| 1 | Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans显示文摘On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more 'human-like'. | 3 | The altered PD-1/PD-L1 pathway delivers the ‘one-two punch’ effects to promote the Treg/Th17 imbalance in pre-eclampsia显示文摘The programmed cell death-1(PD-1)/PD-ligand 1(PD-L1)pathway is critical for normal pregnancy by promoting regulatory T(Treg)cell development and inhibiting the Th17 response.However,the relationship between the PD-1/PD-L1 pathway and the Treg/Th17 imbalance in pre-eclampsia(PE)is an enigma.In this study,decreased PD-1 and PD-L1 expression and a Treg/Th17 imbalance were observed at the maternal-fetal interface in PE.The regulatory effects of the PD-1/PD-L1 pathway on the Treg and Th17 cell quantities were determined in vitro by targeting T-cell proliferation,differentiation and transdifferentiation.First,decreased PD-1 expression might contribute to a higher Th17 cell frequency by promoting proliferation in PE.Second,the percentages of Treg but not Th17 cells differentiated from peripheral naive CD4+T cells were increased by PD-L1 Fc administration.This effect was accompanied by decreased PI3K/AKT/m-TOR and increased PTEN mRNA expression and was completely reversed by PD-1 blockade.Finally,the percentage of IL-17-producing Treg cells increased and was positively associated with the Th17 cell frequency in PE.Increased RORγt and IL-17 but not Foxp3 and IL-10 mRNA expression by Treg cells was observed with PD-1 blockade.Similar findings occurred when Treg cells were exposed to IL-6/IL-23/IL-1βand were reversed by PD-L1 Fc.Taken together,our findings indicate that the PD-1/PD-L1 pathway contributes to the Treg/Th17 imbalance via‘one-two punch’approaches:(i)promoting Th17 cell proliferation,(ii)inhibiting Treg cell differentiation and(iii)enhancing Treg cell plasticity into Th17 cells in PE.The therapeutic value of PD-L1 Fc for PE treatment will be explored in the future. | Yonghong Zhang Zhaozhao Liu Mei Tian Xiaohui Hu Liling Wang Jinlu Ji Aihua Liao | 2018 | Cellular & Molecular Immunology2018,15,7: | 21 |
| 4 | Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. | 6 | Physiological responses of osteoblasts to cyclic stretching and the change of intracellular calcium concentration显示文摘The development of bone tissues is regulated by mechanical stimulation. Cyclic stretching was applied to the osteoblasts that were delivered from rat calvarie. The results showed that stretching at 500 me increased the cell prolifera-tion while loading at 1000 me and 1500 me inhabited cell growth. Loading also increased the adhesive force between cells and substrate as well as spreading areas of osteobalsts. Furthermore, the fluorescence probe Fluo-3/AM was used to investigate the effect of stretching stimulation on the intra-cellular calcium concentration of osteoblasts. The intracellu-lar calcium concentration of osteoblasts that were stretched at 500 me for 5 min was 92.9% higher than the control. After being treated with the panax ontoginseng saponins, the stretched osteoblasts still expressed 28.6% higher intracellu-lar calcium concentration than that of the control, which proved that both the influx of extracellular calcium and the release of intracellular calcium store were involved in the increase of intracellular calcium concentration when os-teoblasts responded to the cyclic stretching. | 13 | Observation of a bi-critical point between antiferromagnetic and superconducting phases in pressurized single crystal Ca0.73La0.27FeAs2显示文摘One of the most strikingly universal features of the high-temperature superconductors is that the superconducting phase emerges in the close proximity of the antiferromagnetic phase, and the interplay between these two phases poses a long-standing challenge. It is commonly believed that, as the antiferromagnetic transition temperature is continuously suppressed to zero, there appears a quantum critical point, around which the existence of antiferromagnetic fluctuation is responsible for the development of the superconductivity. In contrast to this scenario, we report the observation of a bi-critical point identified at 2.88 GPa and 26.02 K in the pressurized high-quality single crystal Ca_(0.73)La_(0.27)FeAs_2 by complementary in-situ high pressure measurements. | 16 | A Method for Alpine Wetland Delineation and Features of Border: Zoigê Plateau, China显示文摘Accurate wetland delineation is the basis of wetland definition and mapping, and is of great importance for wetland management and research. The Zoigê Plateau on the Qinghai-Tibet Plateau was used as a research site for research on alpine wetland delineation. Several studies have analyzed the spatiotemporal pattern and dynamics of these alpine wetlands, but none have addressed the issues of wetland boundaries. The objective of this work was to discriminate the upper boundaries of alpine wetlands by coupling ecological methods and satellite observations. The combination of Landsat 8 images and supervised classification was an effective method for rapid identification of alpine wetlands in the Zoigê Plateau. Wet meadow was relatively stable compared with hydric soils and wetland hydrology and could be used as a primary indicator for discriminating the upper boundaries of alpine wetlands. A slope of less