| 6 | Conservation and Diversity of MicroRNA-associated Copper-regulatory Networks in Populus trichocarpa显示文摘Plants develop important regulatory networks to adapt to the frequently-changing availability of copper (Cu). However, little is known about miRNA-associated Cu-regulatory networks in plant species other than Arabidopsis. Here, we report that Cu-responsive miRNAs in Populus trichocarpa (Torr. & Gray) include not only conserved miR397, miR398 and miR408, but also Populus-specific miR1444, suggesting the conservation and diversity of Cu-responsive miRNAs in plants. Copper-associated suppression of mature miRNAs is in company with the up-regulation of their target genes encoding Cu-containing proteins in Populus. The targets include miR397-targeted PtLAC5, PtLAC6 and PtLAC110a, miR398- targeted PtCSD1, PtCSD2a and PtCSD2b, miR408-targeted PtPCL1, PtPCL2, PtPCL3 and PtLAC4, and miR1444-targeted PtPPO3 and PtPPO6. Consistently, P. trichocarpa miR408 promoter-directed GUS gene expression is down-regulated by Cu in transgenic tobacco plants. Cu-response elements (CuREs) are found in the promoters of Cu-responsive miRNA genes. We identified 34 SQUAMOSA-promoter binding protein-like (SPL) genes, of which 17 are full-length PtSPL proteins or partial sequences with at least 300 amino acids. Phylogenetic analysis indicates that PtSPL3 and PtSPL4 are CuRE-binding proteins controlling Cu-responsive gene expression. | 12 | Copper Adsorption on Olivine显示文摘Copper adsorption on olivine supplied by the A/S Olivine Production Plant at Aheim in western Norway was studied. The factors were evaluated which affect the uptake of copper. It is shown that the equilibrium pH of aqueous solution has the greatest influence on copper adsorption thanks to the competitive adsorption between proton and copper ions, and the adsorption of copper to olivine increases rapidly with pH increasing from 4 to 6. Moreover, initial copper concentration and olivine dose possess significant effect on copper adsorption, for the adsorption rate of copper increases with olivine dose increasing or initial copper concentration decreasing at the same pH. In addition, the ionic strength effect on the adsorption was also investigated, but it owns little effect on the adsorption process of copper due to the formation of inner sphere adsorption of copper on olivine. | 15 | Numerical simulation of characteristics of semidiurnal tidal waves in sea region around Taiwan显示文摘POM97, an oceanic model, has been used for the first time to the numerical study on the tidal waves of the as Regions around Taiwan. In this paper, we have got the result that the semidiurnal tidal waves of these area mainly are the co-operating tides which come from the south of 23’N of the western Pacific. Those semidiurnal tidal waves affecting the Taiwan Straits come respectively from the south and the north entrance of the channel, and the north tidal wave is stronger than the south one. The strongest tidal field is the area from the Meizhou Bay to the Xinhua Bay along the coast of Fujian Province, where the biggest amplitude of the M2 partial tide can reach 240 cm. The strongest tidal cur- rent fields lie in the Penghu watercourse, where the maximum velocity of the M2 partial tide can arrive at 196 m/s. In the horizontal structure of the tidal currets, we have found that there is a stream dot in the north of the channel, besides, there still exist four new ones. | 16 | Proteomic Study Identifies Proteins Involved in Brassinosteroid Regulation of Rice Growth显示文摘Brassinosteroids (BRs) are essential hormones for growth and development of plant. In rice, BRs regulate multiple developmental processes and affect many important traits such as height, leaf angle, fertility and seed filling. We identified brassinosteroid-regulated proteins in rice using proteomic approaches and performed functional analysis of some BR-regulated proteins by overexpression experiments. Using two-dimensional difference gel electrophoresis (2-D DIGE) followed by protein identification by mass spectrometry, we compared proteomic differences in the shoots and roots of the BR-insensitive mutant d61-4 and BR-deficient mutant brd1-3. We identified a large number of proteins differentially expressed in the mutants compared with wild type control. These include a glycine-rich RNA-binding protein (OsGRP1) and a DREPP2 protein, which showed reduced levels in the BR mutants. Overexpression of these two proteins partially suppressed the dwarf phenotype of the Arabidopsis BR-insensitive mutant bri1-5. In contrast to the reduced protein level, the RNA level of OsGRP1 was not significantly affected in the BR mutants or by BR treatment, suggesting BR regulation of OsGRP1 at the posttranslational level.
