_(41)Ti_(14)-Cu_(12.5)Ni_(10)Be_(22.5) bulk glass forming alloy显示文摘The ZrTiCuNiBe alloy is melt and solidified by Bridgman unidirectional solidification on two gravity field orientations (the gravity field orientation is parallel and opposite to solidification direction). Effects of gravity on morphology and microstructure are investigated by scanning electron microscope (SEM) and X-ray diffraction (XRD). 3Observation of a bi-critical point between antiferromagnetic and superconducting phases in pressurized single crystal Ca0.73La0.27FeAs2显示文摘One of the most strikingly universal features of the high-temperature superconductors is that the superconducting phase emerges in the close proximity of the antiferromagnetic phase, and the interplay between these two phases poses a long-standing challenge. It is commonly believed that, as the antiferromagnetic transition temperature is continuously suppressed to zero, there appears a quantum critical point, around which the existence of antiferromagnetic fluctuation is responsible for the development of the superconductivity. In contrast to this scenario, we report the observation of a bi-critical point identified at 2.88 GPa and 26.02 K in the pressurized high-quality single crystal Ca_(0.73)La_(0.27)FeAs_2 by complementary in-situ high pressure measurements. 7Compression behavior of Zr_(41)Ti_(14)Cu_(12.5)Ni_(10)Be_(22.5) bulk metallic glass up to 24 GPa显示文摘The compression of a Zr41Ti14Cu12.5Ni10Be22.5 bulk metallic glass (BMG) is investigated at room temperature up to 24 GPa using in-situ high pressure energy dispersive X-ray diffraction with a synchrotron radiation source. The pressure-induced structural relaxation is exhibited. 8Containerless solidification of Zr_(41)Ti_(14)Cu_(12.5)Ni_(10)Be_(22.5) glass-forming alloy in drop tube显示文摘Droplets of Zr41Ti14Cu12.5Ni10Be22.5 glass forming alloys with different sizes are solidified in a drop tube containerless processing. Glass transition behavior, crystallization kinetics, and the phase evolution during annealing of the Zr41Ti14Cu12.5Ni10Be22.5 glassy spheres are investigated. The experimental results indicate that the apparent activation energy of the glass transition (Eg=435.5 kJ/mol), and the activation energy of the main crystallization reaction (Ep1 = 249.6 kJ/mol) are obviously different from those of bulk glass samples prepared by water quenched (Eg=559.1 kJ/mol and Ep1=192.5 kJ/mol). 10Preparation and characterization of methacrylated gelatin/bacterial cellulose composite hydrogels for cartilage tissue engineering显示文摘Methacrylated gelatin(GelMA)/bacterial cellulose(BC)composite hydrogels have been successfully prepared by immersing BC particles in GelMA solution followed by photo-crosslinking.The morphology of GelMA/BC hydrogel was examined by scanning electron microscopy and compared with pure GelMA.The hydrogels had very well interconnected porous network structure,and the pore size decreased from 200 to 10 mm with the increase of BC content.The composite hydrogels were also characterized by swelling experiment,X-ray diffraction,thermogravimetric analysis,rheology experiment and compressive test.The composite hydrogels showed significantly improved mechanical properties compared with pure GelMA.In addition,the biocompatility of composite hydrogels were preliminarily evaluated using human articular chondrocytes.The cells encapsulated within the composite hydrogels for 7 days proliferated and maintained the chondrocytic phenotype.Thus,the GelMA/BC composite hydrogels might be useful for cartilage tissue engineering.Liling Gu Tao Li Xiongbo Song Xianteng Yang Senlei Li Long Chen Pingju Liu Xiaoyuan Gong Cheng Chen Li Sun 2020Regenerative Biomaterials2020,7,2:0
11Conformation of Organic Chain in Phase Transition of Hybrid(C1_2H_(25)NH_3)_2MnCl_4显示文摘The structural change in phase transition of hybrid(C 12 H 25 NH 3 ) 2 MnCl 4 was investigated.The temperature and the structures of the phase transition is investigated by thermal gravimetry(TG)and differential scanning calorimetry(DSC),infrared spectrum(IR)and X-ray diffraction(XRD).The results suggest that the phase transition is reversible and the structural change arises from the conformation change of the organic chain. 