2Clinical characteristics and effectiveness of antiviral agents in hospitalized children with infectious mononucleosis in China:A multicenter retrospective study显示文摘Importance:The clinical characteristics of infectious mononucleosis(IM)in Chinese children have not been evaluated in multicenter studies,and the effectiveness of antiviral treatment are controversial.Objective:To investigate the clinical characteristics of Chinese children with IM and current status of antiviral therapy for affected patients.Methods:Hospitalized patients with IM were enrolled between 2018 and 2020 in five children’s hospitals in China.The clinical characteristics were compared among four age groups:<3 years,3-<6 years,6-<10 years,and≥10 years.The clinical characteristics of IM and effectiveness of antiviral therapy were compared among patients receiving acyclovir(ACV),ganciclovir(GCV),and no antiviral therapy(i.e.,non-antiviral group).Results:In total,499 patients were analyzed;most patients were 3-<6 years of age.The most common symptoms and signs included fever(100%),lymphadenopathy(98.6%),pharyngitis(86.4%),eyelid edema(76.8%),and snoring(72.9%).There were significant differences in rash,hepatomegaly,and liver dysfunction among the four age groups.Patients aged<3 years had a lower incidence of liver dysfunction and a higher incidence of rash.Among the 499 patients,50.1%were treated with GCV,26.3%were treated with ACV,and 23.6%received no antiviral therapy.Compared with the non-antiviral group,patients in the ACV and GCV groups had longer durations of fever(P<0.001).There were no significant differences in the incidences of complications among the three treatment groups.Interpretation:The incidence of IM in Chinese children peaked at 3-<6 years of age.Clinical features of IM varied according to age.Patients receiving antiviral therapy exhibited more serious clinical manifestations than did patients without antiviral therapy.The effectiveness of antiviral therapy for IM requires further analysis.Huili Hu Huiling Deng Jing Bi Yi Xu Shuangjie Li Yue Xie Xinrong Sun Dongmeng Wang Xufang Li Wenxian Ouyang Bing Hu Yufeng Zhang He Tang Chunxiao Fang Hui Zhang Lingyun Guo Chen Wang Tianyi Wang Fengxia Yang Tao Jiang Zhengde Xie Gang Liu 2021Pediatric Investigation2021,5,3:12

4Hexagonal boron nitride:A metal-free catalyst for deep oxidative desulfurization of fuel oils显示文摘Oxidative desulfurization(ODS)has been proved to be an efficient strategy for the production of clean fuel oil.Numerous metal-based materials have been employed as excellent ODS catalysts,but being hindered by their high-cost and potential secondary pollution.In this work,we employed graphene analogous hexagonal boron nitride(h-BN)as a metal-free catalyst for ODS with hydrogen peroxide(H2O2)as the oxidant.The h-BN catalyst was characterized and proved to be a few-layered structure with relatively high specific surface areas.The h-BN catalyst showed a 99.4%of sulfur removal in fuel oil under the optimized reaction conditions.Besides,the h-BN can be recycled for 8 times without significant decrease in the catalytic performance.Detailed mechanism analysis found that it is the boron radicals in h-BN activated H2O2 to generate·OH species,which can readily oxidize sulfides to corresponding sulfones for separation.This work would provide another choice in choosing metal-free catalysts for ODS.Peiwen Wu Linjie Lu Jing He Linlin Chen Yanhong Chao Minqiang He Fengxia Zhu Xiaozhong Chu Huaming Li Wenshuai Zhu 2020Green Energy & Environment2020,5,2:3
5A novel recombinant DNA vaccine encodingMycobacterium tuberculosis ESAT-6 and FL protects againstMycobacterium tuberculosis challenge in mice显示文摘Mycobacterium tuberculosis 6-kDa early secretory antigenic target(ESAT-6)is a dominant target antigen for cell-mediated immunity in the early phase of tuberculosis.The fms-like tyrosine kinase 3 ligand(FL)that induces potent immune response has been used as an adjuvant in vaccine development.In this study,a new recombinant plasmid(pIRES-epitope-peptides-FL)encoding three T cell epitopes of ESAT-6 and FL was constructed,and the immunogenicity of the DNA vaccine was assessed in C57BL/6 mice immunized with the plasmid DNA vaccine.Additionally,a strategy of intramuscular injection with the DNA vaccine(prime)and intranasal administration of the epitope peptides(boost)was employed to induce higher immune reaction of the mice.The results showed that mice vaccinated with the recombinant plasmid DNA vaccine and boosted with the peptides not only increased the levels of Th1 cytokines(IFN-γand IL-12),the number of IFN-γ+T cells and activities of cytotoxic T lymphocytes as well as IgG,but also enhanced protection against Mycobacterium tuberculosis challenge.In conclusion,these data indicate that the novel recombinant pIRES-epitope-peptides-FL plasmid is a useful DNA vaccine for preventing Mycobacterium tuberculosis infection.Qingtao Jiang Jing Zhang Xia Chen Mei Xia Yanlai Lu Wen Qiu Ganzhu Feng Dan Zhao Yan Li Fengxia He Guangyong Peng Yingwei Wang 2013The Journal of Biomedical Research2013,27,5:2

