1Engineered Functional Surfaces by Laser Microprocessing for Biomedical Applications显示文摘Metallic biomaterials are increasingly being used in various medical applications due to their high strength,fracture resistance,good electrical conductivity,and biocompatibility.However,their practical applications have been largely limited due to poor surface performance.Laser microprocessing is an advanced method of enhancing the surface-related properties of biomaterials.This work demonstrates the capability of laser microprocessing for biomedical metallic materials including magnesium and titanium alloys,with potential applications in cell adhesion and liquid biopsy.We investigate laser-material interaction,microstructural evolution,and surface performance,and analyze cell behavior and the surface-enhanced Raman scattering(SERS)effect.Furthermore,we explore a theoretical study on the laser microprocessing of metallic alloys that shows interesting results with potential applications.The results show that cells exhibit good adhesion behavior at the surface of the laser-treated surface,with a preferential direction based on the textured structure.A significant SERS enhancement of 6×10^3 can be obtained at the laser-textured surface during Raman measurement.Guoqing Hu Kai Guan Libin Lu Jiaru Zhang Nie Lu Yingchun Guan 2018Engineering2018,4,6:5

11A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 13Effect of moxibustion on autophagy and the inflammatory response of synovial cells in rheumatoid arthritis model rats显示文摘OBJECTIVE:To investigate the effect of moxibustion on synovitis and the autophagy of synoviocytes in rheumatoid arthritis(RA).METHODS:Forty Sprague-Dawley rats were randomly divided into a normal group,model group,moxibustion group,cigarette moxibustion group,and medicine group,with eight rats included in each group.The RA model was established by subcutaneous injection of complete Freund's adjuvant into the left posterior toe.Rats in the model group were not interfered with.In the moxibustion group,rats were treated by moxibustion,where a 1-cm diameter moxa stick was applied at the left Zusanli(ST 36)point.The distance of the moxa stick to the skin was 2 cm and moxibustion was completed for 20 min daily for 15 d total.In the cigarette moxibustion group,the moxa stick was replaced by a common cigarette.In the medicine group,rats were treated with a tripterygium glycoside suspension(8 mg/kg)once a day for 15 d total.In each group,the left hind limb toe volume was measured with a toe volume meter;the synovial cells were observed by hematoxylin and eosin staining;the interleukin(IL)-4,IL-6,IL-10,IL-1β,IL-23,IL-17,and tumor necrosis factor(TNF)-αlevels in serum were measured by enzyme-linked immunosorbent assay;the erythrocyte sedimentation rate(ESR)were detected by Westergren sedimentation rate testing;the C-reactive protein(CRP)and rheumatoid factor(RF)levels in serum were detected by rate nephelometry;the expression levels of ULK1,autophagy-associated protein(Atg)3,Atg5,and Atg12 messenger RNA(m RNA)in synovium were detected by real time-quantitative polymerase chain reaction(RT-q PCR);and the protein expression levels of phosphatidylinositol-3-kinase(PI3K),protein kinase B(Akt),mammalian target of rapamycin(mTOR),LC3-Ⅱ,beclin-1,phosphorylated-PI3K(p-PI3K),p-Akt,p-mTOR in synovium were detected by Western blotting.RESULTS:Among the RA model rats,joint swelling,an inflammatory reaction,and the proliferation of synovial tissue were obvious and the signal of the PI3K/Akt/mTOR pathway was active,while autophagy was inhibited.Moxibustion at Zusanli(ST36)or intragastric administration of Tripterygium wilfordii glycosides could alleviate the inflammatory reaction of RA rats;relieve the swelling of the toes;downregulate the levels of ESR,CRF,RF;lower the levels of IL-6,IL-1β,TNF-α,and IL-17;and increase the IL-4 and IL-10.At the same time,the m RNA expression levels of ULK1,Atg3,Atg5,and Atg12 and those of LC3-Ⅱand beclin-1 were increased,while the PI3K,Akt,mTOR,p-PI3K,p-Akt,p-mTOR were decreased.Cigarette moxibustion did not significantly reduce the swelling of the toe joint in RA rats,and was not as good as that of moxibustion or Tripterygium wilfordii polyglycosides in the effects of inflammation relief and the influences of the levels of ESR,CRF,RF.While cigarette moxibustion has a weak effect to affect the expression of corresponding molecules in autophages and the expression level of the autophagy biomaker in synovial tissue.Moxibustion and tripterygium glycosides can significantly reduce the joint swelling,relieve synovitis and synovial hyperplasia,and inhibit the PI3K/Akt/mTOR signaling pathway to increase autophagy in a manner superior to cigarette moxibustion.CONCLUSION:Moxibustion can limit the proliferation of synoviocytes in RA rats by inhibiting the PI3K/Akt/mTOR signaling pathway,promoting autophagy,effectively reducing synovitis,and alleviating joint swelling.HAO Feng WANG Qiang LIU Lei WU Libin CAI Ronglin SANG Jiajia HU Jun WANG Jie YU Qing HE Lu SHEN Yingchao MIAO Yiming HU Ling WU Zijian 2022Journal of Traditional Chinese Medicine2022,42,1:1

