1Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations,respectively,and may benefit from immunotherapy or targeted therapies.We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment.Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show 2020Cell Research2020,30,10:19
2High lake levels on Alxa Plateau during the Late Quaternary显示文摘Field investigations including lake shoreline measurement and 14 C chronology have been carried out on the Alxa Plateau to better understand the formation and evolution of the high lake levels in northwest China during the late Pleistocene.Comprehensive field investigations show that there are at least ten shorelines at different elevations at Juyan Lake,at least four on the northwest side of the Jilantai Salt Lake,and nine shorelines and one terrace on the northeast margin of Zhuye Lake.Stratigraphic correlations and chronological evidence indicate that the paleo-lakes have the highest lake levels in Marine Isotope Stage (MIS) 3.Though the optically stimulated luminescence (OSL) results which have been reported by some scholars do not support this view,the fact that the paleo-lakes have the highest lake levels in MIS 3 can not be denied.Differences between OSL and 14 C results from this work might indicate that high lake levels existed on the Alxa Plateau during two different periods.It also suggests that the high lake levels may exist in both MIS 5 and early MIS 3.This paper also provides the basic hydrological information for further water resources research in this arid region.WANG NaiAng LI ZhuoLun CHENG HongYi LI Yu HUANG YinZhou 2011Chinese Science Bulletin2011,56,17:12

5Characterization and validation of somatic mutation spectrum to reveal heterogeneity in gastric cancer by single cell sequencing显示文摘Gastric cancer(GC) is a highly heterogeneous disease with multiple cellular types and poor prognosis.However, the cellular evolution and molecular basis of GC at the individual intra-tumor level has not been well demonstrated. We performed single-cell whole exome sequencing to detect somatic singlenucleotide variants(SNVs) and significantly mutated genes(SMGs) among 34 tumor cells and 9 normal cells from a patient with GC. The Complete Prediction for Protein Conformation(CPPC) approach directly predicting the folding conformation of the protein 3D structure with Protein Folding Shape Code, combined with functional experiments were used to confirm the characterization of mutated SMGs in GC cells. We identified 201 somatic SNVs, including 117 non-synonymous mutations in GC cells. Further analysis identified 24 significant mutated genes(SMGs) in single cells, for which a single amino acid change might affect protein conformation. Among them, two genes(CDC27 and FLG) that were mutated only in single cells but not in the corresponding tumor tissue, were recurrently present in another GC tissue cohort, and may play a potential role to promote carcinogenesis, as confirmed by functional characterization. 6Implementation of a reference-scaled average bioequivalence approach for highly variable generic drug products of agomelatine in Chinese subjects显示文摘The aim of this study was to apply the reference-scaled average bioequivalence(RSABE)approach to evaluate the bioequivalence of 2 formulations of agomelatine,and to investigate the pharmacokinetic properties of agomelatine in Chinese healthy male subjects.This was performed in a single-dose,randomized-sequence,open-label,four-way crossover study with a one-day washout period between doses.Healthy Chinese males were randomly assigned to receive 25 mg of either the test or reference formulation.The formulations were considered bioequivalent if 90% confidence intervals(CIs)for the log-transformed ratios and ratio of geometric means(GMR)of AUC and C_(max) of agomelatine were within the predetermined bioequivalence range based on RSABE method.Results showed that both of the90% CIs for the log-transformed ratios of AUC and C_(max) of 7-desmethyl-agomelatine and 3-hydroxyagomelatine were within the predetermined bioequivalence range.The 90% CIs for natural logtransformed ratios of C_(max) ,AUC_(0–t)and AUC_(0–∞) of agomelatine(104.42–139.86,101.33–123.83 and97.90–117.94)were within the RSABE acceptance limits,and 3-hydroxy-agomelatine(105.55–123.03,101.95–109.10 and 101.72–108.70)and 7-desmethyl-agomelatine(104.50–125.23,102.36–111.50 and101.62–110.64)were within the FDA bioequivalence definition intervals(0.80–1.25 for AUC and0.75–1.33 for C_(max)).The RSABE approach was successful in evaluating the bioequivalence of these two formulations.Fang Tang Rui Zhou Zeneng Cheng Guoping Yang Aiqiao Chen Zhi Liu Hongyi Tan Shuang Yang Sanwang Li Lingli Mu Peng Yu 2016Acta Pharmaceutica Sinica B2016,6,1:2