8Sublytic C5b-9 triggers glomerular mesangial cell apoptosis in rat Thy-1 nephritis via Gadd45 activation mediated by Egr-1 and p300-dependent ATF3 acetylation显示文摘The apoptosis of glomerular mesangial cells(GMCs)is considered to be an important contributor to the initiation and development of rat Thy-1 nephritis(Thy-1N)and is accompanied by sublytic C5b-9 deposition.However,the mechanism by which sublytic C5b-9 triggers GMC apoptosis has not been elucidated.In this study,functional and histological examinations were performed on GMCs treated with sublytic C5b-9(in vitro)and renal tissues of Thy-1N rats(in vivo).The in vitro studies found that sublytic C5b-9 could trigger GMC apoptosis through upregulating Egr-1,ATF3,and Gadd45 expression.Egr-1-mediated post-transcriptional modulation of ATF3,Egr-1/ATF3-enhanced Gadd45 promoter activity,and p300-mediated ATF3 acetylation were all involved in GMC apoptosis.More importantly,the effective binding elements for Egr-1 and ATF3 to Gadd45β/γpromoters and the ATF3 acetylation site were identified.In vivo,silencing renal p300,Egr-1,ATF3,and Gadd45β/γsignificantly decreased GMC apoptosis,secondary GMC proliferation,and urinary protein secretion in Thy-1N rats.Together,these findings implicate that sublytic C5b-9-induced activation of Egr-1/p300–ATF3/Gadd45 axis plays a critical role in GMC apoptosis in Thy-1N rats.Fengxia He Mengya Zhou Tianyi Yu Dan Zhao Jing Zhang Wen Qiu Yanlai Lu Yu Liu Lulu Wang Yingwei Wang 2016Journal of Molecular Cell Biology2016,8,6:1
9The genetic control of glabrous glume during African rice domestication显示文摘African cultivated rice,Oryza glaberrima,is characterized by its glabrous glumes.During domestication,the pubescent glumes of its wild ancestor,Oryza barthii,lost their trichomes,and in this study,we show that glabrous glume 5(GLAG5),a WUSCHEL-like homeobox transcription factor gene on chromosome 5,is required for trichome development.DNA methylation associated with an hATtransposable element inserted in the promoter region of GLAG5 is found to reduce its expression,leading to the formation of glabrous glumes and leaves in African cultivated rice.Among 82 African cultivated rice varieties investigated in this study,59(approximately 71%)lines exhibit glabrous glumes and harbor the hAT transposon;however,the other 23 varieties(approximately 29%),which exhibit pubescent glumes,lack the hAT transposon,indicating that glag5 had undergone strong artificial selection.Theπ;/π;ratios also show the hAT transposon insertions influence the genetic diversity of an approximately 150-kb interval encompassing the GLAG5 locus.The identification of the GLAG5 gene provides new insights into the domestication of cultivated rice in Africa.We speculate that the selection of varieties with mutations in their promoter regions is an important aspect of crop domestication.Leqin Chang Min Hu Jing Ning Wei He Jiayu Gao Marie-Noelle Ndjiondjop Yongcai Fu Fengxia Liu Hongying Sun Ping Gu Chuanqing Sun Zuofeng Zhu 2022Journal of Genetics and Genomics2022,49,5:1

11Effect of intracranial hypertension on cerebral hemorrhage induced autonomic nerve imbalance显示文摘BACKGROUND: Cerebral hemorrhage can cause the imbalance of nerve function, whereas its mechanism and main impact factors are still not quite clear. OBJECTIVE: To explore the rules about the changes of intracranial pressure in brainstem hemorrhage and internal capsule hemorrhage, and analyze the role of intracranial hypertension in the changes of nerve function caused by cerebral hemorrhage. DESIGN: A self-controlled trial. SETTING: Department of Physiology, Tianjin Medical University. MATERIALS: Sixty-five healthy male Japanese white rabbits with long ears (1.5–1.8 kg) were supplied and fed by the Department of