Application of preoperative artificial neural network based on blood biomarkers and clinicopathological parameters for predicting longterm survival of patients with gastric cancer显示文摘BACKGROUND Because of the powerful abilities of self-learning and handling complex biological information, artificial neural network(ANN)models have been widely applied to disease diagnosis, imaging analysis, and prognosis prediction. However, there has been no trained preoperative ANN(preope-ANN)model to preoperatively predict the prognosis of patients with gastric cancer(GC).AIM To establish a neural network model that can predict long-term survival of GC patients before surgery to evaluate the tumor condition before the operation.METHODS The clinicopathological data of 1608 GC patients treated from January 2011 to April 2015 at the Department of Gastric Surgery,Fujian Medical University Union Hospital were analyzed retrospectively. The patients were randomly divided into a training set(70%)for establishing a preope-ANN model and a testing set(30%).The prognostic evaluation ability of the preope-ANN model was compared with that of the American Joint Commission on Cancer(8th edition)clinical TNM(cTNM)and pathological TNM(pTNM)staging through the receiver operating characteristic curve,Akaike information criterion index,Harrell's C index, and likelihood ratio chi-square.RESULTS We used the variables that were statistically significant factors for the 3-year overall survival as input-layer variables to develop a preope-ANN in the training set.The survival curves within each score of the preope-ANN had good discrimination(P<0.05).Comparing the preope-ANN model,cTNM,and pTNM in both the training and testing sets, the preope-ANN model was superior to cTNM in predictive discrimination(C index),predictive homogeneity(likelihood ratio chi-square),and prediction accuracy(area under the curve). The prediction efficiency of the preope-ANN model is similar to that of pTNM.CONCLUSION The preope-ANN model can accurately predict the long-term survival of GC patients, and its predictive efficiency is not inferior to that of pTNM stage. | Si-Jin Que Qi-Yue Chen Qing-Zhong Zhi-Yu Liu Jia-Bin Wang Jian-Xian Lin Jun Lu Long-Long Cao Mi Lin Ru-Hong Tu Ze-Ning Huang Ju-Li Lin Hua-Long Zheng Ping Li Chao-Hui Zheng Chang-Ming Huang Jian-Wei Xie | 2019 | World Journal of Gastroenterology2019,25,43: | 4
Fast computation of scattering from 3D complex structures by MLFMA显示文摘This paper introduces the research work on the extension of multilevel fast multipole algorithm (MLFMA) to 3D complex structures including coating object, thin dielectric sheet, composite dielectric and con-ductor, cavity. The impedance boundary condition is used for scattering from the object coated by thin lossy material. Instead of volume integral equation, surface integral equation is applied in case of thin dielectric sheet through resistive sheet boundary condition. To realize the fast computation of scattering from composite homoge-neous dielectric and conductor, the surface integral equation based on equivalence principle is used. Compared with the traditional volume integral equation, the surface integral equation reduces greatly the number of unknowns. To compute conducting cavity with electrically large aperture, an electric field integral equation is applied. Some numerical results are given to demonstrate the validity and accuracy of the present methods. | Hu Jun Nie Zaiping Que Xiaofeng Meng Min | 2008 | Journal of Systems Engineering and Electronics2008,19,5: | 3
Efficient analysis of dielectric radomes using multilevel fast multipole algorithm with CRWG basis显示文摘A full-wave analysis of the electromagnetic problem of a three-dimensional (3-D) antenna radiating through a 3-D dielectric radome is preserued. The problem is formulated using the Poggio-Miller-Chang-Harrington-Wu(PMCHW) approach for homogeneous dielectric objects and the electric field integral equation for conducting objects. The integral equations are discretized by the method of moment (MoM), in which the conducting and dielectric surface/interfaces are represented by curvilinear triangular patches and the unknown equivalent electric and magnetic currents are expanded using curvilinear RWG basis functions. The resultant matrix equation is then solved by the multilevel fast multipole algorithm (MLFMA) and fast far-field approximation (FAFFA) is used to further accelerate the computation. The radiation patterns of dipole arrays in the presence of radomes are presented. The numerical results demonstrate the accuracy and versatility of this method. | Que Xiaofeng Nie Zaiping Hu Jun | 2008 | Journal of Systems Engineering and Electronics2008,19,1: | 2