7Anammox bacterial abundance and biodiversity in greenhouse vegetable soil are influenced by high nitrate content显示文摘The anaerobic ammonium oxidizing(anammox) process has been found to play an important role in terrestrial ecosystems in recent years. However,the diversity and abundance of anammox bacteria in nitrogen(N)-rich agricultural soils under high fertilizer greenhouse conditions are still unclear. Two greenhouse fields with different N fertilizer input levels were chosen, and their soil profiles were studied with molecular technologies, including quantitative polymerase chain reaction assay, a clone library, and phylogenetic analysis based on hzsB(encoding anammox hydrazine synthase β-subunit) gene. Molecular analyses suggested that anammox bacteria were at their highest density at 10–20 cm soil depth, and that the anammox bacterial abundance was significantly lower at high N than at low N. Candidatus Brocadia was the sole anammox bacterial genus throughout the soil depth profiles. The highest diversity of anammox bacteria was found at 30–40 cm soil depth, and different phylotypic clusters of Candidatus Brocadia were associated with specific soil environmental factors, such as nitrates, soil depth, and total N. Correlation analyses and redundancy analyses confirmed that high nitrate content associated with high N fertilizer input had a significant negative influence on the abundance and biodiversity of anammox bacteria. 11Historical desertification of the Mu Us Sandy Land: A perspective from the Beidachi section显示文摘Historical desertification of the Mu Us Sandy Land is linked to both environmental changes and anthropogenic activities. This paper reports on an analysis of grain size parameters as indicative of such changes in the southwestern area of the Mu Us Sandy Land. Combined with analysis of chronologies and historical records, our results indicate that Beidachi Lake and a nearby seasonal river have retreated continuously in history and that sand dunes appeared at approximately the end of the Ming dynasty. 14Risk factors for chronic ankle instability after first episode of lateral ankle sprain:A retrospective analysis of 362 cases显示文摘Background:Chronic ankle instability(CAI) is a common sequela following an acute lateral ankle sprain(LAS).To treat an acture LAS more effectively and efficiently,it is important to identify patients at substantial risk for developing CAI.This study identifies magnetic resonance imaging(MRI) manifestations for predicting CAI development after a first episode of LAS and explores appropriate clinical indications for ordering MRI scans for these patients.Methods:All patients with a first-episode LAS who received plain radiograph and MRI scanning within the first 2 weeks after LAS from December 1,2017 to December 1,2019 were identified.Data were collected using the Cumberland Ankle Instability Tool at final follow-up.Demographic and other related clinical variables,including age,sex,body mass index,and treatment were also recorded.Univariable and multivariable analyses were performed successively to identify risk factors for CAI after first-episode LAS.Results:A total 131 out of 362 patients with a mean follow-up of 3.0± 0.6 years(mean ± SD;2.0—4.1 years) developed CAI after first-episode LAS.According to multivariable regression,development of CAI after first-episode LAS was associated with 5 prognostic factors:age(odds ratio(OR)=0.96,95% confidence interval(95%CI):0.93-1.00,p=0.032);body mass index(OR=1.09,95%CI:1.02-1.17,p=0.009);posterior talofibular ligament injury(OR=2.17,95%CI:1.05-4.48,p=0.035);large bone marrow lesion of the talus(OR=2.69,95%CI:1.30-5.58,p=0.008),and Grade 2 effusion of the tibiotalar joint(OR=2.61,95%CI:1.39-4.89,p=0.003).When patients had at least 1 positive clinical finding in the 10-m walk test,anterior drawer test,or inversion tilt test,they had a 90.2% sensitivity and 77.4% specificity in terms of detecting at least 1 prognostic factor by MRI.Conclusion:MRI scanning is valuable in predicting CAI after first-episode LAS for those patients with at least 1 positive clinical finding in the10-m walk test,anterior drawer test,and inversion tilt test.Further prospective and large-scale studies are necessary for validation.Jieyuan Zhang Kai Yang Cheng Wang Wenqi Gu Xueqian Li Shaoling Fu Guoxun Song Jiazheng Wang Chenglin Wu Hongyi Zhu Zhongmin Shi 2023Journal of Sport and Health Science2023,12,5:1