14Thermosensitive injectable hydrogel loaded with hypoxia-induced exosomes maintains chondrocyte phenotype through NDRG3-mediated hypoxic response显示文摘Current clinical treatments cannot effectively delay the progression of osteoarthritis(OA).Consequently,joint replacement surgery is required for late-stage OA when patients cannot tolerate pain and joint dysfunction.Therefore,the prevention of OA progression in the early and middle stages is an urgent clinical problem.In a previous study,we demonstrated that NDRG3-mediated hypoxic response might be closely related to the development and progression of OA.In this study,an injectable thermosensitive hydrogel was established by cross-linking Pluronic F-127 and hyaluronic acid(HA)for the sustained release of hypoxia-induced exosomes(HExos)derived from adipose-derived mesenchymal stem cells.We demonstrated that for OA at the early and middle stages,the HExos-loaded HP hydrogel could maintain the chondrocyte phenotype by enhancing chondrocyte autophagy,reducing chondrocyte apoptosis,and promoting chondrocyte activity and proliferation through the NDRG3-mediated hypoxic response.This novel composite hydrogel,which could activate the NDRG3-mediated hypoxic response,may provide new ideas and a theoretical basis for the treatment of early-and mid-stage OA.Xiongbo Song Liling Gu Qiming Yang Jiarui Wu Junrong Chen Xiaobin Tian Li Sun Long Chen 2023Chinese Chemical Letters2023,34,8:0
15Transitions of amorphous-crystalline-amorphous in bulk metallic glass under HP and HT显示文摘In-situ SR-XRD measurements revealed that the crystallization process in Zr41.2Ti13.8Cu12.5Ni10Be22.5 bulk metallic glass is significantly different from that in tradi-tional glasses. 16Monolithic integration of nanorod arrays on microfluidic chips for fast and sensitive one-step immunoassays显示文摘Here,we present integrated nanorod arrays on microfluidic chips for fast and sensitive flow-through immunoassays of physiologically relevant macromolecules.Dense arrays of Au nanorods are easily fabricated through one-step oblique angle deposition,which eliminates the requirement of advanced lithography methods.We report the utility of this plasmonic structure to improve the detection limit of the cardiac troponin I(cTnI)assay by over 6x 105-fold,reaching down to 33.9fg mL^(-1)(-1.4fM) compared with an identical assay on glass substrates.Through monolithic integration with microfluidic elements,the device enables a flow-through assay for quantitative detection of cTnI in the serum with a detection sensitivity of 6.9 pg mL^(-1)(-0.3 pM)in<6 min,which was 4000 times lower than conventional glass devices.This ultrasensitive detection arises from the large surface area for antibody conjugation and metal-enhanced fluorescent signals through plasmonic nanostructures.Moreover,due to the parallel arrangement of flow paths,simultaneous detection of multiple cancer biomarkers,including prostate-specific antigen and carcinoembryonic antigen,has been fulfilled with increased signal-to-background ratios.Given the high performance of this assay,together with its simple fabrication process that is compatible with standard mass manufacturing techniques,we expect that the prepared integrated nanorod device can bring on-site point-of-care diagnosis closer to reality.Ye Wang Jiongdong Zhao Yu Zhu Shurong Dong Yang Liu Yijun Sun Liling Qian Wenting Yang Zhen Cao 2021Microsystems & Nanoengineering2021,7,4:0

19No evidence of superconductivity in a compressed sample prepared from lutetium foil and H_(2)/N_(2) gas mixture显示文摘A material described as lutetium–hydrogen–nitrogen(Lu-H-N in short)was recently claimed to have“near-ambient superconductivity”[Dasenbrock-Gammon et al.,Nature 615,244–250(2023)].If this result could be reproduced by other teams,it would be a major scientific breakthrough.Here,we report our results of transport and structure measurements on a material prepared using the same method as reported by Dasenbrock-Gammon et al.Our x-ray diffraction measurements indicate that the obtained sample contains three substances:the facecentered-cubic(FCC)-1 phase(Fm-3m)with lattice parameter a=5.03Å,the FCC-2 phase(Fm-3m)with a lattice parameter a=4.755Å,and Lu metal.The two FCC phases are identical to the those reported in the so-called near-ambient superconductor.However,we find from our resistance measurements in the temperature range from 300 K down to 4 K and the pressure range 0.9–3.4 GPa and our magnetic susceptibility measurements in the pressure range 0.8–3.3 GPa and the temperature range down to 100 K that the samples show no evidence of superconductivity.We also use a laser heating technique to heat a sample to 1800 XC and find no superconductivity in the produced dark blue material below 6.5 GPa.In addition,both samples remain dark blue in color in the pressure range investigated.Shu Cai Jing Guo Haiyun Shu Liuxiang Yang Pengyu Wang Yazhou Zhou Jinyu Zhao Jinyu Han Qi Wu Wenge Yang Tao Xiang Ho-kwang Mao Liling Sun 2023Matter and Radiation at Extremes2023,8,4:0
