| 2 | Vaccination of poultry successfully eliminated human infection with H7N9 virus in China显示文摘The H7 N9 viruses that emerged in China in 2013 were nonpathogenic in chickens but mutated to a highly pathogenic form in early 2017 and caused severe disease outbreaks in chickens. The H7 N9 influenza viruses have caused five waves of human infection, with almost half of the total number of human cases(766 of 1,567) being reported in the fifth wave, raising concerns that even more human infections could occur in the sixth wave. In September 2017, an H5/H7 bivalent inactivated vaccine for chickens was introduced, and the H7 N9 virus isolation rate in poultry dropped by 93.3% after vaccination. More importantly,only three H7 N9 human cases were reported between October 1, 2017 and September 30, 2018, indicating that vaccination of poultry successfully eliminated human infection with H7 N9 virus. | 4 | Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. | 5 | Analysis of the CDR3 Length Repertoire and the Diversity of TCRα Chain in Human Peripheral Blood T Lymphocytes显示文摘Analysis of complementarity determining region 3 (CDR3) length of T lymphocyte receptors (TCRs) by immunoscope spectratyping technique has been used successfully to investigate the diversity of TCR in autoimmune diseases and infection diseases. In this study, we investigated the patterns of CDR3 length distribution for all 32 TCR AV gene families in human peripheral blood lymphocytes of four normal volunteers by the immunoscope spectratyping technique. It was found that PCR products exhibited an obscure band on 1.5% agarose gel electrophoresis. Each TCR AV family exhibited more than 8 bands on 6% sequencing gel electrophoresis. The CDR3 spectratyping of all TCR AV families showed a standard Gaussian distribution with different CDR3 length, and the expression frequency of CDR3 was similar among the gene families. Most of CDR3 in TCR AV family recombine in frame. However, some of the CDR3 showed out-of frame gene rearrangement. Additionally, we found that in some of TCR AV families there were 18 amino acid discrepancies between the longest CDR3 and shortest CDR3. | 6 | Spatial-temporal dynamics of desert vegetation and its responses to climatic variations over the last three decades:a case study of Hexi region in Northwest China显示文摘Analysis of spatial-temporal variations of desert vegetation under the background of climate changes can provide references for ecological restoration in arid and semi-arid areas.In this study,we used the Global Inventory Modeling and Mapping Studies(GIMMS)NDVI data from 1982 to 2006 and Moderate Resolution Imaging Spectroradiometer(MODIS)NDVI data from 2000 to 2013 to reveal the dynamics of desert vegetation in Hexi region of Northwest China over the past three decades.We also used the annual temperature and precipitation data acquired from the Chinese meteorological stations to analyze the response of desert vegetation to climatic variations.The average value of NDVImax(the maximum NDVI during the growing season)for desert vegetation in Hexi region increased at the rate of 0.65×10–3/a(P<0.05)from 1982 to 2013,and the significant increases of NDVImax mainly appeared in the typical desert vegetation areas.Vegetation was significantly improved in the lower reaches of Shule and Shiyang river basins,and the weighted mean center of desert vegetation mainly shifted toward the lower reaches of the two basins.Almost 95.32% of the total desert vegetation area showed positive correlation between NDVImax and annual precipitation,indicating that precipitation is the key factor for desert vegetation growth in the entire study area.Moreover,the areas with non-significant positive correlation between NDVImax and annual precipitation mainly located in the lower reaches of Shiyang and Shule river basins,this may be due to human activities.Only 7.64% of the desert vegetation showed significant positive correlation between NDVImax and annual precipitation in the Shule River Basin(an extremely arid area),indicating that precipitation is not the most important factor for vegetation growth in this basin,and further studies are needed to investigate the mechanism for this phenomenon. | YANG Xuemei LIU Shizeng YANG Taibao XU Xianying KANG Caizhou TANG Jinnian WEI Huaidong Mihretab G GHEBREZGABHER LI Zhiqi | 2016 | Journal of Arid Land2016,8,4: | 10 |