Short-term efficacy of robotic and laparoscopic spleen-preserving splenic hilar lymphadenectomy via Huang’s three-step maneuver for advanced upper gastric cancer: Results from a propensity scorematched study显示文摘BACKGROUND Robotic surgery has been considered to be significantly better than laparoscopic surgery for complicated procedures.AIM To explore the short-term effect of robotic and laparoscopic spleen-preserving splenic hilar lymphadenectomy(SPSHL)for advanced gastric cancer(GC)by Huang’s three-step maneuver.METHODS A total of 643 patients who underwent SPSHL were recruited from April 2012 to July 2017,including 35 patients who underwent robotic SPSHL(RSPSHL)and 608 who underwent laparoscopic SPSHL(LSPSHL).One-to-four propensity score matching was used to analyze the differences in clinical data between patients who underwent robotic SPSHL and those who underwent laparoscopic SPSHL.RESULTS In all,175 patients were matched, including 35 patients who underwent RSPSHL and 140 who underwent LSPSHL.After matching, there were no significant differences detected in the baseline characteristics between the two groups.Significant differences in total operative time, estimated blood loss(EBL),splenic hilar blood loss(SHBL),splenic hilar dissection time(SHDT),and splenic trunk dissection time were evident between these groups(P<0.05).Furthermore,no significant differences were observed between the two groups in the overall noncompliance rate of lymph node(LN)dissection(62.9%vs 60%,P=0.757),number of retrieved No.10 LNs(3.1±1.4 vs 3.3±2.5,P=0.650),total number of examined LNs(37.8±13.1 vs 40.6±13.6,P=0.274),and postoperative complications(14.3%vs 17.9%,P=0.616).A stratified analysis that divided the patients receiving RSPSHL into an early group(EG)and a late group(LG)revealed that the LG experienced obvious improvements in SHDT and length of stay compared with the EG(P<0.05).Logistic regression showed that robotic surgery was a significantly protective factor against both SHBL and SHDT(P<0.05).CONCLUSION RSPSHL is safe and feasible, especially after overcoming the early learning curve,as this procedure results in a radical curative effect equivalent to that of LSPSHL. | Jia-Bin Wang Zhi-Yu Liu Qi-Yue Chen Qing Zhong Jian-Wei Xie Jian-Xian Lin Jun Lu Long-Long Cao Mi Lin Ru-Hong Tu Ze-Ning Huang Ju-Li Lin Hua-Long Zheng Si-Jin Que Chao-Hui Zheng Chang-Ming Huang Ping Li | 2019 | World Journal of Gastroenterology2019,25,37: | 2
Tacrolimus reduces scar formation and promotes sciatic nerve regeneration显示文摘A sciatic nerve transection and repair model was established in Sprague-Dawley rats by transecting the tendon of obturator internus muscle in the greater sciatic foramen and suturing with nylon sutures. The models were treated with tacrolimus gavage (4 mg/kg per day) for 0, 2, 4 and 6 weeks. Specimens were harvested at 6 weeks of intragastric administration. Masson staining revealed that the collagen fiber content and scar area in the nerve anastomosis of the sciatic nerve injury rats were significantly reduced after tacrolimus administration. Hematoxylin-eosin staining showed that tacrolimus significantly increased myelinated nerve fiber density, average axon diameter and myelin sheath thickness. Intragastric administration of tacrolimus also led to a significant increase in the recovery rate of gastrocnemius muscle wet weight and the sciatic functional index after sciatic nerve injury. The above indices were most significantly improved at 6 weeks after of tacrolimus gavage. The myelinated nerve fiber density in the nerve anastomosis and the sciatic nerve functions had a significant negative correlation with the scar area, as detected by Spearman's rank correlation analysis. These findings indicate that tacrolimus can promote peripheral nerve regeneration and accelerate the recovery of neurological function through the reduction of scar formation. | Jun Que Quan Cao Tao Sui Shihao Du Ailiang Zhang Dechao Kong Xiaojian Cao | 2012 | Neural Regeneration Research2012,7,32: | 2
