1Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans显示文摘On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more 'human-like'. 3Genomic landscape and genetic manipulation of the black soldier fly Hermetia illucens,a natural waste recycler显示文摘The black soldier fly(BSF),Hermetia illucens(Diptera:Stratiomyidae),is renowned for its bioconversion of organic waste into a sustainable source of animal feed.We report a high-quality genome of 1.1 Gb and a consensus set of 16,770 gene models for this beneficial species.Compared to those of other dipteran species,the BSF genome has undergone a substantial expansion in functional modules related to septic adaptation,including immune system factors,olfactory receptors,and cytochrome P450s.We further profiled midgut transcriptomes and associated microbiomes of BSF larvae fed with representative types of organic waste.We find that the pathways related to digestive system and fighting infection are commonly enriched and that Firmicutes bacteria dominate the microbial community in BSF across all diets.To extend its potential practical applications,we further developed an efficient CRISPR/Cas9-based gene editing approach and implemented this to yield flightless and enhanced feeding capacity phenotypes,both of which could expand BSF production capabilities.Our study provides valuable genomic and technical resources for optimizing BSF lines for industrialization.Shuai Zhan Gangqi Fang Minmin Cai Zongqing Kou Jun Xu Yanghui Cao Liang Bai Yixiang Zhang Yongmao Jiang Xingyu Luo Jian Xu Xia Xu Longyu Zheng Ziniu Yu Hong Yang Zhijian Zhang Sibao Wang Jeffery K.Tomberlin Jibin Zhang Yongping Huang 2020Cell Research2020,30,1:9
4In vitro degradation and biocompatibility of Mg-Nd-Zn-Zr alloy显示文摘In this study,in vitro degradation and biocompatibility of Mg-Nd-Zn-Zr(NZK) alloy were investigated to determine its suitability as a degradable medical biomaterial.Its corrosion properties were evaluated by static immersion test,electrochemical corrosion test,scanning electron microscopy(SEM),and energy dispersive spectroscopic(EDS) analysis,and in vitro biocompatibilities were assessed by hemolysis and cytotoxicity tests.Pure magnesium was used as control.The results of static immersion test and electrochemical corrosion test in simulated body fluid(SBF) demonstrated that the addition of alloying elements could improve the corrosion resistance.The hemolysis test found that the hemolysis rate of calcium phosphate coated NZK alloy was 4.8%,which was lower than the safe value of 5%.The cytotoxicity test indicated that NZK alloy extracts did not significantly reduce MC3T3-E1 cell viability.Hemolysis test and cytotoxicity test display excellent hemocompatibility and cytocompatibility of NZK alloy in vitro.Our data indicate that NZK alloy has excellent biocompatibility and thus can be considered as a potential degradable medical biomaterial for orthopedic applications.WANG YongPing HE YaoHua ZHU ZhaoJin JIANG Yao ZHANG Jian NIU JiaLin MAO Lin YUAN GuangYin 2012Chinese Science Bulletin2012,57,17:8

6Pyrolysis mechanism of glucose and mannose: The formation of 5-hydroxymethyl furfural and furfural显示文摘Fast pyrolysis of biomass will produce various furan derivatives, among which 5-hydroxymethyl furfural(5-HMF) and furfural(FF) are usually the two most important compounds derived from holocellulose. In this study, density functional theory(DFT) calculations are utilized to reveal the formation mechanisms and pathways of 5-HMF and FF from two hexose units of holocellulose, i.e., glucose and mannose. In addition, fast pyrolysis experiments of glucose and mannose are conducted to substantiate the computational results, and the orientation of 5-HMF and FF is determined by ^(13)C-labeled glucoses. Experimental results indicate that C1 provides the aldehyde group in both 5-HMF and FF, and FF is mainly derived from C1 to C5 segment. According to the computational results, glucose and mannose have similar reaction pathways to form 5-HMF and FF with d-fructose(DF) and 3-deoxy-glucosone(3-DG) as the key intermediates. 5-HMF and FF are formed via competing pathways. The formation of 5-HMF is more competitive than that of FF, leading to higher yield of 5-HMF than FF from both hexoses. In addition, compared with glucose,mannose can form 5-HMF and FF via extra pathways because of the epimerization at C2 position. 