than 4.5° could be used as the threshold value for wetland delineation. The normalized difference vegetation index(NDVI) in 434 field sites showed that a threshold value of 0.3 could distinguish grasslands from emergent marsh and wet meadow in September. The median normalized difference water index(NDWI) of emergent marsh remained more stable than that of wet meadow and grasslands during the period from September until July of the following year. The index of mean density in wet meadow zones was higher than the emergent and upland zones. Over twice the number of species occurred in the wet meadow zone compared with the emergent zone, and close to the value of upland zone. Alpine wetlands in the three reserves in 2014 covered 1175.19 km2 with a classification accuracy of 75.6%. The combination of ecological methods and remote sensing technology will play an important role in wetland delineation at medium and small scales. | 17 | Boosting pH-Universal Hydrogen Evolution of Molybdenum Disulfide Particles by Interfacial Engineering显示文摘The design of high-efficiency non-noble and earth-abundant electrocatalysts for hydrogen evolution reaction(HER)is highly paramount for water splitting and renewable energy systems.Molybdenum disulfide(MoS_(2))with abundant edge sites can be utilized as a promising alternative,but its catalytic activity is greatly related to the pH values,especially in an alkaline environment due to the extremely high energy barriers for water adsorption and dissociation steps.Here we report an exceptionally efficient and stable electrocatalyst to improve the sluggish HER process of layered MoS_(2)particles in different pH electrolytes,especially in base.The electrocatalyst is constructed by in situ growing selenium-doped MoS_(2)(Se-MoS_(2))nanoparticles on three-dimensional cobalt nickel diselenide(mCo_(0.2)Ni_(0.8)Se_(2))nanostructured arrays.Due to the large number of active edge sites of Se-MoS_(2)particles exposed at the surface,robust electrical conductivity and large surface area of mCo_(0.2)Ni_(0.8)Se_(2)support,and strong interfacial interactions between Se-MoS_(2)and mCo_(0.2)Ni_(0.8)Se_(2),this hybrid catalyst shows very outstanding catalytic HER properties featured by low overpotentials of 30 and 122 mV at 10 and 100 mA/cm^(2)with good operational stability in base,respectively,which outperforms most of inexpensive catalysts consisting of layered MoS_(2),transition metal selenides and sulfides,and it performs as well as noble Pt catalysts.Meanwhile,this electrocatalyst is also very active in neutral and acidic electrolytes,requiring low overpotentials of 93 and 94 mV at 10 mA/cm^(2),respectively,demonstrating its superb pH universality as a HER electrocatalyst with excellent catalytic durability.This study provides a straightforward strategy to construct an efficient non-noble electrocatalyst for driving the HER kinetics in different electrolytes. | Liling Liao Lun Yang Gang Zhao Haiqing Zhou Fengming Cai Yi Li Xiuzhang Wang Fang Yu | 2021 | Chinese Journal of Chemistry2021,39,2: | 2 |
| 18 | Construction of Dual-Biofunctionalized Chitosan/Collagen Scaffolds for Simultaneous Neovascularization and Nerve Regeneration显示文摘Biofunctionalization of artificial nerve implants by incorporation of specific bioactive factors has greatly enhanced the success of grafting procedures for peripheral nerve regeneration.However,most studies on novel biofunctionalized implants have emphasized the promotion of neuronal and axonal repair over vascularization,a process critical for long-term functional restoration.We constructed a dual-biofunctionalized chitosan/collagen composite scaffold with Ile-Lys-Val-Ala-Val(IKVAV)and vascular endothelial growth factor(VEGF)by combining solution blending,in situ lyophilization,and surface biomodification.Immobilization of VEGF and IKVAV on the scaffolds was confirmed both qualitatively by staining and quantitatively by ELISA.Various single-and dual-biofunctionalized scaffolds were compared for the promotion of endothelial cell(EC)and Schwann cell(SC)proliferation as well as the induction of angiogenic and neuroregeneration-associated genes by these cells in culture.The efficacy of these scaffolds for vascularization was evaluated by implantation in chicken embryos,while functional repair capacity in vivo was assessed in rats subjected to a 10mm sciatic nerve injury.Dual-biofunctionalized scaffolds supported robust EC and SC proliferation and upregulated the expression levels of multiple genes and proteins related to neuroregeneration and vascularization.Dual-biofunctionalized scaffolds demonstrated superior vascularization induction in embryos and greater promotion of vascularization,myelination,and functional recovery in rats.These findings support the clinical potential of VEGF/IKVAV dual-biofunctionalized chitosan/collagen composite scaffolds for facilitating peripheral nerve regeneration,making it an attractive candidate for repairing critical nerve defect.The study may provide a critical experimental and theoretical basis for the development and design of new artificial nerve implants with excellent biological performance. | Guicai Li Qi Han Panjian Lu Liling Zhang Yuezhou Zhang Shiyu Chen Ping Zhang Luzhong Zhang Wenguo Cui Hongkui Wang Hongbo Zhang | 2020 | Research2020,,1: | 2 |