Histones released by NETosis enhance the infectivity of SARS-CoV-2 by bridging the spike protein subunit 2 and sialic acid on host cells显示文摘Neutrophil extracellular traps(NETs)can capture and kill viruses, such as influenza viruses, human immunodeficiency virus(HIV),and respiratory syncytial virus(RSV),thus contributing to host defense.Contrary to our expectation,we show here that the histones released by NETosis enhance the infectivity of SARS-CoV-2,as found by using live SARS-CoV-2 and two pseudovirus systems as well as a mouse model. The histone H3 or H4 selectively binds to subunit 2 of the spike(S)protein,as shown by a biochemical binding assay, surface plasmon resonance and binding energy calculation as well as the construction of a mutant S protein by replacing four acidic amino acids.Sialic acid on the host cell surface is the key molecule to which histones bridge subunit 2 of the S protein. Moreover, histones enhance cell-cell fusion. Finally, treatment with an inhibitor of NETosis, histone H3 or H4,or sialic acid notably affected the levels of sgRNA copies and the number of apoptotic cells in a mouse model. These findings suggest that SARS-CoV-2 could hijack histones from neutrophil NETosis to promote its host cell attachment and entry process and may be important in exploring pathogenesis and possible strategies to develop new effective therapies for COVID-19. | Weiqi Hong Jingyun Yang Jun Zou Zhenfei Bi Cai He Hong Lei Xuemei He Xue Li Aqu Alu Wenyan Ren Zeng Wang Xiaohua Jiang Kunhong Zhong Guowen Jia Yun Yang Wenhai Yu Qing Huang Mengli Yang Yanan Zhou Yuan Zhao Dexuan Kuang Junbin Wang Haixuan Wang Siyuan Chen Min Luo Ziqi Zhang Tiangi Lu Li Chen Haiying Que Zhiyao He Qiu Sun Wei Wang Guobo Shen Guangwen Lu Zhiwei Zhao Li Yang Jinliang Yang Zhenling Wang Jiong Li Xiangrong Song Lunzhi Dai Chong Chen Jia Geng Maling Gou Lu Chen Haohao Dong Yong Peng Canhua Huang Zhiyong Qian Wei Cheng Changfa Fan Yuquan Wei Zhaoming Su Aiping Tong Shuaiyao Lu Xiaozhong Peng Xiawei Wei | 2022 | Cellular & Molecular Immunology2022,19,5: | 2

Central pancreatectomy for benign or low-grade malignant pancreatic tumors in the neck and body of the pancreas显示文摘BACKGROUND For tumors in the neck and body of the pancreas, distal pancreatectomy(DP)has been the standard surgical procedure for the last few decades and central pancreatectomy(CP)is an alternative surgical option.Whether CP better preserves remnant pancreatic endocrine and exocrine functions after surgery remains a subject of debate.AIM To evaluate the safety and efficacy of CP compared with DP for benign or lowgrade malignant pancreatic tumors in the neck and body of the pancreas.METHODS This retrospective study enrolled 296 patients who underwent CP or DP for benign and low-malignant neoplasms at the same hospital between January 2016 and March 2020.Perioperative outcomes and long-term morbidity of endocrine/exocrine function were prospectively evaluated.RESULTS No significant difference was observed in overall morbidity or clinically relevant postoperative pancreatic fistula between the two groups(P=0.055).Delayed gastric emptying occurred more frequently in the CP group than in the DP group(29.4%vs 15.3%;P<0.005).None of the patients in the CP group had new-onset or aggravated distal metastasis, whereas 40 patients in the DP group had endocrine function deficiency after surgery(P<0.05).There was no significant difference in the incidence of diarrhea immediately after surgery, but at postoperative 12 mo,a significantly higher number of patients had diarrhea in the DP group than in the CP group(0%vs 9.5%;P<0.05).CONCLUSION CP is a generally safe procedure and is better than DP in preserving long-term pancreatic endocrine and exocrine functions. Therefore,CP might be a better option for treating benign or low-grade malignant neoplasms in suitable patients. | Yi-Wen Chen Jian Xu Xiang Li Wei Chen Shun-Liang Gao Yan Shen Min Zhang Jian Wu Ri-Sheng Que Jun Yu Ting-Bo Liang Xue-Li Bai | 2022 | World Journal of Gastrointestinal Surgery2022,14,9: | 1