8H7N9 virus infection triggers lethal cytokine storm by activating gasdermin E-mediated pyroptosis of lung alveolar epithelial cells显示文摘The H7N9 influenza virus emerged in China in 2013,causing more than 1560 human infections,39% of which were fatal.A ‘cytokine storm’ in the lungs of H7N9 patients has been linked to a poor prognosis and death;however,the underlying mechanism that triggers the cytokine storm is unknown.Here,we found that efficient replication of the H7N9 virus in mouse lungs activates gasdermin E(GSDME)-mediated pyroptosis in alveolar epithelial cells,and that the released cytosolic contents then trigger a cytokine storm.Knockout of Gsdme switched the manner of death of A549 and human primary alveolar epithelial cells from pyroptosis to apoptosis upon H7N9 virus infection,and Gsdme knockout mice survived H7N9 virus lethal infection.Our findings reveal that GSDME activation is a key and unique mechanism for the pulmonary cytokine storm and lethal outcome of H7N9 virus infection and thus opens a new door for the development of antivirals against the H7N9 virus.Xiaopeng Wan Jiqing Li Yupeng Wang Xiaofei Yu Xijun He Jianzhong Shi Guohua Deng Xianying Zeng Guobin Tian Yanbing Li Yongping Jiang Yuntao Guan Chengjun Li Feng Shao Hualan Chen 2022National Science Review2022,9,1:3
9Preferential expression of cytochrome CYP CYP2R1 but not CYP1B1 in human cord blood hematopoietic stem and progenitor cells显示文摘Cytochrome P450(CYP)enzymes metabolize numerous endogenous substrates,such as retinoids,androgens,estrogens and vitamin D,that can modulate important cellular processes,including proliferation,differentiation and apoptosis.The aim of this study is to characterize the expression of CYP genes in CD34+human cord blood hematopoietic stem and early progenitor cells(CBHSPCs)as a first step toward assessment of the potential biological functions of CYP enzymes in regulating the expansion or differentiation of these cells.CD34+CBHSPCs were purified from umbilical cord blood via antibody affinity chromatography.Purity of CD34+CBHSPCs was assessed using fluorescence-activated cell sorting.RNA was isolated from purified CD34+CBHSPCs and total mononuclear cells(MNCs)for RNA-PCR analysis of CYP expression.Fourteen human CYPs were detected in the initial screening with qualitative RT-PCR in CD34+CBHSPCs.Further quantitative RNA-PCR analysis of the detected CYP transcripts yielded evidence for preferential expression of CYP2R1 in CD34+CBHSPCs relative to MNCs;and for greater expression of CYP1B1 in MNCs relative to CD34+CBHSPCs.These findings provide the basis for further studies on possible functions of CYP2R1 and CYP1B1 in CBHSPCs'proliferation and/or differentiation and their potential utility as targets for drugs designed to modulate CD34+CBHSPC expansion or differentiation.Shuoqi Xu Zhihua Ren Yanan Wang Xinxin Ding Yongping Jiang 2014Acta Pharmaceutica Sinica B2014,4,6:2
10Characterization of a Novel Weak Allele of RGA1/D1 and Its Potential Application in Rice Breeding显示文摘Semi-dwarfing improves the lodging resistance and yield of rice,and the vast majority of modern rice varieties harbor the sd1 allele to decrease plant height,resulting in reduced genetic diversity and negative agronomic traits.Thus,exploring alternative sources of dwarfism is imperative for rice breeding.Here,we identified a novel RGA1 allele,d1-w,from a local indica variety Xiaolixiang(XLX)using a map-based cloning approach.Compared with other rice varieties,RGA1 in XLX contained a unique single nucleotide polymorphism that resulted in an additional transcript and reduced functional RGA1 transcript level.The RGA1 from Nipponbare was introduced into XLX to estimate the value of d1-w in rice breeding.Compared with transgenic XLX plants(XLX^(D1)),XLX exhibited reduced plant height,increased stem strength,lower reactive oxygen species accumulation,delayed senescence,stronger photosynthesis,higher grain yield and quality(including external,milling and nutritional qualities),and enhanced resistance to drought and Rhizoctonia solani.Therefore,we proposed that the d1-w allele has potential as an excellent dwarfism resource for rice breeding.LIU Yantong LI Ting JIANG Zhishu ZENG Chuihai HE Rong QIU Jiao LIN Xiaoli PENG Limei SONG Yongping ZHOU Dahu CAI Yicong ZHU Changlan FU Junru HE Haohua XU Jie 2022Rice science2022,29,6:2
11Biocompatibility of Mg-Nd-Zn-Zr alloy with rabbit blood显示文摘In this study, in vitro blood biocompatibility of Mg-Nd-Zn-Zr (JDBM) alloy was investigated to determine its suitability as a degradable medical biomaterial. Blood biocompatibility was assessed by blood cell aggregation, platelet adhesion, and protein adsorption. A titanium alloy was used as control. The results showed that the JDBM alloy did not induce significant blood cell aggregation, platelet adhesion, and protein adsorption comparison with the titanium alloy (P>0.05).