Based on the valence band E-k relations of strained Si/(111)Si1-xGex, the valence band and hole effective mass along the [111] and [-110] directions were obtained in this work. In comparison with the relaxed Si, the valence band edge degeneracy was partially lifted, and the significant change was observed band structures along the [111] and [-110] directions, as well as in its corresponding hole effective masses with the increasing Ge fraction. | 9 | Progesterone promotes neuronal differentiation of human umbilical cord mesenchymal stem cells in culture conditions that mimic the brain microenvironment显示文摘In this study, human umbilical cord mesenchymal stem cells from full-term neonates born by vaginal delivery were cultured in medium containing 150 mg/mL of brain tissue extracts from Sprague-Dawley rats (to mimic the brain microenvironment). Immunocytochemical analysis demonstrated that the cells differentiated into neuron-like cells. To evaluate the effects of progesterone as a neurosteroid on the neuronal differentiation of human umbilical cord mesenchymal stem cells, we cultured the cells in medium containing progesterone (0.1, 1, 10 μM) in addition to brain tissue extracts. Reverse transcription-PCR and flow cytometric analysis of neuron specific enolase-positive cells revealed that the percentages of these cells increased significantly following progesterone treatment, with the optimal progesterone concentration for neuron-like differentiation being 1 μM. | 10 | Recent developments in multimodality fluorescence imaging probes显示文摘Multimodality optical imaging probes have emerged as powerful tools that improve detection sensitivity and accuracy, important in disease diagnosis and treatment. In this review, we focus on recent developments of optical fluorescence imaging(OFI) probe integration with other imaging modalities such as X-ray computed tomography(CT), magnetic resonance imaging(MRI), positron emission tomography(PET), single-photon emission computed tomography(SPECT), and photoacoustic imaging(PAI). The imaging technologies are briefly described in order to introduce the strengths and limitations of each techniques and the need for further multimodality optical imaging probe development. The emphasis of this account is placed on how design strategies are currently implemented to afford physicochemically and biologically compatible multimodality optical fluorescence imaging probes. | 12 | Preparation and in vitro-in vivo evaluation of intestinal retention pellets of Berberine chloride to enhance hypoglycemic and lipid-lowing efficacy显示文摘Berberine chloride(BBR) is a pharmacokinetic profile of drug with poor bioavailability but good therapeutic efficacy,which is closely related to the discovery of BBR intestinal target.The major aim of this paper is to develop BBR intestinal retention type sustained-release pellets and evaluate their in vivo and in vitro behaviors base on the aspect of local action on intestinal tract. Here,wet milling technology is used to improve dissolution and dissolution rate of BBR by decreasing the particle size and increasing the wettability. The pellets are prepared by liquid layer deposition technology,and then the core pellets are coated with Eudragit~?L30 D-55 and Eudragit~?NE30 D aqueous dispersion. The prepared pellets show high drug loading capacity,and the drug loading up to 93%. Meanwhile,it possesses significant sustained drug release effect in purified water which is expected to improve the pharmacokinetic behavior of BBR. The pharmacokinetics results demonstrate that the halflife of BBR was increased significantly from 24 h to 36 h and the inter-and intra-subject variability are decreased compared to commercial BBR tablets. The retention test results indicate that the pellet size and Eudragit~?NE30 D plays an important role in retention time of the pellet,and it is found that the pellets with small particle size and high Eudragit~?NE30 D coating content can stay longer in the intestine than the pellets with large particle size. | 13 | bHLH104 confers tolerance to cadmium stress in Arabidopsis thaliana显示文摘Cd is a non-essential heavy metal that is toxic to both plants and animals. Here, we