| 1 | In vitro differentiation of adipose-derived stem cells and bone marrow-derived stromal stem cells into neuronal-like cells显示文摘Adipose-derived stem cells and bone marrow-derived stromal stem cells were co-cultured with untreated or Aβ1-40-treated PC12 cells, or grown in supernatant derived from untreated or Aβ1-40-treated PC12 cells. Analysis by western blot and quantitative real-time PCR showed that protein levels of Nanog, Oct4, and Sox2, and mRNA levels of miR/125a/3p were decreased, while expression of insulin-like growth factor-2 and neuron specific enolase was increased. In comparison, the generation of neuron specific enolase-positive cells was most successful when adipose-derived stem cells were co-cultured with Aβ1-40-treated PC12 cells. Our results demonstrate that adipose-derived stem cells and bone marrow-derived stromal stem cells exhibit trends of neuronal-like cell differentiation after co-culture with Aβ1-40-treated PC12 cells. | 3 | Effects of nitrogen sources and glucose on the con- sumption of ethylene and methane by temperate volcanic forest surface soils显示文摘There is limited knowledge with regard to the consumption of ethylene (C2H4) and methane (CH4) in volcanic forest soils containing low microbial carbon-to-organic carbon ratio, and to the responses of both consumptions to nitrogen and carbon additions. Temperate volcanic forest surface soils under three forest stands (e.g. Pinus sylvestris L., Cryptomeria japonica and Quercus serrata) were used to compare CH4 and C2H4 consumption by forest soils, and to study the effects of nitrogen sources and glucose on both consumptions. There was a good parallel between CH4 and C2H4 consumption by for- est soils, but mineralization reduced CH4 consumption rather than C2H4 consumption in forest soils, particularly in a Pinus forest soil. The stimulatory effect of glucose addition on both CH4 and C2H4 consumption by forest soils was increased by increasing the pre-incubation period after glucose addi- tion, and a largest stimulation occurred in the Pinus forest soil. The addition of KNO3-N at the rate of 100 μg·g1 significantly reduced the consumptions of both C2H4 and CH4 by forest soils (P≤0.05). In the presence of urea plus dicyandiamide, the consumption rates of C2H4 and CH4 by forest soils were higher than those in the KNO3-N and urea-N treated soils at the same N rate (P≤0.05), but were similar to those of the control. Hence, under experimental conditions, there was a strong inhibitory effect of NO3 rather than NH4+ addition on the CH4 and C2H4 consumption in these forest soils. When amount of the added NO3-N increased up to more than 2―3 times the soil initial NO3-N concentrations, both C2H4 and CH4 consumption rates were reduced to 10%―20% of the rates in soils without nitrate addition. | 4 | Biodegradation of dissolved organic carbon in soil extracts and leachates from a temperate forest stand and its relationship to ultraviolet absorbance显示文摘The amount and biodegradability of dissolved organic carbon (DOC) in forest floors can contribute to carbon sequestration in soils and the release of CO 2-C from soil to the atmosphere.There is only limited knowledge about the biodegradation of DOC in soil extracts and leachates due to the limitations inherent in degradation experiments.Differences in the biodegradation of DOC were studied in forest soil extracts using cold and hot water and 4 mmol/L CaCl 2 solution and in soil leachates sampled under different conditions over a wide range of DOC concentrations.From these results,we developed a simple and rapid method for determining the biodegradable organic C in forest floors.The hot water extracts and CaCl 2 extracts after CH 3 Cl fumigation contained higher concentrations of biodegradable organic C than the cold water extracts and CaCl 2 extracts before fumigation,with rapid DOC degradation occurring 24-48 h after incubation with an inoculum,followed by slow DOC degradation till 120-168 h into the incubation.During a 7-d incubation with an inoculum,the variation in DOC degradation in the different soil extracts was consistent with the change in special UV absorbance at 254 nm.Relatively higher levels of biodegradable organic C were detected in soil leachates from the forest canopy than in forest gaps between April and October 2008 (P <0.05).Relatively lower concentrations of DOC and biodegradable organic C were observed in soil leachates from N-fertilized plots during the growing season compared with the control,with the exception of the plot treated with KNO 3 at a rate of 45 kg N ha 1 a 1.Around 77.4% to 96.3% of the variability in the biodegradable organic C concentrations in the forest floors could be accounted for by the initial DOC concentration and UV absorbance at 254 nm.Compared with the conventional inoculum incubation method,the method of analyzing UV absorbance at 254 nm is less time consuming and requires a much smaller sample volume.The results suggest that the regression models obtained using the initial DOC concentration and UV absorbance can provide a rapid,simple and reliable method for determining the biodegradable organic C content,especially in field studies involving relatively large numbers of samples. | XU XingKai LUO XianBaot JIANG SongHua XU ZhongJun | 2012 | Chinese Science Bulletin2012,57,8: | 7 |