Animal Experiment of Tianjin Medical University. The RM6240B biological signal collecting and processing system was used. METHODS: The experiments were conducted in the Department of Physiology, Tianjin Medical University from August 2001 to May 2006. ① The rabbits were anesthetized, then fixed onto the brain stereotaxic apparatus, and afterwards fenestration on skull and intubation to lateral ventricle were performed.The dynamic changes of intracranial pressure were monitored continuously. Rabbits were infused with autologous arterial blood (0.3 mL) into midbrain corpora quadrigemina inferior colliculus to induce model of acute brainstem hemorrhage; models of internal capsule hemorrhage were established by infusing autologous arterial blood into internal capsule. ② The dynamic intracranial pressures under the above conditions were recorded continuously with the RM6240B biological signal collecting and processing system. ③ An animal model of persistent intracranial hypertension was established by infusion of physiologic saline into lateral ventricle. ④ The changes of the intensity of autonomic nerve discharge were analyzed, using the biological signal collecting and processing system before and after hemorrhage and under persistent intracranial hypertension. ⑤ Ten animal models of internal capsule hemorrhage and 10 of brainstem hemorrhage were selected respectively, then gross pathological samples were cut open, and the accuracy of hemorrhage models was affirmed. Histological sections in hemorrhage point and around this point were prepared for with hematoxylin and eosin staining, and the pathological changes were observed under light microscope. MAIN OUTCOME MEASURES: ① Changes of intracranial pressures before and after internal capsule hemorrhage and brainstem hemorrhage; ② Changes of the discharge intensity of cervical vagus nerve trunk in animal models of internal capsule hemorrhage, brainstem hemorrhage and persistent intracranial hypertension without hemorrhage; ③ Accuracy of location of internal capsule hemorrhage and brainstem hemorrhage confirmed by gross pathological samples and sections. RESULTS: Totally 65 rabbits were involved in the analysis of results. ① Dynamic state of intracranial pressure: Intracranial pressure increased obviously at 45 minutes after internal capsule hemorrhage and brainstem hemorrhage, the intracranial pressures were (1.31±0.30), (1.82±0.45) kPa, which were obviously higher than those before hemorrhage [(1.04±0.18), ( 1.05±0.19) kPa, P < 0.01]. ② Discharge of vagus nerve: Under intracranial hypertension, the discharge of cervical vagus nerve trunk was enhanced, and the discharge intensity of vagus nerve trunk was significantly different before and after persistent intracranial hypertension [(364.28±78.55), (1252.19±151.75) μV·s, P < 0.01]. The discharges of cervical vagus nerve trunk were significantly enhanced after internal capsule hemorrhage and brainstem hemorrhage (P < 0.01). ③ Validation of hemorrhage sites: The hemorrhage sites were internal capsule and brainstem on histopathological sections. 12Identification of cross-reactive CD8^(+)T cell receptors with high functional avidity to a SARS-CoV-2 immunodominant epitope and its natural mutant variants显示文摘Despite the growing knowledge of T cell responses in COVID-19 patients,there is a lack of detailed characterizations for T cell-antigen interactions and T cell functions.Here,with a predicted peptide library from SARS-CoV-2 S and N proteins,we found that specific CD8+T cell responses were identified in over 75%of COVID-19 convalescent patients(15/20)and an epitope from the N protein,N361-369(KTFPPTEPK),was the most dominant epitope from our selected peptide library.Importantly,we discovered 2 N361-369-specific T cell receptors(TCRs)with high functional avidity that were independent of the CD8 co-receptor.These TCRs exhibited complementary cross-reactivity to several presently reported N361-369 mutant variants,as to the wild-type epitope.Further,the natural functions of these TCRs in the cytotoxic immunity against SARS-CoV-2 were determined with dendritic cells(DCs)and the lung organoid model.We found that the N361-369 epitope could be normally processed and endogenously presented by these different types of antigen presenting cells,to elicit successful activation and effective cytotoxicity of CD8+T cells ex vivo.Our study evidenced potential mechanisms of cellular