Rapid identification of spinal ventral and dorsal roots using a quartz crystal microbalance显示文摘The fast and accurate identification of nerve tracts is critical for successful nerve anastomosis.Taking advantage of differences in acetylcholinesterase content between the spinal ventral and dorsal roots,we developed a novel quartz crystal microbalance method to distinguish between these nerves based on acetylcholinesterase antibody reactivity. The acetylcholinesterase antibody was immobilized on the electrode surface of a quartz crystal microbalance and reacted with the acetylcholinesterase in sample solution. The formed antigen and antibody complexes added to the mass of the electrode inducing a change in frequency of the electrode. The spinal ventral and dorsal roots were distinguished by the change in frequency. The ventral and dorsal roots were cut into 1 to 2-mm long segments and then soaked in 250 μL PBS.Acetylcholinesterase antibody was immobilized on the quartz crystal microbalance gold electrode surface. The results revealed that in 10 minutes, both spinal ventral and dorsal roots induced a frequency change;however, the frequency change induced by the ventral roots was notably higher than that induced by the dorsal roots. No change was induced by bovine serum albumin or PBS.These results clearly demonstrate that a quartz crystal microbalance sensor can be used as a rapid, highly sensitive and accurate detection tool for the quick identification of spinal nerve roots intraoperatively. | Tao Sui Jun Que Dechao Kong Hao Xie Daode Wang Kun Shi Xiaojian Cao Xiang Li | 2013 | Neural Regeneration Research2013,8,8: | 1

Potential risk of mitomycin C at high concentrations on peripheral nerve structure显示文摘Although the local application of mitomycin C may prevent epidural adhesion after laminectomy, mitomycin C can induce neurotoxicity in optic and acoustic nerves at high concentrations. To determine the safe concentration range for mitomycin C, cotton pads soaked with mitomycin C at different concentrations(0.1, 0.3, 0.5, and 0.7 mg/mL) were immediately applied for 5 minutes to the operation area of rats that had undergone laminectomy at L1. Rat sciatic nerves, instead of dorsal nerves, were used in this study. The results showed that mitomycin C at 0.1–0.5 mg/mL did not damage the structure and function of the sciatic nerve, while at 0.7 mg/mL, mitomycin C significantly reduced the thickness of the sciatic nerve myelin sheath compared with lower concentrations, though no functional change was found. These experimental findings indicate that the local application of mitomycin C at low concentrations is safe to prevent scar adhesion following laminectomy, but that mitomycin C at high concentrations(> 0.7 mg/mL) has potential safety risks to peripheral nerve structures. | Tao Sui Jinhong Zhang Shihao Du Changhui Su Jun Que Xiaojian Cao | 2014 | Neural Regeneration Research2014,9,8: | 1

Overexpression of a sugarcane ScCaM gene negatively regulates salinity and drought stress responses in transgenic Arabidopsis thaliana显示文摘Calmodulin(CaM)proteins play a key role in signal transduction under various stresses. In the present study, the effects of a sugarcane ScCaM gene(NCBI accession number:GQ246454)on drought and salt stress tolerance in transgenic Arabidopsis thaliana and Escherichia coli cells were evaluated. The results demonstrated a significant negative role of ScCaM in the drought and salt stress tolerance of transgenic lines of A.thaliana,as indicated by the phenotypes. In addition, the expression of AtP5CS and AtRD29A,two genes tightly related to stress resistance, was significantly lower in the overexpression lines than in the wild type. The growth of E.coli BL21 cells expressing ScCaM showed weaker tolerance under mannitol and NaCl stress.Taken together, this study revealed that the ScCaM gene plays a negative regulatory role in both mannitol and NaCl stresses, and it possibly exerts protective mechanisms common in both prokaryotes and eukaryotes under stress conditions. | JINXIAN LIU JINGFANG FENG CHANG ZHANG YONGJUAN REN WEIHUA SU GUANGHENG WU XIANYU FU NING HUANG YOUXIONG QUE HUI LING JUN LUO | 2023 | BIOCELL2023,47,1: | 0
Recent Progress and Future Direction for the Application of Multiomics Data in Clinical Liver Transplantation显示文摘Omics data address key issues in liver transplantation(LT)as the most effective therapeutic means for end-stage liver disease. The purpose of this study was to review the current application and future direction for omics in LT.We reviewed the use of multiomics to elucidate the pathogenesis leading to LT and prognostication.Future directions with respect to the use of omics in LT are also described based on perspectives of surgeons with experience in omics.Significant molecules were identified and summarized based on omics, with a focus on post-transplant liver fibrosis, early allograft dysfunction, tumor recurrence, and graft failure. We emphasized the importance omics for clinicians who perform LTs and prioritized the directions that should be established. We also outlined the ideal workflow for omics in LT.In step with advances in technology, the quality of omics data can be guaranteed using an improved algorithm at a lower price.Concerns should be addressed on the translational value of omics for better therapeutic effects in patients undergoing LT. | Zhengtao Liu Jun Xu Shuping Que Lei Geng Lin Zhou Adil Mardinoglu Shusen Zheng | 2022 | Journal of Clinical and Translational Hepatology2022,10,2: | 0