| 5 | Effect of vegetation type, wetting intensity, and nitrogen supply on external carbon stimulated heterotrophic respiration and microbial biomass carbon in forest soils显示文摘By using packed soil-core incubation experiments,we have studied stimulating effects of addition of external carbon(C)(glucose,6.4 g Cm^(-2))on heterotrophic respiration and microbial biomass C of a mature broadleaf and Korean pine mixed forest(BKPF)and an adjacent white birch forest(WBF)soil under different wetting intensities(55%and 80%WFPS,water-filled pore space)and nitrogen(N)supply(NH_4C1 and KNO_3,4.5 g Nm^(-2))conditions.The results showed that for the control,the cumulative carbon dioxide(CO_2)flux from WBF soil during the 15-day incubation ranged from 5.44 to 5.82 g CO_2-Cm^(-2),which was significantly larger than that from BKPF soil(2.86 to 3.36 g CO_2-Cm^(-2)).With increasing wetting intensity,the cumulative CO_2 flux from the control was decreased for the WBF soil,whereas an increase in the CO_2 flux was observed in the BKPF soil(P<0.05).The addition of NH_4C1 or KNO_3 alone significantly reduced the cumulative CO_2 fluxes by 9.2%-21.6%from the two soils,especially from WBF soil at low wetting intensity.The addition of glucose alone significantly increased soil heterotrophic respiration,microbial biomass C(MBC),and microbial metabolic quotient.The glucose-induced cumulative CO_2 fluxes and soil MBC during the incubation ranged from 8.7 to 11.7 g CO_2-Cm^(-2)and from 7.4 to 23.9 g Cm^(-2),which are larger than the dose of added C.Hence,the addition of external carbon can increase the decomposition of soil native organic C.The glucose-induced average and maximum rates of CO_2 fluxes during the incubation were significantly influenced by wetting intensity(WI)and vegetation type(VT),and by WI×VT,NH_4Cl×VT and WI×VT×NH_4Cl(P<0.05).The addition of NH_4C1,instead of KNO_3 significantly decreased the glucose-induced MBC of WBF soil(P<0.05),whereas adding NH_4C1 and KNO_3 both significantly increased the glucose-induced MBC of BKPF soil at high moisture(P<0.05).According to the differences in soil labile C pools,MBC and CO_2 fluxes in the presence and absence of glucose,it can be concluded that the stimulating effects of glucose on soil heterotrophic respiration and MBC under temperate forests were dependent on vegetation type,soil moisture,and amount and type of the N added. | WU HaoHao XU XingKai DUAN CunTao LI TuanSheng CHENG WeiGuo | 2015 | Science China Earth Sciences2015,58,8: | 6 |
| 6 | Measurement of ethylene and methane production in a temperate forest soil using inhibition of acetylene and carbon monoxide显示文摘We studied in the laboratory the effects of acetylene (C2H2) concentrations on the accumulation and consumption of ethylene and methane in a temperate pine forest soil, and in situ ethylene and methane production and flush effects of nitrogen sources on both productions in the pine forest stand (Pinus sylvestris L.). The addition of C2H2 at concentrations more than 50 Pa C2H2 in the headspace caused a more than 95% reduction in rates of ethylene and methane consumption in forest soil compared to those with no C2H2. Furthermore, addition of acetylene within a range of 50 to 10, 000 Pa C2H2 induced a similar rate of methane accumulation in forest soil. Hence, it can be concluded that presence of more than 50 Pa C2H2 in the headspace is an effective method to measure methane production in forest soil. The addition of C2H2 at concentrations more than 50 Pa C2H2 induced an increasing concentration of ethylene in the headspace (P≤0.05), indicating the reduction of acetylene to ethylene in forest soil. Using inhibition of 0.5 kPa C2H2 in combination with 5 kPa carbon monoxide that inhibits the reduction of acetylene in a short term, it was observed that there was a larger in situ methane production rate (218 ± 26 μg C m-2 h-1 (μg C per square meter per hour, the same below)) than in situ ethylene produc-tion rate (92 ± 6 μg C m-2 h-1) in the pine forest soil. The addition of nitrogen sources such as urea, urea plus a nitrification inhibitor dicyandiamide, and potassium nitrate, could induce a 5-fold greater in-crease in rates of in situ ethylene and methane production compared to those in the control, particu-larly in the latter (P≤0.05). | 8 | Diversity of metabolite accumulation patterns in inner and outer seed coats of pomegranate:exploring their relationship with genetic mechanisms of seed coat development显示文摘The expanded outer seed coat and the rigid inner seed coat of pomegranate seeds,both affect the sensory qualities of the fruit and its acceptability to consumers.Pomegranate seeds are also an appealing model for the study of seed coat differentiation and development.We conducted nontarget metabolic profiling to detect metabolites that contribute to the morphological differentiation of the seed coats along with transcriptomic profiling to unravel the genetic mechanisms underlying this process.Comparisons of metabolites in the lignin biosynthetic pathway accumulating in seed coat layers at different developmental stages revealed that monolignols,including coniferyl alcohol and sinapyl alcohol,greatly accumulated in inner seed coats and monolignol glucosides greatly accumulated in outer seed coats.Strong expression of genes involved in monolignol biosynthesis and transport might explain the spatial patterns of biosynthesis and accumulation of these metabolites.Hemicellulose constituents and flavonoids in particular accumulated in the inner seed coat,and candidate genes that might be involved in their accumulation were also identified.Genes encoding transcription factors regulating monolignol,cellulose,and hemicellulose metabolism were chosen by coexpression analysis.These results provide insights into metabolic factors influencing seed coat differentiation and a reference for studying seed coat developmental biology and pomegranate genetic improvement. | Gaihua Qin Chunyan Liu Jiyu Li Yongjie Qi Zhenghui Gao Xiaoling Zhang Xingkai Yi Haifa Pan Ray Ming Yiliu Xu | 2020 | Horticulture Research2020,7,1: | 4 |