immunity to SARS-CoV-2,and illuminated potential ways of viral clearance in COVID-19 patients.These results indicate that utilizing CD8-independent TCRs against SARS-CoV-2-associated antigens may provide functional superiority that is beneficial for the adoptive cell immunotherapies based on natural or genetically engineered T cells.Additionally,this information is highly relevant for the development of the next-generation vaccines with protections against continuously emerged SARS-CoV-2 mutant strains.Chao Hu Meiying Shen Xiaojian Han Qian Chen Luo Li Siyin Chen Jing Zhang Fengxia Gao Wang Wang Yingming Wang Tingting Li Shenglong Li Jingjing Huang Jianwei Wang Ju Zhu Dan Chen Qingchen Wu Kun Tao Da Pang Aishun Jin 2022Genes & Diseases2022,9,1:0
13Synthesis and evaluation of peptide-fentanyl analogue conjugates as dualμ/δ-opioid receptor agonists for the treatment of pain显示文摘Novel peptide-fentanyl analogue conjugates were synthesized by the covalent coupling of carfentanyl derivatives to the C-terminus or N-terminus of the conformationally constrained dermorphin tetrapeptide BVD03 via a chemical linker.The carfentanyl-related analogues displayed distinct binding and functional activities atμ/δopioid receptors(MOR/DOR)and antinociceptive effects when conjugated to the peptide.The most potent compound,SW-LJ-11,displayed mixed MOR/DOR agonist properties in the low nanomolar range and significant analgesic efficacy in vivo in four classic mouse models of pain.Interestingly,SW-LJ-11 did not exhibit any physical dependence or respiratory depression,in contrast to an equipotent analgesic dose of morphine or BVD03,indicating that the use of opioid peptide-fentanyl analogue conjugates as dual MOR/DOR agonists may be a promising strategy for obtaining safer opioids.Jing Li Tao Zhang Jialin Sun Fengxia Ren Hongxin Jia Zixing Yu Jingchao Cheng Weiguo Shi 2022Chinese Chemical Letters2022,33,8:0

15Tunable In-Plane Anisotropy in Amorphous Sm Co Films Grown on(011)-Oriented Single-Crystal Substrates显示文摘Amorphous Sm-Co films with uniaxial in-plane anisotropy have great potential for application in information-storage media and spintronic materials.The most effective method to produce uniaxial inplane anisotropy is to apply an in-plane magnetic field during deposition.However,this method inevitably requires more complex equipment.Here,we report a new way to produce uniaxial in-plane anisotropy by growing amorphous Sm-Co films onto(011)-cut single-crystal substrates in the absence of an external magnetic field.The tunable anisotropy constant,kA,is demonstrated with variation in the lattice parameter of the substrates.A kA value as high as about 3.3×10^4J·m^-3 was obtained in the amorphous Sm-Co film grown on a LaAlO3(011)substrate.Detailed analysis indicated that the preferential seeding and growth of ferromagnetic(FM)domains caused by the anisotropic strain of the substrates,along with the formed Sm-Co,Co-Co directional pair ordering,exert a substantial effect.This work provides a new way to obtain in-plane anisotropy in amorphous Sm-Co films.Wenhui Liang Houbo Zhou Jiefu Xiong Fengxia Hu Jia Li Jian Zhang Jing Wang Jirong Sun Baogen Shen 2020Engineering2020,6,2:0
16Prenatal Diagnosis of Dystrophinopathy and Cytogenetic Analysis in 303 Chinese Families显示文摘Objective:This study was to supply information of the Duchenne muscular dystrophy(DMD)mutational spectrum in 303 Chinese families and further offer 5-year clinical experience of DMD/Becker muscular dystrophy genetic counseling and prenatal diagnosis.Methods:In this retrospective cohort study,three hundred and five pregnancies in 303 pregnant women who has a birth history of DMD/Becker muscular dystrophy patients underwent prenatal diagnosis using multiplex ligation-dependent probe amplification followed by Sanger sequencing between January 2014 and December 2018 at Peking Union Medical College Hospital.The mean age of pregnant women was(33.0±4.1)years old.Karyotype analysis was performed to exclude fetal abnormal karyotype.Results:The detection rate of DMD gene mutation in 303 probands was(296/303)97.7%with seven families having a negative genetic diagnosis.The mutational spectrum comprised of large arrangements in 288/303(95.0%)and small mutations in 8/303(2.6%).Carrier testing was performed among 204 pregnant women among whom,108 mothers had the same mutation as family proband.Of the 305 pregnancies underwent prenatal