reveal that the transcription factor bHLH104 positively regulates Cd tolerance in Arabidopsis thaliana. We show that Fe deficiency-responsive genes were induced by Cd treatment, and that their upregulation was suppressed in bhlh104 loss-of-function mutants, but enhanced upon overexpression of bHLH104. Correspondingly, the bhlh104 mutants displayed sensitivity to Cd stress, whereas plants overexpressing bHLH104 exhibited enhanced Cd tolerance.Further analysis suggested that bHLH104 positively regulates four heavy metal detoxification-associated genes, IREG2, MTP3, HMA3 and NAS4, which play roles in Cd sequestration and tolerance. The bHLH104 overexpression plants accumulated high levels of Cd in the root but low levels of Cd in the shoot, which might contribute to the Cd tolerance in those lines. | 15 | Transcriptome Analysis Reveals Clues into leaf-like flower mutant in Chinese orchid Cymbidium ensifolium显示文摘The floral morphology of Cymbidium ensifolium,a well-known orchid in China,has increasingly attracted horticultural and commercial attention.However,the molecular mechanisms that regulate flower development defects in C.ensifolium mutants are poorly understood.In this work,we examined a domesticated variety of C.ensifolium named‘CuiYuMuDan',or leaf-like flower mutant,which lacks typical characteristics of orchid floral organs but continues to produce sepal-to leaf-like structures along the inflorescence.We used comparative transcriptome analysis to identify 6234 genes that are differentially expressed between mutant and wild-type flowers.The majority of these differentially expre ssed genes are involved in membrane-building,anabolism regulation,and plant hormone signal transduction,implying that in the leaf-like mutant these processes play roles in the development of flower defects.In addition,we identified 152 differentially expre ssed transcription factors,including the bHLH,MYB,MIKC,and WRKY gene families.Moreover,we found 20 differentially expressed genes that are commonly involved in flower development,including MADS-box genes,CLAVATA3(CLV3),WUSCHEL(WUS),and PERIANTHIA(PAN).Among them,floral homeotic genes were further investigated by phylogenetic analysis and expression validation,which displayed distinctive spatial expression patterns and significant changes between the wild type and the mutant.This is the first report on the C.ensifolium leaf-like flower mutant transcriptome.Our results shed light on the molecular regulation of orchid flower development,and may improve our understanding of floral patterning regulation and advance molecular breeding of Chinese orchids. | Yonglu Wei Jianpeng Jin Xiani Yao Chuqiao Lu Genfa Zhu Fengxi Yang | 2020 | Plant Diversity2020,42,2: | 4 |
| 16 | BP neural network based flexural strength prediction of open-porous Cu-SnTi composites显示文摘Open-porous Cu-Sn-Ti composites are fabricated by the space holder sintering technique using carbamide particles as space-holder material. Generally, the mechanical properties of open-porous sintered composites,especially the flexural strength affect the machine tools wear significantly. In this paper, a back-propagation(BP)artificial neural network with genetic algorithm(GA) and particle swarm optimization algorithm(PSOA) was then employed to relate the composition parameters(pore size, porosity and concentration of molybdenum disulfide particles) to the flexural strength. Furthermore, a comparison of predicted and experimental results using GA-BP and PSOA-BP models was conducted and good prediction accuracy was obtained. The study showed that PSOA-BP models could achieve better prediction results in aspects of the higher convergence velocity, lower relative errors of the flexure strength utilizing GA-BP models. | 17 | Effects of pore structure and distribution on strength of porous Cu-Sn-Ti alumina composites显示文摘Porous Cu-Sn-Ti alumina composites were fabricated by sintering Cu-Sn-Ti alloy powders, graphite particles, and alumina hollow particles agent. The effects of the pore structure and distribution on the composites strength were evaluated. Different pore distributions were modeled by using finite element analysis to investigate the tensile strength of the composites. Furthermore, a fractal analysis-based box-covering algorithm was