| 10 | Dynamic analysis of heat extraction rate by supercritical carbon dioxide in fractured rock mass based on a thermal-hydraulic-mechanics coupled model显示文摘Heat production from geothermal reservoirs is a typical heat transfer process involving a cold working fluid contacting a hot rock formation.Compared to the thermal-physical characteristics of water,supercritical CO_(2)(scCO_(2))has a higher heat storage capacity over a wide temperature-pressure range and may be favored as a heat transfer fluid.Singularly characteristic of scCO_(2)-based heat extraction is that the hydraulic-thermal properties of the scCO_(2) vary dramatically and dynamically with the spatial pressure gradient during unsteady-state flow along fracture.This highly nonlinear behavior presents a challenge in the accurate estimation of heat extraction efficiency in scCO_(2)-based EGS.In this paper,a thermal-h ydraulic-mechanical(THM)coupled model is developed by considering deformation of the fractured reservoir,non-Darcy flow and the varying thermal-physical properties of scCO_(2).The proposed model is validated by matching the modeling temperature distribution with published data.The results show that during continuous injection of scCO_(2),the fracture first widens and then narrows,ultimately reopening over the long term.The sequential fracture deformation behaviors are in response to the combined impacts of mechanical compression and thermally-induced deformation.By controlling the injection parameters of the scCO_(2),it is found that the heat extraction rate is positively correlated to its pore pressure or mass flow rate.The heat extraction rate can be significantly enhanced,when the inlet temperature of scCO_(2) is below its critical temperature.As a result,the heat increment recovered per unit mass of scCO_(2) decreases as the hot rock is gradually cooled.Meanwhile,the heat increment recovered per unit mass of scCO_(2) decreases by increasing the inlet temperature of scCO_(2) or its mass flow rate,but increases as the outlet pressure rises.Furthermore,multi-linear regression indicates that controlling the inlet temperature of the scCO_(2) can significantly improve the thermodynamic efficiency of heat extraction. | Chunguang Wang Xingkai Shi Wei Zhang Derek Elsworth Guanglei Cui Shuqing Liu Hongxu Wang Weiqiang Song Songtao Hu Peng Zheng | 2022 | International Journal of Mining Science and Technology2022,32,2: | 3 |

The results indicated that the viscosity in the earlier stage calculated by dual-Arrhenius model matched the experimental data. As rising to 400 m·Pas, the viscosity calculated by the improved engineering model was closer to the experimental data. | 13 | Effect of intensity and duration of freezing on soil microbial biomass,extractable C and N pools,and N2O and CO2 emissions from forest soils in cold temperate region显示文摘Freezing can increase the emissions of carbon dioxide(CO_2) and nitrous oxide(N_2O) and the release of labile carbon(C) and nitrogen(N) pools into the soil. However, there is limited knowledge about how both emissions respond differently to soil freezing and their relationships to soil properties. We evaluated the effect of intensity and duration of freezing on the emissions of CO_2 and N_2 O, net N mineralization, microbial biomass, and extractable C and N pools in soils from a mature broadleaf and Korean pine mixed forest and an adjacent secondary white birch forest in northeastern China. These soils had different contents of microbial biomass and bulk density. Intact soil cores of 0–5 cm and 5–10 cm depth sampled from the two temperate forest floors were subjected to -8, -18, and -80°C freezing treatments for a short(10 d) and long(145 d) duration, and then respectively incubated at 10°C for 21 d. Soil cores, incubated at 10°C for 21 d without a pretreatment of freezing, served as control. Emissions of N_2 O and CO_2 after thaw varied with forest type, soil depth, and freezing treatment. The difference could be induced by the soil water-filled pore space(WFPS) during incubation and availability of substrates for N_2 O and CO_2 production, which are released by freezing. A maximum N_2 O emission following thawing of frozen soils was observed at approximately 80% WFPS, whereas CO_2 emission from soils after thaw significantly increased with increasing WFPS. The soil dissolved organic C just after freezing treatment and CO_2 emission increased with increase of freezing duration, which paralleled with a decrease in soil microbial biomass C. The cumulative net N mineralization and net ammonification after freezing treatment as well as N_2 O emission were significantly affected by freezing temperature. The N_2 O emission was negatively correlated to soil p H and bulk density, but positively correlated to soil K_2SO_4-extractable NO_3^--N content and net ammonification. The CO_2 emission was positively correlated to the cumulative net N mineralization and net ammonification.