16Systematic screening reveals synergistic interactions that overcome MAPK inhibitor resistance in cancer cells显示文摘Objective:Effective adjuvant therapeutic strategies are urgently needed to overcome MAPK inhibitor(MAPKi)resistance,which is one of the most common forms of resistance that has emerged in many types of cancers.Here,we aimed to systematically identify the genetic interactions underlying MAPKi resistance,and to further investigate the mechanisms that produce the genetic interactions that generate synergistic MAPKi resistance.Methods:We conducted a comprehensive pair-wise sgRNA-based high-throughput screening assay to identify synergistic interactions that sensitized cancer cells to MAPKi,and validated 3 genetic combinations through competitive growth,cell viability,and spheroid formation assays.We next conducted Kaplan-Meier survival analysis based on The Cancer Genome Atlas database and conducted immunohistochemistry to determine the clinical relevance of these synergistic combinations.We also investigated the MAPKi resistance mechanisms of these validated synergistic combinations by using co-immunoprecipitation,Western blot,qRTPCR,and immunofluorescence assays.Results:We constructed a systematic interaction network of MAPKi resistance and identified 3 novel synergistic combinations that effectively targeted MAPKi resistance(ITGB3+IGF1R,ITGB3+JNK,and HDGF+LGR5).We next analyzed their clinical relevance and the mechanisms by which they sensitized cancer cells to MAPKi exposure.Specifically,we discovered a novel protein complex,HDGF-LGR5,that adaptively responded to MAPKi to enhance cancer cell stemness,which was up-or downregulated by the inhibitors of ITGB3+JNK or ITGB3+IGF1R.Conclusions:Pair-wise sgRNA library screening provided systematic insights into elucidating MAPKi resistance in cancer cells.ITGB3-+IGF1R-targeting drugs(cilengitide+linsitinib)could be used as an effective therapy for suppressing the adaptive formation of the HDGF-LGR5 protein complex,which enhanced cancer stemness during MAPKi stress.Yu Yu Minzhen Tao Libin Xu Lei Cao Baoyu Le Na An Jilin Dong Yajie Xu Baoxing Yang Wei Li Bing Liu Qiong Wu Yinying Lu Zhen Xie Xiaohua Lian 2022Cancer Biology & Medicine2022,19,2:1

20Nonlinear characteristics analyses of particle motion for predicting flow regimes显示文摘Gas-solid flow regimes have a significant impact on particle transport and separation in a fluidized bed reactor.In this study,to determine flow regime transitions in gas-solid fluidized beds,an acoustic technique was used to detect and analyze the behavior of gas and solids.Algorithm complexity,fluctuation complexity,and Shannon entropy analyses of acoustic emission signals were performed to examine non linear system characteristics,and to determine the flow regime transiti on velocities uc,uk,and ufd-Moreover,using the standard deviation of pressure signals,pressure measurements and acoustic measurements were compared.The relative deviations(RDs)between the experimental and empirical values of uk were 8.8%,13.7%,8.8%,and 30.4%for the algorithm complexity,fluctuation complexity,Shannon entropy,and pressure signal standard deviation,respectively,while the respective RDs for Ufd were 15.7%,23.9%,15.7%,and 97.8%.The RDs between the experimental and empirical values of uc were all 6.4%.The experimental values obtained from acoustic signal measurements were therefore closer to the empirical values.In summary,the integration of non-intrusive acoustic measurements,complexity analysis,and Shannon entropy analysis is suitable for identifying flow regime transitions.Panxing Kang Yujian Lu Lei Yang Libin Liu Xiayi(Eric)Hu Xiao Luo Hongbo Chen Yefeng Zhou Rui Zhang 2020Particuology2020,18,6:1