Diversified Application of Barcoded PLATO(PLATO-BC) Platform for Identification of Protein Interactions显示文摘Proteins usually associate with other molecules physically to execute their functions.Identifying these interactions is important for the functional analysis of proteins.Previously,we reported the parallel analysis of translated ORFs(PLATO)to couple ribosome display of full-length ORFs with affinity enrichment of mRNA/protein/ribosome complexes for the “bait”molecules, followed by the deep sequencing analysis of mRNA.However, the sample processing, from extraction of precipitated mRNA to generation of DNA libraries, includes numerous steps, which is tedious and may cause the loss of materials.Barcoded PLATO(PLATO-BC),an improved platform was further developed to test its application for protein interaction discovery. In this report,we tested the antisera-antigen interaction using serum samples from patients with inclusion body myositis(IBM).Tripartite motif containing 21(TRIM21)was identified as a potentially new IBM autoantigen. We also expanded the application of PLATO-BC to identify protein interactions for JQ1,single ubiquitin peptide, and NS5 protein of Zika virus. From PLATO-BC analyses,we identified new protein interactions for these “bait”molecules. We demonstrate that Ewing sarcoma breakpoint region 1(EWSR1)binds to JQ1 and their interactions may interrupt the EWSR1 association with acetylated histone H4.RIO kinase 3(RIOK3),a newly identified ubiquitin-binding protein,is preferentially associated with K63-ubiquitin chain. We also find that Zika NS5 protein interacts with two previously unreported host proteins,par-3 family cell polarity regulator(PARD3)and chromosome 19 open reading frame 53(C19orf53),whose attenuated expression benefits the replication of Zika virus. These results further demonstrate that PLATO-BC is capable of identifying novel protein interactions for various types of “bait”molecules. | Weili Kong Tsuyoshi Hayashi Guillaume Fiches Qikai Xu Mamie ZLi Jianwen Que Shuai Liu Wei Zhang Jun Qi Netty Santoso Stephen JElledge Jian Zhu | 2019 | Genomics, Proteomics & Bioinformatics2019,17,3: | 0
Two antibodies show broad, synergistic neutralization against SARS-CoV-2 variants by inducing conformational change within the RBD显示文摘Continual evolution of the severe acute respiratory syndrome coronavirus(SARS-CoV-2)virus has allowed for its gradual evasion of neutralizing antibodies(nAbs)produced in response to natural infection or vaccination. The rapid nature of these changes has incited a need for the development of superior broad nAbs(bnAbs)and/or the rational design of an antibody cocktail that can protect against the mutated virus strain. Here,we report two angiotensin-converting enzyme 2 competing nAbs—8H12 and 3E2—with synergistic neutralization but evaded by some Omicron subvariants.Cryo-electron microscopy reveals the two nAbs synergistic neutralizing virus through a rigorous pairing permitted by rearrangement of the 472-489 loop in the receptor-binding domain to avoid steric clashing.Bispecific antibodies based on these two nAbs tremendously extend the neutralizing breadth and restore neutralization against recent variants including currently dominant XBB.1.5.Together,these findings expand our understanding of the potential strategies for the neutralization of SARS-CoV-2 variants toward the design of broad-acting antibody therapeutics and vaccines. | Hui Sun Tingting Deng Yali Zhang Yanling Lin Yanan Jiang Yichao Jiang Yang Huang Shuo Song Lingyan Cui Tingting Li Hualong Xiong Miaolin Lan Liqin Liu Yu Li Qianjiao Fang Kunyu Yu Wenling Jiang Lizhi Zhou Yuqiong Que Tianying Zhang Quan Yuan Tong Cheng Zheng Zhang Hai Yu Jun Zhang Wenxin Luo Shaowei Li Qingbing Zheng Ying Gu Ningshao Xia | 2024 | Protein & Cell2024,15,2: | 0