15Influential factors on the repose angle of desert sands显示文摘The repose angle is one of the most significant macroscopic parameters in describing the behavior of granular materials.Under a static condition,the repose angle is the steepest angle at which sediment particles can rest without motion.In this paper,we use existing data and aeolian physics to analyze the main factors that influence the repose angle of sand dunes,and we investigate dif-ferent repose angles involving various states and types of materials.We have determined that different factors have differential in-fluence on the magnitude of the repose angle.Our results show that for powdery(<400-μm diameter) desert sands,the main in-fluential factor on the magnitude of repose angle is the molecular force among particles.Particle size does not influence the repose angle of desert sands directly,but has an indirect impact by affecting the grit sphericity and surface roughness,of which the grit sphericity acts as a major factor.Even at the same average particle size,the repose angle differs with different grain compositions.Furthermore,with increasing unevenness in grain composition,the repose angle increases correspondingly.Sand texture also has a direct influence on the repose angle of desert sands.In two sand samples having the same grain composition but different textures,the repose angles may be different.Water content has a stronger influence on the repose angle than any other factor.However,the relationship between the repose angle and water content is not a simple direct proportion.In fact,with increasing water content,the repose angle first increases and then decreases.These research results will be useful for understanding the mechanisms of dune transport,variations of dune morphology,and the stability and fluidity of dune sands.JianBao Liu NaiAng Wang HongYi Cheng WenTao Yang 2011Research in Cold and Arid Regions2011,3,1:0
166-15 Status Report of On-line Ion Sources in 2015显示文摘In 2015, the service time of the two on-line ion sources of HIRFL-CSR accelerator facility, SECRAL and LECR3, is 3 335.5 and 3 168.5 h, respectively, amouting to 6 504 h. 18POLG Mutations Are Probably Rare in the Han Chinese Population显示文摘Objective Mutations in polymerase gamma gene(POLG)are believed to be an important cause of early and juvenile onset of non-syndromic intractable epilepsy.The aim of this study is to investigate the incidence/prevalence of POLG pathogenic variants in epilespy patients of Han population through sequencing it.Methods Han Chinese patients with seizures prior valproic acid(VPA)exposure at Shanghai Children\Hospital were collected from 2015 to 2019.The clinical diagnosis was based on the 2014 Consensus Statement of Epilepsy by the International League against Epilepsy(ILAE).Blood sampling were performed before VPA treatment.The POLG gene DNA was sequenced by either the first or the next generation sequencing(NGS).The POLG variant burden was illustrated.Liver functions were tested to describe whether they experienced VPA toxicity.Results Ibtally 216 Han Chinese patients were included,aged from 1 month to IS years old,102 were male and 114 were female.The onset age was 1 month old to 13 years old,and the epilepsy course ranged from 2 weeks to about 3 years.VPA treatment was delivered for the generalized or intractable partial seizures at standard dosage.No patient experienced hepatic toxicity following VPA exposure.DNA sequencing data showed no patient had either a homozygous mutation or compound heterozygous mutation of POLG.Single heterozygous mutations of c.1150G>T and p.D384Y were found in 2 patients,and single heterozygous mutation of c.l56_158dupGCA was found in 1 patient.None of these variants showed clinical significance.Conclusions Functional modifying POLG homozygous mutations and compound heterozygous mutations were not detected and VPA toxicity was not seen in the current study.POLG mutation frequency might be rare in Han Chinese,and standard VPA.therapeutic dosage might be safe for Han Chinese patients.Kunfang Yang Linyi Meng Yuanfeng Zhang Yongchen Yang Hongyi Cheng Zhihu Jiang Hong Zhang Yucai Chen 2020Chinese Medical Sciences Journal2020,35,4:0

206-42 Status Report of On-line Ion Sources in 2014显示文摘In 2014, the service time of the two on-line ion sources of HIRFL-CSR accelerator facility, SECRAL and LECR3is 3 594 and 3 457.2 h, respectively, amouting to 7051.2 h. 17 kinds of ion beams have been delivered successfull.The failure time is shrinked down to 8 h in this year, which is the lowest among these years. Table 1 summarizesthe main information about the ion beams delievered by the two ion sources.