diagnosis,55 of 173 male fetuses were affected.We also performed karyotype analysis and found three abnormal karyotypes of trisomy 21.We even found a fetus with DMD gene mutation and trisomy 21 in a same fetus by further analysis.We also identified two times of germline mosaicism.Conclusion:This study demonstrated the distribution and mutation profile of 303 probands and 305 fetuses.Furthermore,considering the possbility of maternl germilne mosaicism,prenatal diagnosis should be suggested to mothers with a proband whether they carry the causative mutation in their blood or not.Mengmeng Li Na Hao Fengxia Yao Weimin Zhang Jing Zhou Li Tan Zhengqing Qiu Juntao Liu 2021Maternal-Fetal Medicine2021,3,4:0

18Unipolar electric-field-controlled nonvolatile multistate magnetic memory in FeRh/(001)PMN-PT heterostructures over a broad temperature span显示文摘Multistate magnetic memory effect in heterostructures composed of FeRh thin films with antiferromagnetic(AFM)-ferromagnetic(FM)phase transition and(001)-oriented PMN-PT substrates has been investigated.Utilizing a unipolar electric field,the nonvolatile change in magnetization was nearly doubled compared with that obtained utilizing a conventional bipolar bias.Four stable nonvolatile magnetic states were obtained over a broad temperature span,from 320 to 390K,by adjusting the amplitude of the unipolar electric pulses,demonstrating the possibility of realizing a multistate nonvolatile magnetic memory in the FeRh/PMN-PT heterostructures.This work provides a new strategy for enhancing the magnetic response by utilizing unipolar electric fields and promotes the utilization of AFM-FM phase transition materials in multifunctional information storage and novel spintronic devices.Kaiming Qiao Fengxia Hu Hu Zhang Ziyuan Yu Xianliang Liu Yuhang Liang Yi Long Jing Wang Jirong Sun Tongyun Zhao Baogen Shen 2022Science China(Physics,Mechanics & Astronomy)2022,65,1:0
19Fabrication of oxygen-defective tungsten oxide nanorods for deep oxidative desulfurization of fuel显示文摘Owing to the increasingly serious environmental issues caused by the sulfur burnt in fuel, desulfurization has become an important topic. In this work, an amphiphilic oxygen-defective tungsten oxide was synthesized by a colloidal chemistry method. The amphiphilic property and oxygen defects were well characterized, and the structure of the oxygen-defective tungsten oxide catalyst was investigated. In addition, the catalyst was employed in oxidative desulfurization system of fuel,and deep desulfurization was achieved. It was found that the very high oxidative desulfurization performance of oxygendefective tungsten oxide catalyst resulted from both the amphiphilic property and oxygen defects. 20Phase transition regulation and caloric effect显示文摘Solid state refrigeration based on caloric effect is regarded as a potential candidate for replacing vapor-compression refrigeration.Numerous methods have been proposed to optimize the refrigeration properties of caloric materials,of which single field tuning as a relatively simple way has been systemically studied.However,single field tuning with few tunable parameters usually obtains an excellent performance in one specific aspect at the cost of worsening the performance in other aspects,like attaining a large caloric effect with narrowing the transition temperature range and introducing hysteresis.Because of the shortcomings of the caloric effect driven by a single field,multifield tuning on multicaloric materials that have a coupling between different ferro-orders came into view.This review mainly focuses on recent studies that apply this method to improve the cooling performance of materials,consisting of enlarging caloric effects,reducing hysteresis losses,adjusting transition temperatures,and widening transition temperature spans,which indicate that further progress can be made in the application of this method.Furthermore,research on the sign of lattice and spin contributions to the magnetocaloric effect found new phonon evolution mechanisms,calling for more attention on multicaloric effects.Other progress including improving cyclability of FeRh alloys by introducing second phases and realizing a large reversible barocaloric effect by hybridizing carbon chains and inorganic groups is described in brief.Yuan LIN Jiazheng HAO Kaiming QIAO Yihong GAO Fengxia HU Jing WANG Tongyun ZHAO Baogen SHEN 2023Frontiers in Energy2023,17,4:0