used on the Cu-Sn-Ti alumina composites topology graphs to better investigate the pore structure and distribution. Results obtained show that different sizes and concentrations of alumina hollow particles could result in different porosities from20% to 50%. A larger pore size and a higher pore concentration reduce the strength, but provide more space for chip formation as a bonding material of a grinding wheel. The body-centered pore structure of the composites shows the highest stress under a tension load. The original composites topology graphs have been transformed to ordered distributed pore graphs based on the total pore area conservation. The information dimension magnitude difference between the original topology graphs and the ordered distributed circulars graphs is found to be linear with the Cu-Sn-Ti alumina composites strength. | 18 | Controllable synthesis of well-ordered TiO_2 nanotubes in a mixed organic electrolyte for high-efficiency photocatalysis显示文摘Well-ordered TiO 2 nanotube arrays (TNAs) were fabricated by electrochemical anodization in a mixed organic electrolyte consisting of ethylene glycol and glycerol. The morphology, structure, crystalline phase, and photocatalytic properties of TNAs were characterized by using TEM, SEM, XRD and photodegradation of methylene blue. It was found that the morphology and structure of TNAs could be significantly influenced by the anodization time and applied voltage. The obtained tube length was found to be proportional to anodization time, and the calculated growth rate of nanotubes was 0.6 m/h. The microstructure analysis demonstrated that the diameter and thickness of the nanotubes increased with the increase of anodization voltage. The growth mechanism of TNAs was also proposed according to the observed relationship between current density and time during anodization. | 19 | Influenza A virus use of BinCARD1 to facilitate the binding of viral NP to importinα7 is counteracted by TBK1-p62 axis-mediated autophagy显示文摘As a major component of the viral ribonucleoprotein(vRNP)complex in influenza A virus(IAV),nucleoprotein(NP)interacts with isoforms of importinαfamily members,leading to the import of itself and vRNP complex into the nucleus,a process pivotal in the replication cycle of IAV.In this study,we found that BinCARD1,an isoform of Bcl10-interacting protein with CARD(BinCARD),was leveraged by IAV for efficient viral replication.BinCARD1 promoted the nuclear import of the vRNP complex and newly synthesized NP and thus enhanced vRNP complex activity.Moreover,we found that BinCARD1 interacted with NP to promote NP binding to importinα7,an adaptor in the host nuclear import pathway.However,we also found that BinCARD1 promoted RIG-I-mediated innate immune signaling by mediating Lys63-linked polyubiquitination of TRAF3,and that TBK1 appeared to degrade BinCARD1.We showed that BinCARD1 was polyubiquitinated at residue K103 through a Lys63 linkage,which was recognized by the TBK1-p62 axis for autophagic degradation.Overall,our data demonstrate that IAV leverages BinCARD1 as an important host factor that promotes viral replication,and two mechanisms in the host defense system are triggered—innate immune signaling and autophagic degradation—to mitigate the promoting effect of BinCARD1 on the life cycle of IAV. | Xuyuan Wang Li Jiang Guangwen Wang Wenjun Shi Yuzhen Hu Bo Wang Xianying Zeng Guobin Tian Guohua Deng Jianzhong Shi Liling Liu Chengjun Li Hualan Chen | 2022 | Cellular & Molecular Immunology2022,19,10: | 3 |
| 20 | Organic solvent-and phase transfer catalyst-free oxidation of cyclohexanol to cyclohexanone with dilute H_2O_2显示文摘The oxidation of cyclohexanol to cyclohexanone with 30% aqueous hydrogen peroxide by using peroxotung-state complexes formed in situ from sodium tungstate di-hydrate and various bidentate organic ligands as the catalysts, without organic solvents, halide and phase transfer catalyst has been carried out. The influence of 13 ligands on the oxidation is investigated. The maximum yield of cyclohexanone is obtained when using 1,10-phenanthroline (96%) and oxalic acid (95%) as the ligand. Very high yields (around 90%) have also been obtained for the instances of using salicylic acid, 3,5-dibromosalicylic acid, and 8-hydro- xylquino-line as ligands.
